Oxalic acid decreases calcium absorption in rats.

Calcium absorption from salts and foods intrinsically labeled with 45Ca was determined in the rat model. Calcium bioavailability was nearly 10 times greater for low oxalate kale, CaCO3 and CaCl2 than from CaC2O4 (calcium oxalate) and spinach (high in oxalates). Extrinsic and intrinsic labeling techniques gave a similar assessment of calcium bioavailability from kale but not from spinach.

Animal models for calcium oxalate kidney stone research.

Calcium oxalate (CaOx) stones are the most prevalent form of nephrolithiasis and are characterized by a substantial risk of recurrence. Their pathogenesis arises through distinct etiological pathways, including idiopathic metabolic susceptibility, monogenic disorders such as primary hyperoxaluria, enteric hyperoxaluria, and toxicant-associated injury, and is shaped by two primary papillary mechanisms: Randall's plaque formation and ductal plugging. A central translational limitation is that most experimental systems, particularly chemically induced rodent models, rely on acute hyperoxaluria. T

The Interaction between Enterobacteriaceae and Calcium Oxalate Deposits.

BACKGROUND: The role of calcium oxalate crystals and deposits in UTI pathogenesis has not been established. The objectives of this study were to identify bacteria present in pediatric urolithiasis and, using in vitro and in vivo models, to determine the relevance of calcium oxalate deposits during experimental pyelonephritis. METHODS: Pediatric kidney stones and urine were collected and both cultured and sequenced for bacteria. Bacterial adhesion to calcium oxalate was compared. Murine kidney calcium oxalate deposits were induced by intraperitoneal glyoxalate injection and kidneys were transu

Exploring calcium oxalate crystallization: a constant composition approach.

Crystal growth rates have been extensively studied in calcium oxalate monohydrate (COM) crystallization, because COM crystals are the principal component in most kidney stones. Constant composition methods are useful for studying growth rates, but fail to differentiate concurrent nucleation and aggregation events. A constant composition method coupled with particle size determinations that addresses this deficiency was previously published for a calcium phosphate system, and this method was extended to COM crystallization in this report. A seeded constant composition experiment was combined wi

Reactive oxygen species, inflammation and calcium oxalate nephrolithiasis.

Calcium oxalate (CaOx) kidney stones are formed attached to Randall's plaques (RPs) or Randall's plugs. Mechanisms involved in the formation and growth are poorly understood. It is our hypothesis that stone formation is a form of pathological biomineralization or ectopic calcification. Pathological calcification and plaque formation in the body is triggered by reactive oxygen species (ROS) and the development of oxidative stress (OS). This review explores clinical and experimental data in support of ROS involvement in the formation of CaOx kidney stones. Under normal conditions the production

Glycine suppresses kidney calcium oxalate crystal depositions via regulating urinary excretions of oxalate and citrate.

An abnormal urine composition is a key reason for kidney stone formation, but little is known about the roles of small metabolites in the urine during kidney stone formation. Here, we found urine glycine in patients with kidney calcium oxalate (CaOx) stone was significantly lower than that in healthy people via 1 H NMR spectra detection, and investigated the role and underlying mechanism of glycine in the regulation of CaOx stone formation. Our results showed that glycine could significantly attenuate ethylene glycol-induced CaOx crystal depositions in rat kidney via decreasing urine oxalate a

Calcium oxalate crystals in the breast. Pathology and significance.

Deposits of birefringent calcium oxalate crystals in the breast constitute a mammographically detectable but often histologically overlooked form of calcification. Unstained by routine dyes, these deposits are important as a cause of discrepant radiographic and microscopic biopsy findings. To elucidate the pathologic changes that give rise to these crystals and the possible significance of these deposits in the breast, we retrospectively studied 119 breast biopsies from 100 women. By polarized light microscopy, we identified 16 cases with calcium oxalate deposits from this group. The crystals

Bladder secretion of inhibitors of calcium oxalate crystal growth.

Differences in calcium oxalate crystal growth inhibition were studied in normally voided urine (bladder urine) and in urine collected directly from the kidney (kidney urine) in nine dogs. Urine samples were collected before and 10 days after bilateral ureterostomies. Calcium oxalate crystal growth inhibition was measured in a standard seeded crystal growth system. The alcian blue-precipitable material of the urine samples was determined. Significantly lower values were observed in kidney urine than in bladder urine for calcium oxalate crystal inhibition (mean difference, 0.07 +/- 0.02 inhibito

Calcium oxalate urolithiasis in a cat with a functional parathyroid adenocarcinoma.

A 9-year-old castrated male domestic shorthair cat with dysuria, anorexia, vomiting, and lethargy was admitted to the veterinary teaching hospital. A large, firm mass was palpable in the ventral cervical region. Hypercalcemia, azotemia, and nonregenerative anemia were evident on serum biochemical analysis and CBC, and multiple uroliths were detected by abdominal radiography. At necropsy, light microscopy of the ventral cervical mass revealed a parathyroid adenocarcinoma. Light microscopy of sections of the kidneys revealed multifocal, chronic, lymphocytic/plasmacytic, tubulointerstitial nephri

Supplemental exposure to polystyrene nanoplastics synergistically amplifies calcium oxalate crystal-induced injury to renal tubular epithelium, accelerating the formation of calcium oxalate kidney stones.

BACKGROUND: With the escalating issue of polystyrene microplastic pollution, microplastic particles have been detected in human urine. While calcium oxalate (CaOx) crystals are well-established mediators of renal stone formation, the role of microplastics, particularly polystyrene nanoplastics (PS-NPs), in promoting CaOx kidney stone formation remains unclear. This study aims to explore whether PS-NPs interact with CaOx crystals to enhance renal tubular epithelial cell injury and facilitate the formation of kidney stones. METHODS: Clinical CaOx kidney stone samples were analyzed using pyrolys

Drivers of calcium oxalate stone formation in the octogenarian population.

INTRODUCTION: American Urological Association (AUA) guidelines suggest metabolic testing via 24-h urine studies in high-risk, interested first-time stone formers, and recurrent stone formers. If metabolic testing is not available or otherwise not feasible, clinicians may need to utilize empiric therapy. Debility and social barriers, particularly in the elderly population, may limit the practicality of metabolic testing, and therefore, empiric therapy is of particular importance. The aim of this study is to identify whether unique urinary metabolic abnormality profiles exist for octogenarians w

The mechanistic insight of polyphenols in calcium oxalate urolithiasis mitigation.

About 12% of world population is affected by different forms of urolithiasis of which the recurrence rate in female is 47-60% and in male is 70-80%. Standard therapeutic agents (allopurinol, citrate, cystone and thiazide diuretics) are used to prevent and treat urolithiasis but these are not universally-effective due to common kidney stone relapse and other side effects. Surgical treatment causes long-term renal damage, hypertension and stone recurrence. Polyphenols, the plant-derived bioactive molecules, have showed protection against cancers, cardiovascular diseases, diabetes, osteoporosis a

Intestinal absorption of calcium and oxalate in magnesium-deficient rats.

To examine the contribution of exogenous calcium and oxalate in magnesium deficiency, intestinal absorption of both calcium and oxalate was studied by preparing brush border membrane vesicles (BBMV). Purity of the BBMV was ascertained biochemically by enrichment of the marker enzyme alkaline phosphatase by 14-fold with a concomitant 90 per cent decrease in the basolateral marker enzyme Na+/K(+)-ATPase in the purified membrane preparation as compared to the respective homogenate in both the groups. Uptake studies were carried out by a rapid filtration technique. The kinetics were studied by mea

Solubility of calcium oxalate monohydrate and hydroxyapatite in EDTA solutions.

The effect of the addition of a buffer to an ethylenediaminetetracetic acid (EDTA) litholytic solution on the solubility of calcium oxalate monohydrate and hydroxyapatite is investigated. The experiments show that the addition of triethanolamine as a buffer to EDTA solutions at pH 8 and 8.5 enhances the solubility of these salts, confirming theoretical predictions. The solubility of calcium oxalate monohydrate at 25C in solutions with varying ratios of buffer to EDTA concentrations and an osmolality of 0.9 was determined. The solutions resulting in the highest solubility were finally tested fo
