Idiopathic Chronic Diarrhea in Rhesus Macaques Is Not Associated with Enteric Viral Infections.

While recent changes in treatment have reduced the lethality of idiopathic chronic diarrhea (ICD), this condition remains one of the most common causes of rhesus macaque deaths in non-human primate research centers. We compared the viromes in fecal swabs from 52 animals with late stage ICD and 41 healthy animals. Viral metagenomics targeting virus-like particles was used to identify viruses fecally shed by each animal. Five viruses belonging to the Picornaviridae, one to the Caliciviridae, one to the Parvoviridae, and one to the Adenoviridae families were identified. The fraction of reads matc

[Gastroenteritis viruses in France and Europe].

Rotaviruses and noroviruses are the main causes of acute gastroenteritis in young children and adults, respectively. Prospective molecular surveillance of rotavirus genotypes in France and Europe shows that circulating strains may vary with the season, locality or country. Rotavirus OK? genotypes G1 and G9 are the most prevalent. Most strains are associated with P[8], showing a certain genotypic stability of rotaviruses currently circulating in Europe and suggesting that vaccination would be effective at least in the short and medium term. However, atypical strains G12 and G8 must be monitored

Sapporo virus: history and recent findings.

Morphologically distinct caliciviruses of human origin were first found in stools of children with gastroenteritis in 1976. Sapporo virus, or human calicivirus Sapporo, with typical surface morphology was first detected during a gastroenteritis outbreak in a home for infants in Sapporo, Japan, in 1977. Since then, morphologically and antigenically identical virus has been detected frequently in the same institution in association with outbreaks of gastroenteritis. Sapporo virus is widely distributed worldwide, as evidenced by the appearance of antigenically or genetically similar viruses and s

Complete genome sequence of a rare recombinant GII.5[P16] norovirus found in Russian Siberia.

Noroviruses (family Caliciviridae) are common causes of acute gastroenteritis worldwide. Multiple polymerase/capsid combinations have been identified among members of norovirus genogroup GII, at least 10 of which contain GII.P16 polymerase. During hospital-based surveillance (2003-2013) in Russia, we identified eight noroviruses with GII.P16 polymerase - five GII.3[P16], two GII.16[P16], and one GII.5[P16]. This is the first report of the nearly complete genome sequence of a rare recombinant GII.5[P.16] norovirus, which was found in the feces of a child in 2010. Phylogenetic analysis revealed

Differential Viral Genome Diversity of Healthy and RSS-Affected Broiler Flocks.

The intestinal virus community contributes to health and disease. Runting and stunting syndrome (RSS) is associated with enteric viruses and leads to economic losses in the poultry industry. However, many viruses that potentially cause this syndrome have also been identified in healthy animals. To determine the difference in the virome of healthy and diseased broilers, samples from 11 healthy and 17 affected broiler flocks were collected at two time points and analyzed by Next-Generation Sequencing. Virus genomes of Parvoviridae, Astroviridae, Picornaviridae, Caliciviridae, Reoviridae, Adenovi

Biological and immunological characteristics of hepatitis E virus-like particles based on the crystal structure.

Hepatitis E virus (HEV) is a causative agent of acute hepatitis. The crystal structure of HEV-like particles (HEV-LP) consisting of capsid protein was determined at 3.5-A resolution. The capsid protein exhibited a quite different folding at the protruding and middle domains from the members of the families of Caliciviridae and Tombusviridae, while the shell domain shared the common folding. Tyr-288 at the 5-fold axis plays key roles in the assembly of HEV-LP, and aromatic amino acid residues are well conserved among the structurally related viruses. Mutational analyses indicated that the protr

Atomic Structure of the Human Sapovirus Capsid Reveals a Unique Capsid Protein Conformation in Caliciviruses.

Sapovirus (SaV) is a member of the Caliciviridae family, which causes acute gastroenteritis in humans and animals. Human sapoviruses (HuSaVs) are genetically and antigenically diverse, but the lack of a viral replication system and structural information has hampered the development of vaccines and therapeutics. Here, we successfully produced a self-assembled virus-like particle (VLP) from the HuSaV GI.6 VP1 protein, and the first atomic structure was determined using single-particle cryo-electron microscopy (cryo-EM) at a 2.9-Å resolution. The atomic model of the VP1 protein revealed a unique

Gut Virome Analysis of Cameroonians Reveals High Diversity of Enteric Viruses, Including Potential Interspecies Transmitted Viruses.

Diarrhea remains one of the most common causes of deaths in children. A limited number of studies have investigated the prevalence of enteric pathogens in Cameroon, and as in many other African countries, the cause of many diarrheal episodes remains unexplained. A proportion of these unknown cases of diarrhea are likely caused by yet-unidentified viral agents, some of which could be the result of (recent) interspecies transmission from animal reservoirs, like bats. Using viral metagenomics, we screened fecal samples of 221 humans (almost all with gastroenteritis symptoms) between 0 and 89 year

Viral diversity in swine intestinal mucus used for the manufacture of heparin as analyzed by high-throughput sequencing.

Heparin is one of the main pharmaceutical products manufactured from raw animal material. In order to describe the viral burden associated with this raw material, we performed high-throughput sequencing (HTS) on mucus samples destined for heparin manufacturing, which were collected from European pigs. We identified Circoviridae and Parvoviridae members as the most prevalent contaminating viruses, together with viruses from the Picornaviridae, Astroviridae, Reoviridae, Caliciviridae, Adenoviridae, Birnaviridae, and Anelloviridae families. Putative new viral species were also identified. The loa

Isolation of a rhesus calicivirus that can replicate in human cells.

Recoviruses (rhesus enteric caliciviruses) are members of the Caliciviridae family. They are a valuable model for studying human caliciviruses such as noroviruses. It has been suggested that some recoviruses may infect humans, which necessitates detailed studies on the cell type tropism of recoviruses. For the recoviruses that have been cultured to date, successful growth has only been reported in monkey kidney cell lines, precluding their use to study virus interactions with human cells. We isolated and characterized a new recovirus, Recovirus Mo/TG30/2012, from monkey stool which grew effici

Characterization of novel intergenogroup and intergenotype recombinant noroviruses from central Greece.

Noroviruses (NoVs) are a major causative agent of acute gastroenteritis in humans. They are members of the Caliciviridae family and based on the genetic analysis of the RdRp and capsid regions, human NoVs are divided into three genogroups (Gs), GI, GII, and GIV. The three genogroups further segregate into distinct lineages called genotypes. The NoV genus is genetically diverse and recombination of viral RNA is known to depend upon various immunological and intracellular constraints that may allow the emergence of viable recombinants. In this study, three Noroviral strains detected in clinical

Stage-Specific Tulane Virus Replication in Zebrafish: Permissive in Embryos, Restricted in Larvae.

Human norovirus (HuNoV), the leading cause of foodborne illness in the United States and acute gastroenteritis globally, replicates in the zebrafish embryo and larvae model. However, due to challenges in obtaining well-characterized HuNoV positive stool specimens for research, demonstrating replication of Tulane virus (TuV), a surrogate of HuNoV, in the zebrafish embryo and larvae model is valuable to study HuNoV infection mechanisms and subsequent prevention and control strategies. Three nL of TuV (9 log RNA copies/mL or 7 log PFU/mL) were microinjected into either fertilized zebrafish eggs (

Human caliciviruses and pediatric gastroenteritis.

Surveillance using molecular diagnostic methods developed in the past decade shows that human caliciviruses, particularly the Noroviruses, are the most important cause of pediatric acute gastroenteritis, next to rotavirus. Noroviruses also cause severe gastroenteritis in children, requiring hospitalization. The Sapovirus, a morphologically and genetically distinct genus of calicivirus, cause mainly milder endemic pediatric gastroenteritis. Noroviruses and Sapoviruses are genetically and antigenetically diverse, and multiple genetic types cocirculate in the same community. Human caliciviruses s

Genetic variation and phylogenetic analysis of rabbit haemorrhagic disease virus (RHDV) strains.

Rabbit haemorrhagic disease virus (RHDV) belongs to the family Caliciviridae and is the etiological agent of the haemorrhagic disease, also known as rabbit plague. Its genome is a linear single-stranded (ss) RNA of 7437 nucleotides and the capsid is built from a single structural protein VP60. In connection with the discovery of new RHDV strains, there is a constant need to investigate the genetic variation of this virus and perform phylogenetic analyses which may show the evolutionary relationships among the RHDV strains. Studies on the divergence of RHDV have shown that it is genetically qui

Structural insights into calicivirus attachment and uncoating.

The Caliciviridae family comprises positive-sense RNA viruses of medical and veterinary significance. In humans, caliciviruses are a major cause of acute gastroenteritis, while in animals respiratory illness, conjunctivitis, stomatitis, and hemorrhagic disease are documented. Investigation of virus-host interactions is limited by a lack of culture systems for many viruses in this family. Feline calicivirus (FCV), a member of the Vesivirus genus, provides a tractable model, since it may be propagated in cell culture. Feline junctional adhesion molecule 1 (fJAM-1) was recently identified as a fu
