Endocrine control of pregnancy and parturition in South American camelids and Old World camels and the resulting possibilities for hormonal pregnancy diagnostics.

For South American camelids (SAC) and, to a lesser extent, Old World camels (OWC), an increasing demand for veterinary services has developed in Central Europe in recent years, with specific knowledge of the reproductive endocrinology of this species being particularly in demand for the management of breeding farms. Compared to many other domestic animal species, relatively little reliable information is available on the endocrine control of pregnancy and parturition in camelids. However, some significant differences to other domestic ungulate species are evident. Knowledge of pregnancy-associ

Seroprevalence of Neospora caninum in Camelids: A Global Systematic Review and Meta-Analysis.

BACKGROUND: Neospora caninum is an apicomplexan parasite that causes significant reproductive losses in livestock, particularly cattle, yet its epidemiological impact on camelids remains poorly understood. This systematic review and meta-analysis aimed to estimate the pooled seroprevalence of N. caninum in camelids worldwide and to compare seroprevalence between Old World camels (OWCs) and New World camelids (NWCs). METHODS: A systematic literature search was performed in various international databases from January 1, 2000 to December 20, 2025. Eligible studies reported serological detection

Male reproductive biotechnologies in South American Camelids Part II: Semen dilution, cryopreservation and artificial insemination.

Even though South American Camelids (SAC) are a rustic species, adapted to harsh environments, their ability to reproduce is low in their natural habitat. Conception and birth rates in camelids vary from 50 % to 90 %. This depends on the mating system used, sire and dam fertility, postpartum interval, environmental conditions, and nutrition. Reproductive biotechnology can reduce generation intervals and quickly spread high-quality genetics, addressing the growing need for genetically superior animals. The advantages of using artificial insemination (AI) and semen preservation are widely known.

High-Throughput Isolation of Nanomouse-Derived VHH Domains: A Practical Guide from Immunization to Nanobody Expression.

Nanobodies are small but specific heavy chain-only antibody fragments. Their small size, relative stability, and ability to access difficult to reach deep-tissue antigens makes them valuable research, diagnostic, and therapeutic tools. Nanobodies are derived from the variable heavy (VH) domain of heavy chain-only antibodies that are unique to camelids, including alpacas, llamas, and camels. The approaches employed to produce nanobodies, have been evolving and expanding since the initial discovery of heavy chain-only antibodies 30 years ago. Traditional nanobody development involves camelid imm

Delving into the innate and adaptive immunity of camelids: a paradigm of interspecies adaptation and evolutionary innovation.

Camelids have evolved to survive some of the most challenging climatic conditions on our planet and are resilient to physiological stress such as temperature extremes, drought, storms, intense ultraviolet radiation, dehydration, and starvation. They exhibit remarkable resistance to several diseases that severely affect other livestock including tetanus, foot-and-mouth disease, and bovine spongiform encephalopathy. The robust nature of the camelid immune system, which maintains functionality under extreme conditions, is remarkable. The special features of the camelid immune system include diffe

Ovarian Dynamics and Pathological Conditions in Camelids.

Domestic camelids comprise Dromedary, Bactrian (old-world camelids, OWC), alpaca, and Lama (new-world camelids, NWC). They possess unique reproductive physiology distinct from other livestock. A key feature is being induced ovulators, triggered by beta nerve growth factor (βNGF) present in the seminal plasma. Proper ovarian function is crucial for reproductive management and biotechnologies. In the absence of ovulation, camelids display overlapping follicular waves, with puberty and sexual receptivity influenced by factors like weight and age. Follicular growth patterns vary among these specie

Prevalence study of Echinococcus granulosus in guanaco (Lama guanicoe) from the Chilean Patagonia unexpectedly reveals the southernmost limit of Taenia omissa.

This study presents the first detection of Taenia omissa metacestodes in guanaco (Lama guanicoe) within the Chilean Patagonia, marking the southernmost record of natural infection in an intermediate host on the continent. Taenia omissa was found in the continental part of the Magallanes region where the top predators are pumas (Puma concolor). Conversely, all metacestodes found in guanacos collected from Tierra del Fuego Island, where no pumas exist, were identified solely as Echinococcus granulosus sensu stricto. Additionally, this research highlights a tissue preference of T. omissa for live

Fast One-Step Ultrasensitive Detection of Toxocara canis Antigens by a Nanobody-Based Electrochemical Magnetosensor.

Human toxocariasis (HT) is a cosmopolitan zoonotic disease caused by the migration of the larval stage of the roundworm Toxocara canis. Current HT diagnostic methods do not discriminate between active and past infections. Here, we present a method to quantify Toxocara excretory/secretory antigen, aiming to identify active cases of HT. High specificity is achieved by employing nanobodies (Nbs), single domain antigen binding fragments from camelid heavy chain-only antibodies. High sensitivity is obtained by the design of an electrochemical magnetosensor with an amperometric read-out. Reliable de

Gastrointestinal parasite diversity in two contrasting guanaco (Lama guanicoe) populations of Argentina: Comparing Chacoan and Patagonian populations.

The guanaco (Lama guanicoe), the largest wild artiodactyl in South America, shows contrasting population trends across its distribution. While more than two million individuals persist in continental Patagonia, only two relict populations remain in the Gran Chaco region. Although gastrointestinal parasites have been documented in some guanaco populations, substantial knowledge gaps persist across their range. This study characterizes the diversity and composition of gastrointestinal parasites in two contrasting populations, one critically endangered and isolated in the Chaco region (Córdoba Pr

Mechanisms and applications of camelid variable heavy-chain nanobodies against bacterial and parasitic protozoal pathogens.

Camelid variable heavy-chain (VHH) nanobodies have emerged as a promising class of single-domain antibodies with significant potential for combating bacterial and parasitic infections. Their small molecular size, high solubility, exceptional physicochemical stability, and ability to recognize cryptic or conformational epitopes inaccessible to conventional antibodies enable unique therapeutic applications. This review summarizes recent advances in the biology, mechanisms of action, engineering, and translational potential of camelid VHH nanobodies against bacterial and parasitic pathogens. In b

Expanding anti-venom strategies: Camelid polyclonal antibodies with high capacity to recognize snake venom.

Camelid immunoglobulins represent a unique facet of antibody biology, challenging conventional understandings of antibody diversification. IgG2 and IgG3 in particular are composed solely of heavy chains and exhibit a reduced molecular weight (90 kDa); their elongated complementarity determining region (CDR) loops play a pivotal role in their functioning, delving deep into enzyme active sites with precision. Serum therapy stands as the primary venom-specific treatment for snakebite envenomation, harnessing purified antibodies available in diverse forms such as whole IgG, monovalent fragment ant

Cytokine responses in camels (Camelus bactrianus) vaccinated with Brucella abortus strain 19 vaccine.

In the present study, we determined the levels of cytokines produced by camel (Camelus bactrianus) peripheral blood mononuclear cells (PBMCs) in response to live attenuated Brucella abortus (B. abortus) S19 vaccine. Seven camels were vaccinated with commercial B. abortus S19 vaccine, and their cytokine responses were determined using a real-time PCR assay. Cytokine responses to B. abortus S19 were examined at 6 hr, 48 hr and 1, 2 and 3 weeks post-vaccination. Serological tests were performed to further confirm these immune responses. The results revealed that IFN-gamma and IL-6 were upregulate

Nanobodies®: new ammunition to battle viruses.

In 1989, a new type of antibody was identified, first in the sera of dromedaries and later also in all other species of the Camelidae family. These antibodies do not contain a light chain and also lack the first constant heavy domain. Today it is still unclear what the evolutionary advantage of such heavy chain-only antibodies could be. In sharp contrast, the broad applicability of the isolated variable antigen-binding domains (VHH) was rapidly recognized, especially for the development of therapeutic proteins, called Nanobodies(®). Here we summarize first some of the unique characteristics an

Reports of Microthoracius mazzai (Phthiraptera: Anoplura) infestations in alpaca (Vicugna pacos) herds from Austria and Germany.

Sucking lice (Phthiraptera: Anoplura) constitute obligate and permanent insect ectoparasites of mammals and exhibit a hematophagous lifestyle. In South American camelids, three species of the genus Microthoracius have been described, however, reports outside South America are scarce. Here we describe infestations with Microthoracius mazzai in three alpaca (Vicugna pacos) herds from Austria (Styria and Salzburg), and Germany (Bavaria). Overall, lice infestations were detected in eight animals. Four of them exhibited a massive generalized infestation. Lice specimens were identified as M. mazzai

Sarcoptic mange outbreak decimates South American wild camelid populations in San Guillermo National Park, Argentina.

Sarcoptic mange epidemics can devastate wildlife populations. In 2014, mange was first detected in vicuñas (Vicugna vicugna) and guanacos (Lama guanicoe) in San Guillermo National Park (SGNP), Argentina. This study describes the temporal dynamics of the outbreak, its effects on the park's wild camelid populations between 2017-2019, and investigates the potential source of the epidemic. From May 2017 to June 2018, transect surveys indicated a sharp decrease in the density of living vicuñas and guanacos by 68% and 77%, respectively. By April 2019 no vicuñas or guanacos were recorded on transect

Use of camel single-domain antibodies for the diagnosis and treatment of zoonotic diseases.

Camelids produce both conventional heterotetrameric antibodies and homodimeric heavy-chain only antibodies. The antigen-binding region of such homodimeric heavy-chain only antibodies consists of one single domain, called VHH. VHHs provide many advantages over conventional full-sized antibodies and currently used antibody-based fragments (Fab, scFv), including high specificity, stability and solubility, and small size, allowing them to recognize unusual antigenic sites and deeply penetrate tissues. Since their discovery, VHHs have been used extensively in diagnostics and therapy. In recent deca

The development of nanobodies for therapeutic applications.

Evolution has been continuously honing the design of antibodies to function as specific molecular markers that are able to alert the immune system to the presence of pathogenic antigens, and to recruit complement- and Fc receptor-bearing effector cells. During the past 25 years, the versatility of antibodies has been applied to several therapeutic applications. The development of new technologies, combined with data obtained using a new generation of antibody reagents, have allowed the adaptation of the design of antibodies to better match drug development requirements. Nanobodies are therapeu

Morphofunctional structure of the lingual papillae in three species of South American Camelids: Alpaca, guanaco, and llama.

The aim of this study was to compare the anatomical and functional characteristics of the lingual papilla among the Camelidae. For this purpose, tongues of alpaca, guanaco, and llama were used. Numerous long and thin filiform papillae were located in the median groove and none were detected on the rest of the dorsal surface of the lingual apex in alpaca. Secondary papillae originated from the base of some filiform papillae on the ventral surface of alpaca tongue. The bases of some filiform papillae of the lateral surface of the lingual apex were inserted into conspicuous grooves in guanaco and

Bacterial osteomyelitis in veterinary orthopaedics: Pathophysiology, clinical presentation and advances in treatment across multiple species.

Bacterial osteomyelitis in veterinary patients can be challenging to diagnose and treat, given limited therapeutic options and reported success rates. Osteomyelitis is frequently associated with surgical implant devices, including those required to optimise stability and healing of fractures. However, management of osteomyelitis sometimes necessitates the removal of these surgical implant devices in order to eradicate infection or limit implant-related osteolysis. The goal of this article is to provide a general and species-specific review of bacterial osteomyelitis in a selection of domestic
