Assessment of a questionnaire for breast cancer case-control studies.

The aim of the present study was to assess criterion validity and external reliability of a questionnaire on risk factors for breast cancer. Materials and Methods. Women with breast cancer diagnosis (the cases) (N=40) and matched individuals without cancer (the controls) (N=40) were asked to fill in a questionnaire twice: on a day of admission to hospital (Q1) and on a day before discharge (Q2), with a time interval of 4-6 days. The questionnaire included questions (N=150) on demographic and socioeconomic factors, diseases in the past, family history of cancer, woman's health, smoking, alcohol

Selecting controls for assessing interaction in nested case-control studies.

BACKGROUND: Two methods for selecting controls in nested case-control studies--matching on X and counter matching on X--are compared when interest is in interaction between a risk factor X measured in the full cohort and another risk factor Z measured only in the case-control sample. This is important because matching provides efficiency gains relative to random sampling when X is uncommon and the interaction is positive (greater than multiplicative), whereas counter matching is generally efficient compared to random sampling. METHODS: Matching and counter matching were compared to each other

Confidence intervals for the excess risk in case-control studies.

Kaufman et al. compute the 'excess risk' of a disease in the presence of an exposure as the product of the incidence rate of the disease in the source population, the complement of the aetiologic fraction and the relative risk minus one. Methods for calculating confidence intervals for this quantity are derived when (as in case-control studies) the relative risk is estimated by the odds ratio, firstly from multiple logistic regression analysis and secondly without adjustment for covariates. For the latter an innovative approach based on confidence bounds for the two exposure parameters is sugg

Differential recall bias and spurious associations in case/control studies.

Consider a case/control study designed to investigate a possible association between exposure to a putative risk factor and development of a particular disease. Let E denote the information required to specify a subject's exposure to the risk factor. We examine the effect that errors in the recorded values of E (which we denote by E*) have on inferences of an association between disease and the risk factor. We concentrate on situations where the errors in recorded exposure are such that exposure is underestimated for controls and overestimated for cases. This phenomenon is referred to as diffe

Assessment of family history information in case-control cancer studies.

Evidence for the genetic basis of cancer has increased in recent years, and thus, assessing familial aggregation may play an important role in epidemiologic studies. To assess the degree to which family history variables are collected in cancer case-control studies, the authors conducted a literature review of studies published in six journals during 1982-1984. Only 25% of these included a family history component. The authors review some of the literature in favor of inclusion of family information and argue for its routine collection in cancer case-control studies.

Observational epidemiologic studies of endemic waterborne risks: cohort, case-control, time-series, and ecologic studies.

Observational studies have assessed endemic waterborne risks in a number of countries. Time-series analyses associated increased water turbidity with increased gastroenteritis risks in several public water systems. Several cohort studies reported an increased risk of gastroenteritis in populations using certain public or individual water systems. Although several case-control studies found increased waterborne risks, they also found increased risks associated with other exposures. An increased risk of campylobacteriosis was associated with drinking untreated water from non-urban areas and some

Evaluation of the Regulatory Required Post-Authorization Safety Study for Propacetamol: Nested Case-Control and Case-Time-Control Studies.

PURPOSE: Following the withdrawal of propacetamol in Europe owing to safety issues, the regulatory authority of South Korea requested a post-marketing surveillance study to investigate its safety profile. MATERIALS AND METHODS: We conducted nested case-control and case-time-control (CTC) analyses of cases and controls identified for outcomes of interest, including anaphylaxis, thrombosis, and Stevens-Johnson syndrome (SJS), using the claims database of South Korea, 2010-2019. Risk-set sampling was used to match each case with up to 10 controls for age, sex, cohort entry date, and follow-up du

Generalized functional linear models for gene-based case-control association studies.

By using functional data analysis techniques, we developed generalized functional linear models for testing association between a dichotomous trait and multiple genetic variants in a genetic region while adjusting for covariates. Both fixed and mixed effect models are developed and compared. Extensive simulations show that Rao's efficient score tests of the fixed effect models are very conservative since they generate lower type I errors than nominal levels, and global tests of the mixed effect models generate accurate type I errors. Furthermore, we found that the Rao's efficient score test st

Reducing selection bias in case-control studies from rare disease registries.

BACKGROUND: In clinical research of rare diseases, where small patient numbers and disease heterogeneity limit study design options, registries are a valuable resource for demographic and outcome information. However, in contrast to prospective, randomized clinical trials, the observational design of registries is prone to introduce selection bias and negatively impact the validity of data analyses. The objective of the study was to demonstrate the utility of case-control matching and the risk-set method in order to control bias in data from a rare disease registry. Data from the International

A method for analysing case-control studies with ordinal exposure variables.

A method is proposed for analysing case-control studies with ordinal or continuous, but unruly, exposure levels. A test is proposed which is an appropriate linear combination of stratum-specific Wilcoxon tests. An estimator for the mean percentile shift between the case and control exposure levels is given. Methods for assessing heterogeneity of the mean percentile shift across strata are also discussed. The performance of the test as a function of the number of controls per case is studied for both local and distant alternatives. The asymptotic relative efficiency compared to the best paramet

Case-control studies of motor neuron disease: association with mechanical injuries.

Two case-control studies of motor neuron disease that involved 712 cases and 158 cases, respectively, showed that (1) mechanical injuries were two to three times more frequent in both sexes, heralding amyotrophic lateral sclerosis, progressive bulbar paralysis, and progressive muscular atrophy; (2) the head, neck, spine, and the extremities were more often traumatized; (3) traumatized parts were not correlated with the initial manifestation of the disease; and (4) more males were traumatized, but males still predominated among uninjured cases. These results suggested that mechanical injuries w

DHEA and polycystic ovarian syndrome: Meta-analysis of case-control studies.

BACKGROUND: Polycystic ovarian syndrome is a heterogenous endocrine disorder characterized by irregular menstrual cycles, hirsuitism and polycystic ovaries. It is further complicated by metabolic syndrome, infertility and psychological stress. Although the etiopathogenesis is unclear, many studies have pointed out the role of stress in this syndrome. DHEA, being a stress marker is being used by scientists to compare the stress levels between polycystic ovarian cases and healthy controls. However, the results obtained from previous studies are equivocal. OBJECTIVE: To perform meta-analysis and

Lower limb immobilisation and venous thromboembolism risk: combined case-control studies.

BACKGROUND: Evidence from randomised controlled trials shows that low-molecular-weight heparin is effective in reducing the risk of venous thromboembolism (VTE) in the clinical setting of temporary lower limb immobilisation. Despite this, international guidelines are non-committal in advocating the use of anticoagulation in this clinical scenario. We determined the risk of VTE associated with lower limb immobilisation and the proportion of VTE events associated with lower limb immobilisation by undertaking a secondary analysis of two case-control studies that had used a similar methodology.
