[Mineral salt-dependent inhibition of light emission from the luminescent microorganism Escherichia coli Z9051].

The influence of some mineral salts on the recombinant strain Escherichia coli Z9051 was investigated. It was shown that the composition (NaCl, Na2SO4, MgCl2 and MgSO4) and concentration (5 and 10%) of the salts substantially affect the expression of genes for the luminescence system of light-emitting bacteria cloned in the plasmid under the control of the lac-promoter. In some cases, the luminescence level of the microorganism in the presence of salts was similar to the luminescence level under catabolite repression by glucose, the more strong influence of the salts exceeding the effect of ca

Anti-mitochondrial autoantibodies of primary biliary cirrhosis as a novel probe in the study of the biosynthetic regulation of the yeast 2-oxo acid dehydrogenase complexes.

Autoantibodies present in the disease primary biliary cirrhosis react by immunoblotting with four major yeast mitochondrial antigens of 58 kDa, 55 kDa, 52 kDa and 45 kDa, tentatively identified as the lipoate acetyl transferases (E2) of the pyruvate dehydrogenase, component X of E2 pyruvate dehydrogenase, E2 of 2-oxo glutarate dehydrogenase and E2 of branched-chain 2-oxo acid dehydrogenase complexes respectively. The synthesis of these antigens is sensitive to catabolite repression. The reactive antigens are present in mit- mutants of yeast which have specific defects in the mitochondrial apoc

The Regulatory Networks That Control Clostridium difficile Toxin Synthesis.

The pathogenic clostridia cause many human and animal diseases, which typically arise as a consequence of the production of potent exotoxins. Among the enterotoxic clostridia, Clostridium difficile is the main causative agent of nosocomial intestinal infections in adults with a compromised gut microbiota caused by antibiotic treatment. The symptoms of C. difficile infection are essentially caused by the production of two exotoxins: TcdA and TcdB. Moreover, for severe forms of disease, the spectrum of diseases caused by C. difficile has also been correlated to the levels of toxins that are prod

Rewiring a methanol-responsive regulatory system improves glucose-methanol co-utilization in Eubacterium limosum.

BACKGROUND: Methanol is a promising one-carbon (C1) feedstock for sustainable bioproduction, and its mixotrophic co-utilization with other substrates can improve product formation. However, mixotrophy often leads to sequential substrate utilization that delays methanol assimilation, and the regulatory basis underlying this phenotype remains unclear. In this study, we aimed to elucidate the regulatory mechanism governing methanol utilization in a methylotrophic acetogen and to determine whether rewiring this system could improve methanol co-utilization. RESULTS: Here, we identify a dual-layer

Regulatory factors affecting alpha-amylase production in bacillus licheniformis.

Possible factors regulating alpha-mylase synthesis in wild-type Bacillus licheniformis and in mutants producing elevated levels of the enzyme were studied in terms of catabolite repression, apparent temperature-sensitive repression, induction, and culture age. The synthesis of alpha-amylase in the parent strain occurred long after the culture reached the stationary phase of growth as a result of de novo protein synthesis, occurred only at high temperature around 50 C and not below 45 C, appeared to be induced in the presence of oligosaccharides with some linkage of alpha-1,4-, beta-1,4, beta-1

The Role of Regulator Catabolite Control Protein A (CcpA) in Streptococcus agalactiae Physiology and Stress Response.

Streptococcus agalactiae is a leading cause of infections in neonates. This opportunistic pathogen colonizes the vagina, where it has to cope with acidic pH and hydrogen peroxide produced by lactobacilli. Thus, in the host, this bacterium possesses numerous adaptation mechanisms in which the pleiotropic regulators play a major role. The transcriptional regulator CcpA (catabolite control protein A) has previously been shown to be the major regulator involved in carbon catabolite repression in Gram-positive bacteria but is also involved in other functions. By transcriptomic analysis, we characte

Tryptophanase disruption promotes insect-bacterium mutualism.

Animal-microorganism symbioses are omnipresent, with both partners often gaining benefits as mutualists. A single mutation in the carbon catabolite repression system in Escherichia coli enables mutualism with the stinkbug Plautia stali. Here we find that this mutation is not present in natural symbioses. Given that the carbon catabolite repression pathway affects the expression of >500 downstream genes, we investigated their role in mutualisms. We find that disruption of a single gene, tnaA, encoding tryptophanase makes E. coli mutualistic to P. stali, resulting in the accumulation of tryptoph

Production and regulation of a thermostable protease by Pseudomonas sp. B45.

A Pseudomonas sp. produced an extracellular thermostable protease which required induction by peptone. Growth of the organism and the production of protease was optimum at 30 degrees C. The enzyme was subjected to catabolite repression by glucose. Both chloramphenicol and rifamycin completely abolished protease production indicating de novo synthesis of the enzyme. Leucine, lysine, histidine and glycine enhanced the protease production considerably and they were the most effective when added during the active period of production. Glucose repression could not be relieved by addition of leucine

The CcpA protein is necessary for efficient sporulation and enterotoxin gene (cpe) regulation in Clostridium perfringens.

Clostridium perfringens is the cause of several human diseases, including gas gangrene (clostridial myonecrosis), enteritis necroticans, antibiotic-associated diarrhea, and acute food poisoning. The symptoms of antibiotic-associated diarrhea and acute food poisoning are due to sporulation-dependent production of C. perfringens enterotoxin encoded by the cpe gene. Glucose is a catabolite repressor of sporulation by C. perfringens. In order to identify the mechanism of catabolite repression by glucose, a mutation was introduced into the ccpA gene of C. perfringens by conjugational transfer of a

Metabolic engineering of Ruminiclostridium papyrosolvens for 2,3-butanediol production from lignocellulose.

Ruminiclostridium papyrosolvens is an anaerobic, mesophilic, and cellulolytic bacterium capable of secreting cellulosomes for efficient lignocellulose degradation, making it a promising candidate for consolidated bioprocessing (CBP). However, its native cellulolytic activity is repressed by cellobiose, and its natural fermentation products are limited to low-value compounds. In this study, we firstly discovered that the cellulosomal cip-cel gene cluster is subject to cellobiose-mediated carbon catabolite repression (CCR). By replacing its native promoter with the endogenous cellobiose/cellodex

A Novel De Novo Biosynthetic Pathway for Efficient Synthesis of 6'-Sialyllactose in Escherichia coli.

6'-Sialyllactose (6'-SL) is an essential human milk oligosaccharide (HMO) that plays a crucial role in infant development with significant industrial potential. In this study, a novel biosynthetic pathway was employed to produce 6'-SL through de novo synthesis from glycerol in E. coli. First, the key enzymes for 6'-SL biosynthesis were expressed to achieve a yield of 6'-SL of 9.72 mg/L in E. coli. Next, to further increase the yield of 6'-SL, lactose degradation was prevented and genes involved in competing pathways were knocked out. Afterwards, by eliminating carbon catabolite repression (CCR

A Limited Number of Amino Acid Permeases Are Crucial for Cryptococcus neoformans Survival and Virulence.

One unique attribute of Cryptococcus neoformans is its ability to procure essential monomers from its surroundings to survive in diverse environments. Preferentially, sugars are the energy sources for this opportunistic pathogenic fungus under the carbon catabolite repression (CCR); however, sugar restriction induces alternative use of low molecular weight alcohol, organic acids, and amino acids. The expression of transmembrane amino acid permeases (Aaps) allows C. neoformans to utilize different amino acids and their conjugates, notwithstanding under the nitrogen catabolite repression (NCR).

CCR4, a RNA decay factor, is hijacked by a plant cytorhabdovirus phosphoprotein to facilitate virus replication.

Carbon catabolite repression 4 (CCR4) is a conserved mRNA deadenylase regulating posttranscriptional gene expression. However, regulation of CCR4 in virus infections is less understood. Here, we characterized a pro-viral role of CCR4 in replication of a plant cytorhabdovirus, Barley yellow striate mosaic virus (BYSMV). The barley (Hordeum vulgare) CCR4 protein (HvCCR4) was identified to interact with the BYSMV phosphoprotein (P). The BYSMV P protein recruited HvCCR4 from processing bodies (PBs) into viroplasm-like bodies. Overexpression of HvCCR4 promoted BYSMV replication in plants. Conversel

A cybernetic model to predict the effect of freely available nitrogen substrate on rifamycin B production in complex media.

It is well-known that secondary metabolite production is repressed by excess nitrogen substrate available in the fermentation media. Although the nitrogen catabolite repression has been known, quantitative process models have not been reported to represent this phenomenon in complex medium. In this paper, we present a cybernetic model for rifamycin B production via Amycolatopsis mediterranei S699 in complex medium, which is typically used in industry. Nitrogen substrate is assumed to be present in two forms in the medium; available nitrogen (S(ANS)) such as free amino acids and unavailable nit

Preassembled Cas9 Ribonucleoprotein-Mediated Gene Deletion Identifies the Carbon Catabolite Repressor and Its Target Genes in Coprinopsis cinerea.

Cre1 is an important transcription factor that regulates carbon catabolite repression (CCR) and is widely conserved across fungi. The cre1 gene has been extensively studied in several Ascomycota species, whereas its role in gene expression regulation in the Basidiomycota species remains poorly understood. Here, we identified and investigated the role of cre1 in Coprinopsis cinerea, a basidiomycete model mushroom that can efficiently degrade lignocellulosic plant wastes. We used a rapid and efficient gene deletion approach based on PCR-amplified split-marker DNA cassettes together with in vitro

The transcription factor FgCreA modulates ergosterol biosynthesis and sensitivity to DMI fungicides by regulating transcription of FgCyp51A and FgErg6A in Fusarium graminearum.

Fusarium head blight (FHB), caused by Fusarium graminearum, is a devastating disease that severely affects crop yield and quality worldwide. The catabolite responsive elements A (CreA) plays a critical role in numerous cellular processes in eukaryotes. In this study, we performed a functional characterization of CreA in F. graminearum. Deletion of FgCreA led to the transcriptional upregulation of FgCyp51A and FgErg6A, consequently resulting in elevated levels of ergosterol. This augmented ergosterol content was observed to improve cellular membrane integrity and diminished sensitivity to demet

Endogenous metabolite bilirubin suppresses esophageal squamous cell carcinoma by activating PERK-dependent translational repression.

Bilirubin, an endogenous heme catabolite with potent antioxidant properties, has been inversely associated with cancer risk across multiple malignancies; however, its clinical relevance and the underlying molecular mechanisms in esophageal squamous cell carcinoma (ESCC) remain incompletely understood. Here, we retrospectively analyzed a cohort of 2011 patients with ESCC and found that higher serum bilirubin levels were associated with improved overall survival and served as an independent favorable prognostic factor. Mechanistically, physiological bilirubin directly engaged protein kinase R (P

Engineering the acid-tolerant yeast Issatchenkia orientalis for efficient lactic acid production via optimized glucose and xylose utilization.

The acid-tolerant yeast Issatchenkia orientalis is a promising platform for the sustainable production of organic acids. However, the inefficient conversion of lignocellulosic biomass-derived sugars, primarily due to carbon catabolite repression (CCR), reduces overall production efficiency and limits its industrial application. In this study, we established a targeted genetic framework for efficient glucose-xylose co-utilization by coordinating hexokinase (HXK) modulation and transport-level engineering. Sequential fed-batch fermentations revealed that a xylose-initiated feeding strategy achie
