The effect of Khat (Catha edulis) as an appetite suppressant is independent of ghrelin and PYY secretion.

BACKGROUND: The leaves of the khat plant (Catha edulis) are chewed for their pleasurable effects. Chewing releases cathinone which may decrease appetite through an unknown mechanism. Levels of the peptide ghrelin increase with hunger and decrease immediately post-prandially, while peptide YY is released following a meal. We hypothesised that the anorexigenic effects of khat may be mediated through changes in these hormones. MATERIALS AND METHODS: Six habitual khat chewers attended on two separate occasions. For a period of 3h they chewed either khat leaves or lettuce. Blood pressure (BP) and

Catha edulis Leaves: Morphological Characterization and Anti-Inflammatory Properties in an In Vitro Model of Gastritis.

Khat leaves, indigenous to eastern Africa, have been chewed for centuries for their stimulant effects, attributed to alkaloids such as cathinone and cathine. Although associated with gastric disorders like gastritis and gastro-oesophageal reflux disease, the underlying molecular mechanisms remain unclear. This study aimed to examine the morpho-anatomy of khat leaves using light microscopy and histochemistry and to assess the effects of leaf extracts and alkaloids on human gastric epithelial cells (GES-1). The study identified specific cells in the palisade-spongy transition zone as storage sit

Rewarding Effect of Catha edulis (Khat) and the Sex Differences to the Responses in Swiss Albino Mice.

BACKGROUND: Burden of substance abuse is becoming a worldwide problem. One of the psychostimulant plants widely consumed in Ethiopia and other East African countries is Catha edulis Forsk (khat). Most of the users claim that its stimulatory effect is the determinant factor that makes them use. However, its rewarding and reinforcing potential and variation between sexes have not been investigated. This study was, therefore, designed to measure the rewarding effect of khat extract (ke) in the addiction mice model of both sexes. MATERIALS AND METHODS: Forty-eight Swiss albino mice of both sexes

Determination of the active principles of Catha Edulis: quali-quantitative analysis of cathinone, cathine, and phenylpropanolamine.

In the last years, all the vegetable material supposed to belong to the Catha Edulis species, seized at the Malpensa and Orio al Serio airports, were analyzed in our laboratory on behalf of the Tribunals of Busto Arsizio and Bergamo, respectively. After a preliminary botanic examination, the quali-quantitative determination of the active principles cathinone, cathine and phenylpropanolamine (PPA) was carried out by means of GC/MS and GC/FID techniques, which meet the requirements of the forensic analyses. We developed a fast, effective and reliable derivatization procedure which allowed to sim

From Psychoactivity to Antimicrobial Agents: Multifaceted Applications of Synthetic Cathinones and Catha edulis Extracts.

The emergence of new psychoactive substances (NPS) in the global drug market since the 2000s has posed major challenges for regulators and law enforcement agencies. Among these, synthetic cathinones have gained prominence due to their stimulant effects on the central nervous system, leading to widespread recreational use. These compounds, often marketed as alternatives to illicit stimulants such as amphetamines and cocaine, have been linked to numerous cases of intoxication, addiction and death. The structural diversity and enantiomeric forms of synthetic cathinones further complicate their de

Cytotoxicity of Khat (Catha edulis) extract on cultured mammalian cells: effects on macromolecule biosynthesis.

A chloroform extract of Khat (Catha edulis) leaves was used to study the cytotoxic activity on KB, 1BR.3, and XP2Bi cells. Log phase cell survival curves showed an LD50 of 40 ng/ml for KB cells. 1BR.3 and XP2Bi cells were biphasic in their response to the extract during log phase, with an LD50 of 20 and 75 ng/ml, respectively. Stationary phase cells were unaffected by the extract. DNA and RNA synthesis inhibition was studied using radiolabeled thymidine or uridine to measure the amount of extract that inhibits the synthesis to 50% of the untreated control cells. DNA synthesis was inhibited by

Forensic analysis of hallucinogenic mushrooms and khat (Catha edulis Forsk) using cation-exchange liquid chromatography.

Hallucinogenic mushrooms (e.g. Psilocybe and Panaeolus species) as well as leaves and young shoots of the khat tree (Catha edulis Forsk) are illicit drugs in many countries. The exact concentration of the hallucinogenic alkaloids psilocin and psilocybin in mushrooms and the sympathomimetic alkaloids cathinone and cathine in khat is usually essential for jurisdiction. Facing an increasing number of mushroom and khat seizures by German customs authorities, a convenient comprehensive quantitative HPLC method based on cation-exchange liquid chromatography for these rather "exotic" drugs has been d

Effects of Catha edulis Extract on Atorvastatin-Induced Myotoxicity in Rats: Biochemical and Histopathological Evidence.

BACKGROUND: Rhabdomyolysis (RML) is a complex disorder caused by muscle cell injury and the subsequent release of intracellular components into circulation. Statins are widely used and generally well tolerated; however, some patients report muscle weakness, particularly in the lower extremities. The concomitant use of statins with other substances, including herbal products such as khat (Catha edulis), may increase the risk of adverse events. Khat chewing is known to cause multiple health problems and has been associated with musculoskeletal weakness. AIM: This study aimed to evaluate the eff

The presumed neurotoxic effects of Catha edulis--an exotic plant now available in the United Kingdom.

Bilateral optic atrophy is reported in two patients who, although they were long-standing users of the leaves of Catha edulis, had chewed larger quantities than usual. Since the leaves of this plant are used widely throughout its native area as the source of a mildly stimulant narcotic, the effect in these cases may be an idiosyncratic reaction to an unusually large dose.

Catha edulis extract and its active principle cathinone induce ipsilateral rotation in unilaterally lesioned rats.

Catha edulis extract, a natural psychostimulant, and its active component, S-(-)-cathinone, were evaluated for their rotational effects in unilaterally lesioned rats. In our earlier study, we demonstrated that commercially available S-(-)-cathinone, the active principle of C. edulis extract, reverses haloperidol-induced catalepsy. In the current study, we evaluated the effect of C. edulis extract or its active principle, cathinone, in 6-hydroxydopamine unilaterally lesioned rats. Evaluation of the rotational behaviour induced by this natural psychostimulant was made upon acute and repeated ora

The neuropsychopharmacological effects of Catha edulis in mice offspring born to mothers exposed during pregnancy and lactation.

Chewing fresh leaves of the khat plant (Catha edulis Forsk) is a deep rooted and widespread habit in East Africa and the Middle East. Although a body of knowledge exists about the adverse effects of khat during pregnancy, data are sparse with regard to the consequences of long-term exposure during pregnancy and lactation. The present work, therefore, was initiated to evaluate the neuropsychopharmacological effects of Catha edulis exposure during pregnancy and lactation in mice at postnatal day 28. To this effect, a lyophilized extract of khat (100 mg/kg, K100 and 200 mg/kg, K200), amphetamine

Association Between Khat (Catha edulis) Consumption and Hepatocellular Enzyme Elevation: A Hospital-Based Case-Control Study.

BACKGROUND: Khat (Catha edulis) is a widely consumed stimulant in East Africa, but its role in hepatocellular alterations remains debated. OBJECTIVES: This study evaluated the association between khat consumption patterns and subclinical liver injury in Somaliland. DESIGN: A hospital-based case-control study. METHODS: The study was conducted at Hargeisa Group Hospital (N = 260). Biochemical markers were compared among current, past, and non-chewing controls. Because khat use was absent among females, multivariable logistic regression was restricted to males (n = 188). Serum AST, ALT, and bi

Flavonoids in a crude extract of Catha edulis inhibit rat intestinal contraction via blocking Ca2+ channels.

BACKGROUND: Animal studies show that Catha edulis inhibits gastrointestinal tract motility. However, there is little or no information on its effect on colon motility and the mechanism of action and active constituents responsible for this effect. This study therefore attempted to discern the effect, suggest the mechanism, and identify the active compounds from the crude extract. METHODS: A crude extract of Catha edulis was fractionated and subfractionated using column chromatography and HPLC. The activity of all fractions and subfractions was evaluated on rat colon longitudinal muscle. Nucle

Acute liver failure secondary to khat (Catha edulis)-induced necrotic hepatitis requiring liver transplantation: case report.

We describe the case of a 26-year-old man with acute liver failure secondary to ingestion of khat (Catha edulis) leaves. In fact, this is the first case of acute liver failure due to khat reported outside the United Kingdom. The combination of specific epidemiologic data (young man of East African origin) and clinical features (central nervous system stimulation, withdrawal reactions, toxic autoimmune-like hepatitis) led to the diagnosis. Mechanisms of action and potential side effects of khat are elaborated on.
