AA1, a newly synthesized monovalent lipophilic cation, expresses potent in vivo antitumor activity.

Certain lipophilic cations have been reported to display anticarcinoma activities because of their selective uptake and retention by mitochondria of cancer cells. Thus, these agents may comprise a unique class of agents directed against carcinoma. After screening more than 1000 lipophilic cations, we found that the monovalent lipophilic cation, 2,6-bis(4-amino-phenyl)-4-[4-(dimethylamino)phenyl]thiopyrylium chloride (AA1), displayed remarkable anticarcinoma activity both in vitro and in vivo. Unlike most other lipophilic cations, AA1 is stable and displays minimal light sensitivity. In vitro t

When muscle Ca2+ channels carry monovalent cations through gating pores: insights into the pathophysiology of type 1 hypokalaemic periodic paralysis.

Patients suffering from type 1 hypokalaemic periodic paralysis (HypoPP1) experience attacks of muscle paralysis associated with hypokalaemia. The disease arises from missense mutations in the gene encoding the α1 subunit of the dihydropyridine receptor (DHPR), a protein complex anchored in the tubular membrane of skeletal muscle fibres which controls the release of Ca2+ from sarcoplasmic reticulum and also functions as a Ca2+ channel. The vast majority of mutations consist of the replacement of one of the outer arginines in S4 segments of the α1 subunit by neutral residues. Early studies have

Interactions of amiloride and small monovalent cations with the epithelial sodium channel. Inferences about the nature of the channel pore.

The voltage dependence of amiloride-induced inhibition of current flow through apical membrane sodium channels in toad urinary bladder was studied at different ionic conditions. The "inert" salt N-methyl-D-glucamine HCl (NMDG HCl) affected neither the apparent inhibition constant (Kl) for the amiloride-induced current inhibition nor the apparent fraction of the transmembrane voltage that falls between the mucosal solution and the amiloride-binding site (delta). When NMDG+ was replaced with Na+, Kl increased, reflecting amiloride-Na+ competition, whereas delta was unchanged. Similar results wer

Characteristics of the passive 45Ca2+ transport by human blood platelets. Their dependence on the monovalent cation composition of the suspension media.

In the suspension of washed human platelets (32 degrees C, up to 30 min) we measured the uptake of 45Ca2+ in various media. We found that the substitution of Na+ by K+ or choline led to the stimulation of the uptake. According to the degree of the Na+ substitution we observed two phases of stimulation. The first one was observed in K(+)-containing media, but not in choline-containing media; the second one was observed in both K(+)- and choline-containing media. The transport in both phases was saturatable by Ca2+ and sensitive to other divalent cations. The saturating Ca2+ concentration and th

A Pb2+-specific DNA G-quadruplex mimic of red fluorescent protein with a monovalent cation ion tolerance.

Nucleic acid mimics of fluorescent proteins (FPs) currently find wide applications in pursuing high-performance sensors and advanced biomedical tools. However, compared to the mimics of green FPs (GFP), the advancement of mimics of red FPs (RFP) remains limited despite their distinct advantages. Herein, we tried to develop a DNA mimic of RFPs based on an originally screened Bibb Lettuce (BL) and a chromophore originally developed for an RNA mimic of RFPs. Pb2+ causes BL to fold into a G-quadruplex (G4) structure that can accommodate the phenolate form of DMHBO+. We found that activation of the

Spatial Synergy of Crown Ether Groups in Covalent Organic Framework Nanochannels Enabling Efficient Monovalent Cation Separation.

Selective ion transport in nanochannels is critical not only for ion extraction but also for analytical applications requiring high-precision ion isolation prior to detection. However, for ions with identical charge and similar size (e.g., Li+, Na+, K+), it is usually difficult to create sufficiently large differences in transport behavior to achieve high selectivity. Here, we report a covalent organic framework (COF) membrane featuring spatially organized 18-crown-6 sites. These uncharged "-CH2-O-CH2-" groups leverage weak yet size-specific coordination with alkali metal ions. The size compat

A solid-state 23Na NMR study of monovalent cation binding to double-stranded DNA at low relative humidity.

We report a solid-state (23)Na NMR study of monovalent cation (Li(+), Na(+), K(+), Rb(+), Cs(+) and NH(4) (+)) binding to double-stranded calf thymus DNA (CT DNA) at low relative humidity, ca 0-10%. Results from (23)Na--(31)P rotational echo double resonance (REDOR) NMR experiments firmly establish that, at low relative humidity, monovalent cations are directly bound to the phosphate group of CT DNA and are partially dehydrated. On the basis of solid-state (23)Na NMR titration experiments, we obtain quantitative thermodynamic parameters concerning the cation-binding affinity for the phosphate

Mechanisms for monovalent cation-dependent depletion of intracellular Mg2+:Na(+)-independent Mg2+ pathways in guinea-pig smooth muscle.

It has been suggested that magnesium deficiency is correlated with many diseases. 31P NMR experiments were carried out in order to investigate the effects of Na+ substitution on Mg2+ depletion in smooth muscle under divalent cation-free conditions. In the taenia of guinea-pig caeci, the intracellular free Mg2+ concentration ([Mg2+]i) was estimated from the chemical shifts of (1) the beta-ATP peak alone and (2) beta- and gamma-ATP peaks. Both estimations indicated that [Mg2+]i decreased only very slowly in Mg(2+)-free, Ca(2+)-free solutions in which Na+ was substituted with large cations such a

The symbiosome-localised aquaporin GmNOD26 functions as a non-selective monovalent cation channel capable of transporting NH4+ in heterologous systems.

Legumes have developed a symbiotic relationship with soil bacteria known collectively as rhizobia, which when present in root nodules fix atmospheric nitrogen into a form usable by the plant. The functioning unit of symbiotic nitrogen fixation is referred to as the symbiosome and the membrane separating symbionts acts to regulate the metabolite exchange within the root nodule cells. The efflux of fixed nitrogen from the symbiosome is through an unidentified non-selective cation channel, which is inwardly rectified by cytosolic Mg2+ and has a preference for ammonium. Expression of Nodulin 26 (N

Characterization of the K+ (Na+)/H+ monovalent cation exchanger in the human red blood cell membrane: effects of transport inhibitors.

The (ouabain + bumetanide + EGTA)-insensitive K+ influx (defined as residual K+ influx) in the human erythrocyte was investigated with respect to the characterization of the recently identified K+(Na+)/H+ exchanger (Richter et al. 1997). In particular, the effects of selected ion transport inhibitors on this flux in physiological ionic strength (high ionic strength, HIS) as well as low ionic strength (LIS) solutions were qstudied. The stimulation of the K+ influx observed in LIS medium was further enhanced when DIDS, phloretin, eosin-5-maleimide, furosemide, DIOA, NPPB, or DCDPC was present at

K+, Na+, Mg2+, Ca2+, and water contents in human skeletal muscle: correlations among these monovalent and divalent cations and their alterations in K+ -depleted subjects.

None of previous studies had simultaneously analyzed the K(+), Na(+), Mg(2+), and Ca(2+) contents in human skeletal muscle. We examined extensively and simultaneously the levels of all these cations and examined water content in vastus lateralis and pectoralis major muscles in 30 northeastern Thai men who were apparently healthy but died from an accident. Specimen collection was performed within 6 h of death. We used atomic absorption or flame photometry to measure the level of muscle cation. Histopathology of muscle and kidney was also evaluated. Huntingtin regulates Ca(2+) chemotaxis and K(+)-facilitated cAMP chemotaxis, in conjunction with the monovalent cation/H(+) exchanger Nhe1, in a model developmental system: insights into its possible role in Huntington׳s disease.

Huntington׳s disease is a neurodegenerative disorder, attributable to an expanded trinucleotide repeat in the coding region of the human HTT gene, which encodes the protein huntingtin. These mutations lead to huntingtin fragment inclusions in the striatum of the brain. However, the exact function of normal huntingtin and the defect causing the disease remain obscure. Because there are indications that huntingtin plays a role in Ca(2+) homeostasis, we studied the deletion mutant of the HTT ortholog in the model developmental system Dictyostelium discoideum, in which Ca(2+) plays a role in recep

Shear stress activates monovalent cation channel transient receptor potential melastatin subfamily 4 in rat atrial myocytes via type 2 inositol 1,4,5-trisphosphate receptors and Ca(2+) release.

KEY POINTS: During each contraction and haemodynamic disturbance, cardiac myocytes are subjected to fluid shear stress as a result of blood flow and the relative movement of sheets of myocytes. The present study aimed to characterize the shear stress-sensitive membrane current in atrial myocytes using the whole-cell patch clamp technique, combined with pressurized fluid flow, as well as pharmacological and genetic interventions of specific proteins. The data obtained suggest that shear stress indirectly activates the monovalent cation current carried by transient receptor potential melastatin

A pore-localizing CACNA1C-E1115K missense mutation, identified in a patient with idiopathic QT prolongation, bradycardia, and autism spectrum disorder, converts the L-type calcium channel into a hybrid nonselective monovalent cation channel.

BACKGROUND: Gain-of-function variants in the CACNA1C-encoded L-type calcium channel (LTCC, Cav1.2) cause type 8 long QT syndrome (LQT8). The pore region contains highly conserved glutamic acid (E) residues that collectively form the LTCC's selectivity filter. Here, we identified and characterized a pore-localizing missense variant, E1115K, that yielded a novel perturbation in the LTCC. OBJECTIVE: The purpose of this study was to determine whether CACNA1C-E1115K alters the LTCC's selectivity and is the substrate for the patient's LQTS. METHODS: The proband was a 14-year-old male with idiopath

Modulation of dopamine receptors by cations in 7315a, MtTW15 and estradiol-induced pituitary tumors.

Modulation of dopamine (DA) receptors by cations (Na+, K+, Mg2+, Ca2+) was compared in 7315a, MtTW15, and estradiol valerate-induced (EV-T) pituitary tumors, and intact adenohypophysis. In 7315a tumors, the affinity of [3H]spiperone binding measured at 25 degrees remained unchanged in the presence of each cation individually or all these cations together (IONS) compared to the affinity obtained using a buffer without ions; the density (Bmax) was not affected by monovalent cations or Mg2+ and was decreased by Ca2+ or IONS. When binding experiments were done at 37 degrees, monovalent cations inc

Possible mechanisms of inhibition and activation of rat N-acetyltransferase (EC 2.3.1.5.) by cations.

The possible mechanisms of inhibition and activation of various cations on rat pineal N-acetyltransferase (NAT) were elucidated. Copper was found to be a partial mixed noncompetitive inhibitor of NAT with respect to both substrates and this inhibition can be considered to result primarily from impairment of tryptamine (serotonin) binding to the enzyme. Both calcium and magnesium were found to activate NAT by a similar mechanism, with calcium being more effective than magnesium. It appears that the activation results from cation binding to the enzyme causing an increased affinity of tryptamine

Study on the Process of Intermolecular Forces and Electrostatic Force Between Cations and Nano-SiO2 Based on Molecular Simulation.

Although low-permeability oil reservoirs boast abundant resources, oil recovery remains relatively low due to the limitations of current water flooding development technology in oilfields. To address the current challenges of low-permeability oil reservoirs, nano-SiO2 particle aqueous solutions, instead of conventional water injection, have been applied to these reservoirs, which can achieve promising results. Nevertheless, due to the simple surface structure of nano-SiO2 particles, the unsaturated hydroxyl groups on their surfaces tend to undergo electrostatic attraction with cations in forma

Monensin A methyl ester complexes with Li+, Na+, and K+ cations studied by ESI-MS, 1H- and 13C-NMR, FTIR, as well as PM5 semiempirical method.

Monensin A methyl ester (MON1) was synthesized by a new method and its ability to form complexes with Li+, Na+, and K+ cations was studied by electrospray ionization-mass spectroscopy (ESI-MS), 1H and 13C nuclear magnetic resonance (NMR), Fourier transform infrared (FTIR), and PM5 semiempirical methods. It is shown that MON1 with monovalent metal cations forms stable complexes of 1:1 stoichiometry. The structures of the complexes are stabilized by intramolecular hydrogen bonds in which the OH groups are always involved. In the structure of MON1, the oxygen atom of the C=O ester group is involv

Evidence that tetrodotoxin and saxitoxin act at a metal cation binding site in the sodium channels of nerve membrane.

The effects of monovalent, divalent, and trivalent cations on the binding of tetrodotoxin and saxitoxin to intact nerves and to a preparation of solubilized nerve membranes have been examined. All eight divalent and trivalent cations tested, and the monovalent ions Li(+), Tl(+), and H(+) appear to compete reversibly with the toxins for their binding site. The ability of lithium to reduce toxin binding is paralleled by its ability to reduce tetrodotoxin-sensitive ion fluxes through the nerve membrane. We conclude that the toxins act at a metal cation binding site in the sodium channel and sugge
