Effector and regulatory B-cell imbalance in systemic sclerosis: cooperation or competition?

B cells play a central role in the pathogenesis of systemic sclerosis (SSc). Most B-cell studies have focused on their pathological role as antibody producers. However, in addition to immunoglobulin secretion, these cells have a wide range of functions in the immune response, including antigen presentation to T cells and cytokine production. Importantly, not all B-cell subsets promote the immune response. Regulatory B cells (Bregs) attenuate inflammation and contribute to the maintenance of immune tolerance. However, effector B cells (Beffs) positively modulate the immune response through the

Rel governs loser elimination during stem cell competition in the Drosophila testis.

In the Drosophila testis, a group of stromal cells termed hub provides multiple niche signals for the surrounding germline and somatic stem cells. Stem cells of both populations compete for physical retention in the niche, and clones unable to transduce any one niche signal are rapidly eliminated from the stem cell pool by differentiation. We have mapped the transcriptomes of isolated somatic cyst stem cells and differentiated cyst cells, and found that the stem cells but not their differentiated progeny exhibit the signature of an innate immune response including the NF-κB transcription facto

Germline Stem Cell Competition, Mutation Hot Spots, Genetic Disorders, and Older Fathers.

Some de novo human mutations arise at frequencies far exceeding the genome average mutation rate. Examples include the common mutations at one or a few sites in the genes that cause achondroplasia, Apert syndrome, multiple endocrine neoplasia type 2B, and Noonan syndrome. These mutations are recurrent, provide a gain of function, are paternally derived, and are more likely to be transmitted as the father ages. Recent experiments have tested whether the high mutation frequencies are due to an elevated mutation rate per cell division, as expected, or to an advantage of the mutant spermatogonial

Influence of antigenic competition on the development of antibody-forming cell clones.

Isoelectric focusing in polyacrylamide gels was used to investigate the anti-sheep red blood cell antibody responses of mice subjected to antigenic competition. A reduction in the number and intensity of antibody bands was found, even in situations where the suppression of IgG antibody titres was minimal, while with large reductions in titre, antibody bands were rarely seen. It thus appeared that the output of individual B-cell clones was severely depressed during competition. It was concluded that inhibition of clonal expansion is an important feature of competition, and that this may reflect

Tumors exploit interclonal Hedgehog-Wnt crosstalk to drive tumor-promoting cell competition.

Intratumoral heterogeneity fuels cancer progression and therapy resistance, yet the cooperative mechanisms between distinct subclones remain poorly defined. Here, we uncover a tumor-promoting form of cell competition wherein malignant clones hijack interclonal Hedgehog (Hh) and Wnt signaling to convert competitive pressure into a pro-tumorigenic force. In Drosophila, we find that Pp1-87B depleted, RAS-mutant "loser" cells undergo Hh-mediated apoptosis while secreting Wingless (Wg). This paracrine Wg signal hyperactivate β-catenin in neighboring RAS "winner" clones, reprogramming them into inva

Cell competition driven by secreted ligands: Modeling liver metastasis of colorectal cancer.

Cell competition in multicellular organisms has been shown to play a critical role during the development of organisms, cancer progression, and in the establishment and maintenance of tissue homeostasis. Various mechanisms of cell competition have been identified, including active elimination via mechanical forces or induced apoptosis, as well as competition for nutrients and other beneficial factors. A recent experiment demonstrated hallmarks of cell competition, associated with cell cycle dynamics, between liver progenitor cells and colorectal cancer cells [A. Krotenberg Garcia et al., iScie

Foxo3-mediated physiological cell competition ensures robust tissue patterning throughout vertebrate development.

Unfit cells with defective signalling or gene expression are eliminated through competition with neighbouring cells. However, physiological roles and mechanisms of cell competition in vertebrates remain unclear. In addition, universal mechanisms regulating diverse cell competition are unknown. Using zebrafish imaging, we reveal that cell competition ensures robust patterning of the spinal cord and muscle through elimination of cells with unfit sonic hedgehog activity, driven by cadherin-mediated communication between unfit and neighbouring fit cells and subsequent activation of the Smad-Foxo3-

Competition within the early B-cell compartment conditions B-cell reconstitution after hematopoietic stem cell transplantation in nonirradiated recipients.

Severe combined immunodeficiency (SCID) is characterized by a complete block in T-lymphocyte differentiation. Most SCID also affects B-cell development or function, although a normal pool of pro-B cells is detectable. Treatment of SCID consists of allogeneic hematopoietic stem cell transplantation (HSCT), but in the absence of a myeloablative conditioning regimen, only T cells, and in some cases, natural killer (NK) cells, are of donor origin, while all other leukocytes subsets are of host origin. We hypothesized that donor B-cell development success could be conditioned by the competitive abi

Determination of intracellular protein-ligand binding affinity by competition binding in-cell NMR.

Structure-based drug development suffers from high attrition rates due to the poor activity of lead compounds in cellular and animal models caused by low cell penetrance, off-target binding or changes in the conformation of the target protein in the cellular environment. The latter two effects cause a change in the apparent binding affinity of the compound, which is indirectly assessed by cellular activity assays. To date, direct measurement of the intracellular binding affinity remains a challenging task. In this work, in-cell NMR spectroscopy was applied to measure intracellular dissociation

Distinct roles of Bendless in regulating FSC niche competition and daughter cell differentiation.

A major goal in the study of adult stem cells is to understand how cell fates are specified at the proper time and place to facilitate tissue homeostasis. Here, we found that an E2 ubiquitin ligase, Bendless (Ben), has multiple roles in the Drosophila ovarian epithelial follicle stem cell (FSC) lineage. First, Ben is part of the JNK signaling pathway, and we found that it, as well as other JNK pathway genes, are essential for differentiation of FSC daughter cells. Our data suggest that JNK signaling promotes differentiation by suppressing the activation of the EGFR effector, ERK. Also, we foun

FGF21 suppresses hepatocellular carcinoma by driving competitive cell-cell interactions.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide. However, the molecular events underlying its early development are poorly understood. Here, using a murine liver cell competition model combined with single-cell and spatial transcriptomic profiling, we show that fibroblast growth factor 21 (FGF21) mediates tumor-suppressive cell competition in the liver. We demonstrated that FGF21, secreted by oncogenic hepatocytes, drives the directional migration of adjacent normal hepatocytes, leading to the mechanical elimination of oncogenic cells. Mechanistical

Competition for survival among developing ciliary ganglion cells.

1. Functionally different subgroups, each innervating a different part of the peripheral target, were defined within the ciliary population of the avian ciliary ganglion by electrical stimulation of the various ciliary nerve branches. 2. Although neurons innervating defined parts of the peripheral target consistently sent their axons through certain nerves, the technique of retrograde horseradish peroxidase (HRP) transport showed that the ganglion cell bodies were not spatially grouped but distributed throughout the ganglion, both before and after the period of naturally occurring cell death.

Molecularly imprinted polymer-based electrochemical sensor for L-tyrosine competition-mediated T cell exhaustion.

Nutrition competition exists in the tumor microenvironment (TME), which is considered as an important factor of T cell exhaustion. Various amino acids are the targets for both tumor cells and T cells, including L-Tyr, which plays an essential role in normal function of T cells. Herein, a novel sensing interface of MIP/Fc/PEDOT:PSS-PPy/graphite/Au established on a PCB substrate, was proposed for L-Tyr detection. This sensor was systematically characterized and showed excellent repeatability, stability, reproducibility, and specificity. Meanwhile, the sensor exhibited good detection capability,

Cell competition time line: winners kill losers, which are extruded and engulfed by hemocytes.

Cell competition is a mechanism that eliminates slow dividing cells from a growing population. It is believed that the genes wasp, psr, and draper are active in the cells that win the competition ("winner cells") and that they are essential in the winner cells for the induction of apoptosis and for the elimination of the "loser cells." Here, we show that lack of those genes in winner cells appears to be dispensable for cell-competition-induced apoptosis and during dmyc-induced supercompetition. Moreover, winner clones do not need those genes in order to preserve their growth advantage. Finally

From fitness to fate: Hippo-YAP-mediated cell competition influence on stem cells activity.

Cell competition is a fundamental quality-control mechanism where fitter 'winner' cells eliminate less-fit 'loser' counterparts, thereby fine-tuning cell populations during development and maintaining adult tissue integrity. This highly conserved, natural cellular process is absolutely crucial for organogenesis, but once dysfunctional, can instead be exploited by 'super-fit' cancer cells to promote tumourigenesis. This review aims to provide an overview of how heterogeneity is the root cause of cell competition, the factors which influence its emergence, the various modes of cell competition,

MECOM promotes leukemia progression and inhibits mast cell differentiation through functional competition with GATA2.

MECOM is a transcription factor critical for the maintenance of hematopoietic stem cells (HSCs) and the pathogenesis of myeloid leukemia. Germline mutations clustered in the C-terminal zinc finger domain (ZFD) of MECOM are known to cause MECOM-associated syndromes, involving bone marrow failure and skeletal anomalies. However, the molecular consequences of these mutations and the precise downstream mechanisms of MECOM remain elusive. Here, we demonstrate that the C-terminal ZFD serves as the dominant DNA-binding module of MECOM, and that disease-associated mutations abrogate its DNA-binding ca

Steatotic liver disease induces YAP/TAZ-driven cell competition that can suppress tumor initiation.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) and its advanced form, metabolic dysfunction-associated steatohepatitis (MASH), are leading risk factors for liver cancer. While inflammation and fibrosis are known to promote tumorigenesis in MASH, the role of early MASLD in cancer initiation is unclear. Here, we investigated how steatosis alone, without significant fibrosis and inflammation, influences liver cancer development in mouse models. METHODS: We fed mice a Western diet to induce early-stage MASLD, characterized by steatosis and mild hepatitis witho

Dietary Interventions Modulate Cell Competition and Locomotor Decline in an Alzheimer's Disease Drosophila Model.

Alzheimer's Disease (AD) is a neurodegenerative disorder characterised by Amyloid-beta 42 (Aβ42) plaque accumulation and cognitive decline, with current treatments focused on symptomatic relief. Emerging therapeutics, such as dietary interventions, can modulate cognitive decline and delay AD progression. Our previous work in Drosophila melanogaster identified cell competition as a key mechanism that eliminates unfit neurons in an AD model, improving locomotion by removing the unfit neurons expressing flowerLoseB and ahuizotl (azot). Here, we explored how diet influences azot-dependent cell com

Cell competition in hematopoietic cells: Quality control in homeostasis and its role in leukemia.

Cell competition contributes to optimal organ function by promoting tissue homogeneity. In the hematopoietic system, cell competition has been described in two distinct cell populations: in hematopoietic stem cells, and in differentiating T lymphocytes, or thymocytes. In hematopoietic stem cells, cell competition was studied in the context of mild irradiation, whereby the levels of p53 determined the outcome of the cellular interactions and the cells with lower p53 were in advantage. In the thymus, cell competition was addressed in thymus transplantation experiments, and found to be a homeosta

Neutral Competition for Drosophila Follicle and Cyst Stem Cell Niches Requires Vesicle Trafficking Genes.

The process of selecting for cellular fitness through competition plays a critical role in both development and disease. The germarium, a structure at the tip of the ovariole of a Drosophila ovary, contains two follicle stem cells (FSCs) that undergo neutral competition for the stem cell niche. Using the FSCs as a model, we performed a genetic screen through a collection of 126 mutants in essential genes on the X chromosome to identify candidates that increase or decrease competition for the FSC niche. We identified ∼55 and 6% of the mutations screened as putative FSC hypo- or hyper-competitor
