Multiciliated cells (MCC) project dozens to hundreds of motile cilia from the cell surface to generate fluid flow across epithelial surfaces or turbulence to promote the transport of gametes. The MCC differentiation program is initiated by GEMC1 and MCIDAS, members of the geminin family, that activate key transcription factors, including p73 and FOXJ1, to control the multiciliogenesis program. To support the generation of multiple motile cilia, MCCs must undergo massive centriole amplification to generate a sufficient number of basal bodies (modified centrioles). This transcriptional program i

Vitamin A and its active metabolite retinoic acid are essential for the development and function of many tissues including the immune system. The induction of mucosal homing receptors on T and B cells by mucosal dendritic cells (DC) depends on the presence of vitamin A. Recent studies indicate that also the differentiation of CD11b+ DC subsets in the mucosa as well as the spleen depend on vitamin A signalling. As CD11b+ DC subsets exert non-redundant functions in anti-bacterial and anti-fungal immune responses, defects in CD11b+ DC differentiation will contribute to the clinical problems obser

The development of the endocrine pancreas and the differentiation of its five cell types, α, β, δ, ε and pancreatic polypeptide (PP) cells, are a highly complex and tightly regulated process. Proper differentiation and function of α- and β-cells are critical for blood glucose homeostasis. These processes are governed by multiple transcription factors and other signalling systems, and its dysregulation results in diabetes. The differentiation of α-cells and the maintenance of α-cell function can be influenced at several stages during development and in the maturing islet. Many transcription fac

The number of recombinant factors which have been shown to regulate B cell activation, proliferation, and differentiation now stands at nine (IL-1, IL-2, IL-4, IL-5, IL-6 (BSF-2), BCGFLOW, IFN-alpha and gamma, and TNF). Several others have been described but are not yet fully characterized or available as recombinant gene products and will not be discussed. Recent work with these factors, especially IL-4, has revealed a remarkable diversity of function. Most of them seem able to act at more than one stage of B cell activation, proliferation and differentiation, thus invalidating the concept of

Hematopoietic stem cells (HSCs) receive a combination of biochemical and biomechanical signals within the bone marrow that guide their differentiation process. These include soluble factor signaling with cytokines, cellular confinement in the stem cell niche, and contact-dependent receptor-ligand interactions with stromal cells. Recreating this complex microenvironment in vitro is a principal engineering challenge for regenerative therapies and tissue engineering. While cytokines can be easily supplemented in vitro, and several systems for confined HSC culture have been developed, integrating

Circadian rhythms regulate diverse immune processes, yet how they influence memory CD8+ T cell differentiation remains unclear. Here, we show that the time of day of antigen encounter shapes CD8+ T cell fate and antiviral immunity. Immunization during the active phase promotes the generation of progenitor-like memory CD8+ T cells and enhances T cell-mediated protection upon viral challenge. Mechanistically, dendritic cell-intrinsic circadian clocks regulate expression of the costimulatory ligand CD70, thereby directing T cell differentiation. These findings uncover a dendritic cell-mediated ci

The mutualistic relationship of gut-resident microbiota and the host immune system promotes homeostasis that ensures maintenance of the microbial community and of a largely non-aggressive immune cell compartment1,2. The consequences of disturbing this balance include proximal inflammatory conditions, such as Crohn's disease, and systemic illnesses. This equilibrium is achieved in part through the induction of both effector and suppressor arms of the adaptive immune system. Helicobacter species induce T regulatory (Treg) and T follicular helper (TFH) cells under homeostatic conditions, but indu

The contribution of stem cells derived from adult tissues to the recovery of pancreatic islets from chemical injury is controversial. Analysis of nonhematopoietic differentiation of bone marrow-derived cells has yielded positive and negative results under different experimental conditions. Using the smallest subset of bone marrow cells lacking immuno-hematopoietic lineage markers, we have detected incorporation and conversion into insulin-producing cells. Donor cells identified by genomic markers silence green fluorescent protein (GFP) expression as a feature of differentiation, in parallel to

A 12-year-old, male castrated Domestic Shorthair cat was presented to Animal Medical Center of Gifu Univeristy with anorexia and vomiting. Physical examination revealed an enlarged left tonsil and right mandibular lymph node (approximately 2-3× the normal size), and a submucosal mass on the right side of the epiglottis (1.5 × 2.0 cm). On computed tomography images, an enlarged left tonsil, and enlarged right mandibular, right pharyngeal, and left and right cervical lymph nodes were observed. Cytologic examination of smears of tonsil and lymph nodes revealed numerous medium- to large-sized neop

The NLRP3 D301N substitution, the murine ortholog of the human D303N variant, causes NLRP3 constitutive activation (NLRP3CA) and inflammasome assembly. Here, we show that priming signals induced by lipopolysaccharide (LPS) are sufficient to trigger GSDMD-dependent pyroptosis in NLRP3CA-expressing bone marrow-derived macrophages (BMDMs), but not in wild-type (NLRP3WT) cells. NLRP3CA mice exhibit elevated IL-1β secretion and LDH release in bone marrow supernatants under both basal and LPS-challenged conditions. Other cytokines (e.g., TNF-α) increased comparably between genotypes, indicating that

BACKGROUND: Goblet cell metaplasia is a pathological feature of airway remodelling in chronic obstructive pulmonary disease (COPD). Basal cell clones with goblet cell metaplasia demonstrate high transient receptor potential cation channel subfamily C member 6 (TRPC6) expression, but its biological function remains unclear. METHODS: TRPC6 expression was analysed by immunostaining in lung tissues and airway basal cells from patients with COPD, and in the airways of mice exposed to cigarette smoke or interleukin-13. In vitro, the role of TRPC6 was assessed by TRPC6 overexpression and depletion in

The potential role of human milk oligosaccharides (HMOs) in regulating intestinal epithelial cell proliferation and differentiation in mammals has attracted considerable attention. Fucosylated HMO can provide fucosylated glycans to the intestine of newborns, an outcome similar to genetic overexpression of fucosyltransferase. However, how HMOs, such as 2'-fucosyllactose (2'-FL), may impact small intestinal fucosylation to regulate intestinal stem cell (ISC)-mediated intestinal epithelial development remains unknown. In the present study, we employed a model of small intestinal fucosylation inhi

The transcription factor promyelocytic leukemia zinc finger (PLZF) is encoded by the BTB domain-containing 16 (Zbtb16) gene. Its repressor function regulates specific transcriptional programs. During the development of invariant NKT cells, PLZF is expressed and directs their effector program, but the detailed mechanisms underlying PLZF regulation of multistage NKT cell developmental program are not well understood. This study investigated the role of acetylation-induced PLZF activation on NKT cell development by analyzing mice expressing a mutant form of PLZF mimicking constitutive acetylation

Objective: It has been demonstrated that the transcription factors TAZ (transcriptional coactivator with PDZ-binding motif), paired box gene 8 (PAX8), and NK2 homeobox 1 (NKX2-1) are coexpressed in the nucleus of thyroid cells. Furthermore, TAZ is known to enhance the transcriptional activity of PAX8 and NKX2-1 as well as the key thyroid-specific gene, thyroglobulin (TG), suggesting a critical role for TAZ in the control of thyroid cell speciation. We previously reported that the small molecule ethacridine, identified as a TAZ activator, was able to induce thyroid-specific transcription in end

CD4+ T helper cells orchestrate the immune response and play a pivotal role during infection, chronic inflammatory, autoimmune diseases, and carcinogenesis. CD4+ T helper cells can be subdivided into different subsets, which are characterized by a specific network of transcriptional regulators and unique cytokine profiles: Th17 cells express RORγt that in turn promotes the transcription of Il17a, Il17f; Th1 cells, expresses T-bet and produces IFN-γ, IL-2, and TNF-α; Th2 cells express GATA-3 and secrete IL-4, IL-5, and IL-13. The two most studied regulatory T cell subtypes are Foxp3+ regulatory

MicroRNAs (miRNAs) are a class of small (approx. 22 nt) noncoding RNAs that are capable of post-transcriptionally silencing mRNAs that contain sequences complementary to the miRNAs' 7- to 8-bp 'seed' sequence. As single miRNAs are often predicted to target up to hundreds of individual transcripts, miRNAs are able to broadly affect the overall protein expression state of the cell. This can translate into global effects on cellular health and differentiation state. Recently, several reports have identified crucial roles for miRNAs in controlling the production, differentiation, and health of mye

It is proposed that epithelial changes induced by gastroesophageal reflux disease are related to the pH environment of the esophageal lumen. We hypothesized that the various types of esophageal epithelium are associated with specific pH environments that induce their formation. The aim of this study was to compare the luminal pH environment to the histology of the distal esophageal epithelium in patients with gastroesophageal reflux disease. A total of 197 symptomatic patients with increased esophageal acid exposure on 24-hour pH monitoring were grouped according to the histology based on biop

Bone morphogenetic protein-6 (BMP-6) is produced by bone marrow-mesenchymal (BMSC) and hematopoietic stem cells, which can differentiate into bone, cartilage, adipose, muscle, hematopoietic, synovial and other tissues. Bmp6-/- null mice have low hepcidin serum levels and an iron overload, resembling hereditary hemochromatosis, which may cause a reduced number of pancreatic beta-cells, increased serum glucose and diabetes. BMP-6 circulates in the normal human plasma and is produced by BMSC prior to differentiation into osteoblasts. Moreover, it is also released by osteoclasts as a key bone coup
