Cell membrane-coating nanotechnology involves dressing synthetic nanoparticles (NPs) with membranes derived from different types of cells to endow the NPs with the properties of a specific cell type and to further achieve precise and effective disease treatment. Of great interest is the fact that cell membranes sourced from different cell sources can give different functions to the NPs. In this review, we comprehensively lay out the major advantages of several typical cell membranes, which are derived from red blood cells (RBCs), platelets, neutrophils, immune cells, stem cells, or cancer cell

Phosphorylation of erythrocyte membrane proteins was determined in patients with homozygous sickle cell disease. After incubation of ghosts with gamma-32P ATP, proteins were submitted to SDS-polyacrylamide gel electrophoresis. Three salient features appeared: (i) a decreased phosphorylation of spectrin bands; (ii) a significantly increased phosphorylation (P less than 0.001) of bands 4(5) and 4(8) in the absence of cAMP and (iii) a significantly increased phosphorylation (P less than 0.001) of bands 7 and 8, both in the absence and the presence of cAMP. Studies on reticulocyte rich blood showe

Antisera were raised in New Zealand White rabbits against non-B, non-T acute lymphocytic leukemia (ALL) cells coated with antilymphocyte serum. Following minimal absorption with chronic lymphocytic leukemia (CLL) cells, the antiserum reacted mainly with non-B, non-T ALL cells. The following numbers of patients had leukemia cells that reacted with the ALL antisera: 13 of 18 with ALL, 3 of 27 with acute myelocytic leukemia, 1 of 8 with chronic myelocytic leukemia (CML), and 0 of 12 with CLL. The positive CML was a patient in CML blast crisis. Normal peripheral blood B- and T-lymphocytes and norm

With the development of nanotechnology, nanoparticles have emerged as a delivery carrier for tumor drug therapy, which can improve the therapeutic effect by increasing the stability and solubility and prolonging the half-life of drugs. However, nanoparticles are foreign substances for humans, are easily cleared by the immune system, are less targeted to tumors, and may even be toxic to the body. As a natural biological material, cell membranes have unique biological properties, such as good biocompatibility, strong targeting ability, the ability to evade immune surveillance, and high drug-carr

We will consider an array of genetic disorders of the red cell membrane. Some affect well-known genes. The mutations of most cases of hereditary spherocytosis (HS) are located in the following genes: ANK1, SPTB, SLC4A1, EPB42 and SPTA1, which encode ankyrin, spectrin beta-chain, the anion exchanger 1 (band 3), protein 4.2 and spectrin alpha-chain, respectively. A dominant form of distal renal tubular acidosis also stems from distinct mutations in the SLC4A1 gene. The mutations responsible for hereditary elliptocytosis (HE) and its aggravated form, poikilocytosis (HP), lie in the SPTA1 and SPTB

Some recent developments in the author's laboratory which rely upon the techniques of radioreceptor assays (RRA) for drugs and hormones are reviewed. These receptor studies have led, for example, to (a) the identification of 17-alpha estrogens that interact with the opiate receptor, (b) the delineation of a unique class of opiate receptor that mediates explosive motor behavior and is resistant to naloxone, (c) the discovery of progesterone derivatives and active tissue fractions that compete for specific binding sites for cardiac glycosides, (d) the observation that both the opiate and digital

The paper deals with analysis of biochemical, morphological and physiological changes in vitamin A-deficiency. On the basis of this analysis a conclusion is drawn that vitamin A is first of all the factor of differentiation. The intercellular zone is considered to be the most probable sphere of its primary effect. Deficiency in vitamin A causes a change in a concentration and ratio of cations and glycosaminglycans in this zone, that results in disturbance of the intercellular interactions. Other changes--in the DNA-RNA system, protein biosynthesis, sulphate, energy and other metabolisms--might

The action of mercuric chloride and methyl mercuric chloride on the membrane lateral domain organization of bovine, equine, and canine erythrocytes was studied. Electron paramagnetic resonance (EPR) spectra of spin-labeled erythrocytes were analyzed with respect to their lateral domain structure. Continuous alteration of the membrane domain populations revealed that mercuric compounds affect the membrane via the evolution of toxic events in the cells.

The complexity of cell membranes makes the resolution of their macromolecular topology one of the more challenging problems in modern molecular and cellular biochemistry. Despite the difficulties inherent in any such analysis, a surprisingly simple yet powerful approach exists that has consistently yielded valuable results. This method is chemical crosslinking, in which cell membranes are treated with crosslinking reagents (usually bifunctional) which produce covalent linkages between membrane components. The resultant complexes are usually then separated and identified by electrophoresis. Thi

Invasion of the colon wall by Entamoeba histolytica during amoebic dysentery entails migration of trophozoites through tissue layers that are rich in extracellular matrix. Transcriptional silencing of the E. histolytica surface metalloprotease EhMSP-1 produces hyperadherent less-motile trophozoites that are deficient in forming invadosomes. Reversible protein phosphorylation is often implicated in regulation of cell motility and invadosome formation. To identify such intermediaries of the EhMSP-1-silenced phenotype, here we compared the phosphoproteomes of EhMSP-1-silenced and vector control t

Abnormal Heinz body formation in the presence of acetylphenyl hydrazine was observed in some patients with red cell membrane abnormalities, such as hereditary red cell membrane high phosphatidyl choline haemolytic anaemia, congenital haemolytic anaemias of unknown origin, acquired hyperlipidaemia and paroxysmal nocturnal haemoglobinuria. No abnormality of haemoglobin composition or of oxidation-reduction activities was noted in 66 patients studied. No abnormal Heinz body formation was seen in hereditary spherocytosis, hereditary elliptocytosis or hereditary stomatocytosis with normal membrane

Infectious cell entry by non-enveloped viruses requires delivery of the viral genome -- in many cases enclosed within a large, subviral particle -- across the membrane of an intracellular compartment. Rotaviruses and other double-strand RNA (dsRNA) viruses introduce into their target cells an inner capsid particle, roughly 700 Å in diameter, that does not uncoat further but instead extrudes capped viral mRNA by virtue of RNA-dependent RNA polymerase and capping activities within it. The delivery agent is an outer protein layer of the virion. We describe here use of cryogenic electron tomograph

Inspired by natural membrane-fusion processes, membrane-fusogenic nanoparticles have emerged as a versatile platform for precise and efficient therapeutic delivery. By fusing directly with target cells, these systems efficiently transfer encapsulated bioactive cargo into the cytoplasm while bypassing endosomal entrapment. Simultaneously, the fusion process enables direct membrane engineering by integrating functional agents into the cell surface through lipid mixing. Leveraging this dual capability, membrane-fusogenic nanoparticles are evolving from simple gene delivery vehicles to versatile t

UNLABELLED: Bacteria use bacteriocins to kill competitors and shape microbial communities. Many bacteriocins are ribosomally synthesized and post-translationally modified peptides (RiPPs), a diverse class of natural products and a source of new antimicrobials. The diversity of bacterial RiPPs stems from the combination of genetically encoded precursor peptides and biosynthetic proteins that post-translationally process the peptides. Characterized RiPP pathways involve soluble precursor peptides and biosynthetic proteins that modify them in the bacterial cytoplasm before secretion across the ce

Cell entry of nonenveloped animal viruses requires translocation of a macromolecular assembly across a cellular membrane. Double-stranded RNA viruses introduce into the target cell an inner capsid particle that does not uncoat further. Instead, it extrudes capped viral mRNA by virtue of polymerase and capping activities within it. As described here, we used cryogenic electron tomography to visualize the full course of rhesus rotavirus entry, from cell attachment and virion uptake to release of the subviral particle. The cryo-tomograms and subtomogram averaging of classified subparticles link h

The reasons for the wide diversity of lipids found in natural cell membranes are still not fully understood but could potentially be exploited in treating disease and infection. This study aims to establish whether charge alone or specific chemical structure of an anionic lipid headgroup determines the structure and properties of model bacterial cell membranes. We compare different compositions of a zwitterionic lipid di-myristoyl phosphatidylethanolamine (DMPE) and two anionic lipids, di-myristoyl phosphatidylglycerol (DMPG) and tetra-myristoyl cardiolipin (TMCL). TMCL has a distinct condensi

It is challenging to achieve selective off to on modulation of the emissive state of a fluorophore within a complex and heterogeneous cellular environment. Herein we show that the dis-assembly of a non-fluorescent aggregate to produce individual fluorescent molecules, termed disaggregation induced emission (DIE), can be utilised to achieve this goal with an amphiphilic BF2-azadipyrromethene (NIR-AZA) probe. Optical near-infrared properties of the NIR-AZA probe used in this study include absorption and emission maxima at 700 and 726 nm respectively when in the emissive non-aggregated state. Key

In contrast to the released/circulating membrane vesicles (extracellular vesicles), cell-bound membrane vesicles are poorly identified and characterized. In this study, cell-bound membrane vesicles on human umbilical vein endothelial cells (HUVECs) and human hepatoma HepG-2 cells were investigated. We identified that cell-bound membrane vesicles are not co-localized with the major markers for extracellular vesicles (e.g. phosphatidylserine, CD63, CD107α, CD31, and DNA fragments for the three well-known types of extracellular vesicles) and for intracellular organelles with similar sizes (e.g. M
