Metastatic effect of LY-6K gene in breast cancer cells.

The over-expression of urokinase receptor in breast cancer cells results in increased tumor invasion, growth and metastasis. As LY-6K belongs to the Ly6/uPAR superfamily, we assessed the association of LY-6K with metastasis in breast cancer cell lines and how the LY-6K gene mediates cancer cell metastasis. The migration ability of cells in which expression of LY-6K gene is up- or down-regulated was investigated by transfecting LY-6K naked DNA and treating LY-6K siRNA. Assays of wound healing, migration and invasion were performed. The assays demonstrated that cell migration significantly incre

Inhibition of melanoma cell motility by the snake venom disintegrin eristostatin.

Eristostatin, an RGD-containing disintegrin isolated from the venom of Eristicophis macmahoni, inhibits lung or liver colonization of melanoma cells in a mouse model. In this study, transwell migration and in vitro wound closure assays were used to determine the effect of eristostatin on the migration of melanoma cells. Eristostatin significantly impaired the migration of five human melanoma cell lines. Furthermore, it specifically inhibited cell migration on fibronectin in a concentration-dependent manner, but not that on collagen IV or laminin. In contrast, eristostatin was found to have no

Rac/Cdc42 inhibitors target pancreatic cancer cells and macrophages in the tumor microenvironment.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the deadliest forms of cancer, with a distinct extracellular matrix and immunosuppressive tumor microenvironment. Therefore, there is a critical need for therapies that simultaneously target metastatic cancer cells and immunosuppressive cells such as tumor associated macrophages (TAMS) that promote pancreatic cancer progression. Ras-activated homologous GTPases Rac and Cdc42 are ideal targets for pancreatic cancer therapy because they regulate cell migration/invasion, cell polarity/morphology, and cell viability/survival in cancer cells an

The piR-31115-PIWIL4 complex promotes the migration of the triple-negative breast cancer cell lineMDA-MB-231 by suppressing HSP90AA1 degradation.

BACKGROUND: P-element-induced wimpy testis (PIWI) proteins bind to PIWI-interactingRNAs (piRNAs) to form the piRNA/PIWI complex, which affects protein regulation. PIWIL4, a member of the PIWI family, has been demonstrated in recent studies to promote the migration of triple-negative breast cancer (TNBC) cell line MDA-MB-231. However, the molecular mechanisms underlying cell migration remain obscure. METHODS: RNA immunoprecipitation and real-time PCR assays were conducted to detect piRNAs binding to PIWIL4. piRNA mimics and inhibitors were employed to modify piRNA expression in MDA-MB-231 cell

UCHL3 Promotes Triple-Negative Breast Cancer Metastatic Potential Through Enhancing Cell Migration and Invasion.

Ubiquitin carboxyl-terminal hydrolase L3 (UCHL3), a key deubiquitinating enzyme of the UCH family, has been implicated in DNA repair in breast cancer; however, its role in regulating cell migration and invasion in triple-negative breast cancer (TNBC) remains poorly understood. The expression pattern of UCHL3 across breast cancer subtypes was analyzed using TCGA data. In TNBC cells, UCHL3 mRNA levels were quantified by qRT-PCR, while its protein expression and epithelial-mesenchymal transition (EMT) markers (Vimentin, N-cadherin, SNAIL, E-cadherin, α-SMA, TWIST) were assessed by Western blot. Uncertainty-aware quantitative analysis of high-throughput live cell migration data.

Cell migration is a fundamental biological process essential for embryonal development, immune function, and cancer metastasis, with migration velocity representing a key parameter of this behaviour. Today, cell migration velocity can be measured in high-throughput assays that generate complex, hierarchically structured datasets with technical noise, batch effects, and biological variability, introducing significant uncertainty in velocity estimates. Current statistical approaches often fail to rigorously quantify this uncertainty, limiting reproducibility and comparisons across independent ex

Migration of gastric cancer is suppressed by recombinant Newcastle disease virus (rL-RVG) via regulating α7-nicotinic acetylcholine receptors/ERK- EMT.

BACKGROUND: Nicotinic acetylcholine receptors (nAChRs) have been reported to be overexpressed in malignancies in humans and is associated with tumorigenesis and cell migration. In previous studies of gastric cancer, alpha7 nicotinic acetylcholine receptor (α7-nAChR) overexpression leads to epithelial-mesenchymal transition (EMT) and promotes the migration of gastric cancer cells. Recombinant avirulent LaSota strain of Newcastle disease virus (NDV) expressing the rabies virus glycoprotein (rL-RVG) may promote apoptosis of gastric cancer cells and reduces the migration of lung cancer metastasis.

Characterizing Cell Migration Within Three-dimensional In Vitro Wound Environments.

Currently, most in vitro models of wound healing, such as well-established scratch assays, involve studying cell migration and wound closure on two-dimensional surfaces. However, the physiological environment in which in vivo wound healing takes place is three-dimensional rather than two-dimensional. It is becoming increasingly clear that cell behavior differs greatly in two-dimensional vs. three-dimensional environments; therefore, there is a need for more physiologically relevant in vitro models for studying cell migration behaviors in wound closure. The method described herein allows for th

Frog-Derived Peptide RL-RF10 Facilitates Gingival Repair and Regeneration Through the Integrin αvβ3/p38/Snail1 Axis.

BACKGROUND: Despite advancements in dental therapies, insufficient gingival tissue remains a significant challenge. Currently, no specific medications promote the regeneration of gingival tissue, with existing treatments primarily redistributing tissue rather than restoring it. Amphibian bioactive peptides show promise but remain underexplored in gingival repair. This study investigates the potential of RL-RF10, a peptide derived from frogs, for gingival tissue repair. METHODS: The localization of RL-RF10 was monitored using fluorescein isothiocyanate labelling. The effects of RL-RF10 on the
