EIF3H Modulates Glycolysis Through LDHA Stabilization in Triple-Negative Breast Cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) lacks effective targeted therapies, and its dependence on glycolysis represents a potential metabolic vulnerability. EIF3H, the largest subunit of the eukaryotic translation initiation factor 3 complex and a putative deubiquitinase, has been implicated in tumor progression, but its role in TNBC metabolism remains unclear. METHODS: EIF3H expression and prognostic value were evaluated in public datasets, clinical TNBC specimens, and cell lines. Functional roles were examined using proliferation, colony formation, migration, and xenograft assays.

Macrophage Enhancement of Tumor Cell Extravasation and Intravasation In Vitro.

Macrophages orchestrate multiple functions in the tumor microenvironment leading to metastatic progression including promotion of tumor cell invasion, intravasation, and extravasation at metastatic sites. In vitro assays have been developed to explore these processes in high resolution. Here we describe an in vitro protocol used to examine macrophage enhanced tumor cell extravasation and intravasation in 3D using a stratified co-culture transendothelial migration (TEM) transwell system. These assays may be generally applied in order to study different aspect of metastatic dissemination, and we

GOLPH3-mediated sialylation of CXCR4 drives malignant progression and M2 macrophage polarization in colorectal cancer.

Sialylation, a post-translational protein modification, correlates with tumor malignancy. This study investigates how GOLPH3 regulates CXCR4 sialylation in colorectal cancer (CRC) and its impact on cancer cell phenotypes. Predicted glycosylation sites on CXCR4 were identified using the NetNGlyc-1.0 tool. The GOLPH3-CXCR4 interaction and GOLPH3-sialyltransferase associations were bioinformatically predicted (BIOGRAD) and validated experimentally via co-immunoprecipitation (Co-IP). CXCR4 sialylation was assessed by immunoprecipitation (IP) and lectin blotting (LB). Immunohistochemical (IHC) asse

FTO-mediated the destabilization of RASGRF1 mRNA impedes thyroid cancer progression and suppresses macrophage M2 polarization.

BACKGROUND: The guanine nucleotide exchange factor RASGRF1 actively acts in a broad range of human cancers, including thyroid cancer (THCA). This study defined the activity of RASGRF1 in THCA progression and elucidated the m6A modification mechanism governing dysregulation of RASGRF1. METHODS: Expression analyses were performed by immunoblotting, immunohistochemistry (IHC) or quantitative PCR. Cell growth was evaluated by colony formation and EdU proliferation assays. Animal experiments tested the function of RASGRF1 in xenograft growth. The conditioned medium (CM) of THCA cells was used to t

Tsc1 is a Critical Regulator of Macrophage Survival and Function.

BACKGROUND/AIMS: Tuberous sclerosis complex 1 (Tsc1) has been shown to regulate M1/M2 polarization of macrophages, but the precise roles of Tsc1 in the function and stability of macrophages are not fully understood. Here we show that Tsc1 is required for regulating the survival, migration and phagocytosis of macrophages. METHODS: Mice with Tsc1 homozygous deletion in myeloid cells (LysMCreTsc1(flox/flox); Tsc1 KO) were obtained by crossing Tsc1(flox/flox) mice with mice expressing Cre recombinase under the control of Lysozyme promoter (LysMCre). The apoptosis and growth of macrophages were de

A micro macrophage migration inhibition test for the detection of cellular immunity in vitro.

A simple, economical, macrophage migration inhibition assay enabling multiple tests to be performed using microculture plates has been developed. The system confers many advantages over the standard in vitro assay of macrophage migration inhibition factor. It requires only 5 X 10(5) macrophages for each assay, small volumes (100 microliter) of test factors are consumed, 96 tests in each culture plate are easily assessed microscopically and the technique is easily and reproducibly executed. An adherent monolayer of activated fresh mouse peritoneal exudate cells (PEC) is established in each micr

Artesunate Suppresses the Migration and Invasion of Thyroid Cancer Cells via Upregulating PTEN to Block M2 Polarization of Tumor-Associated Macrophages.

Immunotherapy holds promise for thyroid cancer (TC) treatment. In the context of our previous findings that artesunate (ART) could inhibit the migration and invasion of TC cells through phosphoinositide 3-kinase/protein kinase B (PI3K/Akt), this study was engineered to investigate whether ART regulates the tumor microenvironment in TC. THP-1 cells were differentiated into M0 macrophages by the induction of 100 ng/mL of phorbol 12-myristate 13-acetate and transfected as needed. M0 macrophages were treated with different concentrations of ART (10 and 20 μM) for 24 h. The co-culture of macrophage

Macrophage chemotaxis to apoptotic Burkitt's lymphoma cells in vitro: role of CD14 and CD36.

In Burkitt's lymphoma (BL), apoptosis occurs at high frequency alongside uncontrolled proliferation. Macrophages infiltrate these tumours in large numbers and engage in the phagocytic clearance of apoptotic cells in situ. Here we tested the hypothesis that apoptosis of BL cells may provide a mechanism for recruitment of macrophages to these tumours. We show that monocytes and macrophages, but not neutrophils, preferentially migrated to apoptotic BL cells in vitro. Transfection of BL cells with the anti-apoptotic gene bcl-2 both prevented apoptosis and abolished macrophage chemotaxis. Macrophag

Exosomes derived from itaconate-treated macrophages promote epithelial-mesenchymal transition, migration, and invasion in MCF-7 breast cancer cells.

Itaconic acid is an immunometabolite that regulates macrophage polarization within the tumor microenvironment and influences tumor progression and invasion. Although its anti-cancer and tumor-promoting effects remain controversial, its role in modulating macrophage-derived exosomes and their impact on breast cancer cells is not well understood. This study investigated the effects of exosomes derived from dimethyl itaconate (DMI)-treated macrophages on epithelial-mesenchymal transition (EMT), apoptosis, migration, and EMT-related miRNAs in MCF7 breast cancer cells. U937 macrophages were treated

Amnion-derived cellular cytokine solution promotes macrophage activity.

Activated macrophages play a significant role in wound healing and infected tissue repair. In this study, we investigate the recruitment of macrophages into the wound, and the effects on the bactericidal/phagocyte activity after exposure to amnion-derived cellular cytokine solution (ACCS). To evaluate the influence of ACCS on the migratory behavior of macrophages, cell migration was assayed quantitatively using a Boyden chamber. Chemotactic migration activity of macrophages through the membrane determined the influence of ACCS. In the presence of ACCS, macrophages demonstrated a statistically
