Acetylcholine receptors in cultured human muscle cells.

Primary cultures prepared from human muscle biopsies were examined for the presence and distribution of acetylcholine receptors, as measured by the binding of 125iodine-labelled alpha-bungarotoxin. The toxin bound to the human muscle cultures with a similar time dependence and specificity as found in muscle cultures from other species. The amount of toxin bound was lower than that obtained for neonatal mouse muscle under similar conditions. The distribution of receptors was similar in cultures derived from the muscles of patients with a variety of neuromuscular disorders. The toxin was located

Osteoblastic cells regulate mi gene expression in cultured bone marrow cells.

The microphthalmia (mi) gene encodes a bHLH-Zip protein and certain mutations in this gene are known to result in osteopetrosis. We examined mi gene expression in murine bone marrow (BM) cells and calvaria-derived osteoblastic (OB) cells. The mi message level in BM cells was three- to four-fold higher than that in OB cells. The mi message level in BM cells alone was not enhanced by 1,25(OH)2 vitamin D3 (vitamin D) which induces TRAP-positive multinucleated cells, but was enhanced two-fold by the addition of conditioned medium obtained from cultures of OB cells. The mi message levels in BM cell

Thiols stabilize cobblestone morphology of cultured mesothelial cells.

Cellular thiols including GSH (glutathione) and L-Cys (L-cysteine) are essential for cell signalling, growth and differentiation. L-Cys is derived from the extracellular thiol pool and is the rate-limiting compound for intracellular GSH biosynthesis. The present study investigated the effect of thiol-supplemented medium on cell growth, phenotype and total GSH of cultured hPMCs (human peritoneal mesothelial cells). Cells were cultured in medium M199 supplemented with 2% serum, with 'plus' or without 'minus' L-Cys and compared with medium supplemented with either β-ME (β-mercaptoethanol) (0.25 m

Electrode assemblies used for electroporation of cultured cells.

Electroporation was initially developed for the introduction of DNA into cells which grow in suspension and was performed in a cuvette with two flat electrodes on opposite sides. Different configurations were subsequently developed for the electroporation of adherent cells in situ, while the cells were growing on nonconductive surfaces or a gold-coated, conductive support. We developed an assembly where the cells grow and are electroporated on optically transparent, electrically conductive indium-tin oxide (ITO). This material promotes excellent cell adhesion and growth, is inert and durable,

Cultured human muscle cells and respiratory chain deficiencies.

Cultured muscle cells are useful in the study of respiratory chain disorders. Muscle tissue is affected in most cases and muscle biopsies are often taken for diagnostic purposes. Small samples of the biopsies can provide large numbers of muscle cells. In contrast with most other cell types, the muscle cells can differentiate in culture. Upon differentiation, multinuclear myotubes are formed by fusion of myoblasts and muscle-specific proteins are synthesized in the myotubes. Muscle cells can be used for diagnostic purposes, e.g. by measurement of the lactate and pyruvate synthesis, although wit

Effects of prolactin on cultured human endometrial cells.

We report the effects of PRL on cultured human endometrial cells (epithelial cells and stromal cells) and discuss the role of PRL in implantation. Essentially similar responses to PRL in cell attachment and growth were obtained both in epithelial and stromal cells: a low concentration of PRL (3-30 ng/ml of PRL) enhanced cell attachment and growth. In contrast, the presence of a high concentration of PRL (over 100 ng/ml) resulted in a decrease in cell attachment and growth in normal culture conditions. Morphological analysis of epithelial glandular reformation on basement membrane extracts indi

Geometrical constraints on the shape of cultured cells.

Previous studies on cultured cells revealed that the values of certain shape descriptors were highly correlated despite the fact that there was little mathematical interdependence (Heckman, CA: In Advances in Cell Culture, Vol. 4, K. Maramorosch, ed, pp 85-156, 1985). The independence of such descriptors was tested by calculating the values of correlations among descriptors for a data set consisting of macroscopic objects. The descriptors were unlinked for this data set, confirming that high correlations reflected specific characteristics of cultured cells. These characteristics were identifie

Hypersensitivity of cultured ataxia-telangiectasia cells to etoposide.

Fibroblasts from patients with ataxia-telangiectasia (A-T) were found to be hypersensitive to killing by the antineoplastic agent etoposide. The A-T fibroblast strains GM5823, GM367, and GM2052 were twofold to threefold more sensitive to killing by etoposide than fibroblasts from normal controls (AG1521, AG1522, and IMR90). A simian virus 40 (SV40)-transformed, immortal human fibroblast line (GM5849) derived from the A-T cell line GM5823 was also studied. GM5849 retained the unusual sensitivity of nontransformed A-T fibroblast lines to x-irradiation, bleomycin, and neocarzinostatin (zinostatin

Alkaline phosphatase isozymes in cultured human cancer cells.

Alkaline phosphatase, an ubiquitous enzyme is known to exist in several isozymic forms. At least three different isozymes have now been identified in humans. Alkaline phosphatase isozymes are among the substances synthesized ectopically by a variety of human tumors and many continuous cell lines derived from different cancers have retained the capacity to produce these membrane-located glycoproteins. This paper reviews the identification of alkaline phosphatase isozymes in cultured tumor cells and relates these findings with recent developments concerning these cell membrane located glycoprote

Metabolic Alterations in Spheroid-Cultured Hepatic Stellate Cells.

Hepatic stellate cells (HSCs) play a vital role in liver fibrosis, and a greater understanding of their regulation is required. Recent studies have focused on relationships between extracellular matrix (ECM) stiffness and gene expression or cellular metabolism, but none have provided a detailed metabolic analysis of HSC changes in spheroid cultures. Accordingly, in the present study, we created an HSC spheroid culture and analyzed changes in gene expression and metabolism. Expression of α-smooth muscle actin (α-SMA) decreased in the spheroids, suppressing proliferation. Gene expression analysi

Interleukin-2 Production by Cultured Human Granulosa Cells.

PROBLEM: Ovarian hyperstimulation syndrome (OHSS) is similar to vascular leak syndrome (VLS), which may be attributable to the massive increase in systemic inflammatory cytokines. The hyperstimulated human ovaries were demonstrated to contain interleukin (IL)-2, which, in turn, was suggested to activate the systemic inflammatory response characteristic of OHSS. As the source of follicular fluid IL-2 is still unclear, in the present study, we sought to validate the presence of IL-2 and IL-2 mRNA expression in human luteinized granulosa cells. METHOD OF STUDY: IL-2 nuclear expression was detect

Generation of iPSCs from cultured human malignant cells.

Induced pluripotent stem cells (iPSCs) can be generated from various differentiated cell types by the expression of a set of defined transcription factors. So far, iPSCs have been generated from primary cells, but it is unclear whether human cancer cell lines can be reprogrammed. Here we describe the generation and characterization of iPSCs derived from human chronic myeloid leukemia cells. We show that, despite the presence of oncogenic mutations, these cells acquired pluripotency by the expression of 4 transcription factors and underwent differentiation into cell types derived of all 3 germ

Karyotype changes in cultured human corneal endothelial cells.

PURPOSE: To examine karyotype changes in cultured human corneal endothelial cells (HCECs). METHODS: HCECs with Descemet's membrane were removed from 20 donors of various ages (range, 2-77 years; average, 43.7+/-26.4 years) and cultured on dishes coated with extracellular matrix produced by bovine corneal endothelial cells (BCECs). Karyotype changes were examined by G-band karyotyping of HCECs at the third passage from 12 donors and the fifth passage from 16 donors. The number of chromosomes was analyzed in 40-50 cells from the third and fifth passages of each HCEC preparation. A detailed kary

Cultured peritoneal mesothelial cells exhibit apical primary cilia.

This study examined primary cilia on cultured human and rabbit peritoneal mesothelial cells (PMC) and investigated factors that influence ciliary expression. Primary cilia were examined with indirect immunocytochemistry, laser scanning confocal microscopy and scanning electron microscopy. Ciliary expression was evaluated in cultures with or without l-cysteine (0.25 mM) or exposure to Ca(2+)-free Krebs-Ringer solution supplemented with EGTA, 0.5 mM. This treatment disrupted cell monolayer integrity. Cilia were counted and normalized to total cell counts using NIH image. Primary cilia were ident

[Reverse cholesterol transport in cultured gallbladder epithelial cells].

Bile is the major route of cholesterol excretion from the body. It is concentrated in the gallbladder, and often results in supersaturation of cholesterol. The high levels of cholesterol in gallbladder bile has clinical implications with respect to cholesterol gallstone formation and cholesterolosis of the gallbladder wall. Gallbladder epithelial cells (GBEC) are exposed to high cholesterol concentrations on their apical surfaces. Therefore, GBEC are uniquely positioned to play an important role in modulating biliary cholesterol concentrations. Recently, it has been documented that the key-tra

Interleukin-2 production by cultured human granulosa cells.

OBJECTIVE: To determine whether cultured human granulosa cells (GC) produce Interleukin (IL)-2 and whether this GC IL-2 production may be regulated by human chorionic gonadotropin (hCG). MATERIALS AND METHODS: Human GC derived from preovulatory follicles during in vitro fertilization (IVF) cycles were cultured in the absence or presence of hCG. Interleukin-2 was measured in tissue-culture medium by an enzyme-linked immunosorbent assay (ELISA) system. RESULTS: Cultured GCs were demonstrated to produce IL-2 in vitro. Moreover, a positive, but not statistically significant, dose-response effect
