Effect of host fruit, temperature and Wolbachia infection on survival and development of Ceratitis capitata immature stages.

The Mediterranean fruit fly, Ceratitis capitata (Diptera: Tephritidae), holds an impressive record of successful invasions promoted by the growth and development of international fruit trade. Hence, survival of immatures within infested fruit that are subjected to various conditions during transportation seems to be a crucial feature that promotes invasion success. Wolbachia pipientis is a common endosymbiont of insects and other arthropods generating several biological effects on its hosts. Existing information report the influence of Wolbachia on the fitness traits of insect host species, in

Natural Oil Lure Outperforms Trimedlure in Capturing Males of the Mediterranean Fruit Fly, Ceratitis capitata (Diptera: Tephritidae).

Females of certain tephritid fruit fly species (Diptera: Tephritidae) pose an enormous agricultural threat, as they oviposit in commercially important fruits and vegetables. Trapping networks are often operated in fruit fly-free areas to detect incipient infestations. Trapping relies largely on male attractants, so-called male lures, with trimedlure (TML) being used to detect invasive Ceratitis spp. Operating large-scale surveillance programs incurs substantial costs for both supplies and labor, and the problem is exacerbated by the fact that trimedlure (as well as other male lures) is effecti

Metabolic and microbial responses of Ceratitis capitata to essential oil-based nano-emulsions: Implications for pest management.

Essential oils (EOs) are a promising alternative to conventional pesticides, but some challenges like high volatility, poor water solubility, and rapid degradation limit their use in Integrated Pest Management (IPM). To overcome these limitations, this study aimed to develop garlic, eucalyptus, and clove EO-based nano-emulsions (EO-NEs) in a bait treatment format through the high-pressure microfluidization technique and investigated the biological activities against Ceratitis capitata. In addition, the adverse effects of the most promising nano-emulsion were evaluated towards a non-target para

Detection of Wolbachia in Natural Populations of Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) Infesting Argan Fruits in Morocco.

The argan tree (Argania spinosa L. Skeels), native to the sub-Saharan region of Morocco, is an endangered agroforestry species renowned for producing one of the world's most expensive and sought-after oils. However, this valuable resource is threatened by the Mediterranean fruit fly (Ceratitis capitata (Wied.)), an invasive pest worldwide. Like other dipteran insects, C. capitata has developed mutualistic interactions with prokaryotic endosymbionts, including Wolbachia, a facultative intracellular bacterium that could play a role in the biology of this pest. Between 2022 and 2024, a field surv

Effect of Wolbachia Infection and Adult Food on the Sexual Signaling of Males of the Mediterranean Fruit Fly Ceratitis capitata.

Sexual signaling is a fundamental component of sexual behavior of Ceratitis capitata that highly determines males' mating success. Nutritional status and age are dominant factors known to affect males' signaling performance and define the female decision to accept a male as a sexual partner. Wolbachia pipientis, a widespread endosymbiotic bacterium of insects and other arthropods, exerts several biological effects on its hosts. However, the effects of Wolbachia infection on the sexual behavior of medfly and the interaction between Wolbachia infection and adult food remain unexplored. This stud

Ultrastructure of the male reproductive accessory glands in the medfly Ceratitis capitata (Diptera: Tephritidae) and preliminary characterization of their secretions.

The morphology and the ultrastructure of the male accessory glands and ejaculatory duct of Ceratitis capitata were investigated. There are two types of glands in the reproductive apparatus. The first is a pair of long, mesoderm-derived tubules with binucleate, microvillate secretory cells, which contain smooth endoplasmic reticulum and, in the sexually mature males, enlarged polymorphic mitochondria. The narrow lumen of the gland is filled with dense or sometimes granulated secretion, containing lipids. The second type consists of short ectoderm-derived glands, finger-like or claviform shaped.

Targeting the autosomal Ceratitis capitata transformer gene using Cas9 or dCas9 to masculinize XX individuals without inducing mutations.

BACKGROUND: Females of the Mediterranean fruit fly Ceratitis capitata (Medfly) are major agricultural pests, as they lay eggs into the fruit crops of hundreds of plant species. In Medfly, female sex determination is based on the activation of Cctransformer (Cctra). A maternal contribution of Cctra is required to activate Cctra itself in the XX embryos and to start and epigenetically maintain a Cctra positive feedback loop, by female-specific alternative splicing, leading to female development. In XY embryos, the male determining Maleness-on-the-Y gene (MoY) blocks this activation and Cctra pro

Quantification of dose-mortality responses in adult Diptera: Validation using Ceratitis capitata and Drosophila suzukii responses to spinosad.

Quantitative laboratory bioassay methods are required to evaluate the toxicity of novel insecticidal compounds for pest control and to determine the presence of resistance traits. We used a radioactive tracer based on 32P-ATP to estimate the volume of a droplet ingested by two dipteran pests: Ceratitis capitata (Tephritidae) and Drosophila suzukii (Drosophilidae). Using blue food dye it was possible to distinguish between individuals that ingested the solution from those that did not. The average volume ingested by C. capitata adults was 1.968 μl. Females ingested a ~20% greater volume of solu

Egg-stage desiccation reduces developmental success and reveals line-dependent Wolbachia-associated costs in the Mediterranean fruit fly, Ceratitis capitata.

The Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann, 1824) is a major agricultural pest, and egg desiccation is a critical constraint during handling and mass-rearing, as even short periods without moisture may compromise developmental success and downstream adult performance. The Wolbachia-medfly symbiosis is a relatively recently established artificial association, generated less than three decades ago using Rhagoletis cerasi as the Wolbachia donor. In this study, we evaluated the effects of egg-stage desiccation on developmental success and subsequent adult performance in th

Fitness improvement of mass-reared sterile males of Ceratitis capitata (Vienna 8 strain) (Diptera: Tephritidae) after gut enrichment with probiotics.

Successful mass rearing is crucial for sterile insect technique programs. It has been shown that the sterilizing process using gammaradiation results in damage to midgut tissue, cellular organelles, and gut microbiota of flies. This can be responsible for the inferiority of sterile males compared with wild males. A bacteria-enhanced diet could contribute to the improvement of the fly's fitness. We investigated ways of increasing the competitiveness of mass-reared Mediterranean fruit fly, Ceratitis capitata (Wiedemann) sterile males. We tested the hypothesis that the addition of beneficial bact

Deep Learning-Based Classification of In Vivo Embryonic Development Stages in Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) Using Real-Time Microscopy Images.

Accurate staging of embryonic development is crucial for applied studies that require precise determination of embryonic age, such as developmental biology research and phenotype-based assessments using microinjection. However, the determination of embryonic development stages in the Mediterranean fruit fly, Ceratitis capitata, still largely relies on manual morphological assessment, which is labor-intensive and susceptible to observer bias. In this study, we evaluated deep learning models for the automatic classification of embryonic development stages in C. capitata using time-lapse microsco

Evaluating impacts of radiation-induced sterilization on the performance and gut microbiome of mass-reared Mediterranean fruit fly (Ceratitis capitata) in Hawai'i.

Sterile insect technique (SIT) is a useful strategy for preventing and mitigative establishment of invasive insect species. SIT of the pest tephritid Mediterranean fruit fly, Ceratitis capitata (Wiedemann, 1824)WiedemannWiedemann, has been effective in preventing population establishment in vulnerable agricultural areas of the United States. However, irradiation-induced sterilization can have detrimental impacts resulting in reduced performance metrics. Mediterranean fruit fly males reared for SIT have been shown to have differences in their microbiomes relative to other population sources, wh

DsiRNA-mediated silencing of Ceratitis capitata transformer or transformer-2 leads to masculinization of XX embryos and systemic gene silencing in ovaries.

Ceratitis capitata (medfly), a major agricultural pest, is predominantly controlled using chemical insecticides, which pose environmental risks. Ecosustainable alternatives, such as the Sterile Insect Technique (SIT), rely on the mass release of sterile male-only progeny. However, sexing of male offspring requires the elimination of females during development. To overcome the loss of 50% of the progeny, the sex reversal of females into XX fertile males at embryonic stages by dsRNA injections was effective but not scalable. This study demonstrates the efficacy of Dicer-substrate small interferi

Use of honey bees (Apis mellifera L.) to detect the presence of Mediterranean fruit fly (Ceratitis capitata Wiedemann) larvae in Valencia oranges.

BACKGROUND: When fruit deteriorates a characteristic profile of volatile chemicals is produced that is different from that produced by healthy fruits. The identification of such chemicals allows the possibility of monitoring the fruit for early signs of deterioration with biological sensors. The use of honey bees and other insects as biological sensors is well known. This study aimed to identify the volatiles produced by oranges infested with larvae of the Mediterranean fruit fly and to test the ability of honey bees, conditioned to this volatile chemical profile, to detect such oranges. Towards improving sterile insect technique: Exposure to orange oil compounds increases sexual signalling and longevity in Ceratitis capitata males of the Vienna 8 GSS.

The Mediterranean fruit fly (medfly), Ceratitis capitata, is a notorious insect pest causing huge economic losses worldwide. The sterile insect technique (SIT) is widely used for its control. Using sexually mature sterilized males of the Vienna 8 (tsl) strain in the laboratory, we explored whether exposure of males to citrus compounds (separately or in a mixture) affects their sexual behaviour and if nutritional conditions and age modulate those effects. Exposed males exhibited increased sexual signalling compared to unexposed ones, particularly when fed a rich adult diet. Interestingly, and f

Analysis of Seasonal Risk for Importation of the Mediterranean Fruit Fly, Ceratitis capitata (Diptera: Tephritidae), via Air Passenger Traffic Arriving in Florida and California.

The Mediterranean fruit fly, Ceratitis capitata (Wiedemann), is one of the most economically damaging pests in the world and has repeatedly invaded two major agricultural states in the United States, Florida and California, each time requiring costly eradication. The Mediterranean fruit fly gains entry primarily in infested fruit carried by airline passengers and, since Florida and California each receive about 13 million international passengers annually, the risk of Mediterranean fruit fly entering the United States is potentially very high. The risk of passengers bringing the pest into Flor

Expression and function of the Drosophila melanogaster ADH in male Ceratitis capitata adults: a potential strategy for medfly genetic sexing based on gene-transfer technology.

The aim of development of a Mediterranean fruit fly Ceratitis capitata genetic sexing strain derives from the large scale SIT programmes being carried out to control this pest. Toward this direction, we present here the male-specific expression of the Drosophila melanogaster alcohol dehydrogenase (ADH) in medfly transgenic adults generated by Minos-mediated germ line transformation. This expression pattern is obtained by using a promoter fragment of the male-specific gene MSSP-alpha2 of the medfly. We show that the heterologous enzyme is functional in the medfly oxidizing both ethanol and 2-pr
