Exposure to Metals Can Be Therapeutic.

Because of the widespread epigenetic changes ensuing from carcinogenesis, structural and chemical features of chromatin provide unique targets for developing safer and more effective anticancer drugs. Metal-based agents have a potential advantage over other small molecular species in that characteristics of coordination geometry, redox state and ligand exchange allow one to fine-tune reactivity and affinity properties in a distinct fashion. This intersection of chromatin biology and bioinorganic medicinal chemistry is the subject of multiple collaborations in and between Switzerland and Singap

Structural characterization of the water-soluble porphyrin complexes [FeII(TPPS) (NO•)]4─ and [μ-O-([FeIII(TPPS)])2]8─.

Iron water-soluble porphyrins have been long used as biomimetic compounds for modelling the active sites found in heme-enzymes. In this regard, the anionic porphyrin [FeIII(TPPS)]3─ and its coordination complexes have been repeatedly chosen as suitable water-soluble platforms for bioinorganic chemistry studies. In this work we report for the first time the crystal structure of the water-soluble nitrosyl complex [FeII(TPPS) (NO•)]4─ along with that of oxodimeric ferric species [μ-O-([FeIII(TPPS)])2]8─.

Zn(II) binding to pramlintide results in a structural kink, fibril formation and antifungal activity.

The antimicrobial properties of amylin, a 37-amino acid peptide hormone, co-secreted with insulin from the pancreas, are far less known than its antidiabetic function. We provide insight into the bioinorganic chemistry of amylin analogues, showing that the coordination of zinc(II) enhances the antifungal properties of pramlintide, a non-fibrillating therapeutic analogue of amylin. Zinc binds to the N-terminal amino group and His18 imidazole, inducing a kink in the peptide structure, which, in turn, triggers a fibrillization process of the complex, resulting in an amyloid structure most likely

The design of inhibitors for medicinally relevant metalloproteins.

A number of metalloproteins are important medicinal targets for conditions ranging from pathogenic infections to cancer. Many but not all of these metalloproteins contain a zinc(II) ion in the protein active site. Small-molecule inhibitors of these metalloproteins are designed to bind directly to the active site metal ions. In this review several metalloproteins of interest are discussed, including matrix metalloproteinases (MMPs), histone deacetylases (HDACs), anthrax lethal factor (LF), and others. Different strategies that have been employed to design effective inhibitors against these prot

A dinuclear nickel peroxycarbonate complex: CO2 addition promotes H2O2 release.

Nickel coordination compounds featuring Ni-O bonds are key structural motifs in both bioinorganic and synthetic chemistries. They serve as precursors for organic substrate oxidation and are commonly invoked intermediates in water oxidation and oxygen reduction schemes. Herein, we disclose a series of well-defined dinuclear nickel complexes that, upon treatment with CO2 and H2O2, afford the first nickel-bound peroxycarbonate. This unprecedented nickel-oxygen intermediate is stabilized by hydrogen bonding templated across the bimetallic core. Contrasting copper and iron analogues, the nickel per

Bioinorganic interactions between PFAS and transition metals and their effects on toxicity and environmental persistence.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants widely detected in water, soil, biota, and human tissues. Although their environmental persistence and toxicity are commonly attributed to the exceptional stability of the carbon-fluorine bond, increasing evidence indicates that transition metals significantly influence PFAS environmental fate, transport, and biological effects. The rationale for examining PFAS in conjunction with transition metals arises from the widespread co-occurrence of these contaminants in natural and engineered systems, where metal-med

Beyond iron: non-classical biological functions of bacterial siderophores.

Bacteria secrete small molecules known as siderophores to acquire iron from their surroundings. For over 60 years, investigations into the bioinorganic chemistry of these molecules, including fundamental coordination chemistry studies, have provided insight into the crucial role that siderophores play in bacterial iron homeostasis. The importance of understanding the fundamental chemistry underlying bacterial life has been highlighted evermore in recent years because of the emergence of antibiotic-resistant bacteria and the need to prevent the global rise of these superbugs. Increasing reports

Reactions of HNO with heme proteins: new routes to HNO-heme complexes and insight into physiological effects.

The formation and interconversion of nitrogen oxides has been of interest in numerous contexts for decades. Early studies focused on gas-phase reactions, particularly with regard to industrial and atmospheric environments, and on nitrogen fixation. Additionally, investigation of the coordination chemistry of nitric oxide (NO) with hemoglobin dates back nearly a century. With the discovery in the early 1980s that NO is biosynthesized as a molecular signaling agent, the literature has been focused on the biological effects of nitrogen oxides, but the original concerns remain relevant. For instan

The Chemistry of Lanthanides in Biology: Recent Discoveries, Emerging Principles, and Technological Applications.

The essential biological role of rare earth elements lay hidden until the discovery in 2011 that lanthanides are specifically incorporated into a bacterial methanol dehydrogenase. Only recently has this observation gone from a curiosity to a major research area, with the appreciation for the widespread nature of lanthanide-utilizing organisms in the environment and the discovery of other lanthanide-binding proteins and systems for selective uptake. While seemingly exotic at first glance, biological utilization of lanthanides is very logical from a chemical perspective. The early lanthanides (L

Topology- and sequence-controlled β-amyloid recognition by a KLVFF/transferrin-functionalized csq porphyrinic zirconium MOF (PCN-222).

Recognition of amyloid-β (Aβ) by structurally defined inorganic scaffolds offers a route to dissecting how framework geometry and surface chemistry govern peptide-material interactions. We report transferrin (Tf)/KLVFF-PCN-222, a bioinorganic recognition platform built by covalently co-grafting the Aβ-homologous pentapeptide KLVFF and transferrin (Tf) onto the csq-topology porphyrinic zirconium MOF PCN-222 via EDC/NHS chemistry. Mol-ecular structure of tris-[(6-bromo-pyridin-2-yl)meth-yl]amine.

Coordination compounds of polydentate nitro-gen ligands with metals are used extensively in research areas such as catalysis, and as models of complex active sites of enzymes in bioinorganic chemistry. Tris(2-pyridyl-meth-yl)amine (TPA) is a tripodal tetra-dentate ligand that is known to form coordination compounds with metals, including copper, iron and zinc. The related compound, tris-[(6-bromo-pyridin-2-yl)meth-yl]amine (TPABr3), C18H15Br3N4, which possesses a bromine atom on the 6-position of each of the three pyridyl moieties, is also known but has not been heavily investigated. Reactions of the Zn Proteome with Cd2+ and Other Xenobiotics: Trafficking and Toxicity.

Understanding the molecular basis of inorganic chemical toxicity has lagged behind the proliferation of detailed mechanisms that explain the biochemical toxicology of many organic xenobiotics. In this perspective, general barriers to explicating the bioinorganic chemistry of toxic metals are considered, followed by a detailed examination of these issues in relation to the toxicology of Cd2+. The hypothesis is evaluated that Cd2+ damages cells by replacing Zn2+ in key Zn proteins. An emerging methodology to assess the speciation of metals among cell proteins is described. Harnessing natures ability to control metal ion coordination geometry using de novo designed peptides.

Advances in protein chemistry and molecular and structural biology have empowered modern chemists to build complex biological architectures using a "first principles" approach, which is known as de novo protein design. In this Perspective we demonstrate how simple three-stranded alpha-helical constructs can be prepared by the sole consideration of the primary amino acid sequence of a peptide. With these well defined systems, we then demonstrate that metal binding cavities can be carved out of the hydrophobic cores of these aggregates in order to bind metal ions such as cadmium with well define
