Chemokine Profiles Are Affected in Serum of Patients with Acute Rejection of Kidney Allograft.

Kidney allograft transplantation improved the prognosis and quality of life of patients with end-stage renal diseases but the occurrence of acute rejection represents a limitation of the final outcome. Noninvasive biomarkers are needed as well as further advancements in the understanding of immune mechanisms of reaction to the allograft. Our study of 138 patients focused on one-year monitoring of serum concentrations of 12 chemokines regulating the recruitment of different immune cells into transplanted allograft and on in vitro regulation of the same chemokines release by interactions of rena

Expression profile of proinflammatory mediators in the placenta of mares during physiological detachment and retention of fetal membranes.

Physiological parturition is characterized by sterile, inflammatory-like processes. During parturition, the placenta expresses various proinflammatory mediators, such as chemokines and IL-17. Nevertheless, inflammatory processes present in the parturient mare are poorly characterized. The aim of this study was to investigate the expression of selected chemokines and IL-17 in the allantochorion and the endometrium of mares that retained fetal membranes (RFM) and expelled them physiologically. We hypothesized that the expression of these mediators may be altered in the placenta of mares with RFM

Longitudinal Profiles of Cytokines and Chemokines in Self-Limiting Hepatitis E.

Hepatitis E virus infection typically results in a self-limited acute viral hepatitis (AVH-E), which is rapidly cleared by the host immune response. In this longitudinal study, temporal cytokine and chemokine profiles were analysed in AVH-E patients' sera using a multiplex immunoassay. HEV RNA became undetectable between 9 and 18 days, with a median of 13 days, occurring 3-20 days after symptom onset. In the AVH-E group, IFN-γ peaked significantly around days 6-9, which is prior to the HEV RNA clearance period, and declined during days 9-18. IL-2, IL-10, and TNF-α increased significantly durin

Spatial characterization of interface dermatitis in cutaneous lupus reveals novel chemokine ligand-receptor pairs that drive disease.

BACKGROUND: Chemokines play critical roles in the recruitment and activation of immune cells in both homeostatic and pathologic conditions. Here, we examined chemokine ligand-receptor pairs to better understand the immunopathogenesis of cutaneous lupus erythematosus (CLE), a complex autoimmune connective tissue disorder. OBJECTIVES: Our objectives were to (1) characterize the cellular and proteomic constitution of interface dermatitis in CLE using blister biopsies, (2) map chemokine:ligand receptor pairs that govern recruitment of immune cells to form interface dermatitis in CLE, and (3) perf

Shared immune-inflammatory mechanisms between ulcerative colitis and periodontitis: a multi-omics analysis.

BACKGROUND: Ulcerative colitis (UC) and periodontitis (PD) are chronic inflammatory diseases with increasing evidence of bidirectional communication through the oral-gut axis. However, the immunological mechanisms underlying their co-occurrence remain largely unclear. METHODS: We conducted a bidirectional Mendelian randomization (MR) analysis to evaluate potential causal relationships between UC and PD. Transcriptomic data from public repositories were integrated to identify shared differentially expressed genes. Immune-related genes were further screened using three machine learning approach

Cancer-Associated Fibroblast-Mediated Cellular Crosstalk Supports Hepatocellular Carcinoma Progression.

BACKGROUND AND AIMS: Cancer-associated fibroblasts (CAFs) are key players in multicellular, stromal-dependent alterations leading to HCC pathogenesis. However, the intricate crosstalk between CAFs and other components in the tumor microenvironment (TME) remains unclear. This study aimed to investigate the cellular crosstalk among CAFs, tumor cells, and tumor-associated neutrophils (TANs) during different stages of HCC pathogenesis. APPROACH AND RESULTS: In the HCC-TME, CAF-derived cardiotrophin-like cytokine factor 1 (CLCF1) increased chemokine (C-X-C motif) ligand 6 (CXCL6) and TGF-β secreti

The CXCL8/IL-8 chemokine family and its receptors in inflammatory diseases.

Chemokines are small proteins that control several tissue functions, including cell recruitment and activation under homeostatic and inflammatory conditions. CXCL8 (interleukin-8) is a member of the chemokine family that acts on CXCR1 and CXCR2 receptors. CXCL1, CXCL2, CXCL3, CXCL5, CXCL6, and CXCL7 are also ELR+ chemokine members that bind to these receptors, especially CXCR2. The majority of studies on the biology of CXCL8 and their receptors have been performed in polymorphonuclear leukocytes. However, many other cells express CXCR1/CXCR2, including epithelial, endothelial, fibroblasts and

Targeting m6A Reader YTHDF1 Enhances Antitumor Immunity and Potentiates Anti-PD-L1 Efficacy in Intrahepatic Cholangiocarcinoma.

The N6-methyladenosine (m6A) reader YTH N6-methyladenosine RNA binding protein 1 (YTHDF1) plays a critical role in the tumorigenesis of intrahepatic cholangiocarcinoma (ICC), but its function in the tumor immune microenvironment remains unclear. RNA sequencing analysis of human ICC samples revealed that, among m6A-related regulators, YTHDF1 exhibited the most significant negative correlation with immune score. In multiple ICC mouse models, Ythdf1 overexpression enhanced the recruitment of myeloid-derived suppressor cells (MDSCs) and suppressed cytotoxic CD8+ T cell responses, promoting ICC pro

CXCL Gene Clusters Regulated by Enhancer-Mediated DNA Looping Alteration in Pancreatic Cancer Cells.

Pancreatic cancer is one of the deadliest cancers. Chemokines affect the progression of pancreatic cancer through various mechanisms. Most of the CXC chemokine genes, CC chemokine genes and CX3C chemokine genes are clustered together within a very short region of chromatin. Transcription activity of gene clusters is usually influenced by the chromatin architecture and spatial organisation. Nevertheless, the chromatin-mediated regulatory mechanism on transcription of chemokine gene clusters has never been studied in pancreatic cancer. Herein, we determined that the expression of C-X-C motif che

Tonsil-derived mesenchymal stromal cells produce CXCR2-binding chemokines and acquire follicular dendritic cell-like phenotypes under TLR3 stimulation.

We previously isolated mesenchymal stromal cells from human tonsils (T-MSCs) and showed the potential of these cells to differentiate into the mesodermal lineage and acquire a follicular dendritic cell (FDC) phenotype under cytokine stimulation. Because these T-MSCs were originally isolated from inflamed tonsillar tissues, we were curious about their activation status in response to innate immune stimuli, such as Toll-like receptors (TLRs). Therefore, we analyzed the expression profile of TLRs in T-MSCs and stimulated the T-MSCs with TLR agonists. TLR3 stimuli induced C-C chemokine receptor ty

CXCL6 exacerbates metabolic dysfunction-associated steatohepatitis by suppressing LPIN1-mediated fatty acid oxidation in hepatocytes.

Progression of hepatic steatosis to metabolic dysfunction-associated steatohepatitis (MASH) is driven by impaired fatty acid (FA) oxidation and subsequent hepatocyte lipotoxicity. While MASH is characterized by upregulation of the neutrophil chemoattractant CXCL6, which functions through CXCR2, the direct impact of this pathway on hepatocyte FA metabolism during MASH progression remains unclear. Here, we demonstrate that hepatic overexpression of Cxcl5 (the murine homolog of CXCL6) inhibits hepatocyte FA oxidation and promotes MASH progression in mice. In contrast, Cxcl5 deficiency conferred p

Macrophages promote cyst growth in polycystic kidney disease.

Polycystic kidney disease (PKD) exhibits an inflammatory component, but the contribution of inflammation to cyst progression is unknown. Macrophages promote the proliferation of tubular cells following ischemic injury, suggesting that they may have a role in cystogenesis. Furthermore, cultured Pkd1-deficient cells express the macrophage chemoattractants Mcp1 and Cxcl16 and stimulate macrophage migration. Here, in orthologous models of both PKD1 and PKD2, abnormally large numbers of alternatively activated macrophages surrounded the cysts. To determine whether pericystic macrophages contribute

CXCL16/SR-PSOX--a friend or a foe in atherosclerosis?

Chemokines, scavenger receptors and adhesion molecules have long been known as important players in the pathogenesis of atherosclerosis. A series of studies conducted in the past few years described CXCL16/SR-PSOX--a new molecule combining those three functions, and suggested that CXCL16/SR-PSOX can be a potential player in atherogenesis. Initial ex vivo studies showed that CXCL16/SR-PSOX is abundant in human and murine atherosclerotic lesions. Following in vitro studies suggested that as an adhesion molecule CXCL16/SR-PSOX might mediate T-cell adhesion to the endothelium, as a chemokine - dri
