Effects of dopamine on the conductance of perfused vascular beds of the chloralosed cat.

1. Dopamine produced vasoconstriction followed by secondary vasodilatation in the perfused cat hindquarters and splanchnic region. Low doses produced only vasodilatation in the splanchnic region.2. Phenoxybenzamine abolished, while despiramine and cocaine potentiated, the vasoconstrictor and secondary dilator actions of dopamine.3. After phenoxybenzamine, dopamine produced only vasodilatation in both the hindquarters and splanchnic region, being more effective in the latter.4. Haloperidol, but not propranolol, atropine or mepyramine, antagonized dopamine-induced vasodilatation.5. Dopamine diff

Hypothalamic stimulation counteracts sympathetically mediated gastrointestinal inhibition in chloralose-anaesthetised cats.

The inhibitory motor effect of the sympathetic gastrointestino-gastrointestinal (GI-GI) reflex was studied in cats with isolated innervated ileal (Thiry-Vella) loops. They were subjected to acute experiments under chloralose-gallamine anaesthesia where gastric volume, arterial blood pressure and heart rate were continuously monitored. The cervical vagi were sectioned and used for efferent stimulation. The hypothalamus was systematically explored by means of stereotaxically guided electrodes. When the abdomen was undisturbed, gastric contractile responses to central stimulation were virtually a

The effects of some phosphodiesterase inhibitors on the conductance of the perfused vascular beds of the chloralosed cat.

1. The vasoactivity of a number of phosphodiesterase inhibitors in the auto-perfused splanchnic region and hindquarters of the cat have been investigated.2. Theophylline, dipyridamole and 4-(3,4-dimethoxybenzyl)-2-imidazolidinone (Ro 7-2956) and its analogues were more potent vasodilators in the splanchnic region than in the hindquarters, whereas papaverine was a potent vasodilator in both these vascular beds.3. The vasodilator response to the phosphodiesterase inhibitors in the splanchnic region and hindquarters was not susceptible to beta-adrenoceptor blockade.4. Infusion of dibutyryl cyclic

Effect of captopril on blood pressure responses to enkephalins in chloralose-anaesthetized rats.

Captopril lowered blood pressure in chloralose-anaesthetized rats and enhanced the depressor responses to metenkephalin and D-Ala-metenkephalinamide to the same extent. Since metenkephalin is a much better substrate than D-Ala-metenkephalinamide for dipeptidylcarboxypeptidase, some mechanism other than inhibition of this enzyme appears to be responsible for the enhancement by captopril of the responses to opioids. When the blood pressure had been lowered by haemorrhage, naloxone was no more effective than saline control in restoring blood pressure. However, when captopril was given after haemo

Development and Validation of a Quantitative UHPLC-MS-MS Method for the Determination of Alpha-Chloralose in Feline Blood and Application on Blood Samples Collected from Cats with Symptoms of Alpha-Chloralose Poisoning.

Alpha-chloralose (AC) is used as a rodenticide as well as an anesthetic agent in laboratory animals. It was previously also used as an avicide. Detection of AC in blood samples or in body tissues collected postmortem is key for the diagnosis of clinical cases and a requirement for surveillance of secondary toxicosis, including potential cases in wild animals. Reports on poisoning of humans and non-laboratory animals confirmed by the detection of AC or its metabolites are available, however poisoning of domestic animals are rarely available. Furthermore, reports on clinical cases in domestic an

Intraventricular infusion of angiotensin II on the hemodynamics and renal function of alpha-chloralose anesthetized cats.

Intraventricular infusion of angiotensin II (50, 100, and 200 ng/kg per min) produced significant elevations of arterial blood pressure (20-25%) in alpha-chloralose-anesthetized cats. The pressor responses were not accompanied by significant changes in heart rate, cardiac output, or contractility and were chiefly due to significant increases in total peripheral resistance. In contrast, pressor responses to intravenous infusion of angiotensin II (100 ng/kg per min) were accompanied by reflex decrease in cardiac activity. While intravenous angiotensin II caused increases in the resistance of skel

Regional blood flow in canine brain during nicotine infusion: pentobarbital vs. chloralose anesthesia.

We compared vasoactive effects of intravenous nicotine (36 micrograms/kg/min) in regional cerebral circulations under pentobarbital and chloralose anesthesia. Experiments were conducted in three groups of dogs: Group I, pentobarbital anesthesia with fixed ventilation; Group II, chloralose anesthesia with fixed ventilation; Group III, chloralose anesthesia with free breathing. Values for regional cerebral blood flow measured with 15 mu radioactive microspheres were used to compute regional cerebral vascular resistance (rCBR). In Group I, nicotine had no effect on rCVR in cerebral cortex, and it

Effects of sodium pentobarbital, ketamine, halothane, and chloralose on brainstem auditory evoked responses.

A parametric study was performed in cats comparing the effects on the brainstem auditory evoked response of four anesthetic agents used either in the operating room or the laboratory: sodium pentobarbital, ketamine, halothane, and chloralose. Each anesthetic agent was administered to each of four cats in varying sequence. Click-evoked brainstem auditory evoked responses (BAER) were compared for differences in wave form configuration, latencies, and amplitudes of the component peaks. Five distinct peaks (I to V) could be identified in the brainstem auditory evoked responses of every animal usin

Determination of Anticoagulant Rodenticides and α-Chloralose in Human Hair. Application to a Real Case.

Anticoagulant rodenticides are the largest group of poisons used to kill harmful rodents. Their fundamental mode of action consists in the inhibition of the vitamin K epoxide reductase, which causes blood-clotting alteration, ultimately leading to hemorrhagic events as the cause of death. In this study, we developed an UHPLC-MS-MS for the simultaneous determination of 10 anticoagulant hydroxycoumarine rodenticides, plus α-chloralose in human hair, with the scope of detecting potential trace of chronological poison exposure in clinical and forensic cases. The method was fully validated and appl

Comparison of urethane/chloralose and pentobarbitone anaesthesia for examining effects of bacterial lipopolysaccharide in mice.

Although anaesthetics are widely used to alleviate stress in endotoxaemic animals, these drugs themselves may interfere with the effects of lipopolysaccharide (LPS). The effects of LPS on serum glucose, biochemical markers of hepatic, renal and pancreatic exocrine function, and lung myeloperoxidase (MPO) activity were compared using anaesthesia with either urethane/chloralose or pentobarbitone. Groups of 10-13 of C57B1/6 mice (22.3 +/- 0.18 g) were treated with 40 mg/kg LPS or the same volume of saline (10 mL/kg, i.p.) at time 0, Animals were anaesthetized either with urethane (1000 mg/kg) and

Effects of pentazocine on coronary hemodynamics under pentobarbital or chloralose-urethane anesthesia in dogs.

The effect of pentazocine on the coronary hemodynamics was studied in anesthetized dogs. Under pentobarbital anesthesia pentazocine did not cause any significant changes in the coronary hemodynamics. Under chloralose-urethane anesthesia, however, it produced a statistically significant increase in heart rate, left coronary blood flow rate, myocardial oxygen consumption as well as myocardial oxygen extraction rate, but a decrease in coronary vascular resistance. These results indicate that the effects of pentazocine on the coronary hemodynamics are modified significantly by the drugs used for b

Alpha-chloralose poisoning in 25 cats: clinical picture and evaluation of treatment with intravenous lipid emulsion.

OBJECTIVES: The aims of this study were to describe the clinical picture and progression in cats with alpha-chloralose (AC) intoxication and to determine if treatment with intravenous (IV) lipid emulsion (ILE) influenced either the serum concentration of AC or the clinical signs. METHODS: Cats with suspected AC poisoning admitted to a university small animal hospital were included. The cats were randomised into two groups: one receiving 20% ILE at a dose of 300 mg/kg as a 2 min bolus, followed by a 1500 mg/kg continuous rate infusion over 30 mins (IL+ group) and the other receiving IV fluid t

Different effects of α-chloralose on spontaneous and evoked GABA release in rat hippocampal CA1 neurons.

The effects of α-chloralose on presynaptic GABA(A) receptors were investigated with respect to spontaneous and evoked GABAergic transmission (sIPSCs and eIPSCs) in rat hippocampal CA1 pyramidal neurons. sIPSCs were recorded in mechanically dissociated CA1 neurons with intact GABAergic terminals, namely the "synaptic bouton preparation." eIPSCs were elicited by focal electrical stimuli of a single GABAergic bouton on an isolated CA1 neuron using the whole-cell patch recording configurations under voltage-clamp condition. We found that α-chloralose potentiated the exogenous GABA-induced Cl(-) re

Effect of beta-adrenergic receptor antagonism on chloralose-induced hemodynamic changes in newborn lambs.

alpha-Chloralose is an anesthetic commonly used in cardiovascular research. Using a chronically instrumented neonatal lamb model, we previously determined that chloralose has important effects on basal hemodynamics and arterial oxygen tension as compared with those of paired conscious control lambs. We wished to determine whether beta-adrenergic receptor stimulation accounted for chloralose-induced hemodynamic effects and to investigate the influence of chloralose and beta-adrenergic receptor antagonism on oxygen metabolism. In paired studies, five lambs were given chloralose intravenously (30

Ventricular fibrillation following canine coronary artery reperfusion: different outcomes with pentobarbital and alpha-chloralose.

We compared the incidence of ventricular fibrillation after release of a 20 min-proximal left anterior descending coronary artery ligation in dogs anesthetized with either pentobarbital (N = 26) or morphine sulfate plus alpha-chloralose (N = 26). Results were analyzed using the logistic risk regression model. In each group, outcome correlated with the amount of myocardium perfused by the ligated vessel (myocardium "at risk") as measured by postmortem coronary injection of monastral dyes. At any given myocardium at risk, animals anesthetized with alpha-chloralose were less likely to fibrillate
