Chlordiazepoxide antinociception: cross-tolerance with opiates and with stress.

Chlordiazepoxide (CDP) has been previously shown to possess antinociceptive properties that are resistant, except at high doses, to the opiate antagonist naloxone. The present study evaluated whether CDP's antinociceptive effects were subject to tolerance following repeated injections and whether cross-tolerance might develop between the antinociceptive action of CDP and that of either morphine or cold water swins. CDP increased flinch-jump thresholds following acute administration and exhibited tolerance following repeated injections. Neither morphine-tolerant nor cold water swim-adapted rats

Treatment of alprazolam withdrawal with chlordiazepoxide substitution and taper.

We describe the first case series (n = 6) of using chlordiazepoxide to accomplish a rapid, well-tolerated withdrawal from alprazolam. After abruptly discontinuing alprazolam, we substituted a 50-mg dose of chlordiazepoxide for each 1 mg of alprazolam (except for one elderly patient where we substituted 25 mg) and gave additional chlordiazepoxide doses (25-50 mg every 4-6 hours) as needed for the first 1-2 days of hospitalization. With this approach, the mean "substitution ratio" of chlordiazepoxide to alprazolam was 86 to 1. We then tapered chlordiazepoxide by an average of 10% each day over a

Anticonflict action of chlordiazepoxide in rats with combined lesions.

Lesions of brain areas thought to promote anxiety do not diminish the anticonflict effects of benzodiazepines (BZDs). After initial training in the lick-suppression conflict test, eight rats received electrolytic lesions of the amygdala, dorsal raphe, locus coeruleus, and mammillary bodies. Ten others received sham lesions. Postoperative testing revealed a significant increase in punished licking at two stages after surgery in lesioned animals when compared with their own preoperative baseline levels and with the punished licking of control animals. Systemic administration of chlordiazepoxide

Maternal aggression in rats: effects of chlordiazepoxide and fluprazine.

Although maternal aggression in rats is confined to a restricted post-partum period, the high and stable aggression level and the constancy of its behavioural structure make it an attractive experimental procedure for studying the behavioural effects of psychotropic drugs. Female rats were tested against naive male intruder rats for 5 or 10 min on post-partum days 3-9, during which aggression is stable. Chlordiazepoxide (CDP; 5, 10 and 20 mg/kg, orally) had a biphasic effect on aggression; it increased aggression considerably at 5 and (to a lesser extent) at 10 mg/kg. At 20 mg/kg aggression re

State-dependent behavioural sensitization: evidence from a chlordiazepoxide state.

The present study investigated the hypothesis that behavioural sensitization to psychomotor stimulants is expressed only if the internal state of the animal is the same as it was at the time of sensitization development. This state-dependency hypothesis has previously been put forward to explain the apparent blockade of sensitization by N-methyl-D-aspartate (NMDA) receptor antagonists. The present study used amphetamine (0.375 mg/kg) as the stimulant and chlordiazepoxide (CDP) as a secondary stimulus. Mice were given double injections of either amphetamine + CDP, amphetamine + saline, CDP + sa

Chronic bretazenil produces tolerance to chlordiazepoxide, midazolam, and abecarnil.

The purpose of the present study was to determine if chronic treatment with a nonsedative benzodiazepine partial agonist would confer tolerance to the rate-decreasing effects of other benzodiazepine ligands in a fixed-interval procedure in rats. A separate group of rats was treated chronically with the sedative benzodiazepine full agonist, chlordiazepoxide, for comparison. It was hypothesized that tolerance would develop rapidly to chlordiazepoxide due to loss of reinforcement density at rate-decreasing doses and that there would probably be cross-tolerance to other rate-decreasing benzodiazep

Chlordiazepoxide, an anxiolytic benzodiazepine, impairs place navigation in rats.

There are separate proposals that the hippocampus is involved in 'spatial memory' and in the control of 'anxiety'. Despite a larger number of common effects of anxiolytic drugs and hippocampal lesions, no effect of anxiolytic drugs has yet been reported in those spatial tasks which are particularly sensitive to the effects of hippocampal lesions. The present study addresses this issue. Separate groups of rats were treated, i.p., with 5 mg/kg chlordiazepoxide (an anxiolytic benzodiazepine), 1 mg/kg scopolamine (a muscarinic antagonist which has previously been shown to impair spatial learning)

Chlordiazepoxide facilitates erections and inhibits seminal emission in rats.

Two experiments have been conducted to clarify the effects of the benzodiazepine chlordiazepoxide (CDP) on the different components of male rat sexual behavior. In the first experiment the effects on penile erectile reflexes (PR) of two doses (10 and 30 mg/kg, IP) of CDP were compared with those of vehicle, no-treatment and baseline pre-test in a repeated measures design. In the second experiment the different components of male sexual behavior - PR, mating behavior and spontaneous seminal emission SSE) - were extensively studied after vehicle, 3 and 30 mg/kg CDP treatments. CDP was found to i

Crossmodal divided attention in rats: effects of chlordiazepoxide and scopolamine.

"Divided attention" is a psychological construct that hinges on assumptions about a fixed finite capacity of subjects to simultaneously process multiple sets of information. A model of a crossmodal divided attention task was developed in rats. Initially, rats were trained consecutively in operant auditory and visual conditional discrimination tasks. The final task consisted of two successive blocks of 20 trials per modality (modality certainty), followed by 60 trials comprising a semi-randomized sequence of stimuli of both modalities (auditory or visual) and qualities (flashing/pulsing or cons

Tolerance to the behavioral effects of chlordiazepoxide: pharmacological and biochemical selectivity.

There is a dynamic interaction between a drug's pharmacological effects and the behavioral context in which it is administered. The present study evaluated the influence of behavioral processes on the development of tolerance and cross-tolerance to the rate-decreasing effects of chlordiazepoxide in rats. Sprague-Dawley rats responded under a fixed-ratio 30 schedule of food delivery. Different groups of rats received 18 mg/kg/day of chlordiazepoxide either before (PRE, n = 8) or after (POST, n = 10) daily experimental sessions for 8 weeks. Cumulative dose-response curves for chlordiazepoxide we

Tolerance to and cross-tolerance among ethanol, pentobarbital and chlordiazepoxide.

The acute administration of ethanol, pentobarbital and chlordiazepoxide impaired, in a dose-dependent manner, the performance of rats on the moving-belt and two-way shuttle-box avoidance tests. Administration of these drugs for three weeks resulted in tolerance to their motor-impairing effects. Tolerance to ethanol or pentobarbital was characterized by a parallel shift of the dose-response curve to the right. Tolerance to chlordiazepoxide, however, was of greater extent and was accompanied by an apparent flattening of the dose-response curve. Symmetrical cross-tolerance developed between ethan

Specific antagonism of the behavioral effects of chlordiazepoxide and pentobarbital in the rat.

Stimulus control was established in rats with 5 mg/kg chlordiazepoxide and saline by employing a two-lever response choice task and an FR10 schedule of food reinforcement. Once trained, administration of chlordiazepoxide doses different from the training dose produced dose-responsive discrimination responding and the chlordiazepoxide discriminative cue was observed to transfer to pentobarbital. Pretreatment with the convulsant bemegride antagonized the pentobarbital transfer but not the chlordiazepoxide cue, whereas the specific benzodiazepine antagonist RO15-1788 decreased the chlordiazepoxid

Effects of chlordiazepoxide on comparable rates of punished and unpunished responding.

Identical overall rates and patterns of key pecking by pigeons were maintained under a multiple fixed-interval schedule of food presentation. In one component, every thirtieth response produced an electric shock (punishment) whereas during the other component the response that produced food had to be preceded by a pause of a minimum (10 or 11 s) duration. Although chlordiazepoxide (1.0--17.0 mg/kg) increased both punished and unpunished responding, greater increases were uniformly obtained with punished responding.

Effects of chlordiazepoxide on tail pinch-induced eating in rats.

The effects of 5, 10, and 20 mg/kg chlordiazepoxide on tail pinch (TP)-induced behavior were investigated. Five mg/kg enhanced TP-induced eating in terms of both latency and duration. Twenty mg/kg had decremental effects. All doses of the drug reduced the incidence of clip-directed behavior, but increased locomotor activity during the TP trials in a dose-dependent manner. On control trials, the drug increased locomotor activity at the low dose and eating at the high dose. The results are examined in terms of the various behavioral actions of the minor tranquilizers. The implications for the be

Solvent-induced photophysical properties and stability of clonazepam and Chlordiazepoxide.

Understanding how solvent environments influence the electronic behavior and molecular stability of nitrogen-based heterocyclic drugs is essential for optimizing their pharmaceutical performance in both ground and excited states. Such knowledge is vital for predicting drug stability, guiding formulation design, and determining suitable storage conditions. In this study, Clonazepam and Chlordiazepoxide-representative nitrogen-based heterocyclic drugs-were systematically investigated in fourteen solvents of varying polarity. Clonazepam exhibited a more pronounced intramolecular charge transfer (

Effects of chlordiazepoxide and putative anxiogenics on conditioned suppression in rats.

This paper reports two experiments. In Experiment 1, the effects of chlordiazepoxide alone and in combination with a series of putative antagonists at various sites on the GABA/benzodiazepine receptor complex on conditioned suppression of operant behavior in rats were assessed. Response rates during presentation of a stimulus associated with shock (CS responding) and when only positive reinforcement is effective (pre-CS responding) were analysed. Chlordiazepoxide (10 mg/kg) significantly increased CS responding. This effect was significantly antagonised by Ro15-1788 (10 mg/kg) and by picrotoxi
