Equilibrium binding experiments have been performed with perchlorate, chloride, and acetate in the presence of horseradish peroxidase. The binding of perchlorate and acetate appears to be like that of nitrate, at a site other than the sixth coordination position of the heme iron. Competitive experiments using both nitrate and cyanide demonstrate that two different binding sites are present on the enzyme. Chloride appears to bind at the sixth coordination position as do both fluoride and cyanide. Temperature jump experiments indicate that it is likely the nitrate anion and not undissociated nit

Association of the physiological substrate cyclopentanedione (CPDO) with chloroperoxidase (CPO) was confirmed from both line-width and longitudinal relaxation time measurements of the proton NMR signals of CPDO. A dissociation constant of 33 mM for the enzyme-substrate complex was calculated from proton NMR relaxation experiments. Chloride ion does not affect the stability of the complex, but iodide ion greatly decreased the stability of the CPO-CPDO complex. Binding of cyanide ion to the heme iron center of CPO also decreased the stability of the CPO-CPDO complex. This result indicates the cr

BACKGROUND: Wide distribution of polyvinyl chloride microplastics (PVC-MPs) has been documented in remote regions such as the Qinghai-Tibet Plateau (QTP). Microbial degradation of plastics is frequently coupled with lignocellulose-degrading enzymatic machinery. As a ubiquitous biological sampler on the QTP, the herbivore plateau pika (Ochotona curzoniae), which harbors diverse lignocellulose-degrading enzymes, represents a promising reservoir for novel PVC-degrading enzymes.
RESULTS: In this study, a PVC-MPs feeding trial of plateau pikas revealed gut microbiota recruitment of plastic degrade

Degradation of methyl mercury (MeHg) and ethyl Hg (EtHg) with reactive oxygens was studied in vitro by using peroxidase-hydrogen peroxide (H2O2)-halide and rose bengal-ultraviolet light A systems. For this purpose, the direct determination method for inorganic Hg was employed. Both systems could effectively degrade EtHg, and MeHg to some extent. Degradation of MeHg and EtHg with the myeloperoxidase (MPO)-H2O2-chloride system was inhibited by MPO inhibitors (cyanide and azide), catalase, hypochlorous acid (HOCI) scavengers (glycine, alanine, serine and taurine), 1,4-diazabicyclo[2,2,2]octane an

Myeloperoxidase (MPO) is a heme peroxidase found in neutrophils, monocytes and macrophages that efficiently catalyzes the oxidation of endogenous chloride into hypochlorous acid for antimicrobial activity. Chronic MPO activation can lead to indiscriminate protein modification causing tissue damage, and has been associated with chronic inflammatory diseases, atherosclerosis, and acute cardiovascular events. Triazolopyrimidine 5 is a reversible MPO inhibitor; however it suffers from poor stability in acid, and is an irreversible inhibitor of the DNA repair protein methyl guanine methyl transfera

Vitamin A deficiency causes a significant decrease in superoxide dismutase, glutathione peroxidase and GSH levels. Simultaneously, it causes a marked increase in Phase I microsomal oxidation (cytochrome P-450, aniline hydroxylase, PNA-O-demethylase) as well as in enzymatic and non-enzymatic lipid peroxidation in lung. Among Phase II enzymes, cytosolic chlorodinitrobenzene glutathione-S-transferase (CDNB-GST) activity showed a significant increase whereas microsomal UDP-glucuronyl transferase and cytosolic p-nitrobenzoyl chloride- and dichloro nitrobenzyl chloride-S-transferase activities showe

Peroxidase in mouse thyroid was inhibited by mercuric chloride but not by methylmercury in in vivo and in vitro systems (Nishida, et al., J. Histochem. Cytochem., 37, 723 (1989)). To identify the reason for the difference, the present study was conducted to examine whether methylmercury is indeed bound within cells or tissues. Mice were given radioactive methylmercury by intubation for 18 d and the tissues were dissected out and vacuum-dried. With this procedure, free methylmercury was evaporated off and the bound mercury remained. The thyroid, liver, kidney and fats examined showed no loss of

The reaction between myeloperoxidase (donor:hydrogen-peroxide oxidoreductase, EC 1.11.1.7), hydrogen peroxide and ethyl hydroperoxide was investigated using the stopped-flow technique. Like other peroxidases, myeloperoxidase forms two sequential peroxide compounds. The pH-dependence of the apparent second-order rate constant of compound I formation shows that there is an acid/base group on the enzyme with a pKa of 4.30 +/- 0.15, which - when protonated - prevents the reaction of the enzyme with peroxides. The rate constants for the formation of compound I by hydrogen peroxide and ethyl hydrope

The aim of the study was the evaluation of cadmium effects on the activity of antioxidant enzymes in rat hepatocytes. The studies were conducted with isolated rat hepatocytes incubated for 1 or 2 hours in a modified (deprived of carbonates with phosphates) Williams' E medium (MWE) in the presence of cadmium chloride (25, 50 and 200 microM). Hepatocytes incubated in the MWE medium without cadmium chloride were used as a control. The application of the modified Williams' E medium allowed for the appearance of cadmium compounds in a soluble form that is indispensable for suitable estimation of it

The chlorination of glycine by the myeloperoxidase-H2O2-Cl- system at acidic pH values yielded N-monochloroglycine and a mixture of HCN and ClCN. HCN was formed as a product of N-dichloroglycine decomposition and cyanogen chloride formation resulted from simultaneous chlorination of HCN by N-chloroglycine or directly by the myeloperoxidase-H2O2-Cl- system. HCN was readily chlorinated by the myeloperoxidase-H2O2Cl- system yielding cyanogen chloride. This dissociation constants of the myeloperoxidase-CN- complex were estimated as 2.5.10(-6)--1.15.10(-5) M within the pH range of 6.2 to 3.4, respe

This study aimed to investigate the effects of zinc compounds on the enzymatic activities of lysozyme, peroxidase, and the glucose oxidase-mediated peroxidase (GO-PO) system and their antifungal activities. Four different zinc compounds (zinc chloride, gluconate, lactate, and sulfate) were incubated with hen egg-white lysozyme (HEWL), bovine lactoperoxidase (bLPO), the GO-PO system, and human unstimulated whole saliva in solution and on a hydroxyapatite surface. Enzymatic activities of lysozyme, peroxidase, and the GO-PO system were measured through the hydrolysis of Micrococcus lysodeikticus,

The effects of dietary zinc on the immune function of equines have not been evaluated in detail so far. In the present study, eight healthy adult ponies and two healthy adult horses were fed a diet supplemented with either zinc chloride hydroxide or zinc methionine in six feeding periods of four weeks each (according to maintenance zinc requirement, 120 mg zinc/kg dry matter, and 240 mg zinc/kg dry matter, for both dietary zinc supplements, respectively). All animals received the six diets, with increasing amounts of zinc chloride hydroxide in the feeding periods 1-3, and with increasing amoun

Halogenation reactions are essential for producing many fine and bulk chemicals. Vanadium-dependent chloroperoxidases (VCPOs) are promising biocatalysts for a more sustainable production of halogenated products because they are cofactor independent, use hydrogen peroxide (H2O2) as oxygen donor, tolerate high H2O2 concentrations, and catalyze all halogenations except fluorination. Despite their outstanding catalytic potential, few VCPOs have been identified yet, and detailed information on their catalytic characteristics is lacking. Here, we report the heterologous expression of three previousl

It is known that activation of neutrophils or monocytes leads to the formation of hydrogen peroxide and the release of myeloperoxidase (MPO). We found that sulfamethoxazole was chlorinated by the combination of MPO, hydrogen peroxide, and chloride. The product, N-chlorosulfamethoxazole, is reasonably stable but reacts rapidly with a variety of compounds. The same product was formed by the reaction between sulfamethoxazole and hypochlorous acid, and dapsone was also N-chlorinated by the MPO system or hypochlorous acid. Although N-chlorination was not observed when sulfamethoxazole or dapsone wa

Iron deficiency and cadmium (Cd) stress causes a rapid change in plant physiology. The aim of this work was to characterize the effects of Fe supplementation on redox reactions in leguminous plants under Cd toxicity. The experiments were performed in hydroponic nutrient media, using mungbean grown under 300 µM cadmium chloride with or without Fe for 10 days. The Fe concentration in plants grown under Fe deficiency was reduced by the presence of Cd; however, it recovered to control levels after Fe was supplied. Similarly, a very high Cd concentration was observed in plants grown under Fe defici

Exposure to heavy metals such as cadmium and mercury is of immediate environmental concern. The present study was aimed at establishing a direct relationship between heavy metal poisoning and thyroid dysfunction. Cadmium and mercury treatment at LD50 levels resulted in severe thyrotoxicosis in the rabbit. Within 24 h of intramuscular administration of cadmium chloride 15 mg.kg-1 body weight (bw) and mercury chloride 20 mg.kg-1 bw, thyroid peroxidase activity increased significantly over the control with a concomitant rise in the triiodothyronine (T3) titre. On the other hand, there was a remar

During anoxia/reoxygenation (A/R) injury, intracellular chloride ion concentration ([Cl(-)](i)) homeostasis may play a role in maintaining the normal physiological function of cardiomyocytes. Various chloride transport systems could have influenced the concentration of chloride ion, but what kinds of chloride transport systems could play an important role in cardiomyocytes subjected to A/R injury and its mechanism are unknown. The aim of our study was to clarify the contributions of various chloride transport systems to anoxia/reoxygenation in rat neonatal cardiac myocytes and further to inves

The catalytic activity of four lyophilized oxidative enzymes-horseradish peroxidase, soybean peroxidase, Caldariomyces fumago chloroperoxidase, and mushroom polyphenol oxidase-is much lower when directly suspended in organic solvents containing little water than when they are introduced into the same largely nonaqueous media by first dissolving them in water and then diluting with anhydrous solvents. The lower the water content of the medium, the greater this discrepancy becomes. The mechanism of this phenomenon was found to arise from reversible denaturation of the oxidases on lyophilization:

The release of proteolytic enzymes and generation of strong oxidants such as the hydroxyl radical by activated neutrophils has been proposed to play an important role in mediating toxin-induced liver injury. The antithyroid drug propylthiouracil protects against liver injury induced by many hepatotoxic agents and markedly reduces mortality in patients with alcoholic liver disease. However, the mechanism(s) by which propylthiouracil protects against liver injury is not well understood. The present studies investigate the effect of antithyroid drugs on proteolytic enzyme activity and on hydroxyl
