Arabidopsis Chloroplast protein for Growth and Fertility1 (CGF1) and CGF2 are essential for chloroplast development and female gametogenesis.

BACKGROUND: Chloroplasts are essential organelles of plant cells for not only being the energy factory but also making plant cells adaptable to different environmental stimuli. The nuclear genome encodes most of the chloroplast proteins, among which a large percentage of membrane proteins have yet to be functionally characterized. RESULTS: We report here functional characterization of two nuclear-encoded chloroplast proteins, Chloroplast protein for Growth and Fertility (CGF1) and CGF2. CGF1 and CGF2 are expressed in diverse tissues and developmental stages. Proteins they encode are associate

Chloroplast SecA functions as a membrane-associated component of the Sec-like protein translocase of pea chloroplasts.

Protein cross-linking studies with a thylakoid membrane translocation intermediate were used to demonstrate that chloroplast SecA functions as a membrane-associated component of the Sec-like ATP-dependent protein translocase of pea chloroplasts. In assays with isolated thylakoids, it was observed that translocation of the 33-kDa protein of the oxygen-evolving complex of photosystem II (OE33) decreased when the ATP concentration was low, and that the protein accumulated as a bound precursor. The bound precursor was able to be translocated into the lumen when the ATP concentration was raised, in

SUMOylation of chloroplast GrpE homolog 1 regulates chloroplast biogenesis through maintaining TIC complex stability.

In Arabidopsis, chloroplast protein import relies on the stromal cpHsp70 motor system, whose nucleotide-exchange factor CGE1 is indispensable, yet its regulatory mechanism remains poorly defined. We integrated bioinformatics prediction with in vitro and in vivo SUMOylation assays, biochemical analyses, confocal microscopy, and genetic complementation to elucidate the functional impact of CGE1 SUMOylation in chloroplast development. CGE1 undergoes SUMO3 conjugation, and Lys-214 is required for efficient CGE1 SUMOylation. The SUMOylation-defective variant CGE1K214R retained stromal localization

CGF1 and CGF2 jointly participate in iron homeostasis in chloroplasts.

Iron (Fe) plays a crucial role in chloroplast function, and its deficiency results in Fe chlorosis, a syndrome characterized by reduced biomass production and plant mortality. In this study, Fe supplementation rescued the dwarfism and chlorotic phenotype of cgf1 cgf2 mutant, suggesting that CGF1 and CGF2 are essential regulators of Fe homeostasis. Subcellular localization analysis revealed that both CGF1 and CGF2 localize to the chloroplast envelope. Their absence led to a decreased photosynthetic protein abundance and reduced photosynthetic supercomplex levels, demonstrating that CGF1 and CGF

Production of a recombinant bacterial lipoprotein in higher plant chloroplasts.

Little is known about the potential of plastids to accomplish post-translational modifications of foreign proteins. In the present study we generated transplastomic tobacco plants that accumulate the outer surface lipoprotein A (OspA)-the basic constituent of the first generation monovalent human vaccine against Lyme disease. The recombinant OspA exhibits a lipid modification typical for bacteria and induced protective antibodies in mice, demonstrating that functionally active bacterial lipoproteins can be processed in plants.

Involvement of chloroplasts in the programmed death of plant cells.

The effect of cyanide, an apoptosis inducer, on pea leaf epidermal peels was investigated. Illumination stimulated the CN--induced destruction of guard cells (containing chloroplasts and mitochondria) but not of epidermal cells (containing mitochondria only). The process was prevented by antioxidants (alpha-tocopherol, 2,5-di-tret-butyl-4-hydroxytoluene, and mannitol), by anaerobiosis, by the protein kinase C inhibitor staurosporine, and by cysteine and serine protease inhibitors. Electron acceptors (menadione, p-benzoquinone, diaminodurene, TMPD, DCPIP, and methyl viologen) suppressed CN--ind

Studies on the reconstitution of o(2)-evolution of chloroplasts.

Extraction of spinach (Spinacia oleracea L.) chloroplasts with cholate-asolectin in the absence of Mg(2+) results in the rapid and selective inactivation of O(2) evolution and a partial (30 to 40%) loss of photosystem II (PSII) donor activity without extraction of thylakoid bound Mn ( approximately 5 to 6 Mn per 400 Chlorophyll). Inclusion of ethylene glycol in the extractions inhibits loss of O(2) evolution and results in quantitative and qualitative differences in proteins solubilized but does not significantly inhibit the partial loss of PSII donor activity. Similarly, in two stage experime

Endonucleases for UV-irradiated and depurinated DNA in barley chloroplasts.

Lysates of barley chloroplasts release more radioactivity into acid soluble form from UV-irradiated and alkylated-depurinated E. coli [3H] DNA than from intact DNA. By means of affinity chromatography on depurinated DNA-cellulose and/or UV irradiated DNA-cellulose and by electrophoresis in polyacrylamide gels, four activities on depurinated DNA were separated. One of these contained activity against heavily UV-irradiated /270 J.m-2/ native DNA. In addition, two other nucleases specific towards UV-DNA were separated. One of them was active on native and heat denatured DNA irradiated with 10 J .

Towards assembling functional cyanobacterial β-carboxysomes in Oryza sativa chloroplasts.

The major limiting factor of photosynthesis in C3 plants is the enzyme, rubisco which inadequately distinguishes between carbon dioxide and oxygen. To overcome catalytic deficiencies of Rubisco, cyanobacteria utilize advanced protein microcompartments, called the carboxysomes which envelopes the enzymes, Rubisco and Carbonic Anhydrase (CA). These microcompartments facilitate the diffusion of bicarbonate ions which are converted to CO2 by CA, following in an increase in carbon flux near Rubisco boosting CO2 fixation process. Inspired by this mechanism, our study aims to improve photosynthetic e

Auxins, the hidden player in chloroplast development.

Throughout decades of plant research, the plant hormones known as auxins have been found to be of vital importance in most plant development processes. Indole-3-acetic acid (IAA) represents the most common auxin in plants and can be synthesized from its tryptophan precursor, which is synthesized in the chloroplast. The chloroplast constitutes an organelle of great relevance to plants since the photosynthesis process by which plants get most of their energy is carried out there. The role of auxins in photosynthesis has been studied for at least 50 years, and in this time, it has been shown that

Chloroplast Signaling Gates Thermotolerance in Arabidopsis.

Temperature is a key environmental variable influencing plant growth and survival. Protection against high temperature stress in eukaryotes is coordinated by heat shock factors (HSFs), transcription factors that activate the expression of protective chaperones such as HEAT SHOCK PROTEIN 70 (HSP70); however, the pathway by which temperature is sensed and integrated with other environmental signals into adaptive responses is not well understood. Plants are exposed to considerable diurnal variation in temperature, and we have found that there is diurnal variation in thermotolerance in Arabidopsis

[Activity of chloroplast RNase under drought].

It is found, that the protein content in chloroplasts remained constant in conditions of drought, while RNase activity in chloroplasts was decreased in 2-2.5 times as compared with total leaves and hyaloplasma. No reliable differences in base composition of DNA and RNA were found in cotton variety "Tashkent 2" under the lack of water.

Chloroplast membranes and coupling factor conformations.

The demonstrated role of proton translocation and resulting electrochemical activity gradients (protonmotive force) in ATP synthesis by chloroplasts is noted. Evidence for the participation of conformational changes in the terminal ATPase (coupling factor, or CF1) is reviewed. Hydrogen exchange into ordinarily cyptic groups of the molecule occurs only when the subtending membranes are put under the stress of a protonmotive force. Since up to 100 hydrogen atoms per mole are involved in the energy-dependent exchange the conformational change permitting tham access to the medium must be a major o

Expression and assembly of largest foreign protein in chloroplasts: oral delivery of human FVIII made in lettuce chloroplasts robustly suppresses inhibitor formation in haemophilia A mice.

Inhibitor formation is a serious complication of factor VIII (FVIII) replacement therapy for the X-linked bleeding disorder haemophilia A and occurs in 20%-30% of patients. No prophylactic tolerance protocol currently exists. Although we reported oral tolerance induction using FVIII domains expressed in tobacco chloroplasts, significant challenges in clinical advancement include expression of the full-length CTB-FVIII sequence to cover the entire patient population, regardless of individual CD4+ T-cell epitope responses. Codon optimization of FVIII heavy chain (HC) and light chain (LC) increas
