The Anti-Tumorigenic Activity of Sema3C in the Chick Embryo Chorioallantoic Membrane Model.

Sema3C protein, a member of the class 3 family of secreted semaphorins, play an important role in tumor development by regulating cell proliferation, migration, invasion, and angiogenesis processes. Depending on the type and malignancy grade of the tumor, Sema3C function remains controversial. In this study, we constructed a stably overexpressing Sema3C glioblastoma cell line U87 MG and tested it on the chicken embryo chorioallantoic membrane (CAM) model with the aim to reveal Sema3C protein function on angiogenesis process in ovo. Our experiments showed that Sema3C not only affects angiogenes

The chick chorioallantoic membrane: a model of short-term study of Dupuytren's disease.

Dupuytren's disease is a progressive fibroproliferative disorder that impairs hand function by altering the normal structures of the palmar fascial bands. Nodules composed almost entirely of myofibroblasts and cords are pathognomonic of Dupuytren's disease. The myofibroblasts express alpha-smooth muscle actin that is especially involved in development of the disease. We aimed to evaluate whether the xenograft of Dupuytren's fibromatosis taken from operating room and transplanted on chick chorioallantoic membrane (CAM) survives with its histological and immunohistological features. Fresh sample

Oral contraceptives prevent the development of endometriosis in the chicken chorioallantoic membrane model.

BACKGROUND: Fundamental and genetic differences between women in the endometrium may cause some to develop endometriosis, whereas others do not. Oral contraceptives (OC) may have an effect on the endometrium, rendering the development of endometriosis less likely. STUDY DESIGN: Endometrium from women using OC (OCE) and menstrual endometrium (ME) from normal cycling women were transplanted onto the chicken chorioallantoic membrane (CAM), and endometriosis-like lesion formation was evaluated. Microarray gene expression profiling was performed to identify differentially expressed genes in the en

[Chorioallantoic membrane of fertilized avian eggs as a substrate for assessment of cancerous invasiveness].

PURPOSE: Invasiveness is a characteristic feature of malignant tumors considerably determining the prognosis of affected patients. For assessment, apart from in vitro procedures with limited validity, tests on animal models have been established which certainly should be replaced by alternative methods whenever possible. The chorioallantoic membrane (CAM) of fertilized avian eggs represents an epithelial-lined membrane composed of all three blastodermic germ layers. In an "in ovo" assay cancer cells can be applied to this membrane after sinking (CAM assay). Tumor growth and invasiveness should

The avian chorioallantoic membrane in ovo--a useful model for bacterial invasion assays.

The aim of this study was to evaluate the practicability of the chick embryo chorioallantoic membrane (CAM) with special regard to the 'natural air sac' technique (NAST) of preparation for in-vivo research on the invasive potential of bacterial strains of various enterobacterial species. It was sought to establish an experimental system more closely resembling in-vivo conditions than cell lines on one hand, and cheaper and easier to handle than established animal models on the other. Fertilized eggs of the domestic fowl were incubated. The CAM was prepared atraumatically at the natural air spa

A chicken chorioallantoic membrane assay for the evaluation of the androgen responsiveness of prostatic tissue.

A biological assay using the chorioallantoic membrane (CAM) of the chicken egg as an in vitro culture system has been developed and standardized in order to assess the androgen responsiveness of normal and abnormal prostatic tissue. In this assay, test tissue is implanted onto the CAM of chicken eggs in the presence or absence of exogenous testosterone treatment, with subsequent serum levels of 45 ng./dl. and greater than 5,000 ng./dl., respectively. The responsiveness of test tissue to low versus high androgen levels was evaluated in this CAM assay using both cellular morphology and mitotic i

Functional involvement of carbonic anhydrase in calcium transport of the chick chorioallantoic membrane.

Carbonic anhydrase activity was demonstrated in the chick-embryonic chorioallantoic membrane and was correlated with the Ca2+-transport activity of the membrane. It is inhibited by sulphonamides and is expressed in the chorioallantoic membrane in an age-dependent fashion during embryonic development. Ca2+ uptake by the chorioallantoic membrane in vivo also increases in a similar age-dependent manner. The temporal increase in these activities is coincident with calcium deposition in the embryonic skeleton. Incubation of the chorioallantoic membrane in ovo with sulphonamides specifically inhibit

Microvascular assembly and cell invasion in chick mesonephros grafted onto chorioallantoic membrane.

Embryonic tissues, in common with other tissues, including tumours, tend to develop a substantial vasculature when transplanted onto the chorioallantoic membrane (CAM). Studies conducted to date have not examined in any detail the identity of vessels that supply these grafts, although it is known that the survival of transplanted tissues depends on their ability to connect with CAM vessels supplying oxygen and nutrients. We grafted the mesonephros, a challenging model for studies in vascular development, when it was fully developed (HH35). We used reciprocal chick-quail transplantations in ord

Morphogenetic maturation of chicken chorioallantoic membrane (CAM) and its disruption by tumor xenografts.

Chorioallantoic membrane (CAM) functions as embryonic "lung" in birds/reptiles and forms through fusion of the chorion and allantois, two extraembryonic membranes conserved in amniotic vertebrates. Modified CAM gives rise to the placenta, performing similar roles in mammalian development. Despite its importance, cellular morphogenesis leading to CAM maturation is poorly understood. In particular, it is unclear how mesoderm-derived capillary cells are positioned within the chorionic ectoderm layer in mature CAM. In this work, using a combination of live imaging, histology, immunofluorescence an

Chorioallantoic membrane (CAM) assay to study treatment effects in diffuse intrinsic pontine glioma.

Diffuse intrinsic pontine glioma (DIPG) is a lethal pediatric brain tumor. While there are a number of in vivo rodent models for evaluating tumor biology and response to therapy, these models require significant time and resources. Here, we established the chick-embryo chorioallantoic (CAM) assay as an affordable and time efficient xenograft model for testing a variety of treatment approaches for DIPG. We found that patient-derived DIPG tumors develop in the CAM and maintain the same genetic and epigenetic characteristics of native DIPG tumors. We monitored tumor response to pharmaco- and radi

Evidence of steroid hormone activity in the chorioallantoic membrane of a Turtle (Pseudemys nelsoni).

Endocrine properties of extraembryonic membranes have traditionally been viewed as a characteristic of placental amniotes. However, our laboratory recently demonstrated that this ability extends to the extraembryonic membranes of two oviparous amniotes (chicken and alligator) indicating that endocrine extraembryonic membranes are not an innovation of placental amniotes and suggesting that this could be a shared amniote characteristic. In this study, we test our hypothesis that the chorioallantoic membrane (CAM) obtained from non-archosaurian obligate oviparous amniotes such as turtles, have th

Organochlorine pesticides in chorioallantoic membranes of Morelet's crocodile eggs from belize.

Recent studies examined the utility of the chorioallantoic membrane (CAM) as a nonlethal, noninvasive indicator of environmental contaminant exposure in oviparous wildlife. The CAM is a highly vascularized extraembryonic membrane that functions as a site for respiration, nutrient transport, and waste storage during embryonic development. After hatching, the CAM is usually discarded with the eggshell and can be used for chemical residue analysis. Chorioallantoic membranes have been used successfully to examine contaminant exposure and predict chemical concentrations in multiple species of birds

Angiostatic activity and metabolism of cortisol in the chorioallantoic membrane (CAM) of the chick embryo.

There is considerable interest in the discovery of compounds which inhibit angiogenesis dependent (neovascular) diseases. The chick embryo, due to the rapid development of an extensive vascular capillary network in the chorioallantoic membrane (CAM), has been used extensively as a model for studying angiogenesis. Angiostatic steroids are a new class of compounds which inhibit the growth of new capillaries in the chick CAM and in other models of neovascularization. Despite the potential therapeutic importance of these compounds, little is known about the ability of the CAM to metabolize these s

Chorioallantoic Membrane Assay to Assess Primary and Metastatic Tumor Growth in Ewing Sarcoma.

The chorioallantoic membrane (CAM) assay provides a rapid, cost-effective, and ethically advantageous in vivo model to investigate tumor growth, angiogenesis, invasion, and metastasis. The CAM of the developing chicken embryo forms a highly vascularized and naturally immunodeficient environment that supports the implantation and proliferation of human or murine cells without the need for immunosuppression. This chapter presents a detailed and standardized protocol for performing tumor xenografts and metastasis analyses on the CAM, including optimized conditions for egg handling, embryo viabili

The effect of early prenatal hypercapnia on the vascular network in the chorioallantoic membrane of the chicken embryo.

Over the last decade, the poultry sector has sought to develop novel ways to monitor chicken embryonic growth, health, and quality to control and optimize egg incubation conditions, particularly the concentration of dissolved gases (O(2), CO(2)). One of the parameters, which may change under different gas concentrations, is the angiogenesis in the chorioallantoic membrane (CAM), the organ for gas exchange of the chicken embryo. In this study, a newly developed methodology was used to quantify the angiogenesis in the CAM under normal and early hypercapnic conditions (i.e., increased CO(2) conce
