Chromogranins (Cg) are large acidic proteins, co-stored with amines or peptides in the secretory granules of a wide variety of paraneurons. By immunohistochemistry performed on serial semithin sections, the distribution of Cg in the gastroenteropancreatic (GEP) system was analyzed in the endocrine pancreas of 10 mammalian species and in enteroendocrine cells of 6 species. Of the Cg families, CgA is primarily localized in GEP endocrine cells. CgA was regularly present in amine containing cells (EC and EC-like cells). In 11 other endocrine cell types, CgA-immunoreactivities showed inter-species

Chromogranin A (CGA) is a peptide secreted by neuroendocrine cells and filtered by the kidneys. Excessive CGA secretion is common in neuroendocrine tumors (NETs) and has been linked to organ dysfunction, though direct kidney injury remains rarely described. We present a woman in her mid-70s diagnosed with metastatic, well-differentiated pancreatic NET who developed progressive renal dysfunction. Initial CGA levels exceeded 86,000 ng/mL and later rose to >500,000 ng/mL. Despite the absence of nephrotoxic exposures, imaging abnormalities, or significant proteinuria, her serum creatinine increase

Medullary thyroid carcinoma (MTC) is a relatively rare thyroid carcinoma of C-cell deviation and produces and secrete calcitonin (CT) and chromogranin A (CgA) into the blood. Thus, both CT and CgA are immunohistochemical and serum markers for MTCs. MTC occurs in both sporadic and inheritable cases and the hallmark of inheritable cases in multiple endocrine neoplasm 2 (NEN2) is MTC. MEN2 cases represent 30% of MTCs through germline RET protooncogene mutation and occur in younger ages involving bilateral thyroid lobes. Sporadic cases are 70% of cases of solitary tumor and occur in older ages. Cg

OBJECTIVES: Meniere's disease (MD) is an inner ear disorder characterized by episodic vertigo, ear fullness, and hearing loss; usually vertigo attacks cluster in specific period. We studied in MD patients the circulating levels of chromogranin A (CgA) and vasostatin-1 (VS-1), secreted by the neuroendocrine system and involved in the regulation of the endothelial barrier function.
METHODS: Serum levels were assessed in 37 MD patients and 36 controls. The ratio between VS-1 and CgA was calculated.
RESULTS: CgA was increased in patients compared to controls (1.46 versus 0.67 nM, p = 0.01) while

New endogenous antimicrobial peptides (AMPs) derived from chromogranin A (CgA) are secreted by nervous, endocrine and immune cells during stress. They display antimicrobial activities by lytic effects at micromolar range using a pore-forming mechanism against Gram-positive bacteria, filamentous fungi and yeasts. These AMPs can also penetrate quickly into neutrophils (without lytic effects), where, similarly to "cell penetrating peptides", they interact with cytoplasmic calmodulin, and induce calcium influx via Store Operated Channels therefore triggering neutrophils activation. BACKGROUND: Chromogranin (Cg) A is expressed in neuroendocrine and neuronal tissues. It is involved in the generation of secretory granules and is cleaved to form biologically active peptides. Targeted ablation of the Chga gene resulted in increased plasma catecholamines, high blood pressure, and decreased size and number of adrenal medullary chromaffin granules. The aim of this study was to determine whether Chga null mice display changes in the morphology and function of the endocrine pancreas.
MATERIALS AND METHODS: Sections of pancreata from Chga-/-, Chga+/- and Chga+/+ mice, were immunos

Chromogranin A (CgA) is a major protein of the parathyroid gland that is costored and cosecreted with parathyroid hormone (PTH). CgA, widely distributed in endocrine and neuroendocrine cells in addition to parathyroid cells, appears to be a precursor of biologically active autocrine or paracrine peptides that include pancreastatin (CgA240-288) and parastatatin (CgA347-419), compounds that strongly inhibit stimulated secretion of PTH and costored CgA. The biosynthesis of CgA and PTH via gene expression and mRNA translation are noncoordinately altered by agents that affect parathyroid cell secre

Several lines of evidence link chromogranin A (CgA), the major soluble protein in catecholamine storage vesicles, with the cholinergic nervous system, abnormalities of which may play a central role in memory deficits in Alzheimer dementia. Because of reported elevations of CgA in Alzheimer brains and its presence in the senile plaque lesions of such brains, we evaluated the concentration of CgA in cerebrospinal fluid of Alzheimer dementia patients and matched controls. CgA was detectable in each sample, but the results in dementia showed substantial overlap with and no significant (p = 0.55) d

Chromogranin A, the most abundant protein in bovine adrenal chromaffin granules, bound calmodulin in a Ca2(+)-dependent manner, and the calmodulin-binding property was utilized to purify chromogranin A. Chromogranin A has been described in the past as a "random-coil polypeptide" with little alpha-helix or beta-sheet conformation. However, circular dichroism measurements with pure, native chromogranin A revealed relatively high alpha-helical contents (40% at the intravesicular pH of 5.5). Fluorescence studies confirmed previous observations that chromogranin A binds Ca2+ with low affinity. Cons

The expression of the neuroendocrine markers synaptophysin and chromogranin A was analyzed by immunohistochemistry in 14,584 samples from 103 different tumor types and subtypes in a tissue microarray format. At least one of these markers was found to be positive in 96.7% of tumors from various subtypes of neuroendocrine neoplasms. In non-neuroendocrine tumors, synaptophysin and/or chromogranin A staining was seen in 6.3% (n = 584), specifically in 41 of 88 non-neuroendocrine tumor entities. Basal cell carcinomas of the skin (50% positive for chromogranin A alone) and adrenocortical carcinomas

Chromogranin A is a member of the granin glycoprotein family that is expressed by the endocrine and neuroendocrine cells of different organs. Intracellularly, chromogranin A contributes to the regulation of secretion and gives several cleavage products after secretion. Some of its cleavage products modify the hormone functions in autocrine and paracrine ways, while the functions of others have not been fully understood yet. Serum chromogranin A level is most prominently used in neuroendocrine tumour diagnostics. In addition, recent studies have suggested that chromogranin A and some of its cle

BACKGROUND: Chromogranin-A (CgA) is a secretory protein processed into peptides that regulate angiogenesis and vascular cells activation, migration and proliferation. These processes may influence arterial inflammation and remodelling in Takayasu arteritis (TA).
METHODS: Plasma levels of full-length CgA (CgA439), CgA fragments lacking the C-terminal region (CgA-FRs) and the N-terminal fragment, CgA1-76 (vasostatin-1, VS-1) were analysed in 42 patients with TA and 20 healthy age-matched controls. Vascular remodelling was longitudinally assessed by imaging. CgA peptides were related to markers

Currently, no effective treatment for malignant pheochromocytoma exists. The aim of our study was to investigate the role of chromogranin A (CgA) as a specific target molecule for immunotherapy in a murine model for pheochromocytoma. Six amino acid-modified and non-modified CgA peptides were used for dendritic cell vaccination. Altogether, 50 mice received two different CgA vaccination protocols; another 20 animals served as controls. In vitro tetramer analyses revealed large increases of CgA-specific cytotoxic T cells (CTL) in CgA-treated mice. Tumors of exogenous applied pheochromocytoma cel

Chromogranin A (CgA) was recently reported as a marker of various stress responses. The aim of this study was to investigate the immunohistochemical distribution of CgA in human tissues in medicolegal autopsy cases as a basis for postmortem investigation of stress responses. The autopsy cases (n=30, within 48 h postmortem) comprised cases of mechanical asphyxia (n=15: strangulation, n=8; hanging, n=7) and acute myocardial infarction/ischemia (AMI, n=15). Routinely formalin-fixed paraffin-embedded tissue sections, including those of the hypothalamus, pituitary gland, cardiac muscle, lungs, live

Chromogranin A is released together with epinephrine and norepinephrine from catecholaminergic cells. Specific endopeptidases cleave chromogranin A into biologically active peptide fragments, including catestatin, which inhibits catecholamine release. Previous studies have suggested that a deficit in this sympathetic "braking" system might be an early event in the pathogenesis of human hypertension. Whether chromogranin A (CHGA) polymorphisms predict end-organ complications of hypertension, such as end-stage renal disease, is unknown. Among blacks, we studied common genetic variants spanning t

In the endocrine pancreas, chromogranin-A and pancreastatin have been suggested to inhibit islet insulin secretion, whereas chromogranin-B has not been studied in this context. Furthermore, a putative effect by chromogranins on IAPP secretion is unknown. We aimed to elucidate the endogenous effect of chromogranin-A, pancreastatin and chromogranin-B on islet insulin and IAPP secretion from pancreatic NMRI mouse islets. In acute experiments, there was a tendency towards an increase in insulin release, which became more manifest after a 48-h exposure. Moreover, 48 h exposure to chromogranin-B ant

We evaluated whether measuring serum levels of chromogranin A, a marker of neuroendocrine tumours, could be useful in the differential diagnosis between pituitary, adrenal and ectopic causes of Cushing's syndrome. Thirty patients with Cushing's syndrome were studied. The localization of the tumours responsible was pituitary in 15, adrenal in 5 and ectopic in 10 patients. Serum concentrations of chromogranin A were measured in all patients. Petrosal sinus sampling for chromogranin A was performed in the cases with pituitary-dependent Cushing's syndrome. Immunohistochemical staining for chromogr
