Reduction of quinones and radicals by a plasma membrane redox system of Phanerochaete chrysosporium.

Quinones which are produced during the mineralization of lignin and xenobiotics by the white rot fungus Phanerochaete chrysosporium were reduced by a plasma membrane redox system of the fungus. Both intracellular enzymes and the plasma membrane redox system were able to reduce 1,4-benzoquinone. However, no quinone reductase activity was observed with the extracellular culture fluid. The intracellular reductase activity had a pH optimum between 6.0 and 7.0 and a Km of 150 microM. Reduction of 1,4-benzoquinone by the plasma membrane redox system had a pH optimum between 7.5 and 8.5 and exhibited

Flavin-containing monooxygenases from Phanerochaete chrysosporium responsible for fungal metabolism of phenolic compounds.

We investigated the cellular responses of the white-rot basidiomycete Phanerochaete chrysosporium against vanillin. Based upon a proteomic survey, it was demonstrated that two flavin-containing monooxygenases (PcFMO1 and PcFMO2) are translationally up-regulated in response to exogenous addition of vanillin. To elucidate their catalytic functions, we cloned cDNAs and heterologously expressed them in Escherichia coli. The recombinant PcFMO1 showed catalytic activities against monocyclic phenols such as phenol, hydroquinone, and 4-chlorophenol. In addition, the product from hydroquinone was ident

H2O2-dependent decolorization of Poly R-481 by particulate fractions from Phanerochaete chrysosporium.

A cell-free preparation from Phanerochaete chrysosporium culture medium decolorized the polymeric dye Poly R-481. The majority of this decolorization activity sedimented when centrifuged at 150,000 X g, indicating that it was associated with a particulate body. The activity was sensitive to heat, azide and cyanide, was stimulated by exogenously added H2O2, and was optimal around pH 4. Electron micrographs of the sedimented culture medium fraction showed the presence of numerous particulate structures. A similar dye decolorization activity from sonicated mycelium also sedimented at 150,000 X g.

Facile green extracellular biosynthesis of CdS quantum dots by white rot fungus Phanerochaete chrysosporium.

This study details a novel method for the extracellular microbial synthesis of cadmium sulfide (CdS) quantum dots (QDs) by the white rot fungus Phanerochaete chrysosporium. P. chrysosporium was incubated in a solution containing cadmium nitrate tetrahydrate, which became yellow from 12h onwards, indicating the formation of CdS nanocrystals. The purified solution showed a maximum absorbance peak between 296 and 298 nm due to CdS particles in the quantum size regime. The fluorescence emission at 458 nm showed the blue fluorescence of the nanoparticles. X-ray analysis of the nanoparticles confirm

1H NMR investigation of manganese peroxidase from Phanerochaete chrysosporium. A comparison with other peroxidases.

1H NMR spectra at 200- and 600-MHz of manganese peroxidase from Phanerochaete chrysosporium and of its cyanide derivative are reported. The spectrum of the native protein is very similar to that of other peroxidases. The assignment of the spectrum of the cyanide derivative has been performed through 1D NOE, 2D NOESY, and COSY experiments. This protein is very similar to lignin peroxidase, the only meaningful difference being the shift of H delta 2 of the proximal histidine. The spectra of the cyanide derivative of these two proteins are compared with those of horseradish peroxidase and cytochr

Voriconazole treatment of pulmonary mycosis caused by Chrysosporium zonatum after treatment for pulmonary tuberculosis.

Chrysosporium zonatum is a soil-dwelling fungus that rarely causes pulmonary infections, and a small number of cases have been reported to date. A 74-year-old man, who had previously been treated for tuberculosis, presented with symptoms of low-grade fever, anorexia, cough, and bloody sputum. Chest computed tomography (CT) showed a thick-walled cavitary lesion in the right upper lobe, in which there was a suspected mycotic mass. Initially, the patient was suspected to have chronic aspergillosis due to positive serum anti-Aspergillus antibodies. However, bronchoscopic culture revealed the growt

Chrysosporium articulatum mimicking Trichophyton spp. infection in a cat: a case presentation and literature review.

BACKGROUND: Dermatophytosis is a common skin infection of cats and many other animals. A reliable diagnosis is crucial because of the zoonotic potential of dermatophytes. The routine mycological diagnostic procedures for dermatophytosis are widely known, but in the case of some isolates, identification based on phenotypic characteristics may be incorrect. Infections caused by Chrysosporium spp. are usually described in reptiles, but in other animals they are uncommon. CASE PRESENTATION: This study presents a description of a cat with dermatological lesions, that was mistakenly diagnosed with

Biodegradation of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) by Phanerochaete chrysosporium in the presence of Cd2.

Aerobic biodegradation of 2,2',4,4'-tetrabrominated diphenyl ether (BDE-47) by Phanerochaete chrysosporium in the presence of Cd2+ was investigated in this study. The results showed that P. chrysosporium could effectively degrade BDE-47, and its extracellular enzyme played an important role in the process of decomposition. BDE-47 biodegradation by fungi was more tolerant than extracellular enzyme in the presence of Cd2+. Also, both of the activity of two typical enzymes, MnP and LiP, descended with ascended Cd2+ concentration. Based on the four mono-hydroxylated PBDEs (5-OH-BDE-47, 4'-OH-BDE-1

Industrial applications of Phanerochaete chrysosporium lignin-degrading enzymes: current status, production challenges, and future directions.

Phanerochaete chrysosporium (Pc) is a white-rot fungus recognized for its highly efficient lignin-degrading enzymes (LDEs), including lignin peroxidase (LiP), manganese peroxidase (MnP), and laccase. These oxidative enzymes possess transformative capabilities across multiple industrial applications, such as biopulping, biofuel production, bioremediation and for the treatment of industrial wastewater. However, the commercial use of these enzymes is limited due to time-consuming scale-up procedures in native hosts, instability in industrial environments, and high production costs. The recent dev

Manganese deficiency can replace high oxygen levels needed for lignin peroxidase formation by Phanerochaete chrysosporium.

The combined effects of Mn and oxygen on lignin peroxidase (LIP) activity and isozyme composition in Phanerochaete chrysosporium were studied by using shallow stationary cultures grown in the presence of limited or excess N. When no Mn was added, LIP was formed in both N-limited and N-excess cultures exposed to air, but no LIP activity was observed at Mn concentrations greater than 13 mg/liter. In oxygen-flushed, N-excess cultures, LIP was formed at all Mn concentrations, and the peak LIP activity values in the extracellular fluid were nearly identical in the presence of Mn concentrations rang

Polyvinyl alcohol-immobilized Phanerochaete chrysosporium and its application in the bioremediation of composite-polluted wastewater.

A novel biosorbent, polyvinyl alcohol (PVA)-immobilized Phanerochaete chrysosporium, was applied to the bioremediation of composite-polluted wastewater, containing both cadmium and 2,4-dichlorophenol (2,4-DCP). The optimum removal efficiency achieved was 78% for Cd(II) and 95.4% for 2,4-DCP at initial concentrations of 20 mg/L Cd(II) and 40 mg/L 2,4-DCP. PPBs had significantly enhanced the resistance of P. chrysosporium to 2,4-DCP, leading to the degradation rates of 2,4-DCP beyond 90% with varying initial 2,4-DCP concentrations. This research demonstrated that 2,4-DCP and secreted proteins mi

Dermatitis and cellulitis in leopard geckos (Eublepharis macularius) caused by the Chrysosporium anamorph of Nannizziopsis vriesii.

An epizootic of ulcerative to nodular ventral dermatitis was observed in a large breeding colony of 8-month to 5-year-old leopard geckos (Eublepharis macularius) of both sexes. Two representative mature male geckos were euthanized for diagnostic necropsy. The Chrysosporium anamorph of Nannizziopsis vriesii (CANV) was isolated from the skin lesions, and identification was confirmed by sequencing of the internal transcribed spacer region of the rRNA gene. Histopathology revealed multifocal to coalescing dermal and subcutaneous heterophilic granulomas that contained septate fungal hyphae. There w

Transcriptional effect of a calmodulin inhibitor, W-7, on the ligninolytic enzyme genes in Phanerochaete chrysosporium.

We investigated the effects of a calmodulin (CaM) inhibitor, W-7, on the expression of lignin peroxidase (LiP) and manganese peroxidase (MnP) genes in Phanerochaete chrysosporium to consider the role of cam gene, which was upregulated in parallel with the total activities of LiP and MnP in our previous transcriptomic analysis. The addition of 100 μM W-7 to the fungal cultures repressed the total activities of LiP and MnP, whereas the addition of 100 μM W-5, which is a control drug of W-7, retained approximately half of them, indicating that the effect of W-7 was attributable to CaM inhibition.

Changes in the gene expression of the white rot fungus Phanerochaete chrysosporium due to the addition of atropine.

We constructed a LongSAGE (Long Serial Analysis of Gene Expression) library from a 3-d culture of Phanerochaete chrysosporium supplemented with atropine, which inhibits the production of lignin-degrading enzymes. The library (the atropine library) contains 13,108 LongSAGE tags and 6,783 unique tags. The gene expression profile represented by the tags was compared with those of two previously constructed libraries, one of which was constructed using 2-d cultures in which the fungus had not yet produced ligninolytic enzymes (the 2-d library) and the other was constructed using 3-d cultures in wh

Hydrogen sulfide and calcium effects on cadmium removal and resistance in the white-rot fungus Phanerochaete chrysosporium.

Hydrogen sulfide (H2S), an emerging gas transmitter, has been shown to be involved in multiple intracellular physiological and biochemical processes. In this study, the effects of hydrogen sulfide coupled with calcium on cadmium removal and resistance in Phanerochaete chrysosporium were examined. The results revealed that H2S enhanced the uptake of calcium by P. chrysosporium to resist cadmium stress. The removal and accumulation of cadmium by the mycelium was reduced by H2S and Ca2+ pretreatment. Moreover, oxidative damage and membrane integrity were alleviated by H2S and Ca2+. Corresponding
