This year marks the 125th anniversary of the beginning of regeneration research in the ascidian Ciona intestinalis. A brief note was published in 1891, reporting the regeneration of the Ciona neural complex and siphons. This launched an active period of Ciona regeneration research culminating in the demonstration of partial body regeneration: the ability of proximal body parts to regenerate distal ones, but not vice versa. In a process resembling regeneration, wounds in the siphon tube were discovered to result in the formation of an ectopic siphon. Ciona regeneration research then lapsed into

The appearance of the notochord represented a milestone in Deuterostome evolution. The notochord is necessary for the development of the chordate body plan and for the formation of the vertebral column and numerous organs. It is known that the transcription factor Brachyury is required for notochord formation in all chordates, and that it controls transcription of a large number of target genes. However, studies of the structure of the cis-regulatory modules (CRMs) through which this control is exerted are complicated in vertebrates by the genomic complexity and the pan-mesodermal expression t

Biological introductions bring into contact species that can still hybridize. The evolutionary outcomes of such secondary contacts may be diverse (e.g. adaptive introgression from or into the introduced species) but are not yet well examined in the wild. The recent secondary contact between the non-native sea squirt Ciona robusta (formerly known as C. intestinalis type A) and its native congener C. intestinalis (formerly known as C. intestinalis type B), in the Western English Channel, provides an excellent case study to examine. To examine contemporary hybridization between the two species, w

Electrophysiological techniques were used to study the role of ion currents in the ascidian Ciona intestinalis oocyte plasma membrane during different stages of growth, meiosis, fertilization and early development. Three stages of immature oocytes were discriminated in the ovary, with the germinal vesicle showing specific different features of growth and maturation. Stage-A (pre-vitellogenic) oocytes exhibited the highest L-type calcium current activity and were incompetent for meiosis resumption. Stage-B (vitellogenic) oocytes showed a progressive disappearance of calcium currents and the fir

The distribution of biomolecules is essential information for understanding biological systems. Here, we present a protocol for 2D and 3D reconstruction of tissue images at the single-cell level by integrating microscopy and mass spectrometry imaging (MSI) data through linear interpolation. We describe steps for sample preparation, MSI, and construction of 2D and 3D single-cell peptide atlases of the cerebral ganglion of Ciona robusta. This approach is also applicable to other animals in which target cells are labeled with endogenous fluorescent proteins. For complete details on the use and ex

Sperm proteins mediating sperm-egg interaction should be exhibited on the sperm surface, or exposed or released when sperm approach an egg. In ascidians (protochordates), sperm undergo a sperm reaction, characterized by enhanced sperm motility and mitochondrial swelling and shedding on contact with the vitelline coat (VC) or by treatment with Ca(2+) ionophore. Here, proteomic analysis was conducted on sperm exudates and sperm surface proteins using ionomycin-induced sperm reaction and cell-impermeable labeling in Ciona intestinalis type A (C. robusta). In the exudate from sperm treated with io

Spermatogenesis is a complex and continuous process of germ-cell differentiation. This complex process is regulated by many factors, of which gene regulation in spermatogenic cells plays a decisive role. Spermatogenesis has been widely studied in vertebrates, but little is known about spermatogenesis in protochordates. Here, for the first time, we performed single-cell RNA sequencing (scRNA-seq) on 6832 germ cells from the testis of adult Ciona intestinalis. We identified six germ cell populations and revealed dynamic gene expression as well as transcriptional regulation during spermatogenesis

BACKGROUND: Left-right asymmetries are a common feature of metazoans and can be found in a number of organs including the nervous system. These asymmetries are particularly pronounced in the simple central nervous system (CNS) of the swimming tadpole larva of the tunicate Ciona, which displays a chordate ground plan. While common pathway elements for specifying the left/right axis are found among chordates, particularly a requirement for Nodal signaling, Ciona differs temporally from its vertebrate cousins by specifying its axis at the neurula stage, rather than at gastrula. Additionally, Cion

The cholecystokinin (CCK)/gastrin family peptides are involved in regulation of feeding and digestion in vertebrates. In the ascidian Ciona intestinalis type A (Ciona robusta), cionin, a CCK/gastrin family peptide, has been identified. Cionin is expressed exclusively in the central nervous system (CNS). In contrast, cionin receptor expression has been detected in the CNS, digestive tract, and ovary. Although cionin has been reported to be involved in ovulation, its physiological function in the CNS remains to be investigated. To elucidate its neural function, in the present study, we analyzed

Gonadotropin-releasing hormone (GnRH) is an important neuropeptide that regulates reproductive function in vertebrates. GnRH-secreting neurons (GnRH neurons) are derived from neurogenic placodes and the neural crest during embryonic development. In the larva of the ascidian Ciona intestinalis type A (Ciona robusta), one of the gnrh genes, gnrh2, is expressed in the protoplacodederived sensory neurons as well as the central nervous system including the brain vesicle, the motor ganglion, and the nerve cord. In this study, we identified gnrh2-expressing cells in the motor ganglion and visualized

The sea squirt Ciona robusta is a model organism characterized by a transparent body, exhibiting peculiar physiologic and evolutionary characters. In vitro fertilization and breeding of sea squirts is possible, in order to preserve consistent genetic pools. However, some aspects of its biology, as the feeding efficiency according to diet quantity and quality, are still scarcely known. Here we test the effects of three experimental diets on survival and growth, to detect physiological and molecular responses to various types of alimentary suspended particles and the effects of feed concentratio

Dopamine is a major neuromodulator synthesized by numerous cell populations in the vertebrate forebrain and midbrain. Owing to the simple organization of its larval nervous system, ascidian tunicates provide a useful model to investigate the anatomy, neurogenesis and differentiation of the dopaminergic neural network underlying the stereotypical swimming behaviour of its chordate-type larva. This study provides a high-resolution cellular analysis of tyrosine hydroxylase (TH)-positive and dopamine-positive cells in Ciona intestinalis embryos and larvae. Dopamine cells are present only in the se

The cerebellum, a major structure of the vertebrate central nervous system, forms the dorsal region of the hindbrain. While evidence from connectomics, neuron classification, and gene expression analyses suggests that the major CNS divisions of forebrain, midbrain, hindbrain, and spinal cord predate the vertebrate/tunicate divergence, it is unknown if the subdivision of the hindbrain into a cerebellum, or cerebellum precursor structure, does as well. In fact, conflicting views exist over whether even the most basal vertebrates, the cyclostomes, have a cerebellum homolog. We report here that th

Mercury is one of the most harmful pollutants in aquatic ecosystems. The present study assessed the effects of gradual concentrations of mercury (1, 10, 50, and 100 μg L-1) on the antioxidant status and fatty acid profiles in the viscera of the solitary ascidian Ciona intestinalis (Linnaeus, 1767). Additionally, histological analyses were conducted on the ovary to evaluate morphological changes induced by mercury exposure. The results revealed an increase in malondialdehyde (MDA), protein carbonyl (PCO), glutathione (GSH) and glutathione S-transferase (GST) levels. Additionally, metallothionei

Due to similarities in iodine concentrations and peroxidase activities, the thyroid in vertebrates is considered to originate from the endostyle of invertebrate chordates even though it is a glandular (mucus-producing) organ for aquatic suspension feeding. Among chordates with an endostyle, urochordates are useful evolutionary research models for the study of vertebrate traits. The ascidian Ciona intestinalis forms an endostyle with specific components of glandular- and thyroid-related elements, and molecular markers have been identified for these components. Since we previously examined a sim

Follicular thyroid cells in vertebrates are considered to have evolved from the endostyle of invertebrate chordates based on the shared features of the concentration of iodine and expression of thyroid-related genes, including Nkx2-1, FoxE, and TPO. We previously demonstrated using gene knockout (KO) experiments on the ascidian Ciona robusta that the thyroid-related transcription factors, Nkx2-1 and FoxE, were associated with endostyle development, and also that Nkx2-1 regulated the expression of TPO via FoxE, even though the expression of Pax2/5/8 homologs was not affected. In the present stu

Invertebrate chordates, such as the tunicate Ciona, can offer insight into the evolution of the chordate phylum. Anatomical features that are shared between invertebrate chordates and vertebrates may be taken as evidence of their presence in a common chordate ancestor. The central nervous systems of Ciona larvae and vertebrates share a similar anatomy despite the Ciona CNS having ~180 neurons. However, the depth of conservation between the Ciona CNS and those in vertebrates is not resolved. The Ciona caudal CNS, while appearing spinal cord-like, has hitherto been thought to lack motor neurons,

Maintenance and breeding of experimental organisms are fundamental to life sciences, but both initial and running costs, and hands-on zootechnical demands can be challenging for many laboratories. Here, we first aimed to further develop a simple protocol for reliable inland culture of tunicate model species of the Ciona genus. We cultured both Ciona robusta and Ciona intestinalis in controlled experimental conditions, with a focus on dietary variables, and quantified growth and maturation parameters. From statistical analysis of these standardized datasets, we gained insights into the post-emb

Antisera specific for mammalian insulin-like growth factor 1 (IGF-1) and mammalian insulin and the double immunofluorescence technique were used for this study. IGF-1-like-immunoreactivity was localized in entero-endocrine cells in the gastro-intestinal tract of the protochordates Ciona intestinalis and Branchiostoma lanceolatum. Some of the specimens also showed IGF-1-like-immunoreactive (-IR) perikarya and fibers in the central nervous system. Whilst in rat endocrine pancreas, IGF-1-IR and insulin-IR occurred in different cell populations, in Ciona and Branchiostoma the vast majority of ente

While the CNSs of the chordate sister clades Tunicata and Vertebrata have unmistakable homology, the extent of their conservation is still being uncovered. We report that the hindbrain of the tunicate Ciona, like those of vertebrates, has functionally-distinct dorsal and ventral domains. The Ciona dorsal hindbrain functions as a relay and processing center for peripheral sensory neurons, while in vertebrates the dorsal hindbrain forms the cerebellum. Despite the different fates of dorsal hindbrains in Ciona versus vertebrates, we present evidence from analysis of gene expression, developmental
