Combined Inhibitory Effects of Citrinin, Ochratoxin-A, and T-2 Toxin on Aquaporin-2.

Aquaporins form a large family of transmembrane protein channel that facilitates selective and fast water transport across the cell membrane. The inhibition of aquaporin channels leads to many water-related diseases such as nephrogenic diabetes insipidus, edema, cardiac arrest, and stroke. Herein, we report the molecular mechanism of mycotoxins (citrinin, ochratoxin-A, and T-2 mycotoxin) inhibition of aquaporin-2 (AQP2) and arginine vasopressin receptor 2. Molecular docking, molecular dynamics simulations, quantum chemical calculations, residue conservation-coupling analysis, sequence alignmen

Effect of feeding graded doses of Citrinin on clinical and teratology in female Wistar rats.

Citrinin is the one of the well-known mycotoxins, which is possibly spread all over the world. The graded doses of citrinin (1, 3 and 5 ppm CIT in feed) in female Wistar rats 10 weeks prior to mating, during mating and during organogenesis resulted in resorptions and post implantation losses, decreased fetal body weights and crown-rump lengths in fetuses of all groups. Various developmental anomalies recorded in fetuses of treated rats included gross (wrist drop, curled tail, stretched forelimb, subcutaneous haematoma), skeletal (incomplete ossification of skull bones, incomplete fusion of ver

Citrinin induces renal PANoptosis by mediating mitochondrial dysfunction through the GSDMD-N/DRP1 pathway.

Citrinin (CTN), a nephrotoxic mycotoxin with high environmental stability, is widely present in contaminated food sources, leading to chronic ingestion in humans and animals. Although CTN exposure is closely associated with Balkan endemic nephropathy (BEN), the molecular mechanisms underlying CTN-induced renal injury remain largely elusive. PANoptosis-a newly recognized form of regulated cell death that integrates pyroptosis, apoptosis, and necroptosis-has emerged as a key contributor to kidney pathology. In this study, we established both in vivo (KM mice) and in vitro (TCMK-1 cells) models o

Dicitrinols A-C: Citrinin Derivatives from Hydrothermal Vent-Associated Fungus Penicillium citrinum TW132-59.

Three new unusual citrinin derivatives with a unique 6/5/7/5 core, dicitrinols A-C (1-3, respectively), were isolated via the fermentation of hydrothermal vent-associated fungus Penicillium citrinum TW132-59. Their structures were unambiguously determined by nuclear magnetic resonance, mass spectrometry, and electronic circular dichroism calculations. Dicitrinols A-C represent a novel cage carbon skeleton with a decahydro-5,9,4-(epipropane[1,1,3]triyl)cycloocta[b]furan ring system. Dicitrinols A-C showed moderate antifungal activity against Candida albicans, Cryptococcus neoformans, and Fusari

Biomonitoring of ochratoxin A, 2'R-ochratoxin A and citrinin in human blood serum from Switzerland.

Biomonitoring of mycotoxins and their metabolites in biological fluids is increasingly used to assess human exposure. In this study, biomarkers of ochratoxin A (OTA) and citrinin (CIT) exposure were determined in a large number of serum samples from healthy blood donors in Switzerland. In 2019, 700 samples from different regions were obtained. From 240 donors, a second sample (taken 2-9 months later) was available for analysis. Moreover, 355 blood donor samples from 2005 from all regions in Switzerland and 151 additional samples from the southern Swiss region of Ticino from 2005 could be analy

Dual fluorescent immunochromatographic assay for simultaneous quantitative detection of citrinin and zearalenone in corn samples.

The presence of multiple mycotoxins in the agricultural products poses a serious threat to the health of humans and animals. Citrinin (CIT) causes slow growth in animals and damages the kidney function. Zearalenone (ZEN) causes chronic poisoning, abnormal functioning and even death in animals. Herein, a dual fluorescent immunochromatographic assay (DF-ICA) based on europium nanoparticles (EuNPs) was developed for the simultaneous detection of CIT and ZEN in the corn samples. After optimization, the limits of detection (LODs), IC50 and average recoveries for the simultaneous determination of CI

Biodegradation characteristics and mechanism of citrinin by Rhodotorula mucilaginosa LWJJ06 from metabolomic and transcriptomic perspectives.

Liupao tea, a type of dark tea produced through pile fermentation, is susceptible to contamination by mycotoxin citrinin (CIT), posing significant health risks due to its nephrotoxic and hepatotoxic properties. In this study, Rhodotorula mucilaginosa LWJJ06, isolated from Liupao tea fermentation samples, completely degraded 10~ 100 µg/mL of CIT within 48 h under optimized conditions (28 °C, pH: 4.0, inoculum size: 1 × 107 CFU/mL). Metabolomic and transcriptomic analyses suggested that extensive metabolic adjustments accompanied CIT degradation, including apparent reallocation of energy resourc

The role of ER stress and ATP/AMPK in oxidative stress meditated hepatotoxicity induced by citrinin.

Citrinin, a secondary metabolite, can pose serious risks to the environment and organisms, but its hepatotoxic mechanisms are still unclear. Histopathological and ultrastructural results showed that citrinin-induced liver injury in Kunming mice, and the mechanism of citrinin-induced hepatotoxicity was studied in L02 cells. Firstly, citrinin mades L02 cell cycle arrest in G2/M phase by inhibition of cyclin B1, cyclin D1, cyclin-dependent kinases 2 (CDK2), and CDK4 expression. Secondly, citrinin inhibits proliferation and promotes apoptosis of L02 cells via disruption of mitochondria membrane po

Citrinin-Induced Hepatotoxicity in Mice Is Regulated by the Ca2+/Endoplasmic Reticulum Stress Signaling Pathway.

Citrinin (CTN) is a mycotoxin found in crops and agricultural products and poses a serious threat to human and animal health. The aim of this study is to investigate the hepatotoxicity of CTN in mice and analyze its mechanisms from Ca2+-dependent endoplasmic reticulum (ER) stress perspective. We showed that CTN induced histopathological damage, caused ultrastructural changes in liver cells, and induced abnormal values of biochemical laboratory tests of some liver functions in mice. Treatment with CTN could induce nitric oxide (NO), malondialdehyde (MDA), and reactive oxygen species (ROS) accum

Endoplasmic reticulum stress promotes oxidative stress: A novel mechanism of citrinin-induced thymus and spleen injury.

Citrinin (CTN) is a mycotoxin that is widespread and can contaminate a wide range of food products, posing a threat to human and animal health. The spleen and thymus are important immune organs of the body, and the damaging effects of CTN on immune organs and their mechanism are still unclear. In this study, we induced spleen and thymus injury in mice by exposure to different doses of CTN (0, 1.25, 5, or 20 mg/kg) and preliminarily investigated the damage mechanisms. It was observed that CTN exposure caused immune damage to the thymus and spleen, which are immune organs in mice. In addition, C

Kidney function of single comb white Leghorn pullets following acute renal portal infusion of the mycotoxin citrinin.

Poultry consuming diets contaminated with citrinin excrete copious quantities of urine and exhibit increased water consumption. The present study was conducted to determine if citrinin acts directly on the kidneys and, if so, to provide a detailed physiological evaluation of the nephrotoxic effects of citrinin. Single Comb White Leghorn pullets were anesthetized and prepared for renal function studies. Ureteral urine was collected during a pre-infusion period (30 min), during unilateral renal portal infusion of 200 ppm citrinin (30 min), and during a recovery period following citrinin infusion

Euphorbia hypericifolia Attenuates Citrinin-Induced Oxidative Stress and Maintains Tight Junction Integrity in Porcine Intestinal Epithelial Cells.

Citrinin (CTN), a mycotoxin commonly found in contaminated food and animal feed, impairs intestinal barrier integrity through oxidative stress and cytotoxicity. However, its link to ferroptosis, an iron-dependent form of regulated cell death, remains unclear. This study investigated whether CTN induces ferroptosis in intestinal epithelial cells and evaluated the protective role of Euphorbia hypericifolia (EH) against CTN-induced oxidative damage and tight junction (TJ) disruption. Using IPEC-J2 cells exposed to CTN, intracellular ferrous ion (Fe2+) levels, reactive oxygen species (ROS) accumul

Synergistic action of the nephrotoxic mycotoxins ochratoxin A and citrinin at nanomolar concentrations in human proximal tubule-derived cells.

Increased ochratoxin A (OTA) or citrinin (CIT) concentrations in food correlate with increased prevalence of tubule-interstitial nephropathy. We tested the hypothesis that co-exposure of human proximal tubule-derived epithelial cells (HK-2) to OTA and CIT promotes synergistic events indicative for inflammation, epithelial-to-mesenchymal-transition (EMT) or fibrosis. We measured markers of inflammation, EMT and fibrosis and investigated the role of MAP-kinases. Only concurrent but not individual exposure to OTA and CIT at nanomolar concentrations led to (i) an increase of TNF protein and mRNA,
