Genome-wide identification of lipid transfer proteins in Sorghum bicolor and discovery of flower-specific promoters.

Sorghum bicolor is a drought- and heat-tolerant cereal widely cultivated in arid regions. However, like other important grasses, it is susceptible to ergot disease caused by Claviceps species, which infect ovary tissues and reduce grain yield. To combat this disease, targeted biotechnological strategies are needed. In this study, we conducted a genome-wide identification and classification of lipid transfer proteins (LTPs) in S. bicolor, uncovering 68 putative genes grouped into five families. In silico analyses of gene expression and promoter sequences identified two LTP genes, SbLTP1.4 and S

[Antony's fire (gangrenous ergotism) and medieval iconography].

Ergotism was known as Holy Fire or St Antony's Fire in the Middle Ages, because of the burning sensations and limb gangrene it entailed. It was a frequent disorder, caused by eating rye flour contaminated by the fungus Claviceps purpurea. The Hospitable Order of St Antony was founded near Vienne in France, and counted 400 establishments in Europe by 1777. Ergotism is the subject of an abundant iconography, including statues and paintings. Woodcuts show the temptations of St Antony, with strange and diabolic scenes, and individuals with gangrenous limbs. Germanic woodcuts of the XVth century sh

Dopamine agonists for the treatment of pituitary tumours: From ergot extracts to next generation therapies.

Dopamine agonists are a key tool in the therapeutic arsenal of endocrinologists worldwide. They exert their effects by binding to dopamine-2 (D2) receptors expressed by pituitary tumour cells to modulate hormonal secretion and tumour size. They are the established first-line treatment for prolactinomas which express high levels of D2 receptors. Growing data support their use as an adjuvant treatment option for other pituitary tumours including growth hormone, adrenocorticotrophic hormones, thyroid hormone secreting adenomas and nonfunctional pituitary tumours, all of which have been shown to e

Human and cattle ergotism since 1900: symptoms, outbreaks, and regulations.

Ergotism in humans and cattle are caused by several species of Claviceps that infect rye and other cereal grains. Symptoms in humans vary greatly and are generally classified as convulsive, gangrenous, or gastrointestinal (enteric). Cattle are particularly susceptible to both gangrenous and hyperthermic ergotism (also called summer syndrome). The prevalence of ergotism has decreased as knowledge of the fungus has increased, mainly through implementation of regulations and advances in milling procedures. However, outbreaks in humans have recently occurred in lower socioeconomic populations of E

Monitoring and Managing of Ergot Alkaloids in Cereal Grains: A Comprehensive Analysis of Analytical Challenges, Sampling Uncertainty, and Processing for Food Safety.

Ergot alkaloids (EAs), toxic secondary metabolites produced by Claviceps purpurea, pose food and feed safety concerns for cereal grains, particularly rye and wheat. While EAs are most frequently associated with rye, their occurrence in other cereals has been increasingly reported across diverse regions. This review critically synthesizes current knowledge on the global occurrence, analytical detection, regulatory control, and processing behavior of EAs, with a particular focus on factors that limit data reliability and risk management. A defining challenge in EA monitoring is the highly hetero

Toxigenic Endophyte-Infected Tall Fescue and Ergot Alkaloids.

"Fescue toxicosis" and reproductive ergotism present identical toxidromes in late-gestational mares and, likely, other equids. Both toxic syndromes are caused by ergopeptine alkaloids (EPAs) of fungal origin, and they are collectively referred to as equine ergopeptine alkaloid toxicosis (EEPAT). EPAs are produced by either a toxigenic endophyte (Epichloë coenophiala) in tall fescue and/or a nonendophytic fungus (Claviceps purpurea), infecting small grains and grasses. EEPAT can cause hypoprolactinemia-induced agalactia/dysgalactia, prolonged gestation, dystocia, and other reproductive abnormal

Ergot alkaloids--biology and molecular biology.

EA have been a major benefit, and a major detriment, to humans since early in recorded history. Their medicinal properties have been used, and continue to be used, to aid in childbirth, with new uses being found in the treatment of neurological and cardiovascular disorders. The surprisingly broad range of pharmaceutical uses for EA stems from their affinities for multiple receptors for three distinct neurotransmitters (serotonin, dopamine, and adrenaline), from the great structural diversity of natural EA, and from the application of chemical techniques that further expand that structural dive

Effects of ergotamine on the central nervous system using untargeted metabolomics analysis in a mouse model.

The ergot alkaloid ergotamine is produced by Claviceps purpurea, a parasitic fungus that commonly infects crops and pastures of high agricultural and economic importance. In humans and livestock, symptoms of ergotism include necrosis and gangrene, high blood pressure, heart rate, thermoregulatory dysfunction and hallucinations. However, ergotamine is also used in pharmaceutical applications to treat migraines and stop post-partum hemorrhage. To define its effects, metabolomic profiling of the brain was undertaken to determine pathways perturbed by ergotamine treatment. Metabolomic profiling id

Natural compounds as potential adjuvants to cancer therapy: Preclinical evidence.

Traditional chemotherapy is being considered due to hindrances caused by systemic toxicity. Currently, the administration of multiple chemotherapeutic drugs with different biochemical/molecular targets, known as combination chemotherapy, has attained numerous benefits like efficacy enhancement and amelioration of adverse effects that has been broadly applied to various cancer types. Additionally, seeking natural-based alternatives with less toxicity has become more important. Experimental evidence suggests that herbal extracts such as Solanum nigrum and Claviceps purpurea and isolated herbal c

Wheat ergot fungus-derived and modified drug for inhibition of intracranial aneurysm rupture due to dysfunction of TLR-4 receptor in Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a form of dementia that strikes elderly people more frequently than it does younger people. The cognitive skills and memory of Alzheimer's sufferers continue to deteriorate over time. Recent studies have shown that patients with AD have greater amounts of inflammatory markers in their bodies, which suggests that inflammation occurs early on in the progression of the disease. There is a possibility that Aß oligomers and fibrils can be recognised by TLRs, in addition to the microglial receptors CD14, CD36, and CD47. When Aß binds to either CD36 or TLR4, it

[About the acting of the brotherhood of St. Anthony in Würzburg].

The brotherhood of St. Anthony, founded in the old French province of Dauphiné in 1095, was elevated to the rank of a monastic order in 1227, complying with the rules of the Augustinian Canons. In their infirmaries the monks took care of the persons attacked by ergot poisoning. If prayers and ointments were of no avail, in most cases the lower leg was amputated, so that the vital organs were not affected by gangrene. After bad harvests this ergotism became an epidemy, as rye-flour was consumed which had been contaminated by the fungus claviceps purpurea. It was as late as the 17th and 18th cen

Presumptive gangrenous ergotism in free-living moose and a roe deer.

Presumptive gangrenous ergotism in 10 moose (Alces alces) and one roe deer (Capreolus capreolus) is reported. Three of the moose came from a municipality in southeastern Norway where the disease occurred as a cluster in 1996. The other moose represented solitary or sporadic cases diagnosed in four municipalities in northwestern Norway between 1996 and 2004. Affected moose (seven calves, three yearlings) were found between October and June, showing distal limb lesions on one to three limbs. The lesions in the moose found during October and November presented as dry gangrene, whereas moose found

A NEW SPECIES OF BOTHRIOCEPHALUS (CESTODA: BOTHRIOCEPHALIDAE) FROM LEPOMIS SPP. (ACTINOPTERYGII: CENTRARCHIDAE) IN NORTH AMERICA.

Based on morphological and molecular data, a new species of tapeworm, Bothriocephalus kupermani n. sp., is described from pumpkinseed, Lepomis gibbosus (Linnaeus, 1758) (type host), and green sunfish, L. cyanellus Rafinesque, 1819 (Actinopterygii: Centrarchidae) in the United States. The new species differs from its North American congeners mainly in the shape of its scolex, which is almost rectangular in dorsoventral and lateral views (nearly parallel margins in the bothrial region), with shallow, wide bothria, and by an extensive field of vitelline follicles that are also variably present, a

[Toxicologic risk assessment and prevention: rational and irrational approaches].

Worldwide, the management and evaluation of risks caused by chemical compounds are handled by the aid of threshold concentrations below of which one can be sure that no biological effect whatsoever can be observed. At the working place, we use the maximally tolerated concentration of the chemicals (MAK) and, in addition, the biologically tolerated concentrations (BTC) of the compounds either in blood or in other body fluids to which a male and/or female worker is exposed. These threshold concentrations should cover any toxic effect including on the one hand mere deviations of clinical chemical

Ergot Alkaloids of the Family Clavicipitaceae.

Ergot alkaloids are highly diverse in structure, exhibit diverse effects on animals, and are produced by diverse fungi in the phylum Ascomycota, including pathogens and mutualistic symbionts of plants. These mycotoxins are best known from the fungal family Clavicipitaceae and are named for the ergot fungi that, through millennia, have contaminated grains and caused mass poisonings, with effects ranging from dry gangrene to convulsions and death. However, they are also useful sources of pharmaceuticals for a variety of medical purposes. More than a half-century of research has brought us extens

Several teenaged girls that were struck by delirious fits and seizures captured the center of the stage. The local physicians, who could not come up with a medical solution, explained the strange phenomenon as witchcraft. The new world's moral code was brought to trial along with 20 innocent men who were accused, convicted and executed for the crime of practicing witchcraft. Mycotoxins in food: Occurrence, health implications, and control strategies-A comprehensive review.

Mycotoxins are secondary metabolites produced by various filamentous fungi, including Aspergillus, Fusarium, Penicillium, Alternaria, Claviceps, Mucor, Trichoderma, Trichothecium, Myrothecium, Pyrenophora, and Stachybotrys. They can contaminate various plants or animal foods, resulting in a significant loss of nutritional and commercial value. Several factors contribute to mycotoxin production, such as humidity, temperature, oxygen levels, fungal species, and substrate. When contaminated food is consumed by animals and humans, mycotoxins are rapidly absorbed, affecting the liver, and causing m
