Generation of Infectious Prions Amenable to Site-specific Click Chemistry.

Prion diseases are a group of fatal neurodegenerative diseases that proceed through the templated conversion of the normal PrP C protein to a self-propagating and infectious form, termed PrP Sc . This conversion process is central to disease progression. However, due to difficulties in producing functional PrP Sc molecules that can be selectively modified with chemical probes, many aspects of PrP Sc biology cannot be directly studied. To overcome this limitation, we substituted p-azido-L-phenylalanine (AzF), a small click chemistry-reactive amino acid, for tryptophan residue 99 of PrP C . W99A

Development of Tetrameric N-Annulated Perylene Diimides Using "Click" Chemistry.

Herein, we report the design, synthesis, and characterization of two novel N-annulated perylene diimide (NPDI) tetramer arrays that were developed using copper catalyzed alkyne-azide cycloaddition. Despite the optoelectronic properties of both tetramers being nearly identical, the two tetramers exhibited very different molecular geometries. The twisted spirobifluorene NPDI tetramer (sbfNPDI4 ) was found to have an extended and flexible geometry, while the planar pyrene NPDI tetramer (pyrNPDI4 ) exhibited a highly congested and conformationally locked geometry. Morphology-Preserving Sensitization of ZnO Nanorod Surfaces via Click-Chemistry.

Films of ZnO nanorods grown by chemical vapor deposition were functionalized with a chromophore in a stepwise process that preserves the surface morphology. In the first step, the ZnO nanorods were functionalized by exposure to prop-2-ynoic acid (propiolic acid) in vacuum, which did bind through the COOH group leading to a ZnO surface functionalized with ethyne moieties (ethyne/ZnO). In the second step, 9-(4-azidophenyl)-2,5-di-tert-butylperylene (DTBPe-Ph-N3) was reacted with the ethyne/ZnO surface via copper-catalyzed azide-alkyne click reaction (CuAAC) in solution to form the DTBPe-function

Shotgun Analysis of Diacylglycerols Enabled by Thiol-ene Click Chemistry.

Diacylglycerols (DAGs) are a subclass of neutral lipids actively involved in cell signaling and metabolism. Alteration in DAG metabolism has been associated with onset and progression of several human-related diseases. The structural diversity of DAGs and their low concentrations in biological samples call for the development of methods that are capable of sensitive identification and quantitation of each DAG species as well as rapid profiling when a biochemical pathway is perturbed. In this work, the thiol-ene click chemistry has been employed to introduce a charge-tag, namely, cysteamine (CA

Protocol for producing phosphoramidate using phosphorus fluoride exchange click chemistry.

Phosphorus fluoride exchange (PFEx) is a catalytic click reaction that involves exchanging high oxidation state P-F bonds with alcohol and amine nucleophiles, reliably yielding P-O- and P-N-linked compounds. Here, we describe steps for preparing a phosphoramidic difluoride and performing two sequential PFEx reactions to yield a phosphoramidate through careful catalyst selection. We then detail procedures for handling and quenching potentially toxic P-F-containing compounds to ensure user safety when conducting PFEx reactions. For complete details on the use and execution of this protocol, plea

Click chemistry inspired highly facile synthesis of triazolyl ethisterone glycoconjugates.

Numerous deoxy-azido sugars 3 were prepared by the reaction of tosyl/bromo sugars with NaN3 in dry DMF under heating condition. The 1,3-dipolar cycloaddition of deoxy-azido sugars 3 with ethisterone 4 to afford regioselective triazole-linked ethisterone glycoconjugates 5 was investigated in the presence of CuI and DIPEA in dichloromethane or CuSO4·5H2O and sodium ascorbate in aqueous medium. All the developed compounds were characterized by spectroscopic analysis (IR, (1)H &(13)C NMR, and MS spectra). Structure of triazolyl ethisterone glycoconjugate 5a has been further confirmed by its Single

Synthesis of water-soluble camptothecin-polyoxetane conjugates via click chemistry.

Water-soluble camptothecin (CPT)-polyoxetane conjugates were synthesized using a clickable polymeric platform P(EAMO) that was made by polymerization of acetylene-functionalized 3-ethyl-3-(hydroxymethyl)oxetane (i.e., EAMO). CPT was first modified with a linker 6-azidohexanoic acid via an ester linkage to yield CPT-azide. CPT-azide was then click coupled to P(EAMO) in dichloromethane using bromotris(triphenylphosphine)copper(I)/N,N-diisopropylethylamine. For water solubility and cytocompatibility improvement, methoxypolyethylene glycol azide (mPEG-azide) was synthesized from mPEG 750 g mol(-1)

Rapid synthesis of Abelson tyrosine kinase inhibitors using click chemistry.

Protein kinases catalyze the phosphorylation of serine, threonine, tyrosine and histidine residues in proteins. Aberrant regulation of kinase activity has been implicated in many diseases including cancer. Thus development of new strategies for kinase inhibitor design remains an active area of research with direct relevance to drug development. Abelson (Abl) tyrosine kinase is one of the Src-family of tyrosine kinases and is directly implicated in Chronic Myelogenous Leukemia (CML). In this article, we have, for the first time, developed an efficient method for the construction of small molecu

RNA Labeling of Nucleolar Transcripts by 5-Ethynyl Uridine Click Chemistry.

The nucleolus is a dynamic membraneless organelle where ribosomal RNA (rRNA) transcription, processing, and ribonucleoprotein assembly occur. Visualizing nascent RNA within nucleolar condensates reveals how RNA metabolism and phase separation reorganize under stress. This chapter provides a practical protocol to label nascent RNA using 5-ethynyl uridine (5-EU), followed by copper-catalyzed azide-alkyne cycloaddition for fluorescent detection. The method is straightforward, works across mammalian cells, and robustly reports nucleolar transcription under basal and stress conditions (heat shock;

Click-chemistry-mediated modulation of CRISPR-Cas12a activity through activator modification.

Chemical modification strategies offer a promising route for spatiotemporal regulation of CRISPR-Cas12a activity in molecular diagnostics. However, existing methods involve CRISPR RNA with photolabile groups that suffer from complexity and RNA instability. To address these limitations, we report a simple and robust strategy using dibenzocyclooctyne (DBCO)-mediated click chemistry to modulate CRISPR-Cas12a activity. The copper-free strain-promoted azide-alkyne cycloaddition reaction enables CRISPR-Cas12a modulation with low toxicity, biocompatibility, and high selectivity. Utilizing azide-modif

Click Chemistry Enables [89Zr]Zr-DOTA Radioimmunoconjugation for Theranostic 89Zr-immunoPET.

There have been predictions that the use of the macrocyclic chelating agent 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) in zirconium-89 (89Zr) immuno-positron emission tomography (89Zr-immunoPET) could enhance the in vivo stability of 89Zr radioimmunoconjugates. However, conjugating [89Zr]Zr-DOTA to a monoclonal antibody (mAb) remains a challenge as the heat treatment required for [89Zr]Zr-DOTA chelation can lead to thermal denaturation of the mAb moieties. We developed a method for synthesizing [89Zr]Zr-DOTA-mAb based on a tetrazine (Tz)-conjugated bifunctional DOTA deriva

Modular click chemistry libraries for functional screens using a diazotizing reagent.

Click chemistry is a concept in which modular synthesis is used to rapidly find new molecules with desirable properties1. Copper(I)-catalysed azide-alkyne cycloaddition (CuAAC) triazole annulation and sulfur(VI) fluoride exchange (SuFEx) catalysis are widely regarded as click reactions2-4, providing rapid access to their products in yields approaching 100% while being largely orthogonal to other reactions. However, in the case of CuAAC reactions, the availability of azide reagents is limited owing to their potential toxicity and the risk of explosion involved in their preparation. Here we repo

In situ click chemistry: probing the binding landscapes of biological molecules.

Combinatorial approaches to the discovery of new functional molecules are well established among chemists and biologists, inspired in large measure by the modular composition of many systems and molecules in Nature. Many approaches rely on the synthesis and testing of individual members of a candidate combinatorial library, but attention has also been paid to techniques that allow the target to self-assemble its own binding agents. These fragment-based methods, grouped under the general heading of target-guided synthesis (TGS), show great promise in lead discovery applications. In this tutoria
