Genome Sequence Analysis of Clostridium chauvoei Strains of European Origin and Evaluation of Typing Options for Outbreak Investigations.

Black quarter caused by Clostridium (C.) chauvoei is an important bacterial disease that affects cattle and sheep with high mortality. A comparative genomics analysis of 64 C. chauvoei strains, most of European origin and a few of non-European and unknown origin, was performed. The pangenome analysis showed limited new gene acquisition for the species. The accessory genome involved prophages and genomic islands, with variations in gene composition observed in a few strains. This limited accessory genome may indicate that the species replicates only in the host or that an active CRISPR/Cas syst

Epidemiology of Clostridium chauvoei infections in southern Chile: Molecular prevalence, diagnostic gaps, and genetic characterization of cctA and nanA virulence genes.

BACKGROUND: Blackleg, a mostly cattle disease caused by Clostridium chauvoei, represents a health challenge for the livestock industry. This study aimed to characterize the epidemiology of C. chauvoei infections in Southern Chile (Los Ríos and Los Lagos regions), where occasional outbreaks persist despite high perceived vaccination coverage. METHODS: A cross-sectional study was conducted on 100 cattle farms (50 beef and 50 dairy), combining a structured survey on management practices with molecular detection (PCR) of C. chauvoei in soil and fecal samples. Additionally, the virulence genes cct…

Genome characterization of a Clostridium chauvoei strain 23CCJK from India and comparative analysis revealing novel insight into species lineages, associated SNPs and virulence factors.

Clostridium chauvoei is a spore-forming bacterium responsible for black quarter (BQ) in cattle, a disease leading to high mortality. Despite its economic significance in India, genomic characterization of C. chauvoei strains from the country remains scarce. This study reports the genome of a C. chauvoei strain, 23CCJK, associated with a black quarter outbreak in Leh, India. The genome, approximately 2.7 Mb, encodes 2557 protein-coding genes including important known virulence determinants such as CctA, NanA, hyaluronidase, and collagenase. Species pan-genome analysis involving 85 global strain…

Blackleg: A deadly disease with a hidden cause.

Blackleg is an infectious disease that mainly affects cattle and rarely affects other ruminants. It is characterized by hemorrhagic blackleg myositis. Clostridium chauvoei is a highly pathogenic anaerobic, endospore-forming Gram- positive bacteria that causes blackleg disease. Blackleg disease was first reported in 1870, but the causative bacterium C. chauvoei was not described until 1887. Clostridium chauvoei is found in grassland, fresh water, silage, soil, and the excrement of healthy animals. Cattle intestines and pasture soil have both been found to contain C. chauvoei spores, suggesting …

Occurrence of blackleg and current preventive measures against outbreaks in the Canton of Bern.

Blackleg is an acute, lethal, infectious disease caused by Clostridium chauvoei. It affects mainly cattle and is characterized by severe necrohemorrhagic myositis and myocarditis. While the disease has caused substantial economic losses in the past, the number of diseased cattle has been decreasing in the last 30 years. Subsequently, the status of blackleg has been officially changed in Switzerland and the disease is no longer reportable since 2022. Considering the potential persistence of C. chauvoei spores in the environment, blackleg may however still be prevalent in certain areas of the Ca…

Pathology of blackleg in cattle in California, 1991-2015.

Blackleg is an infectious disease of cattle and rarely other ruminants, produced by Clostridium chauvoei and characterized by necrotizing myositis. In most cases of blackleg, the large muscles of the pectoral and pelvic girdles are affected, with other skeletal muscles and the heart involved less frequently. We studied 29 blackleg cases selected from the archives of the California Animal Health and Food Safety Laboratory, 1991-2015. Immunohistochemistry was also evaluated to detect C. chauvoei in formalin-fixed, paraffin-embedded (FFPE) tissues of cattle. Nineteen animals had gross and/or micr…

Myonecrosis by Clostridium septicum in a dog, diagnosed by a new multiplex-PCR.

Clostridial myositis is an acute, generally fatal toxemia that is considered to be rare in pet animals. The present report describes an unusual canine clostridial myositis that was diagnosed by a new multiplex-PCR (mPCR) designed for simultaneous identification of Clostridium sordellii, Clostridium septicum, Clostridium perfringens type A, Clostridium chauvoei, and Clostridium novyi type A. A ten-month-old male Rottweiler dog, that had displayed lameness and swelling of the left limb for 12 h, was admitted to a veterinary hospital. The animal was weak, dyspneic and hyperthermic, and a clinical…

Blackleg without skeletal muscle involvement as a cause of sudden death in unvaccinated calves.

Nine, 5-month-old unvaccinated black Angus calves died in less than 48 hours of each other. The calves expressed minimal or no clinical signs prior to death. On-farm post-mortem examinations were carried out on 3 of the deceased calves, and findings included: necrotizing and fibrinosuppurative myositis of the diaphragm muscle, and fibrinosuppurative pleuritis and epicarditis. Fluorescent antibody testing for pathogenic clostridia identified Clostridium chauvoei and a diagnosis of blackleg was made. The remainder of the herd were then vaccinated.

Bacillary haemoglobinuria diagnosis by the peroxidase-antiperoxidase (PAP) technique.

A peroxidase-antiperoxidase (PAP) technique was used to diagnose bacillary haemoglobinuria in formalin-fixed, paraffin-embedded liver tissues of cattle. The PAP method revealed Clostridium haemolyticum in the zone of liver necrosis characteristic of the disease and also in culture smears of this microorganism, but C. novyi type B, C. chauvoei, C. septicum and C. perfringens types B and C remained unstained by the PAP reaction. The PAP technique performed provides a specific, simple and rapid method to diagnose bacillary haemoglobinuria.

Development and evaluation of indirect enzyme-linked immunosorbent assays for the determination of immune response to multiple clostridial antigens in vaccinated captive bred southern white rhinoceros (Ceratotherium simum simum).

BACKGROUND: An overall increase in poaching of white rhinoceros results in captive breeding becoming a significant component of white rhinoceros conservation. However, this type of conservation comes with its own difficulties. When wildlife is captured, transported and/or confined to a boma environment, they are more predisposed to diseases caused by bacterial organisms such as spore forming Clostridium spp. A southern white rhinoceros (Ceratotherium simum simum) population on a captive bred farm was suspected to be affected by Clostridium infections. These endangered animals were apparently e

Rapid identification and differentiation of pathogenic clostridia in gas gangrene by polymerase chain reaction based on the 16S-23S rDNA spacer region.

In cattle, sheep, and other ruminants, clostridial myonecrosis (gas gangrene) is mostly caused by Clostridium chauvoei, C septicum, C novyi and C sordellii. A polymerase chain reaction (PCR) system using common primers designed from multiple alignment of the 16S rRNA and 23S rRNA genes of Clostridium species was developed to identify pathogenic clostridia. The PCR was performed with total DNA from 26 strains which included seven different Clostridia species. These bacteria were differentiated at species level by the different PCR product patterns. To characterise the 16S-23S rDNA spacer region…

Clostridial Infections in Cattle: A Comprehensive Review with Emphasis on Current Data Gaps in Brazil.

Clostridial infections in cattle are a significant concern for Brazilian livestock. These diseases are caused by various species of Clostridium, which are known for their ability to produce potent toxins. Botulism in cattle is a serious and often fatal condition caused by the ingestion of neurotoxins produced by C. botulinum. This bacterium thrives in decomposing organic matter, such as spoiled feed, carcasses, and contaminated water. Tetanus is less common, but it is a serious disease that follows the contamination of wounds with Clostridium tetani spores. It results in muscle stiffness, spas

Gas gangrene in mammals: a review.

Gas gangrene is a necrotizing infection of subcutaneous tissue and muscle that affects mainly ruminants and horses, but also other domestic and wild mammals. Clostridium chauvoei, C. septicum, C. novyi type A, C. perfringens type A, and C. sordellii are the etiologic agents of this disease, acting singly or in combination. Although a presumptive diagnosis of gas gangrene can be established based on clinical history, clinical signs, and gross and microscopic changes, identification of the clostridia involved is required for confirmatory diagnosis. Gross and microscopic lesions are, however, hig…
