Irradiance level and elevation shape the soil microbiome communities of Coffea arabica L.

BACKGROUND: The nexus plant-microbe-environment is essential to understand the ecosystem processes shaping plant health and fitness. Within this triangle, soils and associated microflora are among the key ecosystem's drivers, underpinning plant productivity and evolution. In this study, we conducted a comprehensive analysis (physicochemical properties, enzyme activities, and taxonomic diversity) of soils under the canopy projection of Coffea arabica trees along a gradient of elevation (600, 800, and 900 m) and shade (0, 50, 100%). RESULTS: While shade had no influence on most parameters, alti

In Vitro Conversion of Coffea spp. Somatic Embryos in SETIS™ Bioreactor System.

Somatic embryogenesis (SE) is an excellent example of mass plant propagation. Due to its genetic variability and low somaclonal variation, coffee SE has become a model for in vitro propagation of woody species, as well as for large-scale production of vigorous plants that are advantageous to modern agriculture. The success of the large-scale propagation of an embryogenic system is dependent on the development, optimization, and transfer of complementary system technologies. In this study, two successful SE systems were combined with a SETIS™ bioreactor immersion system to develop an efficient

Potassium Phosphite Activates Components Associated with Constitutive Defense Responses in Coffea arabica Cultivars.

Phosphites have been used as inducers of resistance, activating the defense of plants and increasing its ability to respond to the invasion of the pathogen. However, the mode of action of phosphites in defense responses has not yet been fully elucidated. The objective of this study was to evaluate the effect of potassium phosphite (KPhi) in coffee cultivars with different levels of resistance to rust to clarify the mechanism by which KPhi activates the constitutive defense of plants. To this end, we studied the expression of genes and the activity of enzymes involved in the defense pathway of

NMR Identification and Quantification of Galactinol in Green Coffea arabica: Implications for Traceability and Adulteration.

Among coffee low-molecular-weight carbohydrates, galactinol plays an important role in coffee physiology and quality, having received attention also for its potential use as a marker of geographical origin. Nevertheless, data on the occurrence of this sugar in green coffee beans remain limited. In this study, galactinol was isolated for the first time from green Coffea arabica beans and unambiguously identified by high‑field NMR spectroscopy. A quantitative HPLC-RID screening of 100 green Arabica coffee samples from 16 geographical origins showed that galactinol was detected in 31% of the samp

Metabolic Pathway Reconstruction Indicates the Presence of Important Medicinal Compounds in Coffea Such as L-DOPA.

The use of transcriptomic data to make inferences about plant metabolomes is a useful tool to help the discovery of important compounds in the available biodiversity. To unveil previously undiscovered metabolites of Coffea, of phytotherapeutic and economic value, we employed 24 RNAseq libraries. These libraries were sequenced from leaves exposed to a diverse range of environmental conditions. Subsequently, the data were meticulously processed to create models of putative metabolic networks, which shed light on the production of potential natural compounds of significant interest. Then, we sele

Rhizosphere and fruit microbiome variation across Coffea canephora farms representing conventional, transitional, and organic management.

Robusta coffee (Coffea canephora Pierre ex A. Froehner) is a major global cash crop, and an increasing number of coffee farms are transitioning from conventional to organic management. To investigate how coffee-associated microbiomes vary across different management histories, bacterial communities from the rhizosphere and fruits of three farms representing conventional, transitional organic, and organic management were profiled using 16S rRNA gene amplicon sequencing, differential abundance analysis, and functional prediction. Differential abundance analysis of rhizosphere and fruit bacterial

Genome-wide subgenome-resolved analysis validates chromosome 4 differentiation and prioritizes introgressed Coffea arabica accessions.

Chromosome 4 introgression in Timor hybrid-derived Coffea arabica is established, but the robustness of accession prioritization and the relative strength of cultivated-introgressed differentiation across the canephora-derived (sgC) and eugenioides-derived (sgE) subgenomes remained unclear under explicit subgenome filtering. We reanalyzed public genomic resources from 44 coffee accessions using strict contig-level subgenome filtering, Arabica-only population-structure analysis, SNP-panel sensitivity testing, genome-wide differentiation scans, permutation testing, and direct sequence alignment.

Evaluation of the antidepressant potential of Duloxetine, Coffea canephora, and Nigella sativa in a rat depression model.

Depression is a complex disorder involving neurotransmitter imbalance, oxidative stress, and inflammation of the brain tissue. Traditional antidepressants like duloxetine (Cymbalta®) can produce side effects. Natural extracts like Coffea canephora (green coffee) and Nigella sativa (black seeds) have emerged as possible alternative therapies. The present study aims to evaluate the antidepressant efficacy of Cymbalta, green coffee, and black seeds in a reserpine-induced rat depression model. Thirty adult male albino rats were divided into five groups: (1) Control group, (2) Depression-induced gr

In vitro and in silico studies of alpha glucosidase inhibition and antifungal activity of coffea canephora husk.

The coffea canephora husk, a protected agricultural crop, is abundant in Vietnam. Examining the effects of C. canephora husk compounds on α-glucosidase and antifungal drug activity was the primary objective of this research. A cholestane-type steroid, coffeacanol A (1), was extracted from the ethyl acetate extract. Three cholestane-type derivatives (2-4) and three additional known compounds (5-7) were separated, and we used a variety of chromatographic techniques to identify a total of six substances. We used NMR to determine the chemical structures of these substances. Extensive HR-MS-ESI ana

Genetic diversity and population structure of Coffea arabica in Saudi Arabia revealed by whole-genome resequencing.

Gene banks are essential for conserving plant genetic resources, yet limited genomic characterization constrains their effective use in breeding. Coffea arabica L., an economically important crop in Saudi Arabia, remains poorly characterized at the genome-wide level. We performed whole-genome resequencing of 78 Saudi C. arabica accessions conserved at the Ministry of Environment, Water and Agriculture (MEWA) Seed Center and PGR Bank, generating 86 sequencing libraries, including eight technical replicates. Reads were aligned to the Cara_r6 reference genome, yielding 468, 133 variants (412, 043

Identification of sources of male sterility in the Colombian Coffee Collection for the genetic improvement of Coffea arabica L.

In coffee (Coffea arabica L.), male sterility is a prerequisite for the exploitation of heterosis since it provides an efficient and reliable method for the production of hybrid seeds. Given its relevance, the objective of this study was to identify male-sterile genotypes within the Colombian Coffee Collection that can be used in genetic improvement. For this purpose, Ethiopian germplasm and progenies derived from hybrids between C. arabica x C. canephora were explored between 2017 and 2021. In the first stage, genotypes without visual presence of pollen were preselected in the field, followed

Coffea arabica Extracts and Metabolites with Potential Inhibitory Activity of the Major Enzymes in Bothrops asper Venom.

Background/Objectives: Most snakebite incidents in Latin America are caused by species of the Bothrops genus. Their venom induces severe local effects, against which antivenom therapy has limited efficacy. Metabolites derived from Coffea arabica have demonstrated anti-inflammatory and anticoagulant properties, suggesting their potential as therapeutic agents to inhibit the local effects induced by B. asper venom. Methods: Three enzymatic assays were performed: inhibition of the procoagulant and amidolytic activities of snake venom serine proteinases (SVSPs); inhibition of the proteolytic activ

Stabilization of Anthocyanins from Coffee (Coffea arabica L.) Husks and In Vivo Evaluation of Their Antioxidant Activity.

Coffee (Coffea arabica L.) is one of the most popular and widely consumed products throughout the world, mainly due to its taste, aroma, caffeine content, and natural antioxidants. Among those antioxidants, anthocyanins are one of the most important natural pigments, which can be found in coffee husks. It is widely known that anthocyanins have multiple health benefits partially linked to their antioxidant properties. However, anthocyanins have low stability and are sensitive to all types of changes. In order to prevent its degradation, anthocyanins can be stabilized with nanoparticles. Thus, t

Promoter analysis of the WRKY transcription factors CaWRKY1a and CaWRKY1b homoeologous genes in coffee (Coffea arabica).

The regulation of the CaWRKY1 homoeologous genes were analyzed through the characterization of their promoters. The pW1a promoter is proposed as a new tool for coffee plant biotechnologies. WRKY transcription factors are important elements of the plant immune response. The CaWRKY1 gene from Coffea arabica is induced by several biotic and abiotic stresses, including challenge by the rust fungus Hemileia vastatrix. Two homoeologous CaWRKY1 genes, named CaWRKY1a and CaWRKY1b, were previously identified in the C. arabica allotetraploid genome. To gain insight into the transcriptional regulation of

Effect of phosphate on aluminium-inhibited growth and signal transduction pathways in Coffea arabica suspension cells.

In acid soils, aluminium (Al) toxicity and phosphate (Pi) deficiency are the most significant constraints on plant growth. Al inhibits cell growth and disrupts signal transduction processes, thus interfering with metabolism of phospholipase C (PLC), an enzyme involved in second messenger production in the cell. Using a Coffea arabica suspension cell model, we demonstrate that cell growth inhibition by Al toxicity is mitigated at a high Pi concentration. Aluminium-induced cell growth inhibition may be due to culture medium Pi deficiency, since Pi forms complexes with Al, reducing Pi availabilit

Genetic diversity of cultivated Gayo Arabica Coffee (Coffea arabica L.) based on morphological and microsatellite markers.

BACKGROUND: Indonesia is the fourth largest global producer of coffee (Coffea spp.). The primary production centre for Coffea arabica in the country is the Gayo Highlands of Aceh. The Gayo Highlands in Aceh serve as a key arabica coffee-producing region, covering over 100,000 hectares across three districts: Aceh Tengah, Bener Meriah, and Gayo Lues, situated at elevations ranging from 900 to 2,000 m above sea level (asl). Despite the economic significance of Gayo Arabica coffee, studies on its genetic diversity remain limited. The objective of this study was to evaluate the morphological chara
