Computational memory capacity predicts aging and cognitive decline.

Memory is a crucial cognitive function that deteriorates with age. However, this ability is normally assessed using cognitive tests instead of the architecture of brain networks. Here, we use reservoir computing, a recurrent neural network computing paradigm, to assess the linear memory capacities of neural-network reservoirs extracted from brain anatomical connectivity data in a lifespan cohort of 636 individuals. The computational memory capacity emerges as a robust marker of aging, being associated with resting-state functional activity, white matter integrity, locus coeruleus signal intens

Multidimensional relationships between sensory perception and cognitive aging.

A growing literature suggests that declines in sensory/perceptual systems predate cognitive declines in aging, and furthermore, they are highly predictive for developing Alzheimer's disease and Alzheimer's related dementias (ADRD). While vision, hearing, olfaction, and vestibular function have each been shown to be related to ADRD, their causal relations to cognitive declines, how they interact with each other remains to be clarified. Currently, there is substantial debate whether sensory/perceptual systems that fail early in disease progression are causal in their contributions to cognitive l

Hypothalamic Menin regulates systemic aging and cognitive decline.

Aging is a systemic process, which is a risk factor for impaired physiological functions, and finally death. The molecular mechanisms driving aging process and the associated cognitive decline are not fully understood. The hypothalamus acts as the arbiter that orchestrates systemic aging through neuroinflammatory signaling. Our recent findings revealed that Menin plays important roles in neuroinflammation and brain development. Here, we found that the hypothalamic Menin signaling diminished in aged mice, which correlates with systemic aging and cognitive deficits. Restoring Menin expression in

Temporal Dynamics of Motor Functioning and Cognitive Aging.

BACKGROUND: Because of the possible implications for intervention and thus successful aging, researchers have striven to determine whether the age changes in physical and cognitive functioning are coincident or does functioning in one domain change before, and possibly contribute to, functioning in the other. METHODS: Bivariate dual change score models were applied to four cognitive factors and three motor functioning factors available from 813 adults who participated in the Swedish Adoption/Twin Study of Aging. Participants were aged 50-88 at the first of six waves of testing covering a 19-y

Gonadal hormones and cognitive aging: a midlife perspective.

Gonadal steroids affect a variety of brain processes. Cognitive consequences of hormonal changes associated with menopause are of scientific interest and of relevance to public health. Natural menopause is a normal physiological process that can only be directly studied through observational research. Similarly, surgical menopause in humans is rarely directly amenable to experimental research. Causality with respect to cognitive outcomes is, therefore, difficult to infer. Cross-sectional and longitudinal findings from the Melbourne Women's Midlife Health Project, the Study of Women's Health Ac

Cognitive and functional changes with aging in schizophrenia.

The variation in functional outcome in schizophrenia appears to be exaggerated in late life. The cognitive and functional deficits commonly seen in younger schizophrenic patients appear to worsen in some cases in late life, while others patients appear to have a stable course of illness without functional decline, and still other patients have been reported to have essentially no residual symptoms in their later years. Cognitive and functional deficits appear to worsen more significantly in patients with a lifetime course of severe functional deficit. Despite the profound functional and cognit

Personality and cognitive resilience to accelerated epigenetic aging.

Personality traits are associated with cognitive resilience to dementia-related neuropathology. This study examines whether personality traits are related to cognitive resilience to accelerated epigenetic aging. Participants were adults aged from 50 to 98 years (N = 2926, 58% female, Mean age = 68.72, SD = 9.57) from the Health and Retirement Study (HRS). Data on cognition and epigenetic aging measures (GrimAge and DunedinPoAM38) were obtained in 2016. Data on personality, demographic factors, and clinical, behavioral, and psychological covariates were obtained in 2014/2016. Cognitive resilien

[Mild cognitive impairment - dementia precursor or normal aging?].

BACKGROUND: Mild cognitive change is common with increasing age and may generate concern. Two stages of mild cognitive changes have been defined; subjective cognitive impairment and mild cognitive impairment (MCI), which is confirmed through cognitive testing. Individuals referred to the Landspítali memory clinic and diagnosed with mild cognitive impairment were invited to participate in the study. The aim of the study was to evaluate the risk of individuals with MCI having a disorder leading to dementia. MATERIAL AND METHODS: 214 individuals diagnosed with mild cognitive impairment from 2014

Early Intervention in Cognitive Aging with Strawberry Supplementation.

Late-life dementia is a growing public health concern lacking effective treatment. Neurodegenerative disorders such as Alzheimer's disease (AD) develop over a preclinical period of many years beginning in midlife. The prevalence of insulin resistance, a prominent risk factor for late-life dementia, also accelerates in middle-age. Consumption of berry fruits, including strawberries, has been shown to influence metabolism as well as cognitive performance suggesting potential to mitigate risk for dementia. In this controlled trial, we enrolled overweight middle-aged men and women with insulin res

Cognitive Aging and Dementia: A Life Span Perspective.

This article summarizes empirical findings and theoretical concepts in cognitive aging and late-life dementia research. Major emphases are placed on (1) person-to-person heterogeneity in trajectories of cognitive change over time, (2) how trajectories of child cognitive development determine peak levels of adult cognitive function from which aging-related cognitive declines occur, and (3) how lifelong trajectories of cognitive function relate to the timing of severe cognitive impairments characteristic of dementia. I consider conceptual issues surrounding categorical vs. dimensional models of

Aging Affects Dopaminergic Neural Mechanisms of Cognitive Flexibility.

UNLABELLED: Aging is accompanied by profound changes in the brain's dopamine system that affect cognitive function. Evidence of powerful individual differences in cognitive aging has sharpened focus on identifying biological factors underlying relative preservation versus vulnerability to decline. Dopamine represents a key target in these efforts. Alterations of dopamine receptors and dopamine synthesis are seen in aging, with receptors generally showing reduction and synthesis demonstrating increases. Using the PET tracer 6-[18F]fluoro-l-m-tyrosine, we found strong support for upregulated str

Cognitive Trajectories and Alzheimer Disease Biomarkers: From Successful Cognitive Aging to Clinical Impairment.

OBJECTIVE: Cross-sectional definitions of successful cognitive aging have been widely utilized, but longitudinal measurements can identify people who do not decline. We performed this study to contrast maintenance with declining trajectories, including clinical conversion. METHODS: We included baseline cognitively unimpaired Alzheimer's Disease Neuroimaging Initiative participants with 3 or more cognitive testing sessions (n = 539, follow-up 6.1 ± 3.5 years) and calculated slopes of an episodic memory composite (MEM) to classify them into two groups: maintainers (slope ≥ 0) and decliners (slo

Effects of gait and cognitive task difficulty on cognitive-motor interference in aging.

Although gait-related dual-task interference in aging is well established, the effect of gait and cognitive task difficulty on dual-task interference is poorly understood. The purpose of this study was to examine the effect of gait and cognitive task difficulty on cognitive-motor interference in aging. Fifteen older adults (72.1 years, SD 5.2) and 20 young adults (21.7 years, SD 1.6) performed three walking tasks of varying difficulty (self-selected speed, fast speed, and fast speed with obstacle crossing) under single- and dual-task conditions. The cognitive tasks were the auditory Stroop tas

Cognitive vulnerability and social support among aging women caregivers.

This 15-year longitudinal study examined the association between family caregiving and cognitive function among middle-aged and older adults in South Korea, focusing on gender differences and the moderating role of social support. Using data from 6,481 participants in the Korean Longitudinal Study of Aging (2006-2020), cognitive function was assessed with the Korean Mini-Mental State Examination (K-MMSE). Family caregiving experience was defined as reporting caregiving in at least two survey waves between 2006 and 2020. Repeated caregiving exposure was associated with lower cognitive performan

Differences in Cognitive Aging Assessments Associated With Retest Effects.

IMPORTANCE: Retest effects can inflate longitudinal cognitive test performance and complicate estimation of cognitive change. Whether retest effects influence estimated intervention effects in randomized clinical trials remains unclear. OBJECTIVE: To quantify the magnitude and heterogeneity of differences associated with retesting in longitudinal cognitive testing and examine whether accounting for retesting alters estimated treatment effects in the Aging and Cognitive Health Evaluation in Elders (ACHIEVE) trial. Aging-related changes in cognitive flexibility: fMRI meta-analysis.

Cognitive flexibility-the ability to adaptively shift between different mental processes-is essential for human functioning. This meta-analysis examines age-related changes in neural correlates of cognitive flexibility using two common assessments: the Wisconsin Card Sorting Test (rule-discovery) and Task-Switching Paradigm (rule-retrieval). We synthesized findings from 85 articles comprising 118 experiments with 2246 participants across young, middle-age, and older adult groups. Activation Likelihood Estimation analyses revealed an age-related decrease in neural involvement, particularly in p

Psychosocial Protective Factors in Cognitive Aging: A Targeted Review.

OBJECTIVE: The lack of disease-modifying pharmacological agents for dementia highlights the critical importance of prevention, but known modifiable factors (e.g., education, physical health and health behaviors, depression, and social isolation) do not fully represent potential intervention targets. Positive psychosocial factors predict cognitive aging outcomes above and beyond known risk factors and may also correspond to upstream determinants that open up new avenues for prevention and intervention, as well as for reducing racial/ethnic inequalities in dementia. In this brief report, I summa
