A young child with acute perforated appendicitis due to Comamonas kerstersii: a rare case report.

Comamonas species are rarely associated with human infections. Recent reports found that Comamonas kerstersii was associated with severe diseases such as abdominal infection and bacteremia. However, Comamonas kerstersii may be confused with Comamonas testosteroni using the automatic bacterial identification systems currently available. An 8-year-old boy who had a right iliac fossa pain and classic migration of pain at the temperature of 38.9°C. The positive strain of aerobic and anaerobic bottles of blood cultures was identified. The patient was diagnosed as acute peritonitis and perforated ap

Analyzing the catalytic role of active site residues in the Fe-type nitrile hydratase from Comamonas testosteroni Ni1.

A strictly conserved active site arginine residue (αR157) and two histidine residues (αH80 and αH81) located near the active site of the Fe-type nitrile hydratase from Comamonas testosteroni Ni1 (CtNHase), were mutated. These mutant enzymes were examined for their ability to bind iron and hydrate acrylonitrile. For the αR157A mutant, the residual activity (k cat = 10 ± 2 s(-1)) accounts for less than 1% of the wild-type activity (k cat = 1100 ± 30 s(-1)) while the K m value is nearly unchanged at 205 ± 10 mM. On the other hand, mutation of the active site pocket αH80 and αH81 residues to alani

A new understanding of the regulatory mechanism by which Fe/Mn nanoparticles boost Bisphenol A removal using Comamonas testosteroni.

Green synthesized iron/manganese nanoparticles (Fe/Mn NPs), acted as an exogenous promoter to enhance the lignin-degrading bacteria Comamonas testosteroni FJ17 resulting in more efficient removal of bisphenol A (BPA). Batch experiments demonstrated that removal efficiency of BPA via cells at a BPA concentration of 10 mg·L-1 increased by 20.9 % when exposed to 100 mg·L-1 Fe/Mn NPs after 48 h (93.63 %) relative to an unexposed control group (72.70 %). TEM and 3D-EEM analysis confirmed that the cell membrane thickness increased from 47 to 80 nm under Fe/Mn NPs exposure, and the TB-EPS secretion w

Bacteremia caused by Comamonas kerstersii in a patient with acute perforated appendicitis and localized peritonitis: case report and literature review.

Comamonas kerstersii (C. kerstersii) is a Gram-negative bacterium that was initially thought to be non-pathogenic to humans and is abundant in the environment. In recent years, with the availability of matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) that enable fast and accurate bacterial identification, there have been increasing number of reports of human infections caused by C. kerstersii, indicating that this organism has emerged as human pathogen. In fact, most clinical isolates of C. kerstersii are recovered from peritoneal liquid, and bacterem

Novel Gene Encoding 5-Aminosalicylate 1,2-Dioxygenase from Comamonas sp. Strain QT12 and Catalytic Properties of the Purified Enzyme.

The 1,125-bp mabB gene encoding 5-aminosalicylate (5ASA) 1,2-dioxygenase, a nonheme iron dioxygenase in the bicupin family that catalyzes the cleavage of the 5ASA aromatic ring to form cis-4-amino-6-carboxy-2-oxohexa-3,5-dienoate in the biodegradation of 3-aminobenzoate, was cloned from Comamonas sp. strain QT12 and characterized. The deduced amino acid sequence of the enzyme has low sequence identity with that of other reported ring-cleaving dioxygenases. MabB was heterologously expressed in Escherichia coli cells and purified as a His-tagged enzyme. The optimum pH and temperature for MabB ar

Sediment-isolated Comamonas terrigena strain HJ-2: a novel nitrate-dependent ferrous-oxidizing bacterium with multifunction on pollutant transformation.

Microbially driven Fe(II) oxidation is vital for Fe-cycling processes. In the present study, a novel strain of nitrate-dependent Fe-oxidizing bacteria (FOB) was isolated from the riparian zone sediment of the Hanjiang River, China. It was identified as Comamonas terrigena strain HJ-2. The strain HJ-2 oxidized 2.80 mmol l-1 Fe(II) within 144 h to form Fe(III)/Fe(II) complex on the cell surface using 1.63 mmol l-1 nitrate as an electron acceptor. The formed nitrite from nitrate reduction chemically oxidized Fe(II). Surprisingly, this strain also reduced nitrilotriacetic iron to form 0.5 mmol l-1

An investigation of clinical characteristics and antimicrobial agent susceptibility patterns in clinical Comamonas testosteroni isolates: An increasingly prevalent nosocomial pathogen.

BACKGROUND: Comamonas testosteroni is a gram-negative bacillus, known before 1987 as Pseudomonas testosteroni. Although considered as a rare pathogen, its frequency has been increasing. Data regarding its antibiotic susceptibility are insufficient. To date, forty-four cases have been reported in the literature. In this study, we identified the C. testosteroni infections observed in our hospital and evaluated their antimicrobial agent susceptibility patterns compared with cases reported in the literature. METHODS: For the purposes of the present study, patients admitted to hospital between Nov

Characterization of a high-affinity phenol hydroxylase from Comamonas testosteroni R5 by gene cloning, and expression in Pseudomonas aeruginosa PAO1c.

Comamonas testosteroni strain R5 is a phenol-degrading bacterium which expresses a phenol-oxygenating activity that is characterized by low Ks (the apparent half-saturation constant in Haldane's equation) and low K(SI) (the apparent inhibition constant) values. We have now cloned the gene cluster encoding a phenol hydroxylase (phcKLMNOP) and its cognate regulator (phcR) from strain R5. Transformation of Pseudomonas aeruginosa PAO1c (Phenol Catechol+) with pROR502, a derivative of pRO1614 containing the cloned genes, confers the ability to grow on phenol as the sole carbon source. The Ks and K(

Beyond Bioremediation: A Rare Case of Comamonas testosteroni Bloodstream Infection in a Patient With Stage 5 Chronic Kidney Disease on Maintenance Hemodialysis.

Comamonas testosteroni is a gram-negative aerobic bacillus commonly found in environmental sources and known for its bioremediation potential. Although it is widely distributed in nature, human infection is rare, but it is increasingly being recognized as a nosocomial pathogen. A 68-year-old female with stage 5 chronic kidney disease (CKD V) on maintenance hemodialysis since 2017 presented with fever and decreased consciousness. On examination, a tunneled cuffed catheter (TCC) was noted in the right internal jugular vein. Two sets of blood cultures, one drawn from the TCC and the other from a

Identification of IMP-8-carrying Comamonas thiooxydans strains from a diarrheal sample: genomic insights into plasmid-borne carbapenemase in an environmental bacterium.

BACKGROUND: The proliferation of carbapenem-resistant bacteria, facilitated by carbapenemase enzymes, constitutes a significant public health threat. Although the metallo-β-lactamase gene blaIMP-8 has been identified in various bacterial species, its occurrence in Comamonas thiooxydans within the human intestinal tract is not well understood. METHODS: Three bacterial strains were isolated from an intestinal perforation patient's diarrheal samples using MacConkey agar with meropenem. Species identification was done via MALDI-TOF MS, and the IMP gene's presence was confirmed by PCR. Complete ge

Elevated level of the second messenger c-di-GMP in Comamonas testosteroni enhances biofilm formation and biofilm-based biodegradation of 3-chloroaniline.

The bis-(3'-5')-cyclic dimeric guanosine monophosphate (c-di-GMP) is a ubiquitous second messenger that determines bacterial lifestyle between the planktonic and biofilm modes of life. Although the role of c-di-GMP signaling in biofilm development and dispersal has been extensively studied, how c-di-GMP signaling influences environmental bioprocess activities such as biodegradation remains unexplored. To elucidate the impacts of elevating c-di-GMP level on environmental bioprocesses, we constructed a Comamonas testosteroni strain constitutively expressing a c-di-GMP synthase YedQ from Escheric

Comamonas resistens Co-Producing GES-5 and OXA-17 in Urban Wastewater as a Potential Novel Disseminator of Clinically Relevant β-Lactamases.

Comamonas species have been isolated from different sources, with Comamonas testosteroni and Comamonas resistens commonly related to human diseases and multidrug resistance, respectively. During a surveillance study to monitor carbapenem resistance in bacteria from wastewater samples in Brazil, a carbapenem-resistant strain, named M13, was obtained and identified as C. resistens (ANI 98.90%, dDDH 94.60%) by genomic analysis, being a species distinct from C. testosteroni. It exhibited multidrug resistance and presented small inhibition zones around disks containing novel β-lactams and β-lactam-

Comprehensive summary of steroid metabolism in Comamonas testosteroni TA441: entire degradation process of basic four rings and removal of C12 hydroxyl group.

Comamonas testosteroni is one of the representative aerobic steroid-degrading bacteria. We previously revealed the mechanism of steroidal A,B,C,D-ring degradation by C. testosteroni TA441. The corresponding genes are located in two clusters at both ends of a mega-cluster of steroid degradation genes. ORF7 and ORF6 are the only two genes in these clusters, whose function has not been determined. Here, we characterized ORF7 as encoding the dehydrase responsible for converting the C12β hydroxyl group to the C10(12) double bond on the C-ring (SteC), and ORF6 as encoding the hydrogenase responsible

Acute gastroenteritis caused by Comamonas testosteroni in a patient with hepatobiliary dysfunction: a case report highlighting diagnostic challenges during the pre-monsoon season in India.

Comamonas testosteroni is a rare, environmental Gram-negative Bacillus increasingly recognised as an emerging opportunistic pathogen. It is often misidentified as a Pseudomonas species in routine laboratories due to overlapping phenotypic characteristics. Although generally of low virulence, it can cause severe infections in patients with underlying hepatobiliary or immunocompromised conditions. We report a case of an 18-year-old male from Karnataka, South India, who presented with fever, vomiting, and watery diarrhoea during the early monsoon showers of May 2025. Laboratory evaluation reveale

IncP-1β plasmids of Comamonas sp. and Delftia sp. strains isolated from a wastewater treatment plant mediate resistance to and decolorization of the triphenylmethane dye crystal violet.

The application of toxic triphenylmethane dyes such as crystal violet (CV) in various industrial processes leads to large amounts of dye-contaminated sludges that need to be detoxified. Specific bacteria residing in wastewater treatment plants (WWTPs) are able to degrade triphenylmethane dyes. The objective of this work was to gain insights into the genetic background of bacterial strains capable of CV degradation. Three bacterial strains isolated from a municipal WWTP harboured IncP-1β plasmids mediating resistance to and decolorization of CV. These isolates were assigned to the genera Comamo
