Nitro and Nitroso Metathesis Reactions with Monomeric Zirconium Imido Complexes.

The bis(cyclopentadienyl)(tert-butylimido)zir-conium complex 1 undergoes ambient-temperature metathesis reactions with nitro- and nitrosoarenes, providing a rare nonphotochemical synthesis of cis-azoxy and cis-azo compounds, respectively. 2-Nitro-2-methylpropane also undergoes metathesis to give trans-(tert-butylazoxy)-2-methylpropane. This reaction was studied kinetically, and the rate was found to be first order in both 1 and substrate and inversely proportional to the concentration of tetrahydrofuran, with activation parameters DeltaH(double dagger) = 15 +/- 2 kcal mol(-1) and DeltaS(double

Antifeedant compounds from three species of Apiaceae active against the field slug, Deroceras reticulatum (Muller).

Extracts of volatiles from foliage of three plants in the Apiaceae, Conium maculatum L. (hemlock), Coriandrum sativum L. (coriander), and Petroselinum crispum Mill. (Nym.) (parsley), previously shown to exhibit antifeedant activity in assays with the field slug, Deroceras reticulatum (Muller) (Limacidae: Pulmonata), were studied further to identify the active components. Coupled gas chromatography-mass spectrometry (GC-MS) and neurophysiological assays using tentacle nerve preparations resulted in the identification of 11 active compounds from the three extracts. Quinolizidine and piperidine alkaloid teratogens from poisonous plants and their mechanism of action in animals.

Quinolizidine and piperidine alkaloid teratogens from Lupinus, Conium, and Nicotiana genera have been identified as causes of birth defects in livestock induced by poisonous plants. Many defects now known to be related to poisonous plant ingestion were once thought to have a genetic origin. This supposition delayed diagnosis, reporting, and understanding of such birth defects, because breeders and producers feared the news would make it difficult to sell breeding stock. Defects caused by quinolizidine and piperidine teratogens include cleft palate and contracture-type skeletal defects such as

Practical pH Measurements in Wet Organic Solvents by Gradient NMR: From Charged Amino Acid pKa to Identification of Ionizable Natural Products in Crude Extracts.

We present NMR-based pH measurements in nondried commercial methanol, methanol-OD, ethanol, and acetonitrile. Combining these measurements with pH gradients and spatially selective NMR enables determination of pKa and provides a highly effective tool to analyze complex molecular mixtures and identify unknown organic compounds by the functional groups present. We determine all the pKa values of alanine, glutamic acid, histidine, lysine, proline, tryptophan, and tyrosine in methanol-OD, filling a major gap in the literature for these important polyprotic molecules that exhibit zwitterionic behav

Actions of piperidine alkaloid teratogens at fetal nicotinic acetylcholine receptors.

Teratogenic alkaloids are found in many species of plants including Conium maculatum L., Nicotiana glauca, Nicotiana tabaccum, and multiple Lupinus spp. Fetal musculoskeletal defects produced by alkaloids from these plants include arthrogyropisis, scoliosis, torticollis, kyposis, lordosis, and cleft palate. A pharmacodynamic comparison of the alkaloids ammodendrine, anabasine, anabaseine, anagyrine, and coniine in SH-SY5Y cells and TE-671 cells was made. These alkaloids and their enantiomers were more effective in depolarizing TE-671 cells which express the human fetal-muscle type nicotinic ac

Poisonous Piperidine Plants and the Biodiversity of Norditerpenoid Alkaloids for Leads in Drug Discovery: Experimental Aspects.

There are famous examples of simple (e.g., hemlock, Conium maculatum L.) and complex (e.g., opium poppy, Papaver somniferum L., Papaveraceae) piperidine-alkaloid-containing plants. Many of these are highly poisonous, whilst pepper is well-known gastronomically, and several substituted piperidine alkaloids are therapeutically beneficial as a function of dose and mode of action. This review covers the taxonomy of the genera Aconitum, Delphinium, and the controversial Consolida. As part of studying the biodiversity of norditerpenoid alkaloids (NDAS), the majority of which possess an N-ethyl group

Genetic identification and subsequent LC-QTOF MS analysis of plant remains (Oenanthe spp.) could prove the cause of an undetermined sudden death.

In recent years, recorded cases related to forensic botany and, in particular, of plant poisoning have become rare. We report on the medicolegal characteristics of an undetermined sudden death (USD) of a woman in which scene there were remnants of a vegetal peeling. After the autopsy, macroscopic findings reported multiorgan failure and requested the investigation of the cause of death. Postmortem blood was firstly investigated on cyanide toxicity presumptively coming from a yucca-like root; however, found cyanide levels were under normality. Because of the lack of morphological features of th

[Rhabdomyolysis: have you considered food poisoning from quails?].

Rhabdomyolysis (R) is a complex condition involving the rapid dissolution of damaged or injured skeletal muscle. This leads to the direct release of intracellular components, including myoglobin, creatine kinase, aldolase, and lactate dehydrogenase, as well as electrolytes, into the bloodstream and extracellular space. Clinically, R shows a triad of symptoms: myalgia, limb weakness, and myoglobinuria without hematuria, while myoglobin has been recognized as playing a part in the development of acute kidney injury. Coturnism is a relatively rare disease, mostly found in the European countries b

Fetal muscle-type nicotinic acetylcholine receptor activation in TE-671 cells and inhibition of fetal movement in a day 40 pregnant goat model by optical isomers of the piperidine alkaloid coniine.

Coniine is an optically active toxic piperidine alkaloid and nicotinic acetylcholine receptor (nAChR) agonist found in poison hemlock (Conium maculatum L.). Coniine teratogenicity is hypothesized to be attributable to the binding, activation, and prolonged desensitization of fetal muscle-type nAChR, which results in the complete inhibition of fetal movement. However, pharmacological evidence of coniine actions at fetal muscle-type nAChR is lacking. The present study compared (-)-coniine, (+)-coniine, and nicotine for the ability to inhibit fetal movement in a day 40 pregnant goat model and in

Effect of coniine on the developing chick embryo.

Coniine, an alkaloid from Conium maculatum (poison hemlock), has been shown to be teratogenic in livestock. The major teratogenic outcome is arthrogryposis, presumably due to nicotinic receptor blockade. However, coniine has failed to produce arthrogryposis in rats or mice and is only weakly teratogenic in rabbits. The purpose of this study was to evaluate and compare the effects of coniine and nicotine in the developing chick. Concentrations of coniine and nicotine sulfate were 0.015%, 0.03%, 0.075%, 0.15%, 0.75%, 1.5%, 3%, and 6% and 1%, 5%, and 10%, respectively. Both compounds caused defor

Clinical spectrum of accidental hemlock poisoning: neurotoxic manifestations, rhabdomyolysis and acute tubular necrosis.

In the past, hemlock poisoning was only known for its neurotoxic effects; quite recently non-neurological features, consisting of rhabdomyolysis and acute renal failure, have been also described. Here we report our experience with these clinical findings, which we frequently observe in accidental hemlock poisoning. Between 1972 and 1990 we studied 18 patients: 17 of them were poisoned by conline (an alkaloid of Conium maculatim) in Apulia (Italy), and one by cicutoxin (the active principle of water hemlock) in New Mexico (USA). In the non-rapidly-fatal cases we tested myoglobinuria, serum musc

Spectroscopic investigation of hydrogen bond network stability and microplastic leaching in ethanol-based potentised medicines at extreme dilutions during prolonged plastic storage.

The quality and efficacy of pharmaceutical products stored under proper conditions are critical. This study examined the effects of long-term plastic storage on extremely diluted ethanol-based potentised (EP) medicines using advanced spectroscopic techniques. Four medicines, Arnica montana, Rhus toxicodendron, Conium maculatum, and Belladonna, at ultra-high (200C, 1 M) and moderate-high (30C, 200C) potencies, were stored in glass and plastic containers for one month. Post-storage, antioxidant activity (DPPH assay), pH, and spectroscopic analyses (ATR-FTIR, Raman and DLS) were conducted. Glass-

Favorable fragmentation: river reservoirs can impede downstream expansion of riparian weeds.

River valleys represent biologically rich corridors characterized by natural disturbances that create moist and barren sites suitable for colonization by native riparian plants, and also by weeds. Dams and reservoirs interrupt the longitudinal corridors and we hypothesized that this could restrict downstream weed expansion. To consider this "reservoir impediment" hypothesis we assessed the occurrences and abundances of weeds along a 315-km river valley corridor that commenced with an unimpounded reach of the Snake River and extended through Brownlee, Oxbow, and Hells Canyon reservoirs and dams

Occurrence of neonatal and postnatal mortality in range beef cattle. II. Factors contributing to calf death.

Results are summarized of necropsy findings from 798 calves lost from birth to weaning over a 15-yr period. Autopsies determined cause of death and ascertained anatomical normalcy of the skeletal, musculature and organ systems and functinal status of the lungs. Of the 798 calves, 77.7% were anatomically normal and 22.3% were abnormal (P < 0.01) and 75.0% of the total deaths of abnormal calves occurred by Day 2 postpartum. Birth weights of normal calves averaged 4.2 kg heavier (P < 0.05) than that of abnormal calves. Internal hydrocephalus was identified and confirmed heritable as a lethal rece

What kind of poison was used in "The Name of the Rose"?

In the novel "The Name of the Rose" by Umberto Eco, a fanatical monk laced the pages of a book with a poison, which led to the death of several monks in a medieval monastery. Based on modern toxicological knowledge, an attempt can be made to determine whether there exists a substance meeting the criteria of the poison described in the novel. To this end, toxicological literature on the lethal doses of plant-derived poisons and their duration of action was reviewed. Cowbane (Cicuta virosa), a plant used for preparing poisonous potions, contains cicutoxin which kills at a dose of 500 mg. Similar
