The Iron Deficiency Response of Corynebacterium glutamicum and a Link to Thiamine Biosynthesis.

The response to iron limitation of the Gram-positive soil bacterium Corynebacterium glutamicum was analyzed with respect to secreted metabolites, the transcriptome, and the proteome. During growth in glucose minimal medium, iron limitation caused a shift from lactate to pyruvate as the major secreted organic acid complemented by l-alanine and 2-oxoglutarate. Transcriptome and proteome analyses revealed that a pronounced iron starvation response governed by the transcriptional regulators DtxR and RipA was detectable in the late, but not in the early, exponential-growth phase. A link between iro

De novo tryptophanase-based indole production by metabolically engineered Corynebacterium glutamicum.

Indole has an increasing interest in the flavor and fragrance industry. It is used in dairy products, tea drinks, and fine fragrances due to its distinct floral odor typical of jasmine blossoms. The current production of indole based on isolation from coal tar is non-sustainable and its isolation from plants is often unprofitable due to low yields. To offer an alternative to the conventional production, biosynthesis of indole has been studied recently. A glucose-based indole production was achieved by employing the Corynebacterium glutamicum tryptophan synthase α-subunit (TrpA) or indole-3-gly

Crystal Structure and Functional Characterization of Acetylornithine Aminotransferase from Corynebacterium glutamicum.

The amino acids l-arginine and l-ornithine are widely used in animal feed and as health supplements and pharmaceutical compounds. In arginine biosynthesis, acetylornithine aminotransferase (AcOAT) uses pyridoxal-5'-phosphate (PLP) as a cofactor for amino group transfer. Here, we determined the crystal structures of the apo and PLP complex forms of AcOAT from Corynebacterium glutamicum (CgAcOAT). Our structural observations revealed that CgAcOAT undergoes an order-to-disorder conformational change upon binding with PLP. Additionally, we observed that unlike other AcOATs, CgAcOAT exists as a tet

Isopropanol production using engineered Corynebacterium glutamicum from waste rice straw biomass.

Isopropanol, a well-known biofuel, is a widely used precursor for chemical products that can replace nonrenewable petroleum energy. Here, engineered Corynebacterium glutamicum that can effectively utilize all xylose and glucose in agricultural waste rice straw to produce isopropanol was described. First, codon mutations were introduced into transporters and glycolytic-related genes to decrease the glucose preference of C. glutamicum. A more energetically favorable xylose oxidative pathway was constructed that replaced traditional xylose isomerization pathways, saving twice the number of enzyma

Production of D-lactic acid by Corynebacterium glutamicum under oxygen deprivation.

In mineral salts medium under oxygen deprivation, Corynebacterium glutamicum exhibits high productivity of L-lactic acid accompanied with succinic and acetic acids. In taking advantage of this elevated productivity, C. glutamicum was genetically modified to produce D-lactic acid. The modification involved expression of fermentative D-lactate dehydrogenase (D-LDH)-encoding genes from Escherichia coli and Lactobacillus delbrueckii in L-lactate dehydrogenase (L-LDH)-encoding ldhA-null C. glutamicum mutants to yield strains C. glutamicum DeltaldhA/pCRB201 and C. glutamicum DeltaldhA/pCRB204, respe

Microbial synthesis of the type I polyketide 6-methylsalicylate with Corynebacterium glutamicum.

Type I polyketide synthases (PKSs) are large multi-domain proteins converting simple acyl-CoA thioesters such as acetyl-CoA and malonyl-CoA to a large diversity of biotechnologically interesting molecules. Such multi-step reaction cascades are of particular interest for applications in engineered microbial cell factories, as the introduction of a single protein with many enzymatic activities does not require balancing of several individual enzymatic activities. However, functional introduction of type I PKSs into heterologous hosts is very challenging as the large polypeptide chains often do n

Glucose-6-phosphate dehydrogenase and its deficiency in mutants of Corynebacterium glutamicum.

Corynebacterium glutamicum is a member of a group of taxonomically related glutamate-excreting bacteria which utilize glucose both by the Embden-Meyerhof and the pentose phosphate pathways, the latter sequence accounting for 10 to 38% of the glucose metabolized. Some of the properties of glucose-6-phosphate dehydrogenase in crude extracts of C. glutamicum were studied. The enzyme was rapidly inactivated by dilution in tris (hydroxymethyl)aminomethane-hydrochloride buffer. This inactivation was prevented by the presence of 0.45 m NaCl. Mg(++) was required for enzyme activity, but Mn(++), Ca(++)

The MscS channel from Corynebacterium glutamicum uses a non-canonical gating mechanism.

The function and gating mechanism of Escherichia coli mechanosensitive channel of small conductance (EcMscS) is well understood, but it is unknown whether MscS homologs in other bacteria function the same way. Here, we show that the MscS homolog from Corynebacterium glutamicum (CgMscS) opens at a much higher membrane tension than EcMscS but has otherwise similar functional characteristics. CgMscS is also structurally similar to EcMscS but features an extended transmembrane (TM) helix 2 that forms salt bridges with the cytoplasmic cage. Compared to the closed conformation, the TM1-2 sensor padd

Production of aromatic amino acids and their derivatives by Escherichia coli and Corynebacterium glutamicum.

Demand for aromatic amino acids (AAAs), such as L-phenylalanine, L-tyrosine, and L-tryptophan, has been increasing as they are used in animal feed and as precursors in the synthesis of industrial and pharmaceutical compounds. These AAAs are biosynthesized through the shikimate pathway in microorganisms and plants, and the reactions in the AAA biosynthesis pathways are strictly regulated at the levels of both gene expression and enzyme activity. Various attempts have been made to produce AAAs and their derivatives using microbial cells and to optimize production. In this review, we summarize th

Development of a single-cell GlxR-based cAMP biosensor for Corynebacterium glutamicum.

Cyclic adenosine monophosphate (cAMP) plays a regulatory role as second messenger in many species. In the industrial model organism Corynebacterium glutamicum, cAMP acts as effector of the global transcriptional regulator GlxR, a homolog of enterobacterial Crp. The cAMP-GlxR complex activates or represses the expression of about 200 target genes. CyaB, a membrane-bound class III adenylate cyclase, synthesizes cAMP from ATP, but another yet unknown cAMP-forming enzyme is likely present in C. glutamicum. Recently, we identified the cAMP phosphodiesterase CpdA, which catalyzes the conversion of c

Metabolic engineering strategies for sustainable de novo isobutylamine production by Corynebacterium glutamicum.

BACKGROUND: Short-chain primary amines are a versatile group of molecules in biological systems, derived from the decarboxylation of amino acids or the transamination of carbonyl-containing molecules. Isobutylamine, the decarboxylation product of L-valine, has applications in chemical synthesis of pharmaceuticals, herbicides, fungicides and specialized chemicals, where it is especially interesting as a substitute for petrochemical compounds. Since the chemical synthesis of short-chain primary amines is hazardous and environmentally unsustainable, we established an efficient biosynthetic approa

Artificial metabolic pathway for homoserine biosynthesis enables formaldehyde-dependent growth of Corynebacterium glutamicum.

We designed an artificial homoserine biosynthetic pathway composed of 4-hydroxy-2-oxoheptanedioate aldolase (HpaI) from Escherichia coli B and leucine dehydrogenase (LeuDH) from Lysinibacillus sphaericus and implemented it in a homoserine auxotrophic mutant strain of Corynebacterium glutamicum. The engineered strain grew on glucose plus formaldehyde without homoserine addition. To enhance the metabolic flux of this artificial homoserine biosynthetic pathway, adaptive laboratory evolution of the engineered strain on glucose plus formaldehyde was conducted, and evolved strains exhibiting signifi

Evaluating Cytocompatibility of Corynebacterium glutamicum-poly Vinyl Alcohol Living Biomaterials for Ocular Use.

In ophthalmology, living biomaterials have appeared as promising drug delivery and biosensor devices to tackle dynamic sensing and delivery of compounds. Their living character complicates their assessment with the also dynamic ocular tissues. The use of animal experiments increases complexity, and most animal ocular models are anatomically different from humans. Thus, in vitro ocular systems applied specifically to living biomaterials are required to assess their safety, compatibility and efficacy. Here, we report on an in vitro cornea model for co-cultures with Corynebacterium glutamicum-pol

Biotin concentration affects anaplerotic reactions functioning in glutamic acid production in Corynebacterium glutamicum.

The study investigates the effect of biotin concentration on the role of anaplerotic reactions catalysed by pyruvate carboxylase (PC) and phosphoenolpyruvate carboxylase (PEPC) in glutamic acid production by Corynebacterium glutamicum. C. glutamicum requires biotin for its growth, and its glutamic acid production can be induced by the addition of Tween 40 or penicillin or by biotin limitation. The biotin enzyme PC and the non-biotin enzyme PEPC catalyse two anaplerotic reactions to supply oxaloacetic acid to the TCA cycle in C. glutamicum. Therefore, they are crucial for glutamic acid producti

Functional characterization of Corynebacterium glutamicum catechol 2, 3-dioxygenase induced by aromatic compounds.

In bacteria, phenolic compounds are usually metabolized via the catechol pathway, in which catechol undergoes ring-opening reactions catalyzed by catechol 1, 2-dioxygenase (C12O, ortho cleavage) or catechol 2, 3-dioxygenase (C23O, meta cleavage), eventually entering central carbon metabolism. Corynebacterium glutamicum can utilize phenol as the sole carbon and energy source. The function of its C12O (encoded by ncgl2319) has been confirmed, whereas the putative C23O (encoded by ncgl2007, designated cgc23o) remains uncharacterized. In this study, we functionally characterized CgC23O, a protein

Hypersecretion of OmlA antigen in Corynebacterium glutamicum through high-throughput based development process.

Outer membrane lipoprotein A (OmlA) is a vaccine antigen against porcine contagious pleuropneumonia (PCP), a disease severely affecting the swine industry. Here, we aimed to systematically potentiate the secretory production of OmlA in Corynebacterium glutamicum (C. glutamicum), a widely used microorganism in the food industry, by establishing a holistic development process based on our high-throughput culture platform. The expression patterns, expression element combinations, medium composition, and induction conditions were comprehensively screened or optimized in microwell plates (MWPs), fo

Metabolic engineering of Corynebacterium glutamicum for l-tyrosine production from glucose and xylose.

Microbial production of aromatic compounds is an attractive and sustainable biotechnological approach. With this motivation, here metabolic engineering of Corynebacterium glutamicum for l-tyrosine (l-Tyr) overproduction was attempted by pushing the carbon flux more towards l-Tyr. Translational start codon exchanges of prephenate dehydratase (pheA), anthranilate synthase (trpE), and phenylalanine aminotransferase (pat) genes revealed that reduced expression of pheA was the major contributor to increased l-Tyr titer while codon exchange in trpE was effective to a lower extent. Overexpression of

Impact of LytR-CpsA-Psr Proteins on Cell Wall Biosynthesis in Corynebacterium glutamicum.

UNLABELLED: Proteins of the LCP (LytR, CpsA, Psr) family have been shown to inherit important roles in bacterial cell wall biosynthesis. However, their exact function in the formation of the complex cell wall structures of the Corynebacteriales, including the prominent pathogens Mycobacterium tuberculosis and Corynebacterium diphtheriae, remains unclear. Here, we analyzed the role of the LCP proteins LcpA and LcpB of Corynebacterium glutamicum, both of which localize at regions of nascent cell wall biosynthesis. A strain lacking lcpB did not show any growth-related or morphological phenotype u

Heterologous expression and localization of gentisate transporter Ncg12922 from Corynebacterium glutamicum ATCC 13032.

Ralstonia sp. strain U2 metabolizes naphthalene via gentisate (2,5-dihydroxybenzoate) to central metabolites, but it was found unable to utilize gentisate as growth substrate. A putative gentisate transporter encoded by ncg12922 from Corynebacterium glutamicum ATCC 13032 was functionally expressed in Ralstonia sp. strain U2, converting strain U2 to a gentisate utilizer. Assessing Corynebacterium glutamicum as a surrogate of Mycobacterium tuberculosis for DNA gyrase inhibitor design.

DNA gyrase is an essential bacterial enzyme and a clinically validated target for the treatment of tuberculosis. However, the discovery of new inhibitors remains limited by the many challenges regarding the manipulation on pathogenic mycobacteria. This study validates Corynebacterium glutamicum (Cglu) as a safe, non-pathogenic surrogate for Mycobacterium tuberculosis (Mtb) to investigate DNA gyrase and facilitate the identification of new inhibitors. Using Cglu as a target allows for fast whole-cell screening under safe conditions while ensuring efficient drug uptake. Cglu shares key physiolog
