Cadmium-dependent oxygen limitation affects temperature tolerance in eastern oysters (Crassostrea virginica Gmelin).

Marine ectotherms, including oysters are exposed to variable environmental conditions in coastal shallow waters and estuaries. In the light of global climate change, additional stressors like pollution might pose higher risk to populations. On the basis of the concept of oxygen- and capacity-limited thermal tolerance in aquatic ectotherms (40), we show that a persistent pollutant, cadmium, can have detrimental effects on oysters (Crassostrea virginica). During acute warming from 20 to 28 degrees C (4 degrees C/48 h) standard metabolic rate (SMR) rose in control and cadmium-exposed (50 microg C

A flow cytometric approach to study intracellular-free Ca2+ in Crassostrea gigas haemocytes.

Bivalve haemocytes are essential in defence mechanisms including phagocytosis. They also produce molecules including hydrolytic enzymes and antimicrobial peptides that contribute to pathogen destruction. Although haemocyte activities have been extensively studied, relatively little is known about the intracellular signalling pathways that are evoked during haemocyte activation and especially the role of calcium. Flow cytometry has been used for the first time to define the effect of cell incubation in haemolymph and artificial sea water (ASW) on Pacific oyster, Crassostrea gigas, haemocytes. C

Protective effect of Crassostrea gigas extract on audiogenic seizures in magnesium deficient mice.

Audiogenic seizures associated with loss of weight, prostration, piloerection, palpebral ptosis and motor deficiency were induced after sound stimulation of determined frequency and amplitude in magnesium-deficient DBA/2 mice. These symptoms were maintained when standard diet conditions (1700 ppm Mg2+) were restored. In contrast, mice were protected from audiogenic seizure in a dose related manner when Crassostrea gigas extract (JCOE) were added to the diet for 10 consecutive days. Although a rational explanation for this protective effect has not yet been determined, it is assumed that it mig

Comparing Minimally Lethal Sampling Methods for Genomics in the Eastern Oyster (Crassostrea virginica).

Understanding how selection acts on individual genotypes often requires genotyping at different life stages and tracking their survival under experimental conditions. However, in mollusks, such as the Eastern oyster (Crassostrea virginica), collecting sufficient high-quality DNA without causing mortality remains challenging. While prior studies have explored different noninvasive genotyping methods, an optimal method hasn't been identified. Here, we tested different techniques by combining two minimally invasive accession techniques (relaxation, shell notching), four types of cell sampling (sw

Viral uptake and stability in Crassostrea gigas oysters during depuration, storage and steaming.

More stable than bacteria in environmental samples, enteric viruses are generally related to outbreaks of gastroenteritis caused by the consumption of contaminated oysters. This study evaluated: i) the dynamic processes of enteric viral models bioaccumulation by Crassostrea gigas oysters artificially contaminated; ii) the stability of these viruses in oysters in controlled temperature conditions and iii) the effect of UV light in inactivating these viruses in depurated oysters. Plaque assay (PA) was used to assess the infectivity of both viral models. Cell culture coupled with RT-qPCR (ICC-RT-

Proteomic and metabolomic responses of Pacific oyster Crassostrea gigas to elevated pCO2 exposure.

UNLABELLED: The gradually increased atmospheric CO2 partial pressure (pCO2) has thrown the carbonate chemistry off balance and resulted in decreased seawater pH in marine ecosystem, termed ocean acidification (OA). Anthropogenic OA is postulated to affect the physiology of many marine calcifying organisms. However, the susceptibility and metabolic pathways of change in most calcifying animals are still far from being well understood. In this work, the effects of exposure to elevated pCO2 were characterized in gills and hepatopancreas of Crassostrea gigas using integrated proteomic and metabolo

Stress response in Cu2+ and Cd2+ exposed oysters (Crassostrea gigas): an immunohistochemical approach.

Localization of heat shock proteins (HSPs) and metallothioneins (MTs) was investigated in a marine bivalve (Crassostrea gigas) by immunohistochemical methods. Differential protein expression was demonstrated in digestive gland, gonad and gills, using a polyclonal antibody against C. gigas proteins. Application of this technique showed the cellular and tissue immunolabelling specificity of the two proteins. HSPs and MTs were localized in the epithelium of the digestive gland and gills in contact with the palleal compartment. For the first time, localization of MTs was observed in mature gametes

Physiological and molecular effects of individual and mixtures of drugs of abuse in Crassostrea virginica larvae.

Drugs of abuse (DAs) are increasingly detected in surface waters, yet their effects on marine organisms remain poorly understood. This study evaluated the impacts of environmentally relevant concentrations (100 and 1000 ng L⁻¹) of cannabinol, codeine, and methamphetamine, individually and in mixtures, on the growth and development of Crassostrea virginica larvae. Eight-day exposures significantly reduced larval survival, impaired growth, and altered swimming behavior, with mixtures producing the strongest effects. At 1000 ng L⁻¹ , cumulative survival reached zero in mixture and cannabinol grou

In-situ microcosms, a tool for assessment of pesticide impacts on oyster spat (Crassostrea gigas).

Effects of the herbicide Basamaïs (bentazon) and the fungicide Opus (epoxiconazole) on oyster spat (Crassostrea gigas) were assessed using in-situ microcosms in a field experiment lasting 13 days. Six-week-old hatchery spat (mean size 1.1 mm), previously collected on PVC plates, was immersed in glass bottles filled with 200 mum filtered seawater. Bottles were maintained underwater at 6 m depth and their water content changed every other day. Growth, measured as shell area index increase, was 126 +/- 4% in the control bottles. While no growth differences were observed between control and indivi

Identification and characterization of a testis-specific glycogen phosphorylase in Kumamoto oyster Crassostrea sikamea.

Glycogen serves as the principal energy source for gonadal development in marine bivalves, such as oysters. Glycogen phosphorylase is the rate-limiting enzyme of glycogenolysis by catalyzing the first step of glycogen breakdown. In this study, two different glycogen phosphorylase genes were identified in the Kumamoto oyster (Crassostrea sikamea) genome, including the previously reported GPH and an unknown PYG. C. sikamea GPH and PYG share high mRNA and protein sequence identities, but they have distinct gene structures and show significant sequence variations in their adenine loops. Interestin

Evaluation of toxicity of Deepwater Horizon slick oil on spat of the oyster Crassostrea virginica.

The 2010 explosion of the Deepwater Horizon (DWH) oil rig generated the largest marine oil spill in US history with millions of barrels of crude oil released in the Gulf of Mexico (GoM). The eastern oyster, Crassostrea virginica, is an ecologically and economically important species in the northern GoM. Due to its biological characteristics (sessile, filter feeding), juvenile oysters may have been affected. This study investigated the effects of surface-collected DWH oil prepared as high-energy water-accommodated fraction (HEWAF) on the survival of 2-month-old oyster spat, and evaluated the po

A glycoprotein in shells of conspecifics induces larval settlement of the Pacific oyster Crassostrea gigas.

Settlement of larvae of Crassostrea gigas on shell chips (SC) prepared from shells of 11 different species of mollusks was investigated. Furthermore, the settlement inducing compound in the shell of C. gigas was extracted and subjected to various treatments to characterize the chemical cue. C. gigas larvae settled on SC of all species tested except on Patinopecten yessoensis and Atrina pinnata. In SC of species that induced C. gigas larvae to settle, settlement was proportionate to the amount of SC supplied to the larvae. When compared to C. gigas SC, all species except Crassostrea nippona sho

Facilitated bioaccumulation of cadmium and copper in the oyster Crassostrea hongkongensis solely exposed to zinc.

Exposure to one metal might have significant effects on the bioaccumulation of other metals. In the present study, we examined the possible effects of Zn exposure on the bioaccumulation of Cd and Cu in three populations of the oyster Crassostrea hongkongensis. We found that Zn exposure significantly enhanced the tissue concentrations of Cd and Cu in all populations, and the tissue concentrations of Cd and Cu were highly and positively related to the tissue Zn concentration. Furthermore, the enhanced bioaccumulation of Cd and Cu resulted mainly from their increasing accumulation and distributio

Analysis of genes isolated from plated hemocytes of the Pacific oyster, Crassostreas gigas.

A complementary deoxyribonucleic acid library was constructed from hemocytes of Crassostrea gigas that had been plated on poly-lysine plates for 24 h. From this library, 2,198 expressed sequence tags (ESTs) of greater than or equal to 100 bp were generated and analyzed. A large number of genes that potentially could be involved in the physiology of the oyster hemocyte were uncovered. They included proteins involved in cytoskeleton rearrangement, proteases and antiproteases, regulators of transcription and translation, cell death regulators, receptors and their associated protein factors, lecti

Temperature effects on the nutritional quality in Pacific oysters (Crassostrea gigas) during ultraviolet depuration.

BACKGROUND: Oysters are mainly consumed in the raw form, so it is important to get rid of bacteria and other harmful substances. Ultraviolet (UV) sterilization depuration is a commonly used method and does not produce chemical residues or act directly on shellfish, resulting in minimal adverse effects on flavor. This study simulated the industrial depuration process using UV sterilization to depurate Pacific oysters (Crassostrea gigas). The effects of different temperatures (15, 20, and 25 °C) on the quality and taste components of C. gigas were investigated by measuring changes in physiologic

IgIT2-SHP2 axis inhibits the autophagy through P38-H3K4me3 pathway in oyster Crassostrea gigas.

Inhibitory receptors containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) are generally known to suppress the activation of immune cells through the recruitment of protein tyrosine phosphatase, such as SHP-1/2, SHIP1/2. In our previous study, the inhibitory receptor CgIgIT2 was identified from Crassostrea gigas, and it was composed of four immunoglobulin (Ig) domains, a single fibronectin type III (FN3) domain, a transmembrane region, and a cytoplasmic tail with two ITIM motifs and one immunoreceptor tyrosine-based switch motif (ITSM). Among which, the Ig domains had binding acti

A Caspase-2 involves in regulating LPS-induced haemocyte apoptosis of oyster Crassostrea gigas.

Caspase-2 is an evolutionary conserved cysteine protease that plays multifaceted roles in the DNA damage response, cell cycle regulation, and apoptosis. In the present study, a caspase-2 (designed as CgCaspase-2) from the Pacific oyster Crassostrea gigas was functionally characterized as an important mediator of LPS-induced haemocyte apoptosis. After LPS stimulation, the mRNA expression level of CgCaspase-2 in haemocytes significantly up-regulated, and its protein was translocated from the cytoplasm to nucleus. Transfection of CgCaspase-2 significantly enhanced the apoptosis rate of HEK293T ce

DM9CP-integrin-RhoA pathway promotes the hemocyte phagocytosis in the immune priming of Crassostrea gigas.

DM9 domain-containing proteins (DM9CPs) constitute a novel family of pattern recognition receptors (PRRs) recognizing specific ligands to initiate downstream immune responses. Their involvement in phagocytosis and immune priming is crucial for a comprehensive understanding of their regulatory mechanisms on immune responses. In the present study, the content of CgDM9CP-1 protein in the cell-free hemolymph of Crassostrea gigas was significantly upregulated at 3, 6, and 12 h after Vibrio splendidus stimulation, and CgDM9CP-1 was able to interact with CgIntegrin both in vitro and in vivo. Seasonality of Vibrio parahaemolyticus in the Eastern oyster, Crassostrea virginica, along the coast of Maine, USA.

As the Gulf of Maine warms, characterizing seasonal baseline levels of the human pathogen Vibrio parahaemolyticus (Vp) in Eastern oysters (Crassostrea virginica) is crucial, along with synoptic environmental data, to develop predictive capacity and risk management strategies. In Maine, USA, from May to November 2023, oyster samples were collected from Scarborough River, Maquoit Bay, and the Damariscotta River. Vp was quantified using the U.S. Food and Drug Administration-approved three-tube most-probable-number (MPN) real-time PCR method, detecting the presence of total Vp (i.e., tlh gene) and

The Potential Mechanism of Cuproptosis in Hemocytes of the Pacific Oyster Crassostrea gigas upon Elesclomol Treatment.

Cuproptosis is a novel cell death dependent on mitochondrial respiration and regulated by copper. While the study of it is mainly focused on tumor therapy, in the present study, two key cuproptosis-related genes, ferredoxin (FDX1) and dihydrolipoamide S-acetyltransferase (DLAT) homologs (designated as CgFDX1 and CgDLAT), were identified from Crassostrea gigas. CgFDX1 has a Fer2 domain with a 2Fe-2S cluster forming a unique ferredoxin. CgDLAT is composed of a biotin_lipoyl domain, an E3-binding domain, and a 2-oxoacid_dh domain. CgFDX1 and CgDLAT mRNA were expressed in all the examined tissues.
