Characterization, genomics, and applications of the Cronobacter sakazakii bacteriophage Csp-D17.

Cronobacter sakazakii is an opportunistic pathogen that can contaminate infant formula, resulting in meningitis and neurological disorders in infants. Bacteriophages have been consistently identified as effective inhibitors of pathogenic bacteria. In this study, a lytic phage, Csp-D17, was isolated from environmental wastewater, and its characterization, genomics, and application were analyzed. Transmission electron microscopy revealed that the phage belonged to the order Caudoviricetes within the family Ackermannviridae. The phage Csp-D17 exhibited notable stability across varied thermal and

Effect of polymyxin resistance (pmr) on biofilm formation of Cronobacter sakazakii.

Cronobacter sakazakii (C.sakazakii) has been identified as a wide-spread conditioned pathogen associated with series of serious illnesses, such as neonatal meningitis, enterocolitis, bacteremia or sepsis. As food safety is concerned, microbial biofilm has been considered to be a potential source of food contamination. The current study aims to investigate the ability of biofilm formation of two C. sakazakii strains (wild type BAA 894 and pmrA mutant). Crystal violet (CV), XTT (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino carbonyl)-2H-(tetrazolium hydroxide)] assays, and scanning electron m

New virulence factor CSK29544_02616 as LpxA binding partner in Cronobacter sakazakii.

Cronobacter sakazakii is an opportunistic pathogen that can cause meningitis and necrotizing enterocolitis in premature infants, but its virulence determinants remain largely unknown. In this study, a transposon-mediated random-mutant library of C. sakazakii was used to identify new virulence factors. Compared to wild-type bacteria, a mutant lacking CSK29544_02616 (referred to as labp) was defective in invasion into intestinal epithelial cells (by at least 1000-fold) and showed less phagocytosis by macrophages (by at least 50-fold). The lack of labp in C. sakazakii changed the profile of outer

Specific detection of Cronobacter sakazakii in powdered infant formula using ssDNA aptamer.

Cronobacter sakazakii (C. sakazakii) is a foodborne pathogen associated with bacterial meningitis, sepsis, and necrotizing enterocolitis in premature and immuno-compromised infants. C. sakazakii is typically acquired by ingesting contaminated powdered infant formula (PIF). The growing demand for a safe food supply requires rapid detection of foodborne pathogens for delivering safe-to-consume food to consumers. In the present study, we isolated C. sakazakii-specific aptamers using a centrifugation-based partitioning method (CBPM) instead of systematic evolution of ligands by exponential enrichm

Functional characterization of iucC and iucD in iron acquisition and virulence of Cronobacter sakazakii.

BACKGROUND: Cronobacter is a group of opportunistic pathogens frequently associated with food contamination, particularly in powdered infant formula, posing a serious health risk to neonates and young children. Iron acquisition systems are critical for bacterial survival and virulence, with the iucABCD-iutA operon encoding the aerobactin siderophore synthesis pathway playing a central role. METHODS: Transcriptomic analysis comparing iron-limited and iron-replete conditions was performed to identify iron-responsive genes in Cronobacter. Single-, double-knockout, and corresponding complementati

Protective Effect of Recombinant Proteins of Cronobacter Sakazakii During Pregnancy on the Offspring.

Cronobacter sakazakii is a food-borne pathogen carried in milk powder that can cause severe bacteremia, enterocolitis, and meningitis in newborns, which can lead to death of newborns. Preventing infection by this pathogen is significant to the health of newborns. Since infants and young children are the main target group of C. sakazakii, it is considered that maternal immunity can enhance the protection of newborns. Previous studies showed that two proteins of C. sakazakii (GroEL and OmpX) exhibited high expression levels and elicited strong immune reactions, suggesting their potential as vacc

Characterization and comparative genomic analysis of novel lytic bacteriophages targeting Cronobacter sakazakii.

Cronobacter sakazakii, a foodborne pathogen, can contaminate powdered infant formula (PIF) and cause life-threatening meningitis, necrotizing colitis and meningoencephalitis in infants. Bacteriophages are increasingly considered an efficient approach to target pathogenic microorganisms. In the current study, four virulent phages that can infect C. sakazakii were isolated from sewage samples, and their biological and complete genomic characteristics were analyzed. A comparative genomic analysis was performed to investigate the functional genes and phylogenetic evolution of the four phages. The

Evaluation of commercial probiotic lactic cultures against biofilm formation by Cronobacter sakazakii.

BACKGROUND/AIMS: Cronobacter sakazakii, an emergent pathogen is considered as a major concern to infants and neonates fed on reconstituted powdered infant milk formula. In conjunction with many other factors, biofilm forming capacity adds to its pathogenic potential. In view of the facts that infants are at highest risk to C. sakazakii infections, and emerging antibiotic resistance among pathogens, it is imperative to evaluate probiotic cultures for their efficacy against C. sakazakii. Therefore, pure probiotic strains were isolated from commercial probiotic products and tested for their antim

Hemolysin III drives neuroinvasion and meningitis while impacting desiccation resilience in Cronobacter sakazakii.

UNLABELLED: Cronobacter sakazakii is an emerging pathogen associated with severe neonatal diseases, including meningitis. Its ability to cross the blood-brain barrier (BBB) is critical for meningitis development, yet the underlying mechanisms remain unclear. This study investigates the role of Hemolysin III (Hly III), encoded by the ESA_00432, in neuroinvasion and environmental resilience. A markerless deletion of ESA_00432 (ΔESA_00432) revealed that the mutant exhibited unaltered biofilm formation, increased hydrophilicity, and enhanced desiccation resistance compared to the wild type, sugges

Multiplex Determination of K-Antigen and Colanic Acid Capsule Variants of Cronobacter sakazakii.

Cronobacter sakazakii is associated with the ingestion of contaminated reconstituted powdered infant formula (PIF), resulting in necrotizing enterocolitis, sepsis and meningitis in neonatal infants. Potential virulence determinants include the variable capsular polysaccharides; K-antigen and colanic acid (CA). Strains encoding for the capsule variant K2:CA2 have been strongly associated with neonatal meningitis cases. This study aimed to develop and apply a multiplex PCR assay to determine C. sakazakii K-antigen and colanic acid types. Twenty-six strains of C. sakazakii which had previously be

Identification of potential virulence factors of Cronobacter sakazakii isolates by comparative proteomic analysis.

Cronobacter is a group of important foodborne pathogens associated with neonatal meningitis, septicemia, and necrotizing enterocolitis. Among Cronobacter species, Cronobacter sakazakii is the most common species in terms of isolation frequency. However, the molecular basis involved in virulence differences among C. sakazakii isolates is still unknown. In this study, based on the determination of virulence differences of C. sakazakii G362 (virulent isolate) and L3101 (attenuated isolate) through intraperitoneal injection, histopathologic analysis (small intestine, kidney, and liver) further con

Different factors govern surface adhesion and resilience to gastrointestinal conditions in Cronobacter sakazakii.

Cronobacter species are opportunistic pathogenic bacteria present in many foods that can cause life-threatening infections in newborns and young infants. The infective potential in other vulnerable consumer groups remains unclear. In this study, twenty-eight strains of Cronobacter sakazakii from food and human clinical sources were characterized regarding their surface adhesion ability in 96-well plates at different incubation temperatures, incubation times, and cultivation media. Based on that, eight C. sakazakii isolates were selected and further studied regarding their adhesion ability to n

Inactivation of Lipopolysaccharide-Biosynthesizing Genes Altered Lipids Composition and Intensity in Cronobacter sakazakii.

Gram-negative bacteria possess an asymmetric outer membrane, where the outer leaflet consists of LPSs and the inner leaflet comprises phospholipids. Cronobacter sakazakii, an opportunistic milk-borne pathogen that causes severe neonatal meningitis and bacteremia, displays diverse lipopolysaccharide (LPS) structures. As a barrier of the bacterial cell, LPSs likely influenced C. sakazakii resistance to environment stresses; however, there are no research reports on this aspect, hindering the development of novel bactericidal strategies overcoming the pathogen's resilience. In the present study,

Inactivation of Carotenogenic-Biosynthesizing Genes Altered Lipids Composition and Intensity in Cronobacter sakazakii.

Cronobacter sakazakii, an opportunistic milk-borne pathogen responsible for severe neonatal meningitis and bacteremia, can synthesize yellow pigment (various carotenoids) benefiting for bacterial survival, while little literature was available about the influence of various carotenoids on bacterial resistance to a series of stresses and the characteristics of cell membrane, obstructing the development of novel bactericidal strategies overcoming the strong tolerance of C. sakazakii. Thus in this study, for the first time, five carotenogenic genes of C. sakazakii BAA-894 were inactivated, respec
