Cryptomeria japonica essential oil inhibits the growth of drug-resistant skin pathogens and LPS-induced nitric oxide and pro-inflammatory cytokine production.

In this study, the chemical composition of Cryptomeria japonica essential oil (CJE) was analyzed and its biological activities were tested. CJE was obtained by steam distillation from leaves collected from Jeju Island and analyzed by gas chromatography (GC)-flame ionization detection (FID) and GC-MS. Kaurene (17.20%), elemol (10.88%), gamma-eudesmol (9.41%), and sabinene (8.86%) were the major components in CJE. The antibacterial and anti-inflammatory activities of CJE against drug-susceptible and -resistant skin pathogens have been not reported previously. Thus, we determined the anti-bacteri

Anticholinesterase and Anti-Inflammatory Activities of the Essential Oils of Sawdust and Resin-Rich Bark from Azorean Cryptomeria japonica (Cupressaceae): In Vitro and In Silico Studies.

Alzheimer's disease (AD), a multifactorial neurodegenerative disorder characterized by severe cognitive impairment, affects millions of people worldwide. However, AD therapy remains limited and mainly symptomatic-focused, with acetylcholinesterase (AChE) inhibitors being the major available drugs. Thus, AD is considered by the WHO as a disorder of public health priority. Among several strategies that have been identified to combat AD, the use of natural multi-target drug ligands (MTDLs) appears to be a promising approach. In this context, we previously found that the essential oils (EOs), obta

Oral immunotherapy with transgenic rice seed containing destructed Japanese cedar pollen allergens, Cry j 1 and Cry j 2, against Japanese cedar pollinosis.

Transgenic rice accumulating the modified major Japanese cedar pollen allergens, Cryptomeria japonica 1 (Cry j 1) and Cryptomeria japonica 2 (Cry j 2), which were deconstructed by fragmentation and shuffling, respectively, in the edible part of the seed was generated by transformation of a good-tasting rice variety, 'Koshihikari'. These modified cedar pollen antigens were deposited in ER-derived protein bodies (PB-I), which are suitable for delivery to the mucosal immune system in gut-associated lymphoid tissue when orally administered because antigens bioencapsulated in PB-I are resistant aga

An ecological study on air pollution: changes in annual ring growth of the Japanese cedar and prevalence of respiratory symptoms in schoolchildren in Japanese rural districts.

The effect of air pollution caused by oil-fired electricity-generating stations on the annual ring growth of the Japanese cedar (Cryptomeria japonica, D. Don) and prevalence of respiratory symptoms in schoolchildren were investigated. By retrospective analysis the annual ring growth has been demonstrated to show good agreement with the general trend in air pollution. In addition it was found to be related to the prevalence of respiratory symptoms. The reduction in annual ring growth and increase in the prevalence of respiratory symptoms followed a deterioration in air pollution. Following upgr

Bioactivity of selected plant essential oils against the yellow fever mosquito Aedes aegypti larvae.

The bioactivity of 14 essential oils from five plants has been studied using the brine shrimp lethality test and the Aedes aegypti larvicidal assay. All essential oils screened had LC50 values smaller than 200 microg/ml, showing significant lethality against brine shrimp. In addition, nine of the 14 essential oils tested showed toxicity against the fourth-instar A. aegypti larvae in 24 h (LC50<100 microg/ml). Of these, the leaf and bark essential oils of Cryptomeria japonica demonstrated high larvicidal activity, the most active being the leaf essential oil of C. japonica, with a LC50=37.6 mic

Allergic rhinitis to thuja pollen.

Allergy to pollen of Cupressaceae has been linked to pollens of Cupressus, Juniper and Cryptomeria. The authors report 2 cases of rhinitis and conjunctivitis induced by thuja, another member of the Cupressaceae family. Monosensitization to thuja pollen has been identified as the causal agent: (1) of a long-standing springtime rhinitis in 1 patient with negative skin tests and specific IgE titers to the main inhalants (specificity of the prick test and nasal provocation with the thuja extract was confirmed by a positive RAST) and (2) in a 2nd patient without former history of allergy, who consu

Asian dust exacerbates cedar pollen-induced allergic conjunctivitis in mice.

Asian sand dust (ASD), a type of fine particulate matter originating from arid regions of East Asia, is known to exacerbate respiratory diseases, but its impact on the ocular surface remains poorly understood. Given the increasing prevalence of allergic conjunctivitis in Japan, particularly during the Japanese cedar pollen (JCP) season, we investigated whether ASD exposure aggravates ocular allergic inflammation. Using a mouse model sensitized with JCP, we found that conjunctival instillation of ASD in combination with JCP significantly increased clinical signs such as hyperemia and edema, alo

The soothing effect of phlorotannins on cedar pollinosis in Cry j 1-stimulated mice.

The antiallergic properties of phlorotannins, algal polyphenols, have been widely reported. This study examined the soothing effect of phlorotannin concentrate (PTC) from Eisenia nipponica on cedar pollinosis in Cry j 1-stimulated mice. PTC reduced the mice's sneezing and nasal rubbing, which was attributed to decreased levels of immunoglobulin E and Th2-type cytokines [interleukin (IL)-4, IL-5, and IL-13].

Japanese Cedar Pollen Allergens in Japan.

Pollen from members of the Cupressaceae tree family is one of the most important causes of allergic disease in the world. Cryptomeria japonica (Japanese cedar) and Chamaecyparis obtusa (Japanese cypress) are Japan's most common tree species. The pollen dispersal season is mainly from February to May. The major allergens of Japanese cedar and Japanese cypress exhibit high amino acid sequence similarity due to the phylogenetic relationship between the two species. An epidemiological study has shown that the prevalence of Japanese cedar pollinosis is approximately 40%. Younger children (5 to 9 ye

Inhibition of an Allergen-Antibody Reaction Related to Japanese Cedar Pollinosis Using DNA Aptamers Against the Cry j 2 Allergen.

Japanese cedar pollinosis is one of the most prevalent allergies in Japan. Reducing the allergen content of pollen plays a major role in the alleviation of allergy symptoms. Aptamers, oligonucleotides with an affinity for specific molecules, have great potential for reducing allergic activity. In this study, we report that the anti-Cry j 2 aptamers, CJ2-04 and CJ2-08, inhibited allergen-antibody reactions between Cry j 2, one of the major allergens in Japanese cedar pollen, and immunoglobulin E in serum collected from a patient with Japanese cedar pollinosis. In addition, the suppression of Ca

A sesquiterpenol extract potently suppresses inflammation in macrophages and mice skin and prevents chronic liver damage in mice through JNK-dependent HO-1 expression.

This study aimed to elucidate the anti-inflammatory and hepatoprotective bioactivities of a sesquiterpenol, (1S,6R)-2,7(14),10-bisabolatrien-1-ol-4-one (BSL), isolated from Cryptomeria japonica (Taxodiaceae) wood extract. BSL markedly suppressed TNF-α and IL-6 secretion, PGE(2) production, and mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), in lipopolysaccharide (LPS)-stimulated mouse macrophages. BSL also potently inhibited the 12-O-tetradecanoylphorbol-13-acetate (TPA) induced protein levels of nitrotyrosine and COX-2 in mouse skin with dermatitis. Con

Structural and mechanical roles of wood polymer assemblies in softwood revealed by gradual removal of polysaccharides or lignin.

A deep understanding of the inherent roles of wood polymers such as cellulose, hemicelluloses, and lignin in the hierarchical structure of wood is of key importance for advancing functional wood-based materials but is currently lacking. To address this gap, we clarified the underexplored contributions of wood polymer assemblies to the structural support and compressive properties of wood by chemically removing polysaccharides or lignin from wood blocks of a conifer Cryptomeria japonica. Compositional and structural evaluations revealed that cellulose, hemicelluloses, and lignin contributed to
