A technique for the culture of Barrett's oesophageal cells.

Establishment of cells in tissue culture from Barrett's columnar epithelium has been difficult. The aim of this study was to develop a successful tissue culture method employing a serum-free medium for cultivation of Barrett's epithelial cells. Fragments of Barrett's mucosal tissue were explanted in a 3:1 mixture of Dulbecco's modification of Eagle's medium and Ham's F12, to initiate the outgrowth of epithelial cells. Subsequently, a commercial serum-free medium (formulated for the growth of keratinocytes) was used for the propagation of Barrett's oesophagus cells without fibroblast growth. Ce

Modeling 3D Glioblastoma: A review of spheroid culture techniques and standardization challenges.

Glioblastoma (GB) is among the most aggressive and treatment-resistant cancers, highlighting the urgent need for physiologically relevant preclinical models. Traditional two-dimensional (2D) cultures fall short in replicating the complex microenvironment of GB, whereas three-dimensional (3D) culture models offer improved structural, biochemical, and functional resemblance to in vivo tumors. However, the lack of a standardized protocol for GB spheroid formation leads to significant inter-study variability and limited reproducibility. This review summarizes the key variables affecting spheroid g

Comparison of three blood culture techniques in the diagnosis of human brucellosis.

BACKGROUND: Isolation of Brucella is the gold standard in the laboratory diagnosis of brucellosis. As Brucella is intracellular and the number of circulating bacteria is usually low, removal/dilution of antibacterial substances, concentration of bacteria and optimal culture conditions may enhance the rate of isolation. AIMS AND OBJECTIVES: The objective of the following study was to compare the lysis concentration (LC), clot culture and conventional Castaneda blood culture techniques for the isolation rate and recovery time in the diagnosis of human brucellosis. MATERIALS AND METHODS: Blood

Human diseases associated with mycoplasms. With an appendix on simple culture techniques.

The mycoplasmas (formerly called pleuropneumonia-like organisms, or pplo) are a group of pleomorphic micro-organisms characterized by lack of cell wall and ability to form colonies on agar resembling tiny fried eggs. They have been recognized as pathogens of lower mammals since 1898. Of the more than 40 known veterinary species, many are pathogens, commonly causing pneumonia, arthritis or arteritis. Of the mycoplasmas found in man, Mycoplasma pneumoniae is the only well established human pathogen. It is responsible for a variety of respiratory syndromes, of which the most frequently recognized

Intestinal microbial ecology in premature infants assessed with non-culture-based techniques.

OBJECTIVES: To use high throughput techniques to analyze intestinal microbial ecology in premature neonates, who are highly susceptible to perturbations of the luminal environment associated with necrotizing enterocolitis (NEC) and late-onset sepsis. STUDY DESIGN: With non-culture-based techniques, we evaluated intestinal microbiota shortly after birth and during hospitalization in 23 neonates born at 23 to 32 weeks gestational age. Microbiota compositions were compared in 6 preterm infants in whom NEC, signs of systemic inflammation, or both developed with matched control subjects by using 1

Detection of malignant cells in histologically normal bone marrow using culture techniques.

Cellular material retained on screens used to filter aspirated bone marrow for future autologous marrow transplantation was studied using long-term culture techniques in 20 consecutive patients. All patients had marrow aspirate and biopsy specimens that were normal histologically. Cultures from five patients grew malignant cells similar to those of the known underlying malignancy. Two types of culture methods were used in these studies. Method I consisted of a long-term bone marrow culture system which predominantly favors adherent cells, and Method II consisted of a suspension type culture sy

Comparison of polymerase chain reaction and culture techniques for detection of Chlamydia trachomatis.

The aim of this study was to compare culture and polymerase chain reaction techniques in detection of Chlamydia trachomatis in clinical specimens. Two hundred clinical specimens previously examined for C. trachomatis by culture were analysed blindly by polymerase chain reaction. A 144 bp fragment of DNA from the MOMP of C. trachomatis was amplified. In addition, a 250 bp segment of beta-globin gene was amplified as an internal control. In 27 culture negative specimens, the beta-globin amplicon was not detected. Of the 173 specimens assessable by PCR, 24 (13.8%) were positive by both methods. F

Canine prostatic fluid: techniques of collection, quantitative bacterial culture, and interpretation of results.

Specimens of prostatic fluid were collected by ejaculation from 104 dogs after collection of a specimen of urethral contents and after collection of a urine specimen by antepubic cystocentesis. For purposes of data analysis, results of bacterial culture of the specimens were grouped according to individual colony counts in each specimen group (ie, urine, urethra, prostatic fluid). For purposes of this study, prostatic infection was defined as greater than or equal to 2 log10 of one or more bacterial species in prostatic fluid over the number of colonies of the same species in the paired urethr

Microbial film formation: dental plaque deposition on acrylic tiles using continuous culture techniques.

A chemostat system has been developed to model the attachment of oral bacteria, and the subsequent development of plaque film, to acrylic surfaces immersed in steady state cultures. Plaque was removed from the teeth and gingival margin of volunteers who refrained from oral hygiene for at least 72 h. Samples were pooled and inoculated into a complex growth medium maintained at 37 degrees C. Glucose-limited continuous culture was established at a dilution rate of 0.05/h and at pH 7.0. Microbiological analysis of the culture indicated that a complex community of oral bacteria was established, typ

Laboratory Culturing Techniques and Maintenance of Vibrio cholerae.

Cholera is a severe diarrheal disease caused by the consumption of food or water contaminated with the aquatic gram-negative bacterium Vibrio cholerae. Infected hosts will experience vomiting and severe watery diarrhea and if not treated properly will ultimately succumb to death by dehydration. Due to the global prevalence and severity of cholera, V. cholerae has been extensively studied in both environmental and laboratory settings. Herein, we describe proper V. cholerae maintenance, in addition to classical and El Tor biotype culturing in a laboratory setting.

Establishment of Nile Tilapia Primary Cell Culture Methods and In Vitro Cell Knockdown Techniques.

As an important aquaculture species and research model, Nile tilapia (Oreochromis niloticus) has not yet been systematically studied for the isolation, culture, and in vitro gene manipulation techniques of primary cells from various tissues. This study aimed to explore methods for isolating primary cells from various tissues, as well as developing in vitro gene manipulation techniques in Nile tilapia. Four different Nile tilapia tissues were enzymatically digested and separated using trypsin or collagenase. Collagenase (0.1%) was used for the digestion of the gonads, liver, and heart, while tr

[Comparative evaluation of culture techniques and ELISA test in detection of Helicobacter pylori infection].

The paper evaluates the correlation between culture tested for Helicobacter pylori from biopsy material obtained from patients diagnosed with stomach ulcer, duodenal ulcer and gastritis, and the level of specific IgG antibodies measured by the ELISA test. As compared with culture, the ELISA test yielded a higher percentage of positive assays. Antibody titer was particularly raised in patients with duodenal ulcer, which accounted for 83% of cases. The highest frequency of positive cultures (50%) was also obtained in this group of patients. Essentially, the results are in agreement with findings
