Phencyclidine-like discriminative stimulus properties of opioids in the squirrel monkey.

The opioids SKF 10047, dl-cyclazocine, and dextrorphan have been shown to have phencyclidine (PCP)-like discriminative stimulus properties in the rat. In order to extend the generality of this observation, the stimulus effect of these and other opioids were evaluated in squirrel monkeys trained to discriminate between IM injections of saline and 0.25 mg/kg of PCP in a two-choice discrete-trial avoidance paradigm. Stimulus control of behavior was characterized by the reliable completion of at least 22 trials of a 25-trial session on the appropriate choice lever after an injection of saline or P

Oxilorphan (l-N-cyclopropylmethyl-3,14-dihydroxymorphinan): a new synthetic narcotic antagonist.

Oxilorphan is a fully synthetic morphinan derivative containing the 14-hydroxy group characteristics of naloxone and naltrexone. As a narcotic antagonist, oxilorphan was 4 times more potent than dl-cyclazocine, equipotent to naloxone and about one-fourth as potent as naltrexone parenterally. Duration studies in rodents were inconclusive, but in antagonism of morphine analgesia and miosis in the dog, oxilorphan was longer acting than naloxone and equivalent to dl-cyclazocine. Oxilorphan was inactive in the conventional animal thermal analgesic tests. However, oxilone did exhibit relatively weak

Antagonism of N-methyl-D-aspartate-induced transmitter release in the rat striatum by phencyclidine-like drugs and its relationship to turning behavior.

Representatives from several chemical classes known to share behavioral properties with phencyclidine [(1-phenylcyclohexyl)piperidine] (PCP) have been shown to antagonize the excitatory effects of N-methylaspartate on spinal neurons selectively. This study compared the abilities of several of these PCP-like drugs to inhibit N-methyl-D-aspartate (NMDA)-stimulated efflux of acetylcholine in rat striatum. PCP completely inhibited this parameter with an IC50 value of 68 nM. The IC50 values (nanomolar) found for the other drugs tested were as follows: etoxadrol (98), (-)-cyclazocine (120), N-allyln

Therapeutic usefulness of hallucinogenic drugs as a function of their chemical structure.

D-lysergic acid diethylamide (LSD) displays (1) the phenylethylamine pattern present in mescaline, cyclazocine and catecholamines and (2) the 4-substituted tryptamine structure of psilocybin which is a serotonin analog. Hence (a) Naloxone--a blocker of the LSD-like side effects of cyclazocine--should (and does) block effects of LSD, and (b) cross-tolerance may be present between LSD and cyclazocine but not between mescaline and psilocybin. Even though LSD binds subcortically, its effect on regional perfusion of the brain and, presumably, function is primarily cortical and, since the perfusion

Morphine discrimination in the pigeon using a color tracking procedure.

Pigeons were trained to discriminate 5.0 mg/kg morphine from saline. After morphine, subjects tracked the location of red response keys and after saline, the location of green keys. When stimulus generalization to other drugs was investigated dl-methadone produced morphine-like responding and this response generalization was primarily due to the l-isomer. Pretreatment with 1.0 mg/kg naloxone shifted the morphine generalization curve 10-fold to the right but only shifted the rate suppression curve 3-fold to the right. dl-Cyclazocine generated dose-related increases in responding on the red key

Antitussive effect of the optical isomers of mu, kappa and sigma opiate agonists/antagonists in the cat.

The optical isomer of mu and kappa opiates, when given i.v., inhibited the cough reflex in the lightly anesthetized cat, the levoisomers being, in general, 2 to 14 times more potent than the dextro-isomers. The optical isomers of the sigma agonist, SKF 10,047, did not show any antitussive activity up to near lethal or lethal doses (5 mg/kg i.v.). Naloxone (1 mg/kg i.v.) did not block or reverse the antitussive effects of (-)- and (+)-codeine but completely antagonized the effects of an ED84 of (-)-, (+)-morphine, (-)-, (+)-methadone, levomethorphan and dextromethorphan. Toxic interactions of stimulants, narcotics and narcotic antagonists.

This study examines the lethality of combinations of central nervous system stimulants, narcotics and narcotic antagonists in rodents housed individually. The lethality of mice and rats from d-amphetamine was potentiated by methadone, cyclazocine and morphine, in that order of potency. Naloxone did not enhance lethality from d-amphetamine in mice, whereas it did antagonize lethality from morphine-d-amphetamine interactions. Naloxone did slightly enhance lethality from d-amphetamine in rats, but at a higher dose than those found to antagonize the lethality from morphine-d-amphetamine combinatio

Effects of mu, kappa and sigma opioids on fixed consecutive number responding in rats.

Responding in rats was maintained under a fixed consecutive number 20 schedule. Under this schedule, at least 20 consecutive responses were required on one lever before a response on a second lever produced food. Morphine, buprenorphine, ethylketocyclazocine (ECK), N-allylnormetazocine (NANM) and d-cyclazocine all caused dose-dependent decreases in response rate. With the exception of buprenorphine and EKC, each drug also produced a decrease in the percent of reinforced runs. Differences among the drugs were more apparent, however, on the basis of the conditional probability of switching to th

Effects of phencyclidine-like drugs on punished behavior in rats.

The effects of phencyclidine (PCP) and a number of PCP-like drugs on punished responding in rats were compared to those of chlordiazepoxide and morphine. Twenty rats, trained to lever-press for food reinforcement, were tested under a modified Geller-Seifter procedure using a multiple schedule with responding in one component reinforced under a fixed-interval 60-sec schedule while each response in the other component resulted in both food and brief electric shock presentation. Chlordiazepoxide, PCP, etoxadrol, (+)-N-allylnormetazocine and (-)-beta-cyclazocine all resulted in selective increases

Evaluation of narcotic and narcotic antagonist interactions in primates.

Multiple and single drug interactions were studied in morphine-dependent monkeys whose dependency was maintained by self-infusion. Naloxone, naltrexone, and cyclazocine precipitated abstinence syndrome which the animals generally controlled with increased morphine intake. Methadone and L-alpha-acetylmethadol (LAAM) initially reduced, then increased, the morphine intake. Multiple interactions were studied using ethanol, seconal, diazepam, amphetamine, and diphenylhydantoin.

Effects of kappa opioid agonists on cocaine- and food-maintained responding by rhesus monkeys.

There is accumulating evidence that kappa opioid agonists attenuate cocaine's behavioral effects, and we recently reported that the kappa opioid agonists ethylketocyclazocine (EKC) and U50-488 decreased cocaine self-administration by rhesus monkeys. In the present study, we first examined the effects of acute intramuscular administration of six kappa opioid agonists on responding maintained by food under an FR30 schedule. Each kappa agonist produced dose-dependent decreases in schedule controlled behavior, and the relative potencies were enadoline >/= bremazocine > Mr2033 >/= cyclazocine = spi

5-Hydroxytryptamine and narcotic-analgesics interactions in the intestine.

The effects of 5-hydroxytriptamine (5-HT), 5-HT blocking agents, morphine, narcotic-antagonists and ganglionic blocking agents were tested in the dog intestine by close intra-arterial injection. Morphine, as 5-HT, induced an immediate increase in the intestinal tonus followed by phasic contractions. When 5-HT was injected immediately after cessation of morphine induced phasic contractions, a significant potentiation of the 5-HT induced intestinal contraction could be observed. The potentiation could be demonstrated for other narcotic-analgesics like dextromoramide and it is specific for 5-HT.

Benzomorphan interactions with acetylcholine receptor complexes from Torpedo.

Benzomorphan interactions with acetylcholine receptor complexes from Torpedo californica electric organ were investigated using radiolabelled probes. All of the benzomorphans had minimal affinity for the acetylcholine binding site. Four benzomorphans (N-allylnormetazocine, cyclazocine, phenazocine and pentazocine) inhibited the binding of tritiated perhydrohistrionicotoxin and phencyclidine to ion channel sites with IC50 values between 0.4 and 5 microM. Two other benzomorphans, ketazocine and ethylketocyclazocine, were much less active. Thus, ion channel affinity is not limited to benzomorphan

Examination of the kappa agonist and antagonist properties of opioids in the rat drug discrimination procedure: influence of training dose and intrinsic efficacy.

In a two-choice drug discrimination procedure, rats were trained to discriminate either 0.056 (low dose) or 0.17 (high dose) mg/kg of the kappa opioid bremazocine from saline. Substitution and antagonism tests were then conducted with a variety of opioids. The opioids U50, 488, ethylketocyclazocine, spiradoline, enadoline, and U69,593 substituted completely (>/=80% bremazocine-appropriate responding) for the bremazocine stimulus in both training groups, a finding consistent with their high-efficacy kappa profile. In contrast, the other opioids examined could be classified into different subset

Dopaminergic drugs antagonize the psychotomimetic effects of partial-agonist analgesics.

The psychotomimetic actions of the partial-agonist analgesic drugs cyclazocine and levallophan have been demonstrated using a quantitative behavioural test in young rats which measures lateral head movements and pivoting on the hind paws. This drug induced behaviour is antagonized by low doses of the dopaminergic agonists apomorphine and piribedil, the dopamine releasing drug amphetamine, the dopamine reuptake blocking agent benztropine and by large doses of the dopamine precursor L-Dopa. Naloxone antagonize the behaviour, but only at one hundred times the analgesic antagonist dose. These resu

Serum levels of butorphanol by radioimmunoassay.

A radioimmunassay was developed for the analysis of butorphanol in human serum. The procedure involves extraction of serum with hexane containing 2% isoamyl alcohol, followed by evaporative solvent removal and radioimmunoassay of the reconstituted residue. The antibody significantly cross-reacts with unidentified butorphanol metabolites but not with two known metabolites, hydroxybutorphanol and norbutorphanol. Extraction eliminated interference from the butorphanol metabolites normally present in serum. The antibody also cross-reacts with pentazocine and cyclazocine but not with morphine, hydr
