Knowledge about intracellular signal transduction cascades is largely based on investigations of cultured cells whose responses to different stimuli are typically quantified via RIA, ELISA, or immunoblots. These techniques, which require relatively large amounts of biological material, are performed with homogenized cells and therefore do not allow localization of the molecules under investigation. We describe a protocol for recording dose-response curves directly from immunocytochemical preparations using rat pinealocytes as a model system. The cells were exposed to beta-adrenergic stimuli in

Elevated concentrations of cyclic AMP (cAMP) in the human myometrium may promote uterine quiescence during pregnancy by protein kinase A (PKA)-mediated phosphorylation and subsequent inactivation of myosin light-chain kinase, as well as by the phosphorylation and activation of cAMP-dependent transcription factors. In this context, we show that the altered expression of cAMP response-element binding protein (CREB), cAMP response-element modulator protein (CREM) and activating transcription factor 2 (ATF2) are implicated in the maintenance of myometrial quiescence during fetal maturation and the

In the rat pineal gland neuronal signals determine the rhythmic synthesis of the hormone melatonin. Norepinephrine (NE) is the principal neurotransmitter that drives hormone synthesis by activating the cAMP signaling pathway. This activation depends on transcriptional and posttranscriptional regulatory mechanisms. The cAMP-dependent transcriptional regulation of the rate-limiting enzyme of melatonin synthesis, arylalkylamine-N-acetyltransferase (AA-NAT) involves the activating transcription factor (TF) CREB and the inhibitory TF ICER. By silencing elements of this cAMP-dependent neuroendocrine

Analysis of mice with targeted disruptions of fosB or the gene encoding dopamine beta-hydroxylase suggests that FosB and adrenergic signaling play critical roles in maternal nurturing behavior. The majority of neonates born to null females from either mutation fail to thrive, and virgin mutant females of both lines exhibit impaired pup retrieval. Considering whether FosB and adrenergic signaling might share a signaling pathway important for maternal behavior, we examined the role of a potential intermediary, cyclic AMP response element-binding protein (CREB). Here we report that approximately

Cation channel sperm-associated protein 4 (CATSPER4) is a subunit of the sperm-specific cation/calcium channel, CatSper, located in the principal piece of the sperm flagellum. It is expressed during the late stages of spermatogenesis, and disruption of the gene encoding this protein leads to male infertility. Mutations in Catsper4 are linked to asthenozoospermia. However, the molecular mechanisms regulating Catsper4 expression remain unclear. Here, we present a detailed molecular characterization of the Catsper4 promoter in mice, focusing on the role of the cAMP-responsive element modulator is

The anticipation of day length and duration of darkness is necessary and advantageous for animals to survive and requires a photoperiodic memory. In the Syrian hamster this adaptation to photoperiod is mirrored by seasonal changes in the animal's reproductive state and its liver metabolism. Both events are linked to season-dependent alterations of the nocturnally elevated synthesis of the pineal hormone melatonin. To decipher molecules that are involved in this temporal gating, hamsters were exposed to long photoperiod (16 hr light:8 hr darkness; LP), or short photoperiod (8 hr light:16 hr dar

Natural killer (NK) cells are critical components of the innate immune system, renowned for their ability to recognize and eliminate malignant and infected cells without prior sensitization. NK cell immunotherapy encompasses various approaches, including adoptive transfer of ex vivo expanded NK cells, cytokine stimulation to enhance their activity, and genetic modifications to improve persistence and specificity. CREM is a cAMP-responsive transcription factor that modulates gene expression in response to receptor- and cytokine-driven signaling. Recent evidence now shows that IL-15 and CAR stim

Fusion genes between the FET (EWSR1/FUS) and CREB (CREB1, ATF1, CREM) families characterize many tumor types, including mesenchymal entities. Herein, we tested the diagnostic utility of CREM (C-terminus) immunohistochemistry using 51 CREB-rearranged mesenchymal tumors and 159 tumors of 14 mimicking entities. Staining was considered positive if nuclear staining of moderate or strong intensity was observed in at least 10% of the tumor cells. Among the 51 CREB-rearranged tumors, CREM was at least focally positive in 39 tumors (76.5%), diffusely (≥ 50%) positive in 31 tumors (60.8%), and diffuse s

Cyclic AMP-responsive genes are regulated both positively and negatively by a number of constitutively expressed nuclear proteins. These proteins bind to cAMP-responsive DNA elements in their target genes and they are activated by protein kinase A-mediated phosphorylation. The cAMP response element modulator gene encodes for several constitutively expressed products. However, a second intronic promoter within the gene is inducible and produces another negatively acting transcription factor, inducible cAMP early repressor (ICER). ICER shows a diurnal pattern of expression in the pineal gland, b

Melatonin synthesis in rodents is tightly regulated at the transcriptional level by stimulatory and inhibitory transcription factors. Among them, phosphorylated cAMP-related element binding protein (pCREB) and inducible cAMP early repressor (ICER), a strong inhibitor of cAMP-related element-driven genes, have an antagonistic action in activating/inhibiting the transcription of the Aa-nat gene, which is an important enzyme in melatonin synthesis. In the Syrian hamster, a rodent displaying a seasonal control of reproduction, melatonin synthesis is strongly gated to the second part of the night.

OBJECTIVE: To investigate the effect of miR-199b-5p knockout on the expression of the CREM gene in the semen and testis of mice.
METHODS: We selected 8 miR-199b-5p knockout (KO) male mice (the KO group) and another 8 wild-type (WT) C57BL/6 male mice (the WT group), and observed the changes in their body weight, testicular index, testis structure and sperm morphology. We detected sperm concentration under the microscope, determined the mRNA and protein expression levels of CREM in the semen and testis by RT-qPCR and Western blot respectively, and predicted the targeting relationship between CR

BACKGROUND: Hypertriglyceridaemia (HTG) encompasses a spectrum from common multifactorial disorders to rare monogenic conditions. Variants in cyclic AMP-responsive element-binding protein 3-like protein 3 (CREB3L3), encoding the transcription factor cyclic AMP-responsive element-binding protein H (CREB-H), have emerged as important regulators of triglyceride metabolism.
CASE SUMMARY: A 33-year-old woman presented with severe HTG (peak triglycerides 11.4 mmol/L) in the absence of secondary causes. She had marked obesity [body mass index (BMI) 39 kg/m2] and elevated fat mass (48.6%). Imaging re

BACKGROUND: Intracranial mesenchymal tumors, FET::CREB fusion-positive (ICMT), show fusions involving FET RNA-binding protein family genes (EWSR1 or FUS) and CREB family of transcription factors (ATF1, CREB1, or CREM). The methylation signature(s), gene expression characteristics, and clinical behavior of this important tumor type require further characterization.
METHODS: We study the methylation profiles of 81 ICMT cases (61 newly profiled cases and 20 cases from publicly available sources). Clinicopathologic and genomic data were recorded for each case when available.
RESULTS: ICMT showed

Glucagon-like peptide 1 (GLP-1) is an incretin hormone that is secreted from the enteroendocrine L-cells of the gut in response to nutrient ingestion. GLP-1 enhances both insulin secretion and insulin gene expression in a glucose-dependent manner via activation of its putative G-protein-coupled receptor on pancreatic beta-cells. In the presence of DMSO (0.5-2.5%), these functional responses were enhanced significantly (2- to 2.5-fold) in a concentration-dependent manner in the beta-cell line INS-1, although basal levels were not affected. Rat insulin 1 (rINS1) promoter activity appeared to be

Chronic morphine inhibits interleukin-2 (IL-2) at both the transcriptional and protein synthesis levels. The molecular mechanisms by which morphine decreases IL-2 are not fully understood. The production of IL-2 is tightly regulated by several transcription factors that bind to the IL-2 promoter. Herein, we show that chronic morphine treatment results in an increase in cAMP levels with a concurrent up-regulation of the cAMP inducible repressor inducible cAMP early repressor (ICER)/cAMP response element modulator (CREM) and down-regulation of p-cAMP-response element-binding protein (CREB) in ac

The encoding of photoperiodic information ensues in terms of the daily profile in the expression of cyclic AMP (cAMP)-inducible genes such as the arylalkylamine N-acetyltransferase (AA-NAT) gene that encodes the rate-limiting enzyme in melatonin formation. In the present study, we compared the influence of the photoperiodic history on the cAMP-inducible genes AA-NAT, inducible cyclic AMP early repressor (ICER), fos-related antigen-2 (FRA-2), mitogen-activated protein kinase phosphatase-1 (MKP-1), nerve growth factor inducible gene-A (NGFI-A) and nerve growth factor inducible gene-B (NGFI-B) in

Regulatory T cells (Tregs) are essential for immune tolerance, with cAMP metabolism central to their suppressive function. Inducible cAMP early repressor (ICER) is a negative regulator of cAMP signalling. However, its role in Treg biology is not fully understood. Here we generated Foxp3-specific ICER knockout mice to assess ICER's function in Tregs. We found that ICER deficiency does not impair Treg development or suppression of effector T cell proliferation under homeostatic conditions. However, ICER-deficient Tregs display enhanced Foxp3 stability, increased glycolysis and proliferation but

Nutrition is a key determinant of health and may be regarded as a form of preventive medicine, as an adequate supply of vitamins, fats, proteins, and trace elements is essential for proper immune function. In recent decades, vegetarian and vegan diets have become increasingly popular but may increase the risk of trace element deficiencies if not carefully planned. Zinc deficiency can impair immune responses and reduce resistance to infections. While previous research has mainly focused on animal models or elderly populations, little is known about younger adults. Thus, we investigated the impa

The methylation pattern of non-imprinting genes was little studied, although it is widely known that the abnormal methylation levels of imprinting genes are associated with different forms of male infertility. The purpose of this research was to assess the CREM gene's methylation status and seminal characteristics in infertile individuals who were potential intracytoplasmic sperm injection (ICSI) candidates. A total of 45 semen samples (15 normospermia, 15 asthenospermia, and 15 oligoasthenoteratospermia) were examined. Using aniline blue (AB) staining, we carried out conventional semen analys
