The contrasting epidemiology of Cyclospora and Cryptosporidium among outpatients in Guatemala.

We compared epidemiologic characteristics of Cryptosporidium and Cyclospora in surveillance data from outpatient departments in Guatemala. Routinely-submitted stool specimens were screened by microscopy. Age, sex, and symptom data were collected. Cyclospora was detected in 117 (2.1%) and Cryptosporidium in 67 (1.2%) of 5,520 specimens. The prevalence of Cyclospora peaked in the warmer months, while Cryptosporidium was most common in the rainy season. Both affected children more than adults, but Cryptosporidium affected children at a younger age than Cyclospora (median age 2 years versus 5 year

Cyclospora cayetanensis or Clostridium difficile: Who Caused the Diarrhea?

Cyclospora cayetanensis, a parasite causing human enteritis, has been rarely reported from healthcare laboratories despite its universal presence. While it is usually responsible for self-limiting diarrhea in immunocompetent individuals, we present a case where the patient suffered from severe, unrelenting diarrhea despite having no obvious immune defects. Additionally, there was no history of international travel or consumption of outside food or water. A week after testing positive only for Cyclospora, the patient tested positive for Clostridium difficile as well, and her diarrhea showed no

Fuchsin fluorescence and autofluorescence in Cryptosporidium, Isospora and Cyclospora oocysts.

Cryptosporidium parvum and Isospora belli oocysts stained with carbol-fuchsin, as in a modified Ziehl Neelsen technique, fluoresce bright red under green light (546nm). Cryptosporidium oocysts tend to fluoresce more brightly the less intensely stained they appear under transmitted light; this is not the case with Isospora. Fuchsin-stained Cyclospora cayetanensis oocysts fluoresce rather dimly, but those not taking the dye retain their typical autofluorescence. Cryptosporidium and Isospora oocysts are also autofluorescent, appearing violet under u.v. light (365 nm), and green under violet (405

Cyclospora in AIDS patients: not always an agent of diarrhoic syndrome.

The present study reports the clinical features of two AIDS patients infected by Cyclospora. According to our observations, Cyclospora in AIDS can be responsible both for gastroenteritis and for asymptomatic infections with spontaneous rapid clearance. In addition, and undiagnosed circulation of this agent in Italy could be hypothesized: neither patient had a past history of foreign travel. These are the first two cases in AIDS described in Italy.

Understanding intestinal spore-forming protozoa: cryptosporidia, microsporidia, isospora, and cyclospora.

OBJECTIVES: To summarize recent information about the "new" gastrointestinal protozoal pathogens (cryptosporidia, microsporidia, isospora, and cyclospora) and to help practicing clinicians integrate this information into their clinical databases by emphasizing the similarities among these organisms. DATA SOURCES: Relevant English-language articles published between 1988 and 1995 were identified through a MEDLINE search done using the names of the intestinal spore-forming protozoa. Articles cited in the bibliographies of these and other articles were searched manually. STUDY SELECTION: Studie

[Observations on Blastocystis hominis and Cyclospora cayetanensis in routine parasitological examinations].

We report some observations made from routine parasitological examinations on feces. The methods of Faust et al. and of spontaneous sedimentation in water are not enough to identify Blastocystis hominis. Significant percentage presence of this protozoan was found, especially when staining with iron hematoxylin was performed. Cyclospora cayetanensis was found in 0.7% of the cases, which suggests that this parasite should also routinely be investigated by appropriate techniques.

Cyclospora Genotypic Variations and Associated Epidemiologic Characteristics, United States, 2018-2021.

Seasonal cyclosporiasis outbreaks occur in the United States every year. To better understand the disease, the Centers for Disease Control and Prevention developed a novel genotyping system that successfully clusters nonclonal eukaryotes. We examined temporal-geographic distributions of Cyclospora cluster consensus genotypes (CCGs) and applied regression analyses to identify correlations between Cyclospora spp. parasites and clinical manifestations or epidemiologic risk factors, using data collected during 2018-2021. No CCG was uniquely associated with or consistently detected in a state durin

Sources and Prevalence of Cyclospora cayetanensis in Southeastern U.S. Growing Environments.

Recent cyclosporiasis outbreaks associated with fresh produce grown in the United States highlight the need to better understand Cyclospora cayetanensis prevalence in U.S. agricultural environments. In this study, C. cayetanensis occurrence was assessed in municipal wastewater sludge, on-farm portable toilets, irrigation pond water, and spent packing house dump tank water in a Southeastern Georgia growing region over two years. Detection of the C. cayetanensis 18S rRNA qPCR gene target in pond samples was 0%, 28%, and 42% (N = 217) depending on the detection definition used, and ≤1% in dump ta

Development of a workflow for identification of nuclear genotyping markers for Cyclospora cayetanensis.

Cyclospora cayetanensis is an intestinal parasite responsible for the diarrheal illness, cyclosporiasis. Molecular genotyping, using targeted amplicon sequencing, provides a complementary tool for outbreak investigations, especially when epidemiological data are insufficient for linking cases and identifying clusters. The goal of this study was to identify candidate genotyping markers using a novel workflow for detection of segregating single nucleotide polymorphisms (SNPs) in C. cayetanensis genomes. Four whole C. cayetanensis genomes were compared using this workflow and four candidate marke

Co-infection with Cryptosporidium parvum and Cyclospora cayetanensis in immunocompromised patients.

Forty nine stool specimens collected from severe diarrheic patients. Eight were suffering from Hodgkin's lymphoma, and the rest were suffering from acute lymph plastic leukaemia. All were examined microscopically for protozoan parasites mainly, Cryptosporidium parvum and Cyclospora cayetanensis. Of the patients, 34 (69.4%) were positive and 15 (30.6%) were negative by both microscopy and nested PCR. An additional 12 (24.5%) who were negative by microscopy were positive by nested PCR. Stool examination revealed 16 cases with C. parvum, and 6 with C. cayetanensis, and 3 cases showed mixed infect

Cyclospora cayetanensis comprises at least 3 species that cause human cyclosporiasis.

The apicomplexan parasite Cyclospora cayetanensis causes seasonal foodborne outbreaks of the gastrointestinal illness cyclosporiasis. Prior to the coronavirus disease-2019 pandemic, annually reported cases were increasing in the USA, leading the US Centers for Disease Control and Prevention to develop a genotyping tool to complement cyclosporiasis outbreak investigations. Thousands of US isolates and 1 from China (strain CHN_HEN01) were genotyped by Illumina amplicon sequencing, revealing 2 lineages (A and B). The allelic composition of isolates was examined at each locus. Two nuclear loci (CD

Genotyping genetically heterogeneous Cyclospora cayetanensis infections to complement epidemiological case linkage.

Sexually reproducing pathogens such as Cyclospora cayetanensis often produce genetically heterogeneous infections where the number of unique sequence types detected at any given locus varies depending on which locus is sequenced. The genotypes assigned to these infections quickly become complex when additional loci are analysed. This genetic heterogeneity confounds the utility of traditional sequence-typing and phylogenetic approaches for aiding epidemiological trace-back, and requires new methods to address this complexity. Here, we describe an ensemble of two similarity-based classification

[Newly established causes of diarrhea: the protozoan Cyclospora cayetanensis (Coccidia, Apicomplexa)].

UNLABELLED: Coccidia Cyclospora cavetanesis has been considered a saprophytic or accidental infection without special medical significance until 1984. However, epidemics of diarrhea first in Peru and later in USA. Haiti and Nepal have aroused interest in pathogenic characteristics of this parasite and diseases it causes. The study first deals with biological cycle of parasite development as well as clinical picture and treatment, diagnostics and epidemiology of this parasitic disease. CLINICAL PICTURE: On average the incubation period was 2-11 days with frequent stools (3 or more times a day)

Current Knowledge and Future Directions for Cyclospora cayetanensis Research and Its Surrogates.

Cyclospora cayetanensis is a foodborne protozoan parasite that causes cyclosporiasis, a disease transmitted by the consumption of sporulated oocysts, often via contaminated produce. Since 2018, outbreaks traced back to domestically grown produce in the United States have raised growing concern. Despite its public health significance, research is hindered by methodological challenges, including inability to culture in vitro/in vivo and limited genomic characterization. This review examines current knowledge on its occurrence and transmission, detection methods, host-parasite interactions, genet

Investigation of Cyclospora spp. in farmed fur animals based on PCR-RFLP.

Cyclospora spp., foodborne intestinal protozoa with potential zoonotic risk, are distributed across the globe. However, their presence in animals bred for fur has not been extensively studied, raising concerns about zoonotic transmission. To assess the prevalence of Cyclospora spp. in fur animals in China, we collected fresh fecal samples from 862 animals, including 352 blue foxes, 275 minks, and 235 raccoon dogs, across multiple provinces. The small subunit (SSU) rRNA gene of Cyclospora spp. was amplified using nested PCR, followed by restriction fragment length polymorphism (RFLP) analysis w

Molecular Prevalence of Cyclospora spp. in Tibetan Antelope (Pantholops hodgsonii) in China.

Cyclospora spp. is a globally distributed foodborne intestinal parasite with potential zoonotic risk. The Tibetan antelope (Pantholops hodgsonii), an iconic wildlife species in China, is classified as a national first-class protected animal. However, data on Cyclospora spp. infections in Tibetan antelope remain limited. This study employed polymerase chain reaction-restriction fragment length polymorphism analysis targeting the small subunit ribosomal RNA gene of Cyclospora spp. on 503 fecal samples collected from Tibetan antelope in the Tibet Autonomous Region, Xinjiang Uygur Autonomous Regio

Aqueous Ozone Exposure Inhibits Sporulation in the Cyclospora cayetanensis Surrogate Eimeria acervulina.

Ozone is a potent disinfecting agent used to treat potable water and wastewater, effectively clearing protozoa such as Giardia and Cryptosporidium spp. It is unclear whether ozone treatment of water or fresh produce can reduce the spread of the emerging parasite Cyclospora cayetanensis, which causes cyclosporiasis in humans. Obtaining viable C. cayetanensis oocysts to evaluate inactivation methods is challenging because we lack the means to propagate them in vitro, because of delays in case reporting, and because health departments typically add inactivating fixatives to clinical specimens.
