Use of N-chlorosuccinimide/urea for the selective cleavage of tryptophanyl peptide bonds in proteins. Cytochrome c.

The conditions and utility of the N-chlorosuccinimide/urea (NCS/urea) reagent for the selective cleavage of tryptophanyl peptide bonds in proteins is demonstrated with cytochrome c. At low concentrations of NCS/urea the oxidation of thioether side chains in cytochrome c is the predominant reaction. Methionyl residues are oxidized to sulfoxide and the heme-thioether bridge is partially cleaved. At 10-fold excess of NCS/urea reagent, cleavage of the tryptophanyl peptide bond is optimal at approximately 50% yield in several species of cytochrome c studied. Analytical data on isolated horse cytoch

Biosynthesis of vasopressin in vitro and ultrastructure of a bronchogenic carcinoma. Patient with the syndrome of inappropriate secretion of antidiuretic hormone.

Tumors from patients with the syndrome of inappropriate secretion of antidiuretic hormone (SIADH) have been found to contain large amounts of the antidiuretic hormone vasopressin. A lung tumor from a patient with hyponatremia most likely due to SIADH was removed at surgery and found to contain 23.5 mU vasopressin/g wet weight by radioimmunoassay Slices of this tumor were incubated with phenylalanine-(3)H. Arginine vasopressin-(3)H was purified from the incubate by Sephadex G-25 column chromatography in two different systems, performic acid oxidation, and gradient elution column chromatography

Metabolic Reprogramming in Primary Microglial Cell and Extracellular Vesicle Triggered by Aβ Exposure.

Microglia, key immune cells in the brain, play a pivotal role in brain homeostasis and immune responses. Emerging evidence suggests their critical involvement in Alzheimer's disease (AD) pathogenesis and propagation. The propagation of AD pathology is related to the extracellular matrix of microglia, including extracellular vesicles (EV). Recently, microglia-derived EVs are implicated in inflammatory processes and neuronal death. This study aimed to extensively profile and propose the metabolic role of microglial EVs in AD. Accordingly, we determined the significant alterations of the EV metab

Key locations of oxidative damage in human hair keratins after heat and ultraviolet light exposure.

OBJECTIVE: This study investigated the locations of amino acid modifications within two major human hair keratins (Type I K31 and Type II K85) with probable implications for protein and hair structural component integrity. The particular focus was on cysteine modifications that disrupt intra-protein and inter-protein disulphide bonds. METHODS: Human hair was exposed to accelerated, sequential heat or UV treatments, simulating effects resulting from the use of heated styling tools and environmental exposure over a time frame approximating one year. The proteins were extracted and detected usin

Oxidation of cysteine and glutathione by soluble polymeric MnO2.

The kinetics of reduction of soluble polymeric MnO2 by cysteine and glutathione has been studied in the pH range of 4.0-9.0. The concentration of thiols was varied between 1 and 2 mM, while the MnO2 concentration was varied between 2 and 12 microM. In this pH range, the reaction products were identified as Mn(II) and the corresponding disulfides (cystine and glutathione disulfide). Cysteic or cysteine sulfonic acid was formed only when pH < 2. Experimental data indicate that the rate law over the pH range of 4-9 is first-order in both MnO2 and thiol concentration. Eyring plots for both thiols

Hypohomocysteinemic effect of cysteine is associated with increased plasma cysteine concentration in rats fed diets low in protein and methionine levels.

Rats were fed diets with and without 0.5% L-cysteine supplement for 14 d or shorter periods to clarify the mechanism by which dietary cysteine elicits its hypohomocysteinemic effect. Cysteine supplementation significantly decreased plasma homocysteine concentration with an increase in plasma cysteine concentration in rats fed 10% casein diet (10C) or 15% soybean protein diet (15S) but not in rats fed 25% casein diet (25C) or 25% soybean protein diet. Cysteine supplementation also significantly suppressed hyperhomocysteinemia induced by choline-deprived 10C with an increase in plasma cysteine c

In vitro study of the antiseborrheic activity of zinc L-cysteate, a novel zinc compound, on rat preputial gland.

The antiseborrheic effect of zinc L-cysteate, a new zinc compound, was evaluated in vitro by determining the lipidic metabolic activity of rat preputial glands as measured by incorporation of 14C-sodium acetate. At 10(-3) and 10(-4) mol/l, zinc L-cysteate was more active than S-carboxymethyl L-cysteine used as reference in corresponding concentrations (p < 0.05 and p < 0.01, respectively). The pharmacologic results seem promising for clinical studies in dermatology.

Proteomic analysis of oxidative modifications in human atherosclerotic plaques and control arteries.

Cardiovascular disease is a major cause of worldwide mortality. A prime underlying cause is atherosclerotic plaque formation at specific arterial locations. Considerable evidence links plaques with chronic inflammation and leukocyte activation, with this potentially resulting in enhanced oxidant-mediated damage to arterial proteins. In this study, human carotid artery plaques and control tissue (superior thyroid artery) were analyzed by liquid chromatography-mass spectrometry (LC-MS/MS) for the presence of post-translational modifications (PTMs) generated by redox reactions on proteins. Taurine reduction in anaerobic respiration of Bilophila wadsworthia RZATAU.

Organosulfonates are important natural and man-made compounds, but until recently (T. J. Lie, T. Pitta, E. R. Leadbetter, W. Godchaux III, and J. R. Leadbetter. Arch. Microbiol. 166:204-210, 1996), they were not believed to be dissimilated under anoxic conditions. We also chose to test whether alkane- and arenesulfonates could serve as electron sinks in respiratory metabolism. We generated 60 anoxic enrichment cultures in mineral salts medium which included several potential electron donors and a single organic sulfonate as an electron sink, and we used material from anaerobic digestors in com

Multimode MALDI-MSI deciphers matrine-induced metabolic reprogramming in prostate cancer xenografts: spatial mapping of low-molecular-weight compound alterations.

BACKGROUND: Matrine, a bioactive isoquinoline alkaloid, exhibits antitumor efficacy by modulating multiple signaling pathways to suppress cancer cell proliferation, migration and invasion. However, its metabolic regulatory mechanisms in prostate cancer intervention require systematic characterization. METHODS: We implemented matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) for spatial metabolomic profiling of prostate tissues, integrated with multivariate analytical approaches including principal component analysis, Pearson correlation-based clustering heatmap

The effect of cysteine oxidation on conformational changes of SARS-CoV-2 spike protein using atomistic simulations.

The SARS-CoV-2 Spike (S) protein plays a central role in viral entry into host cells, making it a key target for therapeutic interventions. Oxidative stress, often triggered during viral infections, can cause oxidation of cysteine in this protein. Here we investigate the impact of cysteine oxidation, specifically the formation of cysteic acid, on the conformational dynamics of the SARS-CoV-2 S protein using atomistic simulations. In particular, we examine how cysteine oxidation influences the transitions of the S protein's receptor-binding domain (RBD) between "down" (inaccessible) and "up" (a

Organosulfur compound-driven mobilization of colloidal metals and organic matter from acid mine drainage-contaminated paddy soils.

Although organosulfur compounds dominate the speciation of sulfur in acid mine drainage (AMD)-contaminated paddy soils, their impact on the migration and transformation of heavy metals remains unclear. We conducted soil incubation experiments to investigate organosulfur compound effects on the speciation of mobilized dissolved and colloidal cadmium (Cd), copper (Cu) and organic matter. The addition of cysteine (Cys) as a model thiol or cysteic acid (Cya) as a model sulfonate significantly increased the mobilization of Cd and Cu from the iron (Fe)-manganese (Mn) oxides fraction of soils compare

The neurotoxicity of sulfur-containing amino acids in energy-deprived rat hippocampal slices.

The rat hippocampal slice preparation and its electrophysiology were used to assess the toxicity of two sulfur-containing amino acids, L-cysteate (CA) and L-cysteine (CYS). Both compounds were innocuous under normal conditions but became toxic in energy-deprived (lack of oxygen or glucose) slices. CA and CYS toxicity was apparent as both reduced the number of slices that normally recover their neuronal function (evoked CA1 population spike) after a standardized period of hypoxia or glucose deprivation (GD). The competitive N-methyl-D-aspartate (NMDA) antagonist DL-2-amino-5-phosphonovalerate b

Gut microbiota dysbiosis exacerbates polystyrene microplastics-induced liver inflammation via activating LPS/TLR4 signaling pathway in ducks.

Ubiquitous microplastics can bioaccumulate in organisms, resulting in detrimental health impacts, such as liver inflammation. Nonetheless, the exact mechanism by which polystyrene microplastics (PS-MPs) trigger liver inflammation via the gut-liver axis in ducks remains unclear. The purpose of this study was to clarify the impact of PS-MPs exposure to liver inflammation through the gut-liver axis in ducks. Our investigation indicated that exposure to PS-MPs markedly upregulated the levels of MDA and ROS in the liver tissue and enhanced the release of pro-inflammatory cytokines (TNF-α, IL-6, and

Antisickling Drugs Targeting βCys93 Reduce Iron Oxidation and Oxidative Changes in Sickle Cell Hemoglobin.

Sickle cell disease is a genetic blood disorder caused by a single point mutation in the β globin gene where glutamic acid is replaced by valine at the sixth position of the β chain of hemoglobin (Hb). At low oxygen tension, the polymerization of deoxyHbS into fibers occurs in red blood cells (RBCs) leading to an impaired blood vessel transit. Sickle cell hemoglobin (HbS), when oxidized with hydrogen peroxide (H2O2), stays longer in a highly oxidizing ferryl (Fe4+) form causing irreversible oxidation of βCys93 to a destabilizing cysteic acid. We have previously reported that an antisickling dr
