Diet and microbiome shape small-molecule cytokinin pools in mammals.

Cytokinins (CKs) are adenine-derived metabolites traditionally characterized as plant hormones, yet their origin, distribution, and functions in mammalian systems remain largely undefined. Using integrated metabolomics, microbiome, and metagenomics approaches, we provide a systematic characterization of CK occurrence and potential sources in mammals. Serum profiling across five animal species revealed consistent detection of multiple CK derivatives, with concentrations markedly lower than in plant tissue. The CK storage form, zeatin-O-glucoside, predominated in mammalian sera, followed by tran

TOR coordinates cytokinin and gibberellin signals mediating development and defense.

Plants constantly perceive and process environmental signals and balance between the energetic demands of growth and defense. Growth arrest upon pathogen attack was previously suggested to result from a redirection of the plants' metabolic resources towards the activation of plant defense. The energy sensor Target of Rapamycin (TOR) kinase is a conserved master coordinator of growth and development in all eukaryotes. Although TOR is positioned at the interface between development and defense, little is known about the mechanisms by which TOR may potentially regulate the relationship between th

Cytokinin receptors from structural, evolutionary, and functional perspectives: a review.

Cytokinins are a major class of plant hormones that have been recognized as key regulators of plant growth and development since their biological activity was first described in the 1950s. However, cytokinin receptors were identified only about twenty-five years ago, opening a new field of research into their molecular and physiological functions. Since then, extensive studies have examined receptor ligand-binding properties, evolution, distribution, subcellular localization, hormone cross-talk, and biological roles, including their involvement in plant-microbe interactions. Because of their c

Phytohormone crosstalk research: cytokinin and its crosstalk with other phytohormones.

As signal molecules produced within the plant, phytohormones (plant hormones) control plant growth and development through the regulation of gene expression. They play also a crucial role in the regulation of cellular activities including elongation, cell division and differentiation, organogenesis, pattern formation, reproduction and responses to abiotic and biotic stress conditions. Cytokinins, auxins, gibberellins, abscisic acid and ethylene are five classic phytohormones that had been discovered by the middle of the twentieth century, while strigolactones, brassinosteroids, jasmonates and

Rescue of a human mRNA splicing defect by the plant cytokinin kinetin.

The defective splicing of pre-mRNA is a major cause of human disease. Exon skipping is a common result of splice mutations and has been reported in a wide variety of genetic disorders, yet the underlying mechanism is poorly understood. Often, such mutations are incompletely penetrant, and low levels of normal transcript and protein are maintained. Familial dysautonomia (FD) is caused by mutations in IKBKAP, and all cases described to date involve an intron 20 mutation that results in a unique pattern of tissue-specific exon skipping. Accurate splicing of the mutant IKBKAP allele is particularl

The stability and biological activity of cytokinin metabolites in soybean callus tissue.

The activity, uptake and metabolism of cytokinin metabolites was determined in soybean (Glycine max (L.) Merr.) callus tissue. The following activity sequence was established: zeatin riboside (ZR)>zeatin (Z)>O-glucosides of Z, ZR and their dihydro derivatives>lupinic acid (an alanine conjugate of Z)>7- and 9-glucosides of Z which were almost inactive. The 7- and 9-glucosides and lupinic acid were taken up very slowly by the callus tissue and showed great metabolic stability, but some degradation to 7-glucosyladenine, 9-glucosyladenine and the 9-alanine conjugate of adenine occurred. The cytokinin requirement for cell division in cultured Nicotiana plumbaginifolia cells can be satisfied by yeast Cdc25 protein tyrosine phosphatase: implications for mechanisms of cytokinin response and plant development.

Cultured cells of Nicotiana plumbaginifolia, when deprived of exogenous cytokinin, arrest in G2 phase prior to mitosis and then contain cyclin-dependent protein kinase (CDK) that is inactive because phosphorylated on tyrosine (Tyr). The action of cytokinin in stimulating the activation of CDK by removal of inhibitory phosphorylation from Tyr is not a secondary downstream consequence of other hormone actions but is the key primary effect of the hormone in its stimulation of cell proliferation, since cytokinin could be replaced by expression of cdc25, which encodes the main Cdc2 (CDK)-Tyr dephos

Cytokinin regulates flowering time in Arabidopsis thaliana through the gibberellin pathway.

Cytokinin and gibberellin are two plant hormones promoting flowering in Arabidopsis thaliana grown under short-day conditions. The activities of both hormones depend on similar signaling components, including several elements of the age pathway. However, to date, the interaction between cytokinin and gibberellin in regulating the transition to flowering remains unknown. In this work, we show that exogenous treatment of late-flowering or non-flowering cytokinin mutants by gibberellin restores their ability to flower. In contrast, the promotion of flowering by cytokinin is suppressed by mutation

Auxin and cytokinin regulate growth dynamics underlying carpel initiation in Arabidopsis.

Plant organ initiation requires precise spatial and temporal coordination of cellular behaviors. In Arabidopsis, the gynoecium, the female reproductive structure formed by two fused carpels, is initiated after the termination of the floral meristem. Proper initiation of the carpels is crucial for ovule protection, successful fertilization, and the formation of diverse fruit structures. While the phytohormones auxin and cytokinin are known to regulate organogenesis, their interplay during the earliest stages of carpel development is not fully understood. In particular, how auxin-cytokinin cross

Meta-Topolin as an Aromatic Cytokinin for In Vitro Propagation of Thymus vulgaris L.

Effective in vitro propagation of medicinal and aromatic plants such as Thymus vulgaris is often limited by the choice of optimal cytokinin sources, which are critical for successful shoot proliferation and overall regeneration. Since their discovery, topolins have been recognized as alternatives to traditional cytokinins (CKs) such as benzyladenine (BA) and kinetin (Kin) in plant tissue culture (PTC). This study investigated the influence of three aromatic CKs (meta-topolin (mT), BA, and Kin), on shoot proliferation of thyme, with the goal of improving current PTC protocols. Of all the tested

Investigation of cytokinin-deficient phenotypes in Arabidopsis by ectopic expression of orchid DSCKX1.

The plant hormone cytokinin plays a major role in regulating plant growth and development. Here we generated cytokinin-reduction Arabidopsis plants by overexpressing a heterologous cytokinin oxidase gene DSCKX1 from Dendrobium orchid. These transgenic plants exhibited reduced biomass, rapid root growth, decreased ability to form roots in vitro, and reduced response to cytokinin in growing calli and roots. Furthermore, the expression of KNAT1, STM, and CycD3 genes was significantly reduced in the transgenic plants, suggesting that cytokinin may function to control the cell cycles and shoot/root

Cytokinins Promote Rust Susceptibility in Poplar by Suppressing Defence Signalling, Reducing Defence Accumulation and Reprogramming Primary Metabolism.

Cytokinins (CKs), classically recognised as plant growth regulators, are increasingly implicated in plant-microbe interactions. Studies in herbaceous plants suggest that CKs can function as signalling molecules with dual roles in plant defence, either restricting or facilitating pathogen infection. However, their roles in tree-pathogen interactions remain largely unexplored. Here, we investigated CK induction, metabolism, and functions in black poplar (Populus nigra) during infection by the rust fungus Melampsora larici-populina, an obligate biotrophic pathogen. We found that most isoprenoid C

Exploring the Potential of Plant Cytokinins Against Common Human Pathogens: In Vitro Assessment and In Silico Insights.

Cytokinins, plant hormones derived from adenine, are best known for regulating growth and stress responses in plants. Recent findings suggest they may also influence microbial viability, yet their direct antimicrobial potential remains underexplored. This study evaluates the antimicrobial activities of four natural cytokinins (iPA, B, K, and p-T) and their N9-ribosides (iPAR, BR, KR, and p-TR) against selected human pathogens. Using the broth microdilution method, we assessed their effects on Gram-positive and Gram-negative bacteria, as well as fungal strains. While Gram-negative species showe

A member of the maize isopentenyl transferase gene family, Zea mays isopentenyl transferase 2 (ZmIPT2), encodes a cytokinin biosynthetic enzyme expressed during kernel development. Cytokinin biosynthesis in maize.

Cytokinins (CKs) are plant hormones that regulate a large number of processes associated with plant growth and development such as induction of stomata opening, delayed senescence, suppression of auxin-induced apical dominance, signaling of nitrogen availability, differentiation of plastids and control of sink strength. In maize, CKs are thought to play an important role in establishing seed size and increasing seed set under normal and unfavorable environmental conditions therefore influencing yield. In recent years, the discovery of isopentenyl transferase (IPT) genes in plants has shed ligh

Abscisic Acid-Cytokinin Antagonism Modulates Resistance Against Pseudomonas syringae in Tobacco.

Phytohormones are known as essential regulators of plant defenses, with ethylene, jasmonic acid, and salicylic acid as the central immunity backbone, while other phytohormones have been demonstrated to interact with this. Only recently, a function of the classic phytohormone cytokinin in plant immunity has been described in Arabidopsis, rice, and tobacco. Although interactions of cytokinins with salicylic acid and auxin have been indicated, the complete network of cytokinin interactions with other immunity-relevant phytohormones is not yet understood. Therefore, we studied the interaction of k

Amyloplast formation in cultured tobacco BY-2 cells requires a high cytokinin content.

When cytokinin-autonomous tobacco BY-2 cell cultures are transferred into 2,4-dichlorophenoxyacetic acid (2,4-D)-deprived medium, amyloplast development is initiated. Using this in vitro amyloplast-inducing system, the role of cytokinins in amyloplast formation was investigated. We show that addition of lovastatin, an inhibitor of mevalonate synthesis, to amyloplast-inducing medium reduced starch accumulation. Microscopic observation also revealed that lovastatin treatment decreased starch deposition; however, the overall morphologies of cells and plastids were less affected than control cell

Transcriptomic and physiological analyses reveal that cytokinin is involved in the compound leaf development of alfalfa.

Alfalfa is one of the primary forages, and its yield is largely dependent on the development of its leaf. In this study, to elucidate the mechanism of compound leaf development, we isolated and examined the alfalfa phenotype Chuancao No.7, exhibiting five leaflets. The agronomic traits of the Chuancao No.7 including the leaf blade area, leaf/stem ratio, total fresh weight, and dry weight showed significant increases compared to those of the wild-type. Analyses of forage quality traits indicated significant differences in crude protein (CP), acid detergent fiber (ADF), crude fat (CF), water-sol

Cytokinin Plant Hormones Have Neuroprotective Activity in In Vitro Models of Parkinson's Disease.

Cytokinins are adenine-based phytohormones that regulate key processes in plants, such as cell division and differentiation, root and shoot growth, apical dominance, branching, and seed germination. In preliminary studies, they have also shown protective activities against human neurodegenerative diseases. To extend knowledge of the protection (protective activity) they offer, we investigated activities of natural cytokinins against salsolinol (SAL)-induced toxicity (a Parkinson's disease model) and glutamate (Glu)-induced death of neuron-like dopaminergic SH-SY5Y cells. We found that kinetin-

Cytokinins mediate resistance against Pseudomonas syringae in tobacco through increased antimicrobial phytoalexin synthesis independent of salicylic acid signaling.

Cytokinins are phytohormones that are involved in various regulatory processes throughout plant development, but they are also produced by pathogens and known to modulate plant immunity. A novel transgenic approach enabling autoregulated cytokinin synthesis in response to pathogen infection showed that cytokinins mediate enhanced resistance against the virulent hemibiotrophic pathogen Pseudomonas syringae pv tabaci. This was confirmed by two additional independent transgenic approaches to increase endogenous cytokinin production and by exogenous supply of adenine- and phenylurea-derived cytoki
