Comparative DNA ploidy measurements were carried out by flow cytometry and by image analysis on cells in 71 bladder washing specimens from 50 patients with past histories of bladder tumors. Among the specimens classified as diploid or questionable by flow cytometry, 14 showed the presence of aneuploid DNA values documented by image analysis. In 18 of the 50 patients, recurrent tumors were observed during a relatively brief period of follow-up. In 15 of them the DNA pattern was aneuploid and in three it was questionable. Concerning the content of DNA, there are significant differences between nuclei of cells from the adenoma and those from the carcinoma of the prostate. As it is possible to measure these differences by using flow-through cytophotometry, this method might also be capable of automatizing the cytology of this organ. We collected prostatic glands from 50 unselected autopsies at the Pathology Institute and compared their histologic sections with cytologic preparations and with results of photometric measurements of isolated prostatic cells and isolated nuclei. The results obtained with single cell photometry and flow-through cytophotometry proved to be comparable with one another and with the results of the cytologic and histologic studies. With these methods of cytophotometry we could differentiate inflammatory conditions, microcarcinomas and frank carcinomas from normal and hyperplastic prostatic tissue.

Cytophotometry has been used to study DNA content in oral epithelial cells of Feulgen-stained specimens from a total of 43 patients: 3 with erythema exudativum multiforme (EEM), 5 with pemphigus, 3 with stomatitis aphtosa, 5 with lichen ruber planus, 8 with leukoplakia, and 19 with carcinomas. The prognostic value of cellular DNA content measured by static cytophotometry was evaluated in 69 children with acute lymphoblastic leukemia (ALL) using the pretreatment distribution of the DNA content in blast cells of bone marrow and peripheral blood. By flow microfluorometry the DNA distribution curve of the cell nuclei of 39 adnex tumors, mainly ovarian carcinomas, was measured. 47% of the malignant tumor cell populations were diploid, the other ones were heteroploid (i.e. polyploid, tetraploid, aneuploid stem lines). Heteroploid tumors had the worse prognosis. In 8 cases effusions were analysed (7 ascites, 1 pleura). It is possible to diagnose a malignant tumor from effussion fluid by impulse cytophotometry. Feulgen-DNA cytophotometry was carried out in skin imprint preparations of patients with mycosis fungoides, reticulum-cell sarcoma, lymphomatoid papulosis and Sézary's syndrome. These patients proved to have aneuploid and polyploid DNA histograms. In tumour stage of mycosis fungoides a more bimodal distribution of the DNA values was found. Of 79 cases of suspected malignant reticulosis of the skin, 32 showed an abnormal DNA histogram with 5 percent or more tetraploid and hypertetraploid cells. In 55 patients with urothelial carcinoma of the renal pelvis or ureter, the ploidy, the DNA heterogeneity and the counts of cell cycle phases in the tumor were examined by means of single-cell DNA cytophotometry in order to find more prognostic factors than those already known (stage and grade). Follow-up periods ranged from 1 to 6 years. At the time of first diagnosis, 42 (76%) of the patients had tumors of the renal pelvis, 13 (24%) of them had ureteral tumors. 23 (42%) patients were in stage pT 1 N 0, 15 (27%) in stage pT 2 N 0, 12 (22%) in stage pT 3 N 0, and 5 (9%) were in stage pT 3 N+.

The present study reports results obtained with DNA ploidy measurement by Feulgen cytophotometry and in situ hybridization (ISH) on the same sections of 36 cervical lesions containing human papillomavirus (HPV) DNA. Searching for bladder cancer, 1268 flow-through-cytophotometric determinations of DNA in nuclei of cells were done using the ICP 11. 134 urines (36.2%) and 15 bladder washings (2.9%) could not be evaluated. Lymphocyte transformation with phytohemagglutinin (PHA) was analyzed by karyometry and cytophotometry of single cells. The former showed the distribution of various-sized transforming cells, and the latter the increase of DNA and RNA. Morphometric parameters were measured and specific light optic features of epitheliocyte nuclei examined in the cytologic preparations from 67 patients with various abnormalities of the cervix uteri multi-layer squamous epithelium by a present-day long-distance microimage analyzer.
