3H-labelled R 1881 (methyltrienolone) was bound with high affinity (Kd, 2 . 10(-9) M) and low capacity (11--15 fmol/mg protein)in prostate cytosol from rats castrated 18 h earlier. The binding was inhibited by 5alpha-dihydrotestosterone, testosterone, cyproterone acetate and LEO 1727, but not by estradiol-17beta, R 5020 or corticosterone. The 3H-labelled R 1881-receptor complex was excluded from a Sephadex G=200 column, had a sedimentation coefficient of 3.5 S, was eluted from a DEAE-cellulose column at 0.2 M KCl concentration and precipitated between 20--40% of saturation of ammonium sulfate.

Thyroxine (T4) and triiodothyronine (T3)-binding interactions in preparations of rat anterior pituitary gland have been studied. T4 is bound primarily to extranulcear binding sites located in the cytosol fraction of the cell. The mammalian cytosolic sulfotransferases (SULTs) catalyze the sulfation of endocrine hormones as well as a broad array of drugs, environmental chemicals, and other xenobiotics. Analyses of steroid hormone receptors were performed using a dextran-coated charcoal technique in cytosolic preparations from 40 cases of benign prostatic hyperplasia (BPH) and 10 normal human prostates. The 5 alpha-Dihydrotestosterone (DHT) receptor in cytosol prepared from foreskin fragments was identified and measured by polyacrylamide gel electrophoresis (PAGE). 14 normal subjects, 4 boys with hypospadias and two patients with male pseudohermaphroditism (MPH) were studied. Specimens from 344 cases of endometrial carcinoma, 289 cases of ovarian carcinoma, 177 cases of cervical carcinoma, 10 vulva and 4 fallopian tube carcinoma were assayed for cytosol estrogen and progesterone receptor (ER, PR) contents with dextran-coated charcoal (DCC) method. This report describes the measurement of estrogen and progesterone receptors in cytosols and nuclear fractions from endocervical tissue components. Unoccupied cytosol estrogen receptor levels as determined by Scatchard analysis of [3H]-estradiol binding data indicated a single class of high affinity binding sites for the epithelial-stromal complex (KD = 0.74 x 10(-9) M). Binding was specific for estrogen (estradiol greater than estriol greater than estrone) and unaffected by desoxycorticosterone, dihydrotestosterone and progesterone. Assays for total estrogen receptor verified that 71.6 +/- 5.

The synthetic progestogen R5020 (17,21-dimethyl-19-norpregna-4,9-diene-3,20-dione) binds with high affinity (Ka = 8.8 X 10(8) l/mol at 0 degree C) to the progesterone receptor from rat uterine cytosol.
