Transcriptomic analysis suggests the inhibition of DNA damage repair in green alga Raphidocelis subcapitata exposed to roxithromycin.

Macrolide antibiotics are common contaminants in the aquatic environment. They are toxic to a wide range of primary producers, inhibiting the algal growth and further hindering the delivery of several ecosystem services. Yet the molecular mechanisms of macrolides in algae remain undetermined. The objectives of this study were therefore to: 1. evaluate whether macrolides at the environmentally relevant level inhibit the growth of algae; and 2. test the hypothesis that macrolides bind to ribosome and inhibit protein translocation in algae, as it does in bacteria. In this study, transcriptomic an

Prevalence of the crayfish plague pathogen Aphanomyces astaci in populations of the signal crayfish Pacifastacus leniusculus in France: evaluating the threat to native crayfish.

Aphanomyces astaci, the crayfish plague pathogen, first appeared in Europe in the mid-19(th) century and is still responsible for mass mortalities of native European crayfish. The spread of this parasite across the continent is especially facilitated by invasive North American crayfish species that serve as its reservoir. In France, multiple cases of native crayfish mortalities have been suggested to be connected with the presence of the signal crayfish Pacifastacus leniusculus, which is highly abundant in the country. It shares similar habitats as the native white-clawed crayfish Austropotamo

Biogeographic and phylogenetic diversity of thermoacidophilic cyanidiales in Yellowstone National Park, Japan, and New Zealand.

Members of the rhodophytan order Cyanidiales are unique among phototrophs in their ability to live in extreme environments that combine low pH levels ( approximately 0.2 to 4.0) and moderately high temperatures of 40 to 56 degrees C. These unicellular algae occur in far-flung volcanic areas throughout the earth. Three genera (Cyanidium, Galdieria, and Cyanidioschyzon) are recognized. The phylogenetic diversity of culture isolates of the Cyanidiales from habitats throughout Yellowstone National Park (YNP), three areas in Japan, and seven regions in New Zealand was examined by using the chloropl

An integrated and portable optoelectronic biosensing system for rapid post-extraction on-site quantification of Heterosigma akashiwo.

Rapid, species-level identification of harmful algal blooms (HAB) remains constrained by laboratory-based molecular workflows and bulky fluorescence instrumentation. We report an integrated portable optoelectronic biosensing system for post-extraction quantification of Heterosigma akashiwo 18S rDNA. The key innovation is not the GO-based recognition chemistry itself, but the analytical signal-reading and conversion strategy: GO-regulated fluorescence recovery is captured by a sealed lens-filter-PIN photodiode module and converted through a resistor-based current-to-voltage circuit into an ampl

LSU rDNA based RFLP assays for the routine identification of Gambierdiscus species.

The Gambierdiscus genus is a group of benthic dinoflagellates commonly associated with ciguatera fish poisoning (CFP), which is generally found in tropical or sub-tropical regions around the world. Morphologically similar species within the genus can vary in toxicity; however, species identifications are difficult or sometimes impossible using light microscopy. DNA sequencing of ribosomal RNA genes (rDNA) is thus often used to identify and describe Gambierdiscus species and ribotypes, but the expense and time can be prohibitive for routine culture screening and/or large-scale monitoring progra

Redox modulation of chloroplast DNA replication in Chlamydomonas reinhardtii.

We constructed a plasmid probe containing DNA sequences unique for chloroplast (Cp) genome and nuclear genome of Chlamydomonas reinhardtii. Using this probe and quantitative Southern blot analyses, we determined the content ratio of Cp DNA/nuclear DNA in total DNA isolated from cells grown in different conditions. Algal cells grown photoheterotrophically with acetate as an added carbon source contain the highest amount of Cp DNA compared with cells grown in other conditions tested. We investigated the effect of nitrogen limitation, 5-fluorodeoxyuridine treatment, cadmium exposure, photoautotro

Connectivity of toxigenic Pseudo-nitzschia species assemblages between the Northeast U.S. continental shelf and an adjacent estuary.

Pseudo-nitzschia harmful algal blooms have recently caused elevated domoic acid in coastal environments of the Northeast United States. In 2017, the toxigenic species P. australis was observed in Narragansett Bay, Rhode Island, a temperate estuarine ecosystem, for the first time since 2009 when DNA monitoring for Pseudo-nitzschia species began. This highly toxic species likely contributed to toxin-related shellfish harvest closures and is hypothesized to have been introduced by an offshore source. Little is known about offshore Pseudo-nitzschia spp. populations in the Northeast Continental She

Using Algal Virus Paramecium bursaria Chlorella Virus as a Human Adenovirus Surrogate for Validation of UV Treatment Systems.

Adenovirus is among the most UV-resistant waterborne human pathogens. There is a need to identify nonpathogenic surrogates for adenovirus for the water treatment industry. In this study, the feasibility of using the algal virus Paramecium bursaria chlorella virus (PBCV-1) as an adenovirus surrogate for validation of UV reactors was evaluated. The UV dose-response behavior of PBCV-1 to monochromatic UV radiation at 254 nm and action spectrum for wavelengths ranging from 214 to 289 nm were measured. A culture-based infectivity assay was used to evaluate viral inactivation, and a quantitative PCR

Unraveling the mechanisms underlying the fluorescent probe detection of microcystin-LR and its binding with CT-DNA.

Cyanobacteria blooms are concerning due to algal toxins like microcystin-leucine arginine (MC-LR). Despite progress in detecting MC-LR and understanding its toxic effects, including calf thymus DNA (CT-DNA) damage, the mechanisms for fluorescent probe detection of MC-LR and its binding to CT-DNA are poorly understood. In this study, we designed three fluorescent probes for MC-LR detection. Probe 1, with an acidic recognition site, is effective but influenced by solution pH. Probe 2, featuring a benzene ring structure, shows stable detection regardless of pH. Probe 3 offers the best performance

Granulomatous pneumonia caused by Pythium insidiosum in a central American jaguar, Panthera onca.

A 7-month-old, male jaguar presented with dyspnea and leukocytosis unresponsive to antibiotic therapy. Radiographs revealed unilateral pulmonary consolidation. An exploratory thoracotomy was performed, and the left lung, which contained a large multilobular mass with extensive fibrosis and numerous caseonecrotic foci, was removed. Microscopically, eosinophilic granulomatous inflammation surrounded broad (4.4-8.3 microm) rarely septate hyphae. A diagnosis of Pythium insidiosum infection was confirmed by immunohistochemistry, immunoblot serology, culture, and polymerase chain reaction. Dyspnea r

The Genome of Bangia fuscopurpurea Illustrates Early Establishment of Multicellularity and Intertidal Adaptation in Red Algae.

The billion-year-old fossil red alga Bangiomorpha pubescens represents the oldest taxonomically resolved multicellular eukaryote, morphologically indistinguishable from the extant intertidal alga Bangia fuscopurpurea. Here, we report a chromosome-level 107.55 Mb genome assembly of B. fuscopurpurea. Phylogenomic analysis confirms its placement as the most basal multicellular red algal lineage, diverging ∼973.40 million years ago. The genome reveals lineage-specific expansion of Osvaldo-type LTR retrotransposons that influence generation-specific gene expression. Metabolically, B. fuscopurpurea

Infection Dynamics and Coexistence of Two Novel Arctic Phytoplankton Viruses.

Marine algal viruses exhibit a high level of diversity, and closely related viruses targeting the same algal host species can stably coexist. Here we report an example of a single virus-host system concealing hidden complexity. We discovered two double stranded (ds) DNA viruses infecting the Arctic picophytoplankter Micromonas polaris coexisting in culture for over a decade. Genomic sequencing of the lysate originally characterized as MpoV-44T revealed that it comprises two distinct prasinoviruses with ~203-204 kb genomes (MpoV-44T.A and MpoV-44T.B), of which conserved regions only accounted f

A Case of Prototheca zopfii Genotype 1 Infection in a Dog (Canis lupus familiaris).

Protothecosis is a rare disease caused by environmental algae of the genus Prototheca. These are saprophytic, non-photosynthetic, aerobic, colorless algae that belong to the Chlorellaceae family. Seven different species have been described. Prototheca zopfii genotype 2 and P. wickerhamii are most commonly involved in pathogenic infections in humans and animals. The objective of this work is to describe, for the first time, a case of protothecosis caused by P. zopfii genotype 1 in a dog. The dog, a 4-year-old mix bred male, was presented to a veterinary clinic in Montevideo, Uruguay, with multi

Breaking the cell wall for efficient DNA delivery to diatoms.

Diatoms produce 20% of the world's fixed organic carbon yet remain underutilized as cell factories due to limited genetic engineering tools. Here, we present optimized electroporation and polyethylene glycol (PEG) transformation methods for the model diatom Phaeodactylum tricornutum, enabling delivery of DNA and Cas9 ribonucleoprotein complexes with high efficiency. Transformants are recovered with as little as 1 ng of DNA, and linear or circular episomes as large as 55.6 kb are successfully introduced. The optimized electroporation protocol also reveals an unexpected capability: episomes can

Effect of ozone on algal organic matters as precursors for disinfection by-products production.

The effect of ozone dose on algae (Microcystic aeruginosa), algal extracellular organic matters (EOM), humic acids (HA) and four model compounds: bovine serum albumin (BSA), starch, deoxyribonucleic acid (DNA) and fish oil as precursors for disinfection by-products (DBPs) production was investigated. Algae showed the highest DBPs formation (71.8 microg mg-1 total organic carbon (TOC)) than other samples. Only BSA showed lower chloroform yield (5.9 microg mg-1 TOC) than haloacetic acids, HAAs (11.2 microg mg-1 TOC). Algae, EOM, starch, DNA, fish oil and HA all showed higher chloroform yields (4

Molecular and microscopic assessment of the effects of caffeine, acetaminophen, diclofenac, and their mixtures on river biofilm communities.

The authors examined effects of three common contaminants, caffeine (CF), acetaminophen (AC), and diclofenac (DF), as well as their mixtures on the development, functioning, and biodiversity of river biofilm communities. Biofilms were cultivated in rotating annular reactors. Treatments included AC, CF, DF, AC + CF, AC + DF, CF + DF, AC + CF + DF at 5 µg/L, and their molar equivalent as carbon and nutrients. Incubations using ¹⁴C-labeled AC, DF, and CF indicated that 90% of the CF, 80% of the AC, and less than 2% of the DF were converted to CO₂. Digital imaging revealed a variety of effects on

Toward an inordinate fondness for stars, beetles and Lobophora? Species diversity of the genus Lobophora (Dictyotales, Phaeophyceae) in New Caledonia.

Until the recent use of molecular markers, species diversity of Lobophora, an ecologically important brown algal genus with a worldwide distribution in temperate and tropical seas, has been critically underestimated. Using a DNA-based taxonomic approach, we re-examined diversity of the genus from New Caledonia in the Southwest Pacific Ocean. First, species were delineated using general mixed Yule coalescent-based and barcoding gap approaches applied to a mitochondrial cox3 data set. Results were subsequently confirmed using chloroplast psbA and rbcL data sets. Species delimitation analyses agr
