Withanolides from Datura stramonium: two unprecedented B-ring-seco derivatives and anti-inflammatory active analogues.

UV-guided separation of the Datura stramonium L. extract led to the elucidation of thirteen previously unreported withanolides (1-13), together with nine known analogues (14-22). Compounds 1 and 2 are unprecedented withanolides featuring B-ring cleavage and bearing two formyl (-CHO) groups. The structures of the new withanolides were elucidated on the basis of UV, HRESIMS, and NMR data. The inhibitory effects of the isolated compounds on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in BV2 microglial cells were evaluated. Compounds 16 and 22 exhibited potent in vitro anti-infla…

A case of pediatric age anticholinergic intoxication due to accidental Datura stramonium ingestion admitting with visual hallucination.

Datura stramonium (DS) is a hallucinogenic plant that can produce anticholinergic toxicity because of its significant concentrations of toxic alkaloids, such as atropine, hyoscyamine, and scopolamine. DS grows in both rural and urban areas in Turkey. Clinical findings of toxicity are similar to those of atropine toxicity. DS abuse is common among adolescents because of its hallucinatory effects. However, accidental DS poisoning from contaminated food is very rare. Accidental poisonings are commonly seen among children. Children are more prone to the toxic effects of atropine; ingestion of even…

Integrated serum metabolomics and machine learning for classification of Datura stramonium L. exposure using plant part-specific metabolic signatures.

Datura stramonium L. (DS) poisoning remains a significant toxicological concern in clinical and forensic contexts. In forensic casework, leaf ingestion is mainly associated with accidental exposure, whereas seed ingestion is more frequently linked to suspected intentional poisoning. However, conventional toxicological analysis based on tropane alkaloids cannot distinguish between these exposure sources, limiting forensic interpretation. This study developed a serum metabolomics strategy integrating machine learning to viably discriminate between seed- and leaf-derived Datura exposure using met…
