The growth of sulfate-reducing bacteria (SRB) and associated hydrogen sulfide production can be problematic in a range of industries such that inhibition strategies are needed. A range of SRB can reduce metal ions, a strategy that has been utilized for bioremediation, metal recovery, and synthesis of precious metal catalysts. In some instances, the metal remains bound to the cell surface, and the impact of this coating on bacterial cell division and metabolism has not previously been reported. In this study, Desulfovibrio desulfuricans cells (1g dry weight) enabled the reduction of up to 1500

Microbial conversion of inorganic mercury (IHg) to methylmercury (MeHg) is a significant environmental concern because of the bioaccumulation and biomagnification of toxic MeHg in the food web. Laboratory incubation studies have shown that, despite the presence of large quantities of IHg in cell cultures, MeHg biosynthesis often reaches a plateau or a maximum within hours or a day by an as yet unexplained mechanism. Here we report that mercuric Hg(II) can be taken up rapidly by cells of Desulfovibrio desulfuricans ND132, but a large fraction of the Hg(II) is unavailable for methylation because

Dietary carrageenan (CGN) is a widely used food additive, yet its microbial metabolism in the human gut and health implications remain unclear. This study elucidated a syntrophic pathway wherein Bacteroides thetaiotaomicron (B. theta) degrades CGN via specific sulfatases and glycoside hydrolases, releasing inorganic sulfate. This sulfate fuels the growth and metabolic activity of the sulfate-reducing bacterium Desulfovibrio desulfuricans (D. desulfuricans). In vitro co-culture experiments demonstrated that this cross-feeding interaction significantly enhanced hydrogen sulfide (H2S) production.

Biofilms can methylate mercury (Hg) at higher rates than unattached bacteria and are increasingly recognized as important Hg methylation sites in the environment. Our previous study showed that methylation rates in biofilm cultures were up to 1 order of magnitude greater than those in planktonic cultures of a sulfate-reducing bacterium. To probe whether the differential Hg methylation rates resulted from metabolic differences between these two cultures, Hg methylation assays following molybdate or chloroform inhibition (a specific inhibitor of the acetyl-CoA pathway) were conducted on biofilm

We propose the use of Desulfovibrio desulfuricans ND132 as a model species for understanding the mechanism of microbial Hg methylation. Strain ND132 is an anaerobic dissimilatory sulfate-reducing bacterium (DSRB), isolated from estuarine mid-Chesapeake Bay sediments. It was chosen for study because of its exceptionally high rates of Hg methylation in culture and its metabolic similarity to the lost strain D. desulfuricans LS, the only organism for which methylation pathways have been partially defined. Strain ND132 is an incomplete oxidizer of short-chain fatty acids. It is capable of respirat

The effect of bacterial growth phase is an aspect of mercury (Hg) methylation that previous studies have not investigated in detail. Here we consider the effect of growth phase (mid-log, late-log and late stationary phase) on Hg methylation by the known methylator Desulfovibrio desulfuricans ND132. We tested the addition of Hg alone (chloride-complex), Hg with Suwannee River natural organic matter (SRNOM) (unequilibrated), and Hg equilibrated with SRNOM on monomethylmercury (MMHg) production by ND132 over a growth curve in pyruvate-fumarate media. This NOM did not affect MMHg production even u

The levels of sulfate-reducing bacteria (SRB), including Desulfovibrionaceae, in the gut increase following a fat-enriched diet. Endotoxins from gut microbiota contribute to the inflammation process, leading to metabolic diseases. Thus, we sought to characterize the lipid A structures of Desulfovibrionaceae lipopolysaccharides (LPS) that are associated with the microbiota inflammatory properties. LPS variants were obtained from two SRB isolates from the gut of a single individual. These LPS variants shared similar lipid A moieties with Enterobacterial LPS, but differed from one another with re

Various genera of sulphate reducing bacteria (SRB) have been found in the human digestive tract. It is suggested that some of SRB species may be responsible for the development of the clinical symptoms of ulcerative colitis and other disease of large intestine. Sulphasalazine (salicyl-azo-sulphapyridine, SAS) is commonly used to treat patients with ulcerative colitis and Crohn disease. Above 30 samples of faeces or biopsy specimens from 25 patients (age 45 +/- 14 years; M/F, 13/12) suffering from gastrointestinal disorders were used for isolation of Desulfovibrio desulfuricans species. The mor

A 2.3-kb plasmid present in about 20 copies per genome was identified in extracts of Desulfovibrio desulfuricans G100A and designated pBG1. It appears to be unable to replicate in Escherichia coli. Although composite plasmids of pBG1 inserted into pTZ18U are stable in E. coli, few if any pBG1-specific transcripts are detectable. The plasmid sequence reveals several features typical of the origin of replication of non-ColE1 enterobacterial plasmids as well as several potential open reading frames. This small replicon has been shown to support the replication of recombinant plasmids in D. desulf

Glutathione is the metal stress protector and changes of its level in the sulfate-reducing bacteria cells under the influence of heavy metal salts have not been studied yet. CdCl2, Pb(NO3)2, CuCl2, and ZnCl2 influence on the total glutathione level in cell-free extracts of sulfate-reducing bacteria Desulfovibrio desulfuricans IMV K-6 was studied. The research has been carried out using Ellman, Lowry methods, statistical processing of the results. It was shown that the glutathione level depends on the heavy metal salts concentration in the medium. The total glutathione level was the highest und

Sulfate-reducing bacteria have been suggested to have an etiological role in the development of inflammatory bowel diseases and ulcerative colitis in humans. Traditionally. bismuth compounds have been administered to alleviate gastrointestinal discomfort and disease symptoms. One mechanism by which this treatment occurs is through binding bacterial derived hydrogen sulfide in the intestines. With the addition of bismuth-deferiprone, bismuth-citrate and bismuth subsalicylate to reactions containing cells of D. desulfuricans ATCC 27774, the oxidation of H2 with sulfate as the electron acceptor w

The effects of mercury added as Hg(2+) and selenium as selenite to cultures of the sulfate reducing bacterium Desulfovibrio desulfuricans were investigated under controlled laboratory conditions. There was no significant difference in the growth curves in comparison to control except in the 0.5 μM Hg-6.3 μM Se combined system in which Hg methylation was significantly reduced. A significant decrease in the production of methylmercury indicates a disruption of the methylation process due to the presence of the relatively high concentrations of Se in the system, suggesting a modification of the b

Natural dissolved organic matter (DOM) affects mercury (Hg) redox reactions and anaerobic microbial methylation in the environment. Several studies have shown that DOM can enhance Hg methylation, especially under sulfidic conditions, whereas others show that DOM inhibits Hg methylation due to strong Hg-DOM complexation. In this study, we investigated and compared the effects of DOM on Hg methylation by an iron-reducing bacterium Geobacter sulfurreducens PCA and a sulfate-reducing bacterium Desulfovibrio desulfuricans ND132 under nonsulfidic conditions. The methylation experiment was performed

BACKGROUND: Desulfovibrio desulfuricans (D. desulfuricans), a commensal anaerobic gram-negative rod endemic to the soil environment and human gastrointestinal tract, rarely causes bloodstream infections. We report two rare cases of bacteremia caused by D. desulfuricans in which the intestinal tract was the portal of entry. In addition, we summarize findings on D. desulfuricans.
CASE PRESENTATION: Case 1: A 51-year-old man presented to the emergency department with the chief complaints of fever and right lower abdominal pain. He was admitted to the hospital with ascending colonic diverticuliti

Cultures of the sulfate reducing bacteria Desulfovibrio desulfuricans were grown under anoxic conditions to study the effect of added sulfide, selenite and mercuric ions. A chemical trap consisting in a CuSO4 solution was used to control the poisoning effect induced by the bacterial production of hydrogen sulfide via the precipitation of CuS. Following the addition of Hg(2+), the formation of methylmercury (MeHg) was correlated to bacterial proliferation with most of MeHg found in the culture medium. A large fraction (50-80%) of added Hg(2+) to a culture ended up in a solid phase (Hg(0) and li

The versatility and reaction specificity of a novel bioinorganic catalyst is demonstrated in various reactions. Palladized cells (bioPd) of the sulphate-reducing bacterium Desulfovibrio desulfuricans showed an increased product selectivity and a catalytic activity comparable to a commercial Pd catalyst in several industrially relevant hydrogenations and hydrogenolyses (reductive dehalogenations). The ability of palladized cells to promote the reductive debromination of a polybrominated diphenyl ether (PBDE #47) is demonstrated, although chemically reduced Pd(II) and commercial Pd(0) were more

The sulphate-reducing bacteria (SRB) of genus Desulfovibrio are a group of prokaryotes associated with autism spectrum disorders (ASD). The connection between the elevated numbers of Desulfovibrio in the gut of children with ASD compared with healthy children remains unresolved. A conceivable consequence of SRB overgrowth in the gut is the conversion of bioavailable iron into low-soluble crystalline iron sulphides, causing iron deficiency in the organism. In this study, we report the draft genome sequence and physiological features of the first cultivable isolate from a patient with ASD, Desul

Desulfovibrio spp. are gram-negative anaerobes phylogenetically related to Bacteroides spp., which are rarely isolated and which are mostly isolated from intra-abdominal abscesses. Desulfovibrio desulfuricans clinical isolate D3 had a clavulanic acid-inhibited beta-lactam resistance profile and was resistant to some expanded-spectrum cephalosporins. A beta-lactamase gene, bla(DES-1), was cloned from whole-cell DNA of isolate D3 and expressed in Escherichia coli. Purified beta-lactamase DES-1, with a pI value of 9.1, had a relative molecular mass of ca. 31 kDa and a mature protein of 288 amino
