Identification of the peroxisomal beta-oxidation enzymes involved in the degradation of long-chain dicarboxylic acids.

Dicarboxylic acids (DCAs) are omega-oxidation products of monocarboxylic acids. After activation by a dicarboxylyl-CoA synthetase, the dicarboxylyl-CoA esters are shortened via beta-oxidation. Although it has been studied extensively where this beta-oxidation process takes place, the intracellular site of DCA oxidation has remained controversial. Making use of fibroblasts from patients with defined mitochondrial and peroxisomal fatty acid oxidation defects, we show in this paper that peroxisomes, and not mitochondria, are involved in the beta-oxidation of C16DCA. Additional studies in fibrobla

Dicarboxylate and dicarboxylic acid appended supramolecular self-associating amphiphiles as antimicrobial agents against high priority bacterial pathogens.

Antimicrobial resistance remains a significant global health challenge. Supramolecular self-associating amphiphiles (SSAs) represent a promising class of compounds for development as antimicrobial agents, offering tuneable self-assembly properties alongside phospholipid membrane-targeting capabilities. Here we evaluate a series of 10 chiral SSAs, incorporating either urea-dicarboxylic acid or urea-dicarboxylate functional groups for the first time. This series of compounds has been designed to investigate how structural features including chirality, positioning of multiple polar functionalitie

Dicarboxylic Acid Excretion in Normal Formula-Fed and Breastfed Infants.

PURPOSE: Infant formulas are often supplemented with medium-chain triglycerides (MCTs) to optimize calories for small for gestational age or preterm infants. High amounts of MCTs have been associated with an increase in dicarboxylic acid (DCA) in the urine. Elevated DCA in the urine is also a clinical indicator for fatty acid metabolism disorders. The purpose of this study was to identify if there is an amount of MCTs that can be provided without elevating urinary DCA excretion. METHODS: A metabolic screening laboratory provided urinary DCA excretion data for 175 infants. It was verified that

Recent Advances in the Biosynthesis of Mid- and Long-Chain Dicarboxylic Acids Using Terminally Oxidizing Unconventional Yeasts.

As high-performance monomers for the manufacture of polyamide materials, mid- and long-chain dicarboxylic acids (DCAi, i ≥ 6) have received extensive attention from researchers. Biosynthesis is gradually replacing chemical synthesis due to its outstanding advantages in the industrial production of mid- and long-chain dicarboxylic acids, which is mostly achieved by using the strong terminal oxidation ability of nonmodel microorganisms such as Candida tropicalis to oxidize hydrophobic substrates such as alkanes. Here, we first summarize the metabolic pathways of oxidative alkane conversion into

Multicomponent ionic crystals of diltiazem with dicarboxylic acids toward understanding the structural aspects driving the drug-release.

Diltiazem (DIL) is a calcium channel blocker antihypertensive drug commonly used in the treatment of cardiovascular disorders. Due to the high solubility and prompt dissolution of the commercial form hydrochloride (DIL-HCl) that is closely related to short elimination drug half-life, this API is known for exhibiting an unfitted pharmacokinetic profile. In an attempt to understand how engineered multicomponent ionic crystals of DIL with dicarboxylic acids can minimize these undesirable biopharmaceutical attributes, herein, we have focused on the development of less soluble and slower dissolving

Reactive uptake of N2O5 by aerosols containing dicarboxylic acids. Effect of particle phase, composition, and nitrate content.

Reactive uptake coefficients for loss of N(2)O(5) to micron-size aerosols containing oxalic malonic, succinic, and glutaric acids, and mixtures with ammonium hydrogen sulfate and ammonium sulfate, are presented. The uptake measurements were made using two different systems: atmospheric pressure laminar flow tube reactor (Cambridge) and the Large Indoor Aerosol Chamber at Forschungszentrum Juelich. Generally good agreement is observed for the data recorded using the two techniques. Measured uptake coefficients lie in the range 5 x 10(-4)-3 x 10(-2), dependent on relative humidity, on particle p

An FDA‑approved dicarboxylic acid allosterically targets PNPLA3 to resolve osteoarthritis.

UNLABELLED: Osteoarthritis (OA) lacks disease‑modifying therapies. The infrapatellar fat pad (IPFP) is hypothesized to regulate joint metabolism, but its molecular effectors are unknown. We investigated whether IPFP‑derived metabolites protect cartilage via lipid‑metabolizing enzymes. METHODS: Non‑targeted metabolomics, IPFP excision/transplantation, single‑cell RNA‑seq, molecular dynamics, enzyme kinetics, and AAV‑mediated gene silencing were used in patient samples and the DMM mouse model. RESULTS: Azelaic acid (AA) was the most significantly reduced metabolite in OA IPFP, and its levels c

Towards sustainable long-chain dicarboxylic acid production: challenges and opportunities.

Long-chain dicarboxylic acids are versatile aliphatic compounds with two terminal carboxyl groups and hydrocarbon chains of 12 or more carbons, offering tunable thermal, mechanical, and chemical properties. They are widely used in polymers, lubricants, adhesives, pharmaceuticals, and other industrial chemicals. Conventional chemical synthesis relies on fossil feedstocks, harsh reagents, and energy-intensive processes, generating hazardous by-products and raising sustainability concerns. Microbial production has emerged as a greener alternative, exploiting α- and ω-oxidation pathways in oleagin

Acquired deficiency of peroxisomal dicarboxylic acid catabolism is a metabolic vulnerability in hepatoblastoma.

Metabolic reprogramming provides transformed cells with proliferative and/or survival advantages. Capitalizing on this therapeutically, however, has been only moderately successful because of the relatively small magnitude of these differences and because cancers may further adapt their metabolism to evade metabolic pathway inhibition. Mice lacking the peroxisomal bifunctional enzyme enoyl-CoA hydratase/3-hydroxyacyl CoA dehydrogenase (Ehhadh) and supplemented with the 12-carbon fatty acid lauric acid (C12) accumulate the toxic metabolite dodecanedioic acid (DDDA), which causes acute hepatocyt

Grease trap waste, a renewable resource for biosynthesis of long-chain dicarboxylic acids using Candida tropicalis ATCC 20962.

Long-chain dicarboxylic acids (LCDAs) are valuable chemicals with a variety of industrial applications. However, since they are typically fossil-derived, there is a need for renewable feedstocks. This study investigated grease trap waste (GTW), an underutilised waste stream, for LCDA production using Candida tropicalis. Several experiments optimised glucose and GTW feed rates and pH, resulting in a final titre of 34.7 g/L LCDAs with a productivity of 0.42 g/(L·h) and a yield of 76 % after 140 h. Optimal conditions included a glucose feed rate of 0.38 g/(L·h), as lower feed rates resulted in gl

Urine dicarboxylic acids change in pre-symptomatic Alzheimer's disease and reflect loss of energy capacity and hippocampal volume.

Non-invasive biomarkers will enable widespread screening and early diagnosis of Alzheimer's disease (AD). We hypothesized that the considerable loss of brain tissue in AD will result in detection of brain lipid components in urine, and that these will change in concert with CSF and brain biomarkers of AD. We examined urine dicarboxylic acids (DCA) of carbon length 3-10 to reflect products of oxidative damage and energy generation or balance that may account for changes in brain function in AD. Mean C4-C5 DCAs were lower and mean C7-C10 DCAs were higher in the urine from AD compared to cognitiv

The anterior midgut of larval yellow fever mosquitoes (Aedes aegypti): effects of amino acids, dicarboxylic acids, and glucose on the transepithelial voltage and strong luminal alkalinization.

Isolated anterior midguts of larval Aedes aegypti were bathed in aerated mosquito saline containing serotonin (0.2 micromol L(-1)) and perfused with NaCl (100 mmol L(-1)). The lumen negative transepithelial voltage (V(te)) was measured and luminal alkalinization was determined through the color change of luminal m-cresol purple from yellow to purple after luminal perfusion stops. Addition of 10 mmol L(-1) amino acids (arginine, glutamine, histidine or proline) or dicarboxylic acids (malate or succinate) to the luminal perfusate resulted in more negative V(te) values, whereas addition of glucos

Optimization of protocol for analysis of dicarboxylic acids in ambient aerosol samples using GC-MS: method comparison and application.

Dicarboxylic acids (DCAs), with their deliquescence and hygroscopic nature, can function as cloud condensation nuclei (CCN) and ice nuclei (IN), affecting rainfall patterns. DCA analysis can serve as organic molecular markers for anthropogenic and biogenic sources. Very few studies deal with the optimization of the protocol for qualitative and quantitative analysis of DCAs using gas chromatography-mass spectrometry (GC-MS). In this study, we have optimized the extraction of DCAs from aerosol samples by employing several organic solvents with differing relative polarities. Extraction efficienci

Extracellular lipids of Camelina sativa: Characterization of cutin and suberin reveals typical polyester monomers and unusual dicarboxylic fatty acids.

Camelina sativa is relatively drought tolerant and requires less fertilizer than other oilseed crops. Various lipid- and phenolic-based extracellular barriers of plants help to protect them against biotic and abiotic stresses. These barriers, which consist of solvent-insoluble polymeric frameworks and solvent-extractable waxes, include the cuticle of aerial plant surfaces and suberized cell walls found, for example, in periderms and seed coats. Cutin, the polymeric matrix of the cuticle, and the aliphatic domain of suberin are fatty acid- and glycerol-based polyesters. These polyesters were in

N-hydroxysulfosuccinimide active esters: bis(N-hydroxysulfosuccinimide) esters of two dicarboxylic acids are hydrophilic, membrane-impermeant, protein cross-linkers.

We have synthesized and characterized N-hydroxysulfosuccinimide, a new hydrophilic ligand for the preparation of active esters. We have incorporated this ligand into two new protein cross-linking reagents, 3,3'-dithiobis(sulfosuccinimidyl propionate) and bis(sulfosuccinimidyl) suberate. In experiments with rabbit muscle aldolase, it is demonstrated that both of these reagents are highly efficient protein cross-linkers at physiological pH and that 3,3'-dithiobis(sulfosuccinimidyl propionate) is quantitatively cleavable by reduction under mild conditions. In experiments with intact human erythro

Synthesis of Succinic Acids via Electrochemical Dicarboxylation of Aryl Terminal Alkynes with CO2.

An environment-friendly and efficient method for the synthesis of succinic acids has been developed through the electrochemical dicarboxylation of aryl terminal alkynes with CO2. The reaction proceeds smoothly at a certain constant current in acetonitrile with manganese(II) carbonate (MnCO3) as the additive, giving the desired succinic acids with up to 85% isolated yield. The system is applicable to various aryl terminal alkynes, exhibiting good functional group tolerance. This method may open up a window for the application of the electrochemical strategy in the synthesis of multicarboxylic a

The chemical conditions on the parent body of the Murchison meteorite: some conclusions based on amino, hydroxy and dicarboxylic acids.

Amino and hydroxy acids have been identified in the Murchison meteorite. Their presence is consistent with a synthetic pathway involving aldehydes, hydrogen cyanide and ammonia in an aqueous environment (Strecker-cyanohydrin synthesis). From the various equilibrium and rate constants involved in this synthesis, four independent estimates of the ammonium ion concentrations on the parent body at the time of compound synthesis are obtained; all values are about 2 x 10(-3) M. Succinic acid and beta-alanine have also been detected in the Murchison meteorite. Their presence is consistent with a synt

Coordination Compounds of Cu, Zn, and Ni with Dicarboxylic Acids and N Donor Ligands, and Their Biological Activity: A Review.

Complexes of carboxylic acids are very often studied due to their interesting structural, spectral, and magnetic properties. This review is focused on complexes of four dicarboxylic acids, namely, 2,2'-thiodioacetic, 3,3'-thiodipropionic, 3,3'-dithiodipropionic, and fumaric acid. Many of the complexes were characterized by single crystal X-ray analyses. Without the analyses, it is very difficult to predict the coordination mode of carboxylate groups or nitrogen ligands on central atoms. Thus, structural properties are also discussed, as well as potential applications.
