Pharmacological analysis of rebound excitation in large intestine of piebald mouse model for Hirschsprung's disease.

Pharmacological agents that might influence the poststimulus rebound contractile response (PSRR) through an action on the intrinsic inhibitory innervation of the circular muscle coat were studied in the large intestine of the piebald mouse model for Hirschsprung's disease. Tetrodotoxin (0.3-3 microM) suppressed, in a dose-dependent manner, the amplitude, duration, and area under the contractile traces of the PSRR and increased the latency for the response in ganglionated segments of the large intestine of piebald and normal mice. Sensitivity to tetrodotoxin was greater in the megacolon of pieb

Evidence for a respiration-modulated cholinergic action on the activity of medullary respiration-related neurons in the rabbit. An iontophoretic study.

Effects of the iontophoretically administered cholinergic agonists acetylcholine, bethanechol and DMPP on the activity of medullary respiration-related neurons were examined in urethane-anaesthetized rabbits. Inhibitory effects prevailed over excitatory effects. Analysis of cholinergic effects by cycle-triggered averaging revealed three major types of neuronal responses: (i) constant alterations of spike-density throughout the whole period of activity ("constant effects"), (ii) effects increasing during the progression of the burst of discharge or effects restricted to a particular fraction of

Alterations of epidermal lectin binding sites in acute contact dermatitis.

We investigated alterations of epidermal lectin binding sites, as well as of pemphigus and bullous pemphigoid antigens, in 28 human patch test reactions, both allergic (nickel, formaldehyde, N,N'-1,3-dimethylbutyl-N'-phenylenediamine) and irritant (sodium lauryl sulfate). The epidermal reactivity to a panel of lectins and human antisera to pemphigus vulgaris and bullous pemphigoid antigens was compared with samples obtained from normal skin and from skin under tape occlusion. We observed selective perturbations of lectin and antibody binding in acute contact dermatitis, whether allergic or irr

Pharmacologic potency and selectivity of a new bronchodilator agent, 2,3-dihydro-7-methyl-9-phenyl-1H-pyrazolo [1,2-a] indazolium bromide (FKK) on canine tracheal preparations "in vivo" and "in vitro".

In tracheal preparations of dogs anesthetized with pentobarbital, the bronchodilating activity and selectivity of FKK were compared with those of aminophylline and isoproterenol. When administered by either the i.a., i.v. or i.d. route in blood-perfused or nonperfused tracheal preparations, FKK produced a dose-dependent and long-lasting tracheal dilatation through all the administration routes, but unlike aminophylline and isoproterenol it hardly affected systemic blood pressure (SBP), heart rate (HR) and tracheal blood flow (TBF). The ability of i.a. and i.v. FKK to reduce the tracheal intral

The actions of bretylium: adrenergic neurone blocking and other effects.

Bretylium caused a specific and lasting depression of many excitatory and inhibitory responses evoked by electrical stimulation of the peripheral sympathetic nervous system, probably by impairing conduction of impulses in adrenergic neurones with consequent failure of noradrenaline and adrenaline release. This effect, which will be referred to as the adrenergic neurone blocking action, was preceded by weak sympathomimetic effects. In the presence of bretylium the effects of adrenaline and noradrenaline were increased, as after sympathectomy. Concentrations producing blocking of adrenergic neur

Synthesis and pharmacology of alkanediguanidinium compounds that block the neuronal nicotinic acetylcholine receptor.

Taking as models the polyamine toxin fraction FTX from the funnel-web spider venom, and the guanidinium moiety of guanethidine, a series of azaalkane-1, omega-diguanidinium salts were obtained. Some of them blocked ion fluxes through the neuronal nicotinic receptors for acetylcholine (nAChR). The blockade was exerted at submicromolar concentrations, suggesting a highly selective interaction with the nAChR. In fact, the active compounds on the nAChR ion channel did not recognize the voltage-dependent Na+ or Ca2+ channels of bovine adrenal chromaffin cells. Therefore, these compounds may be usef

Identical patterns of somatostatin secretion from isolated antrum and fundus of rat stomach.

The patterns of somatostatin secretion from the fundus, the main source of somatostatin, and the antrum, the site of paracrine regulation of gastrin secretion, were examined using perifused antral and fundic segments from rat stomach. Gastrin secretion fundic segments from rat stomach. Gastrin secretion could be obtained from antral segments only. Indirect intestinal stimulatory effects of heroin: direct action on opiate receptors.

The effects of bolus intra-arterial doses of heroin and other stimulant drugs were studied in vascularly perfused isolated segments of dog small intestine. Heroin caused dose-related increases in intraluminal pressure similar in appearance to those caused by morphine. Perfusion with Krebs bicarbonate solution containing naloxone selectively abolished intestinal responses to heroin. Perfusion with cinanserin, a 5-hydroxytryptamine (5-HT) antagonist, decreased intestinal responses to 5-HT and heroin without affecting responses to dimethylphenylpiperazinium (DMPP) or bethanechol (BeCh). Supersensitivity induced by the toxin of Brazilian scorpion venom (Tityus serrulatus Lutz e Mello).

The purified toxin of the Brazilian scorpion venom (TsTX) enhances the pharmacological actions of directly and indirectly acting substances on several preparations. The enhancing action of TsTX is more pronounced for the indirectly acting drugs, which is highly suggestive of a prejunctional site of action. Secondarily, TsTX does also produce an unspecific increase of sensitivity at the level of the pharmacological receptor itself. The mechanism by which these changes are brought about remains to be determined.

Modulation of memory processing in the cingulate cortex of mice.

To evaluate the possible role of the cingulate cortex in memory processing for training using a noxious stimulus, we trained mice on foot shock avoidance in a T-maze. Cholinergic, GABAergic, serotonergic, and glutamatergic agonists and antagonists were administered into the cingulate cortex immediately after training. Retention for the foot shock avoidance training was tested 1 week later. The results indicate that muscarinic and nicotinic agonists improved retention, while antagonists impaired it. GABA and serotonin agonists impaired retention, while antagonists improved it. Drugs acting on G

Stimulation of acetylcholine release from guinea-pig ileal synaptosomes by cyclic nucleotides and forskolin.

Various agents which are known to affect intracellular levels of cAMP have been assessed for their ability to induce the release of [3H]acetylcholine ([3H]ACH) from a synaptosomal preparation derived from the guinea-pig ileum myenteric plexus. 8-Bromo-cAMP increased the release of [3H]ACh above basal levels. While 8-bromo-cGMP also increased the release, this nucleotide was far less potent than 8-bromo-cAMP. Comparison of the release caused by the cyclic nucleotides to the release induced by the nicotinic agonist dimethylphenylpiperazinium (DMPP) suggested that there is some relationship, as y

[Effects of selective applications of drugs affecting ganglionic transmission to the hypogastric ganglion of the guinea pig (author's transl)].

Effects of selective applications of drugs to the guinea pig hypogastric ganglion were studied on the contractile response of the vas deferens to the hypogastric nerve stimulation or to ganglionic stimulating drugs. Both acetylcholine (ACh) and dimethylphenylpiperazinium (DMPP) contracted the vas deferens. Neostigmine potentiated the ACh-induced contractions but not those which were DMPP-induced. Hemicholinium-3 (HC-3) reduced the response to nerve stimulation (R-NS) but affected neither the ACh-induced contractions nor the DMPP-induced contractions. Morphine blocked R-NS but not the ACh-induc

Nicotinic cholinergic agonists inhibit androgen biosynthesis by cultured rat testicular cells.

The actions of cholinomimetics on androgen biosynthesis were investigated using a primary culture of testicular cells derived from adult hypophysectomized rats. Testicular cells were maintained in serum-free medium and treated with hCG and/or drugs on day 8 of culture. Media were collected 2 days later for measurement of steroids and cAMP. Treatment with hCG increased testosterone production 50-fold over control values, whereas treatment with cholinomimetics alone failed to increase androgen production. Concomitant treatment of testicular cells with nicotinic cholinergic agonists (lobeline, ni

Cardiovascular responses to cholinergic agonists in sinoaortic denervated rats.

1. The cardiovascular effect of systemic nicotinic and muscarinic cholinergic stimulation were studied in conscious sham operated and sinoaortic denervated (SAD) rats, 7 days after the corresponding operation. 2. The administration of the nicotinic ganglionic agent, 1,1-dimethyl-4-phenylpiperazinium (DMPP, 50-100 micrograms.kg-1, i.v.) induced a fall of heart rate that was significantly higher in SAD rats than in sham rats. DMPP induced in sham rats an increase of arterial pressure but in SAD animals a biphasic response: an initial hypotension followed by an increase of arterial pressure. 3. U

Differential effects of prostaglandin E2 on contractions of airway smooth muscle.

In an in vitro muscle bath, the active tension generated by strips of canine tracheal smooth muscle responding to cumulative additions of either histamine (10(-8) to 10(-3) M) or acetylcholine (10(-9) to 10(-3) M) was measured in the absence and presence of prostaglandin E2 (PGE2) (10(-6) to 10(-5) M). When contractile responses of equal magnitude were compared, the contractions elicited by acetylcholine were resistant to the inhibitory effects of PGE2, relative to comparable contractions elicited by histamine. Otilonium: a potent blocker of neuronal nicotinic ACh receptors in bovine chromaffin cells.

1. Otilonium, a clinically useful spasmolytic, behaves as a potent blocker of neuronal nicotinic acetylcholine receptors (AChR) as well as a mild wide-spectrum Ca2+ channel blocker in bovine adrenal chromaffin cells. 2. 45Ca2+ uptake into chromaffin cells stimulated with high K+ (70 mM, 1 min) was blocked by otilonium with an IC50 of 7.6 microM. The drug inhibited the 45Ca2+ uptake stimulated by the nicotinic AChR agonist, dimethylphenylpiperazinium (DMPP) with a 79 fold higher potency (IC50 = 0.096 microM). 3. Excitatory 5-hydroxytryptamine receptors in the cat urinary bladder are of the M- and 5-HT2-type.

In anaesthetized cats, a biphasic contraction of the urinary bladder, consisting of an initial spike followed by a longer-lasting contracture, was elicited by 5-hydroxytryptamine (5-HT) in doses of 3-30 micrograms/kg, i.a. Administration of MDL 72222 (30, 100 and 300 micrograms/kg, i.v.), which specifically antagonizes the M-type 5-HT receptors, selectively blocked the spike phase of the 5-HT response. Role of K+ channels in adrenal catecholamine secretion in anesthetized dogs.

We examined the role of K+ channels in the secretion of adrenal catecholamine (CA) in response to splanchnic nerve stimulation (SNS), acetylcholine (ACh), 1,1-dimethyl-4-phenyl-piperazinium (DMPP), and muscarine in anesthetized dogs. K+ channel blockers and the cholinergic agonists were infused and injected, respectively, into the adrenal gland. The voltage-dependent K+ channel (KA type) blocker mast cell degranulating (MCD) peptide infusion (10-100 ng/min) enhanced increases in CA output induced by SNS (1-3 Hz), but it did not affect increases in CA output induced by ACh (0.75-3 micrograms),
