Acute diquat poisoning in children: a single-center retrospective observational study.

BACKGROUND: Acute diquat poisoning can cause multi-organ dysfunction and even death. The dose-effect relationship and multi-organ injury patterns of diquat poisoning in children remain unclear. OBJECTIVE: This study analyzed 58 pediatric cases to systematically characterize clinical features and investigate the relationship between intake dose, organ injury patterns and prognosis. METHODS: Patients were divided into three groups according to the volume of diquat ingested: group A (≤20 mL, n = 24), group B (21-50 mL, n = 15), and group C (>50 mL, n = 19). General clinical information, inciden

Diquat exposure causes brainstem demyelination by upregulating the mitochondrial calcium uniporter.

Diquat (DQ) is a widely used new herbicide that poses a great threat to the environment, ecological systems and human health. Although the central nervous system (CNS) is a sensitive target of DQ exposure, the major brain regions, pathological changes and underlying mechanisms of DQ damage to the CNS remain obscure. We demonstrated that the brainstem was the primary region where DQ damaged the CNS. DQ exposure damaged both neurons and glial cells and disrupted neurotransmitter metabolism. DQ caused brainstem demyelination, as indicated by the loss of myelin sheaths, decreased levels of myelina

Diquat induces renal proximal tubule injury in glutathione reductase-deficient mice.

Reactive oxygen species (ROS) have been associated with many human diseases, and glutathione (GSH)-dependent processes are pivotal in limiting tissue damage. To test the hypothesis that Gr1(a1Neu) (Neu) mice, which do not express glutathione reductase (GR), would be more susceptible than are wild-type mice to ROS-mediated injury, we studied the effects of diquat, a redox cycling toxicant. Neu mice exhibited modest, dose- and time-dependent elevations in plasma alanine aminotransferase (ALT) activities, 126+/-36 U/l at 2 h after 5 micromol/kg of diquat, but no ALT elevations were observed in di

Exposure Impacts of Diquat dibromide herbicide formulation on amphibian larval development.

Many anthropogenic chemicals in general, and specifically aquatic herbicide formulations have the potential to modulate the thyroid pathways of the endocrine system of aquatic organisms, because they are normally applied directly into the aquatic system, to manage aquatic weeds. These thyroidal effects have been widely linked with disruption in developmental and reproductive processes. In fact, the exposure impacts of many of these substances on metamorphic organisms could produce a precocious metamorphosis. Using Xenopus Metamorphosis Assay (XEMA) protocol, this study assessed the thyroidal e

Intrastriatal pre-treatment with L-NAME protects rats from diquat neurotoxcity.

INTRODUCTION: Contact herbicide diquat (DQ), redox cycling compound, mediates its systemic toxicity throughout the enlarged production of free radicals. Target organs are liver and kidney in humans. To-date, the mechanism of DQ-induced neurotoxicity has not been rationalized. OBJECTIVE: The objectives of the study were to examine the ability of DQ to induce oxidative stress (OS) and/or nitrosative stress (NS) upon intrastriatal (i.s.) administration and to investigate the role of nitric oxide (NO(x)) using NG-nitro-L-arginine methyl ester (L-NAME), a non-selective inhibitor of nitric oxide sy

Diquat increases cysteine proteinase inhibitors greatly in rat plasma and tissues.

Biochemical and gross pathological effects of diquat were studied with special attention to cysteine proteinase inhibitor level which was often increased in acute and chronic disorder. Diquat was fed continuously to rats at the dose of 1000 ppm in the diet. After 10 days, anorexia and severe diarrhea were observed but epistaxis and hypokinesia were not apparent. The rats were killed after feeding the diet for 13.5 days and plasma components such as acute phase reactant proteins and some vitamins which act as antioxidants were examined. The results showed that alpha-cysteine proteinase inhibito

Liver membrane calcium transport in diquat-induced oxidative stress in vivo.

Hepatic necrosis is produced rapidly by 0.1 mmol/kg diquat in male Fischer-344 rats but not Sprague-Dawley rats, yet massive oxidant stress is caused by diquat in both strains of rat. Liver plasma membrane calcium uptake was unaltered by diquat treatment in either strain. However, diquat inhibited ATP-dependent calcium sequestration by hepatic microsomes from Fischer rats by 33% (33 +/- 2 versus 50 +/- 2 nmol/mg/20 min), whereas liver microsomal calcium uptake in Sprague-Dawley rats was not decreased by diquat treatment. Microsomes of diquat-treated Fischer rats showed marked increases in calc

Toxicological effects of diquat on the central nervous system and associated treatment challenges.

BACKGROUND: Diquat is a highly toxic pyridine-based herbicide, and its central nervous system (CNS) toxicity is a major factor contributing to the high mortality rate and poor neurological outcomes in poisoned patients. Compared with other organ injuries, the complex molecular mechanisms underlying diquat-induced CNS damage and effective neuroprotective strategies remain poorly understood. METHODS: A systematic search and screening of relevant literature on diquat-induced CNS toxicity were conducted. Based on predefined inclusion and exclusion criteria, 21 articles were selected from 424 retr

Acute diquat poisoning resulting in toxic encephalopathy: a report of three cases.

INTRODUCTION: Diquat-related acute kidney injury is well-known. However, neurological disorders caused by diquat are often underestimated, and changes in the imaging findings are rarely reported. We present three cases of acute diquat poisoning resulting in toxic encephalopathy. CASE REPORT: In the first case, a 20-year-old previously healthy man ingested approximately 80-100 mL of diquat. He developed acute renal failure, neurological disorders, and respiratory failure. Central pontine myelinolysis was considered by magnetic resonance imaging (MRI), 18 days after ingestion. In the second cas

Redox cycling and hepatotoxicity of diquat in aging male Fischer 344 rats.

The aim of this study was to determine the influence of aging on diquat-induced redox cycling in liver microsomes and diquat hepatotoxicity in rats. Diquat-stimulated production of superoxide anion radical and NADPH-cytochrome c (P-450) reductase activity were measured in liver microsomes prepared from male Fischer 344 rats at ages representing young adulthood (5-6 months), middle age (15-16 months), and old age (24-27 months). Both activities were decreased substantially (40%) in old rats. Diquat-induced liver damage was assessed 6 hr after the administration of diquat (0.1 mmol/kg, ip) on th

Selenium yeast alleviates diquat-induced oxidative stress and testicular damage in roosters.

Diquat (DQ) is a pro-oxidant that generates free radicals in cells through redox reactions, leading to the induction of oxidative stress. During the processes of growth and reproduction, poultry are particularly vulnerable to oxidative stress. Selenium yeast (SeY) serves as an organic selenium source characterized by high activity and low toxicity, imparting antioxidant effects. The objective of this study was to assess the protective effects of SeY against DQ-induced oxidative stress in rooster testicles. The results demonstrated that SeY pretreatment mitigated DQ-induced oxidative damage in t

Lysine succinylation-mediated energy metabolism imbalance drives brain injury in acute diquat poisoning.

Neurological damage induced by diquat (DQ) poisoning has garnered significant attention from researchers in recent years. This study represents the first systematic investigation into the molecular mechanisms underlying acute DQ-induced brain injury by integrating quantitative proteomics with lysine succinylation proteomics in male C57BL/6 J SPF mice exposed to 200 mg/kg DQ dibromide via a single oral gavage. The study revealed that DQ poisoning significantly downregulated lysine succinylation levels in mouse brain tissue, thereby inhibiting key mitochondrial energy metabolism pathways and dis
